Abstract

The study was  conducted to develop methods for  planting stock production by shoot cuttings. The objectives of the study were to identify the effect of parent tree age, shoot age and  rooting media on the growth of shoot cuttings. The study was conducted in 2 experimental steps. The first was  to examine the  effect of  parent tree age and shoot age  on the groeth of cutting: parent tree age class of 5 years (KU-1), 6-10 years (KU-2), 11-15 years (KU-3) and 16-20 years  (KU-4), and shoot ages of  3 and 4 months. The second  was to test 6  rooting media types  (volcanic sands, soil, rice husk charcoal, coconut husk, sand + rice husk charcoal/4:1 and sand + coconut husk/4:1). The results showed that shoot cuttings from root cuttings grew successfully. It reached 90-100% of survival rate. Parent tree age  and shoot age significantly influenced the  number of roots with the highest root number was 5,42 from 4 months shoot taken from KU-4 root cuttings.  Types  of media  significantly affected  rooting rate with the best was 98% shown  in sand + rice husk charcoal (4:1) media.

Abstract

Santalum album is one of the extinct species which need to be conserved. Therefore, the information of reproductive biology of this species, especially the pollination system can produce important clues in order to conserve it.  Preliminary study of pollination system of S. album was conducted at ex-situ conservation plot at Watusipat, Gunung Kidul, Yogyakarta. The observations showed that S. album flowers categorised as protandrous, which structured acropetally; this structure may lead self-pollination within a spike. However, result from artificial pollination showed that S. album was a predominantly out crossing species. Very low level of seeds setted from self-pollinated flowers indicated that pollinator was needed for reproduction process. Observation revealed that European honey bee (Apis mellifera) was one of potential pollinating agents.

Abstract

The aim of this experiment was to investigate effect of  media growth composition and NPK fertilizer dosage toward growth ability of A. angustoiloba from Pendopo Provenance, Muara Enim, South Sumatera. Completely Randomized Design of factorial with two factors (growth media composition and fertilizer dosage) and four levels of each factors were used. Four replications were used with five seedlings per replication. The effects of those treatments were evaluated through seedling height growth, stem diameter growth and number of leaves. The result showed that media composition variations gave very significant effect on height growth and number of leaves of  A. angustiloba seedling for 4 months. The best height growth was produced of top soil + compost (1:1) media (48,51 cm) and the best number of leaves was produced of top soil media (19,89). The fertilizer dosage variations gave very significant effect on height growth, stem diameter and number of leaves of  A. angustiloba seedling for 4 months. The best height growth (49,58 cm), stem diameter (5,93 mm) and number of leaves (20,24) were produced by 1 g NPK fertilizer dosage. Interaction of media composition and fertilizer dosage gave significant influence on height growth and stem diameter of  A. angustiloba seedling for 4 months.The best height growth (52,7 cm) was produced by interaction of top soil + compost (1:1) media and 1 g dosage of NPK fertilizer and the best stem diameter (6,5 mm) was produced  by interaction top soil and 0.5 g dosage of NPK fertilizer.

Abstract

Aren tree or Sugar Palm (Arenga pinnata Merr) is one of bioethanol-producer which has advantages compared to other plant producing bioethanol. This study is  aimed to determine the genetic diversity of aren tree populations in its natural distribution. Genetic diversity of four populations were investigated by isozyme markers with 4 enzyme systems namely Esterase (EST), Glutamate oxaloacetate transaminase (GOT), Diaphorase (DIA) and 6-Phospogluconate dehydrogenase (6Pg). The number of alleles identified by 9 alleles are distributed on   four polymorphic loci. The research revealed that mean alleles per locus was 2,2500. The mean effective number of alleles per polymorphic locus was 1,8377. All loci were of  polymorphic. The mean genetic diversity within population was 0,4381and the proportion of genetic variation among populations was 0,0702. The UPGMA cluster analysis based on Nei’s standard genetic distance reflected two main clusters. The first cluster consisted of Central Java and South Kalimantan populations; while second cluster included the populations of Bengkulu and North Sulawesi.

Abstract

This research was conducted to support tree improvement program for sengon (Falcataria moluccana (Miq.) Barneby & J.W. Grimes). Polymorphic fragments were surveyed in 288 RAPD primers using 24 materials from Okinawa, Japan. Carefully selections of the polymorphic fragments were applied to overcome a problem of low reproducibility of RAPD markers. The first screening using 4 materials found that 285 out of 288 primers successfully produced fragments. The second screening using 16 materials finally obtained 48 polymorphic fragments. High discrimination ability (DP=0.483, in average) was obtained from all the selected fragments. The selected fragments can be used to support tree improvement program of the species for genetic diversity study and to facilitate a development of more specific markers such as Sequence Characterized Amplified Region (SCAR) marker.

Abstract

Aquilaria sp, belong to Family Thymelaeaceae, is one of several species whose can produce agarwood or gaharu. Quality of gaharu of this species is better than the other species. With high economic value, demand of Aquilaria sp increase and also increase exploitation of the species without counterbalanced by enough planting activity. Therefore, conservation and breeding program of the species become important to carry out. Two important Aquilaria sp.  are A. malaccensis and A. microcarpa. However, both species only can be distinguished by generative characteristics such as flower and fruit. The most appropriate and rapid method to identify species is DNA marker, especially using RAPD markers. In order to obtain DNA marker to species identification, 2 selection steps should be carried out. The first step was using 2-3 samples per species. From 180 RAPD primers, only 5 primers produced loci those can identify both species, those are D-20, A-17, G-12, R-15 and U-13. The second selection step was carried out by increasing number of samples per species (24 samples in total). From the five primers obtained from first selection, only U-13 700 basepairs that can be selected as species-specific DNA markers to identify both Aquilaria sp.
