Abstract

Phosphorus in the soil is essential macronutrient for plant growth. Phosphate solubilizing bacteria are capable of managing phosphate availability in its soluble forms and are often found in the rhizosphere. This study aimed to know the level of total density and characteristics of phosphate solubilizing bacteria isolated from the rhizosphere of maize plants at three dry land locations in Banten.  The bacteria were isolated using serial plate dilution method. Results showed that four isolated phosphate solubilizing bacteria were obtained. Two isolates were obtained from Curug, one from Walantaka, and one from Petir. The total density of phosphate solubilizing bacterial colonies from those dry land locations were  1.9 x 1010 CFU/ml for Curug 1, 1.9 x 1012 CFU/ml for Curug 2, 6x1012 CFU/ml for Walantaka, and 4,8x1010 CFU/ml for Petir.

Abstract

Phytophthora sp. is one species of fungus that attacks the seeds of the cocoa plant. Control may use biological agents and Bokashi. This study aimed to determine the effect of Trichoderma sp. and Bokashi on the growth of plants and pathogens Phytophthora sp on cocoa plant in nurseries. The treatment was Trichoderma sp at a doses that was 0 gram, 25 grams, 50 grams and 75 grams, and Bokashi at a doses of 0 gram, 25 grams, 37.5 grams and 50 grams. The study was designed using a completely randomized design with three replications. The results showed that administration of Trichoderma sp. and Bokashi jointly improve the interaction effects canopy height, canopy fresh weight and shoot dry weight, and suppress the intensity of the disease. A dose of 75 grams of Trichoderma sp + 50 grams Bokashi is best dose increase canopy height, canopy fresh weight and shoot dry weight. A dose of 50 grams Bokashi without giving Trichoderma sp. can reduce the disease intensity to 14.95%, and a dose of 25 grams of Trichoderma sp, with or without the provision of Bokashi can reduce the disease intensity to 0%.

Abstract

 The research was aim to study the effect of NaCl on callus of sugarcane varieties Bululawang. This experiment has been conducted in Tissue Culture Laboratory of the Faculty of Agiculture, University of Jember from December 2012 to Mei 2013. The experiment which consists of four levels concentration of NaCl : 0 g/l (K0), 4 g/l (K1), 8 g/l (K2), 12 g/l (K3), and 16 g/l (K4). Each treatment consists of four replications. The experiment consisted of three stages: callus induced, salinity strees of callus, and shoot regeneration of sugarcane on various rate NaCl. Observations conducted qualitative description of the parameters of the color of callus , callus structure and shoot regeneration. The results of this experiment indicated that the callus of sugarcane varieties Bululawang experienced salinity stres of NaCl at 4 g/l showed symptom degradation of color to yellow with sticky crumb structure. Increasing consentrations of NaCl 8-16 g/l lead callus color being tawny until black with sticky crumb structure. Salinity stres with consentrations of NaCl at 4 g/l caused callus is not able to regenerate to form buds and increase callus cell death.

Abstract

Development of upland rice is one of the answers in increasing rice production, but the productivity of upland rice in Banten is still very low. One reason is the low use of improved varieties. But of the many  upland rice varieties are not all suitable to be developed in a region. To  find suitable varieties are developed in a region need to be  an assessment of the introduction of varieties to obtain location-specific adaptive varieties. Assessments conducted in Cisehreheun Village, Cigeulis Sub district, Pandeglang District, during  June-September 2013. Assessments using a  randomized block design with upland rice varieties as treatment used are  Inpago-4, Inpago- 5, Inpago- 6,  Situbagendit, Stone Tegi and local varieties as a comparison, each variety was repeated  4 times.  To determine the feasibility of technology to analyze revenue and cost  (B/C), while for  measuring the level of technological excellence  applied to farmers to use  marginal benefit cost ratio analysis (MBCR). The results showed that varieties Inpago 5 generate  the highest production (3.5 t/ha). The results of financial analysis shows Inpago-5 provide the highest  profits  with a gross value of B/C 2.18  and the marginal value of B / C of 4.69.. Based on the results of the study, varieties Inpago-5 is a decent variety of adaptive and developed by farmers.

Abstract

This research aimed to assess the effect of alternative plant growth regulators to the growth of stum dorman see rubber Klon BPM 24. This research was done at Lubung Saung Village, District And Banyuasin III Regency South Sumatera Province. This research used a randomized block design with five treatment and five replications. The treatments studied were B0 = 0 ml L-1 control, B1 = 2 ml L-1 of water, B3 = 6 ml L-1  of water, B4 = 8 ml L-1  of water. Each treatment consisted of 10 plants, with a long soaking the whole treatment was 15 minutes. Thus obtained 25 combined treatment. Each unit experiments were 3 examples of plans. Parameters namely speed sprout observations (HST), shoot length (cm), number of leak stalk (strands), the percentage of seedlings grown (%), weight wet plant (g),  dry weight of plant (g), weight of wet roots departure (g), and root dry weight (g). Based on result obtained it can be concluded that, the concentration of growth regulators treatment 0f 8 ml L-1 water tends to give the highest yield and better seed to grow that percentage to 80 %, the speed of growth was 20.40 HST shoots, shoot length was 30.79 cm, weight was plants dry weight 7.10 g, wet weight was 1.94 g root, root dry weight was 0.88 g.

Abstract

The research was conducted to explore and determine the presence of antagonistic bacteria on banana plants (Musa spp) to inhibit (Fusarium oxysporum f.sp. cubense). The research was conducted in the laboratory of Agroekoteknologi, Faculty of Agriculture, Sultan Ageng Tirtayasa University. The experiment was arranged in non-factorial Randomized Completely Design (RCD) using 3 replication. The treatment consisted of 9 rhizobacteria isolates acquired from rhizosper of roots of banana on first screening. The test results showed antagonistic in vitro testing is able to inhibit the mycelium growth of Foc where the greatest inhibition was shown by BR 45 and BR 15 with the largest percentage inhibition of 88.90% and 87.78%. Observations on colony morphology showed 7 rhizosphere bacterial, genus observed as Bacillus sp. and that besides it was not identified. Research showed that the bacteria could potentially control the banana root disease caused by Fusarium.

Abstract

The aim of this research was to know In Vitro initiation on shoots of marasi (Curculigo latifolia) by different concentration of  NAA(Naphthalene Acetic Acid) And BAP (Benzyl Amino Purine). This research has been done at Biotechnology Laboratory of Agriculture Faculty, Sultan Ageng Tirtayasa University since August to October 2014. This research used Randomized Complete Design (RCD) with two factors. The first factor was NAA concentration with 4 levels, they were  mg/l; 0,2 mg/l; 0,4 mg/l; 0,6 mg/l. The second factor was BAP concentration with 4 levels, they were 0 mg/l; 0,5 mg/l; 1 mg/l; 1,5 mg/l. The research showed that the NAA concentration gave significant effect on plant height at 4, 5, 6 and 7 WAP (Week after planting), and the  BAP concentration gave significant effect on plant height at 7 WAP. There were not combination effect on NAA and BAP concentration on bud initiation time and plant height. All the living explants during the observation reached 83,5% with the number of contaminating explants was 60% and browning explants 87,5%.

Abstract

Tomato plants require intensive cultivation techniques to growth and high production. The use of fertilizers is one of the techniques that need to be developed. This study aimed to determine the effect of bokashi and urea fertilizer on the growth and production of tomato plants. The treatments tested is bokashi the dose of 0 or without bokashi, 10 ton Bokashi ha-1, and 20 ton bokashi ha-1, and urea that doses of 0 or without urea, 200 kg urea ha-1 and 400 kg urea ha-1. The study was designed using a randomized block design, two factors with three replications. The results showed that administration of bokashi fertilizer and urea together can cause interaction effects increased plant height, number of fruits per plant and fruit weight per plant, and a dose combination bokashi and urea best is 20 tons bokashi ha-1 and 400 kg urea ha-1.

Abstract

Corn corp (Zea mays L.) is the second commodity food after rice. Corn crop is a plant that in its growth and development process need environmental factor, such as land. Land factor has big contribution in supporting growth and development of plants. Therefore, land has to be evaluated as of the commodities that will be cultivated can provide optimal results. The purpose of this research was to assess land suitability classes and provide direction zoning commodity corn crops in Lebak regency and provide alternative improvement of the limiting factors in an effort to increase corn production. Descriptive method based on desk study activities and survey was used in this research.  Principal of that method was based on approach of landscape mapping and the interpretation of aerial photographs or satellite images, then land evaluation of that land units. Agro-ecological zones parted as 5 zones and 18 sub zones. Zones that being in research areas were zone I (forestry), II (annual plants/plantation), III (annual plants and crops), IV (crops) and VII (forestry/pasture). Based on its land suitability for corn crops were in agro-ecological zone III and IV. In research area agro-ecological zone III and IV that suitable for corn commodities planting were 7 sub zones, there were III/Df;h;ei (S2-tc/wa/rc), III/Dhf;sp (S2-tc/wa/rc), IV/Df;h;ei (S2-tc/rc), IV/Dfs,hv/Wr (S2-tc/rc), IV/Dhf/Wr (S2-tc/rc), IV/Dfe/Wr (S2-tc), and IV/Wr/Dhf (S2-tc/rc), with land area reaches 62.877 ha or about 18,92%  from the total of Lebak regency land area is  332.371 ha.

Abstract

Studying plant growth hormone such as Indole-3-butyric acid (IBA), one of auxin family is important for potato seed development. Unfortunately, simple, quick, and accurate extraction and analysis method to support the study of hormone in potato plant still limited. This study aim to develop extraction and analysis method of IBA from potato fresh tissue. For analysis method development, this study used IBA standard (purity 99.9%) as sample. The Analysis used reversed-phase high performance liquid chromatography (RP-HPLC) with ultraviolet detector (wavelength 280 nm), C18 column, and room temperature for chromatography condition. To get the best analysis method, this study used methanol, water, and acetic acid as mobile phase with several different compositions. This study also used several different flow rate of mobile phase. The study tested the best analysis method by linearity standard curve procedure. The concentrations of the standard IBA solution for linearity were 50 ppm, 100 ppm, and 150 ppm. This study used the best analysis method to confirm extraction result of IBA hormone from potato fresh tissue by maceration procedure. Several compositions of methanol and temperatures used to find the best extraction procedure of the hormone. The result showed for the best analysis method by RP-HPLC, the composition of mobile phase was methanol: water = 80: 20 (v/v) with 0.8 ml/min in flow rate. The result showed, the best analysis method of IBA has linearity equation: y = 7175x + 17734 with correlation coefficient value (R2) = 0.992. For the best extraction method of IBA, the best composition of sample and methanol was 2.5 gram: 10 ml (b/v) under controlled temperature (10oC).  The conclusions, the studies were successfully developing extraction and analysis method of IBA hormone from potato fresh tissue.
