Abstract

Sandalwood (Santalum Album Linn) is one of high economic value foret commodities. However, over exploitation without appropriate rehabilitation has serious degradation of the natural populations caused. Therefore, an ex situ conservation of this species has been established at Watusipat, Gunung kidul in 2000. It comprised of 6 populations collected from East Nusa Tenggara .  The genetic diversity of these populations was investigated using isozyme genetic marker with 3 enzymes.Shikimate  dehydrogenase  (SHD), Esterase (EST), and Diaphorase (DIA). Thirteen alleles were identified on fife polymorphic loci. A relatively high genetic diversity was reflected by paramaters, such as the mean number of alleles per polymorphic locus (A=2.1333); the mean effective number of alleles per polymorphic locus (v= 1.6302); the mean percentage of polymorphic loci (PPL=83.333%); the mean observed heterozygosity (Ho= 0.3951) and the mean expected heterozygosity (HE = 0.3166).  Most of genetic diversity (95.62%) was distributed within population. Cluster analysis using UPGMA based on Nei S standard genetic distance reflected two main clusters : Palakahembi, Belu and Soebela constructed the first cluster; whereas Bama, Balela and Helangdohi formed the second cluster.

Abstract

An easy, Cheap and acacurate identification method is needed to ensure the identity of teak clones used in planation. Molecular markers offer an alternative technique for clonal identivication . one such method is SCAR markers. Seventy teak clones of which 20 clones were produced by tissue cultures and claimed to be originated from the some parent. And 50 clones from buddings were used as materials. Analysis was conducted using 6 highly polymorphic SCAR primers. The results showed that 20 teakclones Originated from tissue cultures clustered into 3 groups, Whereas the rest 50 samples from buddings clustered into 4 groups. SCAR markers were able to differentiate the identity of these clones. These SCAR markers can applied to prove the claim abaout the identity of chones in the market and to ovoid labeling errors in the field  for Vegetatively propagated plants.

Abstract

The Study was conducted to examine the sprouting response of four local varieties of bread-fruit to pruning and application of NPK fertilizer. Four local varieties of bread-fruit namely from bone, Banten, Sukabumi and Kediri. The experiment lay out was Randomimized Complete Design with 3 treatment factors. The result did not show significant difference among local varieties. However there are significant differences in budding response . For number of buds. Best treatment was fertilization 20 g and pruning at 50 cm. For buds growth, best treatment was NPK of 30-50 g, and best pruning at 20 and for bud diameter the best NPK dosage was 20 g.

Abstract

Micropropagation technique is applied to produce large number of plants. Juvenile explants and culture medium are two crucial factors that influence the success. The aims of this study were to determine culture medium that was effective to induce shoot and to produce plantlests from embryo culture of Pinus merkusii. Screening of five cytokines and five basal medium were carried aut. Then, explants wee cultured on the best cytokine and basal medium for plant regeneration. The results showed that BAP was the best cytokine type; LP and GD medium were the best medium compositions for shoots induction. GD medium supplemented by high consentration BAP showed the best medium for shoots induction. Solid GD medium containing 0,25uM BAO was effective for multiple shoots. Liquid GD medium without plant hormone was the best composition for shoots elongation. Half strength of GD medium without plan hormone gave the best result for rooting.

Abstract

It is important tu understand the floral visitor as preliminary study of aspecies in order to manage seed orchard. This study was aimed  to observe the floral visitors and nectar  nectar     production of M. alternifolia flowers at second generation seeding seed orchard, West Wyalong, New Sounth Wales, Australia.  One Five-minute count of insect visiations was carried out each morning between 10.00 and 11.00  am and each afternoon between 03.00 and 04.00 pm over five consecutive days during peak flowering. The numbersss of inflorescences for each sample branch, and all insect visits to each inflorescence, were recorded and collected. Insect traps were also placed on every sample tree to assist observation of the varieties of small insect. Insect traps were placed in the two partial exclusion treatments, i.e: (1) inside the fiberglass fly-screen with 2 mm apertures; (2) on the branch located in the middle of each tree without netting. Results revealed that flowers produced only a small amount of nectar which was indicated by a shiny appearance at the base of the floral cup in the morning, but this quckly disappeared as a result of evaporation. Thrips and flies and flies were dominat floral visitor of M. alternifolia, while several other types of insects, e.g. beetles, wasps, plant bugs, butteflies, predatory shield bugs and leafhopper were only infrequent visitors.
