Abstract

Chrysanthemum is an ornamental plant that contains many secondary metabolites such as flavonoids and various volatile compounds that have the potential as medicinal ingredients. Production of secondary metabolites from plants can be carried out in vitro through callus culture. The purpose of this study was to determine the effect of Plant Growth Regulators (PGR) combination on callus formation from explants of leaves and stems of Chrysanthemum plantlets in Murashige & Skoog (MS) media. The study was conducted using a Completely Randomized Design (CRD) with a single factor in the form of 10 combinations of 2,4-D + kinetin  for callus induction from stem explants and 12 combinations of NAA + BAP  for callus induction from leaf explants. Observations were conducted on two months after planting. Observation data were analyzed descriptively. The results showed that callus formation occurred both from stem and leaf explants in all treatment combinations.In the group of stem explant, a combination of 3 `ppm 2,4-D + 2 ppm kinetin resulted in the highest average of callus size, wet weight, and dry weight, ie 13 (clay models); 1.186 grams; 0.316 grams respectively with a compact texture and light green, brownish dark green.Pada kelompok ekpslan daun, kombinasi 1 ppm NAA + 1 ppm BAP rata-rata ukuran, berat basah dan berat kering kalus tertinggi  berturut-turut sebesar 17 (skala clay models); 1,3921 gram dan 0,1024 gram dengan tekstur kompak dan berwarna hijau muda.In the leaf explant group, a combination of 1 ppm NAA + 1 ppm BAP, the highest average of size, fresh weight and dry weight of callus at 17 (clay models); 1,3921 grams and 0.1024 grams with compact texture and light green color.   Keywords: Chrysanthemum morifolium, calli, NAA, BAP, kinetin

Abstract

Mathematics is an important lesson, especially because mathematics can be used in everyday life to solve problems. But this is not the case when met in the field, one of the 'popular' subjects as a less desirable subject is mathematics. Furthermore, creative thinking is the ability to make new combinations, based on data, information or elements that exist. The objectives in this study were: To determine the improvement of creative thinking skills and positive attitudes of students by using the realistic mathematical education (RME) approach in learning mathematics at the Trisakti Private Middle School Pematangsiantar T.A. 2018/2019. This research is a quasi-experimental study. The population in this study were all seventh-grade students of Pematangsiantar Trisakti Private Middle School, which consisted of 3 parallel classes with an average number of students per class of 30 people. From 3 class VII parallel, students were selected as a sample of 1 class to be used as Experimental class, namely Class VII-A. Hypothesis testing uses Paired-Samples t-Test with a significance level of 0.05. Test criteria if α> Sig. (2-tailed) then reject Ho and Ha accepted. The results showed that there was a significant increase in positive attitudes with RME  in social arithmetic material in class VII of Pematangsiantar Trisakti Private Middle School, there was a significant increase in the ability to think creatively with the RME approach to social arithmetic material in class VII Pematangsiantar Trisakti Private Middle School.  Keywords : Positive Attitudes, Creative Thinking Skills, Realistic Mathematical Education

Abstract

Hibiscus rosa-sinensis L. has been long used as medicine, ornamental plants, and foodstuffs. The plants uses as foodstuffs have nutritional or improve the taste, while the plants used as medicine have biocytivity compounds. This article was written based on a review of vary scientific articles and books published online and offline related to the use of H. rosa-sinensis. Hibiscus rosa-sinensis is a plant originating from China and has around 300 hybrids, which structures of corolla has vary in color and shape. In the traditional medicine of H. rosa-sinensishas been used to cure of the hypertension, diabetes mellitus, wounds, menstrual disorders, and abortion. The bioessay of H. rosa-sinensis has activity as recovery of heart disolder, hypertension, diabetes mellitus, antioxidants, antifertility, anti ulceration, and kidney disorders.   Keywords: Hibiscus rosa-sinensis, hypertension, antifertility, abortion.

Abstract

Xylanase is one type of enzyme that has an important role in the field of industry. One way that can be done to improve the thermostability of an enzyme is by protein engineering . Mutation of these proteins can be done by studying the structure of proteins through molecular dynamics simulation approach. In this research, thermal stability analysis on the structure of Aspergilus niger Wild Type xylase (AnX) was performed. This study aims to study the thermal stability characteristic of Aspergilus niger xylanase enzyme through molecular dynamics simulation approach. Molecular dynamics simulations of AnX were performed using NAMD (Not Just Another Molecular Dynamic) software at at a temperature of 300-500 K. This study focused on studying the thermal stability characteristic of the enzymes to obtain information on residues that are responsible for the characteristic. Selection of residues to be mutated, based on the results of Hydrophobic Interactions. Based on the analysis results, the design of mutant Xylanase enzyme that is more thermostable than the Wild Type Xilanase Enzyme, so it can provide suggestions a more stable Xilanase mutation design that can be implemented into wet experiments to genetically engineer the Aspergilus niger xylanase enzyme. Xylanase Aspergillus niger enzyme is unfolded at 500 K at 9.5 ns. The residues responsible for the thermal stability of Xilanase Aspergilus niger enzyme based on hydrophobic interaction analysis are Alanine at residue 60. This residue is in segment / chain 3. The best mutant that can increase thermal stability is indicated by the Alanin 60 residue replaced with Methionin. The mutant Alanin 60 residue that replaced with a methionine mutant obtained ΔΔGsolv value of -21.10345. Thus the Ala60Met mutant is the most stable mutant supposed to increase thermal stability of Aspergillus niger Xilanase Enzyme.  Keywords : enzyme, thermostability, mutation,  molecular dynamics

Abstract

Differential equations are mathematical equations for functions of one or more variables, which relate the value of the function itself and its derivatives in different order. As a prospective teacher, students are required to master the cognitive domain of differential equations. Based on the observations made where tested the initial knowledge of students in the course of differential equations and found 80% of students are not biased deserve to be said to pass. Therefore, this research is aimed at 1. Developing instructional of differential equations based on brown model and cooperative learning model as a tool in learning process; 2. Testing the effectiveness of teaching materials of developed differential equations; 3.Test whether there is a good improvement in learning outcomes in the teaching and learning process using the teaching aid tool. The method used in this research is development research. From this result Researchers believe that in the course of teaching materials is very effective tool process of course in the classroom.   Keywords: Course Tools, Differential Equations.

Abstract

The achievement of the graduates profile which is the goal of Mathematics Education majors Teachers College UPH is to prepare teacher candidates who have the character, calling, and competence also the graduates can role as a Mathematics teacher know the concept of knowledge. However, in the achievement of the graduates profile has not been satisfactory, because results of learning are low in some mathematics courses, especially in Calculus II course. Based on the problem, major Teachers College do a structured academic activity that is response learning. The research method used is control group posttest only design. The sample of the research are the students of Mathematics Education major that take the Calculus II course in the academic year 2015/2016 as the control group and who take the Calculus II course in the academic year 2016/2017 as the experimental group. The improvement of this reponse learning result is seen from t-test analysis, obtained the decision of hypothesis test is exist significant difference between the control class and the experimental class.  Keywords : response, calculus II, learning result

Abstract

An x-ray experiments require sophisticated and advanced apparatus which is very expensive resulting some laboratories in universities have not been able to accommodate these stuffs. In fact, a simple and homemade X-ray apparatus may be designed easily and it is quite cheap even the rill x-ray apparatus that equipped by many vendors. Basically, x-ray is generated from x-ray tube following two mechanisms which are characteristics x-ray and Bremstrahlung x-ray. In this study, we designed a simple x-ray apparatus which may generate Bremstrahlung x-ray. The designed simple x-ray apparatus consists of 2x2A rectifier vacuum tube and 25 kV high voltage power supply. For radiation protection and dosimetry, we conducted the experiment in a lead-shielded bunker and used Unfors Raysafe X2 as well as Digilert 100 surveymeter. We measured background radiation dose of 0,49 μGy/h. Abstract

In this research, There were 9 images for training data and 26 images for test data. This image processing studied first carried out a cropping process on training data images. Then Pre-processing was also done to eliminated noise using the Salt and Pepper Noise method. After that, the RGB image results obtained were converted to grayscale images. Then the color segmentation process was based on the mean and standard deviation values. The edge detection process was then carried out using the sobel edge detection method and the results showed visible edge thickening on plasmodium. Through the color segmentation process, the percentage of plasmodium in the blood was <9%. In the process of identifying 26 test image data, 20 successful images and 6 images were declared failed to be identified. So the percentage of success of color segmentation in identification was 76.9% for 26 input data.   Keywords : Sobel edge detection,color segmentation, plasmodium, image processing.

Abstract

This study aims to improve the ability to ask questions and problem solving skills of learners in the learning of physics through Problem Based Learning. This research is a classroom action research with two cycles in learning using problem based learning. Data collection techniques used to determine the ability to ask is by observation while for problem-solving skills using the test. Subjects in this study were students of Senior High School X class at St. Seminary Rafael Oepoi Kupang Academic Year 2017/2018. The results showed that: (1) there was an increase in the ability to ask learners in the second cycle; (2) there is an increase of mean value of problem solving ability of learners gradually that is from 68 at initial test, 72 at cycle I and 77 in cycle II. Abstract

Noise is one of the waveforms that can be heard by human hearing. Truth is the activation of sound that can not be done. The effect of sound on humans not only affects the auditory organ, but also can affect other organs, such as narrowing of blood vessels and heart system. Truth caused by transportation is very disturbing but not all of it is caused by transportation. Wisdom can also occur in public places or facilities such as hospitals and childrens playgrounds, one of the childrens and childrens favorite theme parks is TimeZone Mall. You can do research to measure and learn the level of playground at TimeZone Mall Taman Anggrek and TimeZone Tamini Square. The average measured result of 6 readings with calculations per 60 minutes in the orchid malls time zone is 70.5 dB and the tamini square time zone is 65.33 dB. In the time zone of the orchid mall park and the tamini square time zone there is no value beyond the threshold limit (85 dBA). With no measurement results exceeding the workspace limits for uninterrupted or preventive work.  Keywords: Noise, Timezone Mall, playground, sound
