Abstract

Penelitian ini bertujuan untuk mengetahui imunogenisitas antigen whole cell Aeromonas hydrophila dari beberapa strain bakteri A. hydrophila yang diberikan pada lele dumbo (C. gariepinus). Penelitian ini menggunakan metode eksperimen. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 6 perlakuan, yaitu A : strain GPW-02, B : strain KLK -11, C : strain GLR-08, D : strain KLK-14, E : strain KLK-05, F : kontrol (PBS pH 7,0) dengan 3 kali ulangan. Ikan uji yang digunakan adalah lele dumbo berumur sekitar 2 bulan dengan ukuran panjang 10-13 cm dan berat antara 12,2-14,5 g. Parameter utama yang diamati adalah titer antibodi dan uji reaksi silang, sedangkan parameter pendukungnya adalah parameter kualitas air, meliputi suhu air, pH, dan oksigen terlarut (Dissolved Oxygen = DO) yang diamati setiap minggu. Titer antibodi diamati 3 kali, yaitu sebelum divaksinasi, seminggu setelah vaksinasi booster dan dua minggu setelah vaksinasi booster. Kemudian dilakukan uji reaksi silang pada antigen dengan titer antibodi yang tinggi, yaitu ≥ 25. Data titer antibodi dianalisis menggunakan Analisis Sidik Ragam (Analysis of Variance/ANOVA), dan apabila berbeda nyata dilanjutkan dengan uji DMRT (Duncan Multiple Range Test) pada taraf uji 5% (Steel & Torrie, 1993), sedangkan data kualitas air dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa antigen whole cell A. hydrophila berpengaruh nyata (P<0,05) terhadap imunogenisitas ikan lele dumbo. Strain yang memiliki imunogenisitas paling tinggi adalah strain A (antigen whole cell A. hydrophila strain GPW-02). Parameter kualitas air selama penelitian dijaga pada kisaran optimal, yaitu suhu air 25-28°C, pH air 7,1-7,9, dan DO 6,6-6,8 mg/L.

Abstract

These research were conducted to isolate, characterize, and identify bacterium Aeromonas sp. causative agents of Motile Aeromonas Septicemia (MAS) in gouramy. Samples of sick gouramy with MAS attack symptoms were taken random from tree regencies, i.e. Banyumas regency (Pliken and Lemberang), Purbalingga regency (Padamara and Jompo), and Banjarnegara regency (Kaliwinasu and Blimbing). Ten isolates were isolated from ren of gouramy on Glutamat Starch Phenile (GSP). Koch Postulat Test was conducted to determine pathogenic bacteria by intramuscular injection to gouramy (10-12 cm of total length) suspension bacteria 0.1 ml at 109 cell/fish, and control with 0.1 ml sterile PBS pH 7.0.  Then, the pathogenic bacteria were identified with morphological and biochemical test. Results indicated that 10  isolates were pathogenic bacteria and morphological and biochemical tests suggested that the pathogenic bacteria could be identified to be Aeromonas hydrophila.

Abstract

The aims of this research were to determine effect vaccination with extra cellular product and intracellular product of A. hydrophilic by 1-2 times booster with 2nd week time interval on African catfish immune response. The research was constructed as CRD with five treatments and three replicates, i.e. (1) E1: vaccination with extra cellular product of A. hydrophilic with one time booster; (2) E2: vaccination with extra cellular product of A. hydrophila with two times booster; (3) I1: vaccination with intracellular product of A. hydrophila with one time booster; (I2) vaccination with intracellular product of A. hydrophilic with two times booster, and (5) K: control. African catfish of 13-15 cm length and 18-21.5 g of weight were used, with the number of 15 fish for each treatment. Intramuscular injection method was conducted for vaccination and booster of 0.1 ml vaccine into the body, with the dosage of 5 µg/fish. Booster time interval was two weeks. Data were taken for fifteen weeks. The parameters to observe were titer antibody and water quality. Furthermore, they were analyzed by Analysis of Variance and Duncan’s Multiple Range Test (DMRT). The result showed that vaccination with extra cellular product and intracellular product of A. hydrophila by 1-2 times booster with 2nd week time interval could increase the adaptive immune response through the increase titer of antibody (P<0.05) on African catfish. Vaccination with intracellular product of A. hydrophila by two times booster (I2) was the most effective method of vaccination.

Abstract

The research conducted n gouramy fish with size among 12-15 cm in length and average of 30 g in wight. the treatment used levamisol and their combination with vaccine Aeromonas hydrophila which applied by immersion during 6 hours. Methods of research used experimentally laboratories using completely randomized design with 6 replication. The fishes placed in aquaria, each of its 12 fishes. The data’s observed consisted of growth both of length and weight, number of neutrophyl, survival rate, and water quality. The aim of the research was to know the effect of levamisol and its combination with vaccine on the respons against their growth, neutrophyl responses and survival rate of gouramy fishes. The results showed, treatment of levamisol and their combination with vaccine could increase on the growth and the best result revealed on P1, as 1.2 cm and 0.68 g for their length and weight respectively. Cellular response of neutrophyl showed better reached at P3 and P4 as number of 5.3 neutrophyl cell between on day H18 and H24. Higher survival rate reached on P2 and P3. During researched, water quality was on qualified standard requirements for larvae rearing.

Abstract

Penelitian ini bertujuan untuk mengungkap dua masalah, yaitu (1) mengetahui efektivitas vaksin sitoplasma sel A. hydrophila untuk pengendalian penyakit MAS pada gurami, dan (2) mengetahui dosis optimal vaksin sitoplasma sel A. hydrophila untuk pengendalian penyakit MAS pada gurami. Penelitian dilaksanakan menggunakan Rancangan Acak Lengkap (RAL) dengan lima perlakuan tiga ulangan, yaitu : (1) K : kontrol yang divaksin dengan larutan PBS steril, (2) D1: vaksin protein sitoplasma sel  A. hydrophila dosis 2,5 µg/ekor, (3) D2 : vaksin protein sitoplasma sel  A. hydrophila dosis 5 µg/ekor, (4) D3 : vaksin protein sitoplasma sel  A. hydrophila dosis 7,5 µg/ekor, (5) D4 : vaksin protein sitoplasma sel  A. hydrophila dosis 10 µg/ekor  Gurami yang digunakan berukuran panjang 10-12 cm dengan berat 25,8-28,5 g. Vaksinasi dilakukan secara suntikan intramuskular. Satu minggu setelah vaksinasi dilakukan vaksinasi booster, dan uji tantang dilakukan dua minggu setelah vaksinasi booster. Parameter yang diamati meliputi titer antibodi, sintasan, tingkat perlindungan relatif (Relative Percent Survival/RPS), dan rerata waktu kematian (RWK). Data dianalisis dengan analisis varian dan Duncan’s Multiple Range Test (DMRT). Hasil penelitian menunjukkan bahwa vaksin protein sitoplasma sel  A. hydrophila efektif untuk pengendalian penyakit MAS pada gurami. Vaksin ini dapat meningkatkan produksi titer antibodi, sintasan, dan RPS (P<0,5) gurami. Dosis vaksin protein sitoplasma  sel  A. hydrophila optimum adalah 7,4 mg/ekor yang memberikan nilai RPS tertinggi sebesar 89,31 %.

Abstract

The research was carried out with the experiment method, to look for the value of Minimal Inhibitory Concentration (MIC or KHM) and LD50.The MIC determination was carried out in two stages. Firstly, it determined the dose of the concentration 0, 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 mg/ml.  The research showed that MIC was found 16,8 mg/ml and for determining of LD50 was continously decided by up to dose 16.8; 17.0; 17.2; 17.4; 17.6 mg/ml with 10 replication. MIC’s data analyzed with Completely Randomize Design with 9 treatment and 5 replication. The analysies used F test and than BNT with 5 % significantly. Results of the research showed the treatment of the dose 16.6 mg/ml could inhibite the A. hydrophila growth with the diameter in general as big as 0.72 mm,.whereas on the LD50 the consentration revealed 16.8 mg/ml.

Abstract

The aims of this research was to determine effect vaccination of cell debris of A. hydrophila by 1-3 times booster with 2nd week time interval on African catfish immune response. The research was constructed as CRD with five treatments and three replicates, i.e. K : control; D1 : without booster; D2 : once booster; D3: twice booster, and D4 : three times booster. African catfish of 10-13 cm length and 12-16 g of weight were used, with the number of 15 fish for each treatment. The result showed that cell debris A. hydrophila vaccination by 1-3 times booster with 2nd week time interval could increase the adaptive immune response through the increase titer of antibody (P<0.05) on African catfish. Vaccination with two time booster (D3) was the most  effective method of vaccination.

Abstract

The aim of research were to know effect of fasting method on growth of aquatic crayfish (Cherax quadricarinatus) and feed efficiency. The experiment used Completely Randomized Design (CRD), consist of 4 treatments and 3 replicates. The treatments were P1 (aquatic crayfish were fed one day and fasted one day), P2 (aquatic crayfish were fasted every Monday and Thursday), P3 (aquatic crayfish were fed two days and fasted two days), and P4 (aquatic crayfish were fed everyday/control). Aquatic crayfish with initial average body weight of 5 g used in this experiment. Aquatic crayfish were cultivated in cement pond (100 x 60 x 50 cm3) with density 20 crayfishes/pond for 40 days. Aquatic crayfish were fed at 5% of body weight. Results of research indicate that fasting method had effect on growth of aquatic crayfish and feed efficiency, but had no effect on survival rate of aquatic crayfish. Treatment P1 showed highest growth of aquatic crayfish and feed efficiency.

Abstract

The aims of this research was to know prevalency of protozoa ectoparasites in fish fry of nila (Oreochromis niloticus). Experiments held at two different ponds, Local Hatchery Center (BBI) Pandak and Local Hatchery Center (BBI) Sidabowa, Banyumas regency. The research method used survey and the location site was determined with purposive sampling method. The samples were taken randomly (random sampling) from two locations. Total fishes samples were 135, as follow 75 of fishes from BBI Pandak and other 60 fishes from BBI Sidabowa, with twice replication of taking samples.  The datas was analyzed descriptively and continued with t test to compare the prevalency of ectoparasite in   Pandak and  Sidabowa. The result of this research showed the type of ectoparasite and infection level that nila fish fry at Pandak which found in the surface and gill were Trichodina sp. (80% & 18,67%), Ichtyophthirius multifiliis (58,67% & 24%), Chilodonella sp. (29,33% & 5,33%), Epistylis sp. (49,33% & 0%), Vorticella sp. (15% & 0%), while type of ectoparasite and infection level which found in the surface and gill at Sidabowa were Trichodina sp. (86,67% & 63,33%), Ichtyophthirius multifiliis (53,33% & 23,33%), Chilodonella sp. (18,33% & 3,33%), Epistylis sp. (23,33% & 0%). The t test compare the prevalency showed inexistence differences of ectoparasite infection level (P>0,05) between Pandak and Sidabowa.

Abstract

This research was conducted to know  the effect of methyltestosteron dietary on sex ratio of guppy and determine the effective dosage of methyltestosteron. This research used experimental method with Completely Randomize Design condist of 4 treatments and replicates. The treatmens were P1 (0 mg/kg), P2 (10 mg/kg) P3 (20 mg/kg), and P4 (30 mg/kg), respectively. Observation of phenotype sex had done by identified of body style and morphologys variety. Percentage of sex ratio and survival rate of guppy were analyzed by ANOVA (Analysis of Variance) and LSD (Least Significant Difference Test). Results of the research showed that  methyltestosteron dietary had effect on sex ratio of guppy (P<0,1), but had no effect on survival rate of guppy (P>0,05). The dosage of 10 mg/kg feed) result the highest percentage of male (61.69%).
