Abstract

The study was carried out to investigate the effect of follicle stimulating hormon (FSH) into in vitro maturation medium to increase oocytes maturation and 4 cell stadium embryo development of Bligon goat. Goat ovaries were obtained from a slaughterhouse and transported to the laboratory in a flask of NaCl at temperature of 31 – 34°C. Oocytes were aspirated from 2 – 6 mm of follicles into a 3 mL syringe (23G needle) that contained Dulbecco’s Phosphate-Buffered Saline. Oocytes were divided into three groups, i.e tissue culture medium (TCM) with FSH supplementation 0, 50, and 100 IU/mL. Oocytes were put into those medium and incubated on 39°C, 5% CO2, and 95% humidity for 24 hours. Matured oocytes were fertilized with capacitated frozen thawed-semen and incubated on 39°C, 5% CO2, and 95% humidity for 5 hours. Fertilized oocytes were washed for 3 times in TCM and incubated in the same condition for embryo culture. The data of FSH supplementation and embryo development were analyzed using randomized completely one way classification. The results showed that the percentages of mature oocytes from FSH supplementation 0, 50, and 100 IU/mL were 70,48±23,22, 78,48±15,80, and 80,29±12,86%, respectively. Cleavage rate of the two cells stage were 36,00±14,22, 44,00±33,94, and 57,45±31,78%, respectively, and for the 4 cells stage were 27,33±22,04, 35,33±40,73, and 39,45±20,38%. It is concluded that supplementation of FSH in the maturation medium could not increase the percentages of in vitro maturation and embryo development.

Abstract

This study aimed to know the effect of AGP on growth performance, duodenal histological structure and pectoralis muscle of Kampung chicken. This research was conducted by designing 3 groups with each group consisting of 20-day old chicks (DOC) of Kampung chickens. The control group was given basal feed without AGP bacitracin, group 2 was treated with 0.125 g of bacitracin /kg of basal feed and group 3 was treated with 0.25 g bacitracin/kg of basal feed until 12 days old. The parameters observed were chicken body weight on days posthatch, 3, 5, 7, 9, and 12, morphometry, visceral organ weight, duodenal organ morphology and muscle performance of Pectoralis thoracicus at 12 days old. Data analysis used one way ANOVA test followed by Tukey test with significance of P≤ 0,05. The result showed that the morphology of the doudenum and the myofiber area of group 3 indicated significant differences compared to the control group. The conclusion of this study revealed that administration of AGP bacitracin with 0.25 g/ kg dose of basal feed may increase the growth performance of Kampung chicken.

Abstract

This research aimed to evaluate the effect of different protein sources (plant or animal source) as substitution of 50% soybean meal protein in the flushing diet on early reproduction performances of doeling. The treatments were three types of flushing diet, i.e., Control diet containing soybean meal (C), diet containing Indigofera meal (IM), and diet containing cricket meal (CM). Diet IM and CM were substitution of 50% soybean meal protein in the ration. Completely randomized design was used with three treatments and four replications by using twelve Sapera doeling. The experiment with flushing diet was done started from three weeks before mating (including adaptation), during mating and following one month after pregnancy. Parameters measured were nutrient consumption, BCS, plasma glucose, cholesterol, progesterone hormone and reproductive performance. The results showed that treatments of utilization of different protein sources were not significantly affected the feed consumption, BCS and reproductive performances of doeling, but it had significant effect (P<0.05) on plasma glucose and cholesterol. The protein source of Indigofera and cricket meal showed in improving of progesterone status to support until 30 days pregnancy. Diet containing cricket meal could improved onset estrous and duration of estrous. It was concluded that cricket meal can be used as an alternative protein source to substitute of 50% soybean meal protein in the Sapera doeling flushing diet and it can improve the reproductive performances.

Abstract

The objective of this study was to examine the effect of seminal plasma on viability of Boer goat spermatozoa and effectiveness of sugar palm juice as an alternative extender during preservation at 5ºC. Semen of two Boer goats were collected using an artificial vagina. Fresh semen were evaluated and divided in equal volume into four tubes. Semen in the first and second tubes diluted with 80% sugar palm juice + 20% egg yolk (P1) and Andromed (P2), respectively. Semen in the third and fourth tubes were centrifuged with 3,000 RPM for 20 minutes, and the supernatant removed. diluted with 80% sugar palm juice + 20% egg yolk (P3) and Andromed (P4), respectively. Diluted-semen were preserved in refrigerator at 5oC, and quality of the spermatozoa including motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) were evaluated every day for four days. Results of this study showed that at day-2 preservation, mean percentages of MS, LS, and IPM for P2 (72, 83.4, and 83.4%), P3 (72, 82.6, and 82.2%), and P4 (72, 83, 83.8%) were significantly (P<0.05) higher than P1 (3, 24.8, and 25.2%). At day-3 preservation, mean percentages of MS, LS, and IPM for P2 (57, 65.6, and 69.6%) was significantly (P<0.05) higher than P3 (21, 34.8, and 31.8%), P4 (22, 33.6, and 31.2%), and P1 (0, 0, and 0%). In conclusion, semen of Boer goat to be preserved with extender containing egg yolk should be removed seminal plasma. Sugar palm juice containing egg yolk could be used as an extender for Boer goat semen, but should be applied in the AI program immediately after the semen is diluted.

Abstract

The aim of this research was to investigate the quality of physicochemistry and microbiology of Pegagan duck meat preserved with various concentrations of liquid smoke and storage time. This study used a complete randomized design (CRD) with a 4x2 factorial pattern. The first factor was the concentration of liquid smoke solution consisted of 4 levels, namely 2; 4; 6, and 8% (v/v). The second factor was the storage time which includes 2 levels ie 12 and 24 hours. The replication used was 4 times. The observed variables were physical quality of meat (water holding capacity, cooking loss, and tenderness), chemical quality (moisture content, protein content, and fat content), and microbiological quality (total microbes, pH, acid total, and total phenol). The data were processed by analysis of variance, followed by Duncans multiple range test (DMRT). The results showed that there was a significant interaction (P<0.05) between liquid smoke concentration and storage time to water holding capacity, cooking loss, tenderness, fat content, total phenol, total microbial, total acid, and pH in Pegagan duck meat but did not show a significant interaction with other variables. It could be concluded that the optimum liquid smoke concentration was 4% and the optimal storage time was 12 hours.

Abstract

The research was aimed to evaluate the effects of substitution level of corn with palm kernel meal (PKM) untreated and palm kernel meal (PKM) treated by enzyme on production performance and carcass quality of broilers. The method used in this research was experiment using nested of completely randomized design with 2 factors, the main factor was type of palm kernel meal factors consisting of PKM without enzyme (B1) and PKM by mannanase enzyme (B2), and substitution corn level L0 (without substitution), L1 (12.5%), L2 (25%), L3 (37.5%) and L4 (50%). The different among the treatments were tested by Duncan’s multiple range test. The results showed that corn substitution with PKM untreated and PKM treated by enzymes was significantly effect (P<0.01) on feed conversion ratio (FCR), tenderness, meat cholesterol of broilers and income over feed cost (IOFC) and significant effect (P<0.05) on feed intake, but there is no significant difference on body weight, percentages of abdominal fat and water holding capacity (WHC). Furthermore, effect of level corn substitition with PKM nested to PKM untreated and PKM treated by enzyme was significantly effect (P<0.01) on feed intake, body weight, FCR, and tenderness and significant effect (P<0.05) on IOFC, but there is no effect on percentages of abdominal fat, WHC and meat cholesterol. It can be concluded that the corn substitution with PKM treated by enzymes was better than the PKM untreated on production performance and carcass quality of broilers. The optimal level of PKM untreated as corn substitution was 12.5%. While replacement with PKM treated by enzyme can be used up to 25%.

Abstract

The purpose of this study was to obtain information on the readiness of the development of beef cattle using ranch system in Bintuni District, West Papua through the approach of four local foundations in terms of technical, social and economic. Research respondents were 62 families from four local foundations in Bintuni. Determination of technical feasibility of land used based on carrying capacity for each land. The technical feasibility of raising cattle was observed based on the knowledge of the community. Socio-economic feasibility was investigated based on food security and poverty level of society. Business feasibility was determined according to economic indicators namely B/C ratio, NPV, and IRR. The results showed that the aspect of land provided by the four foundations was feasible with the average carrying capacity based on fresh weight, dry matter and TDN basis were 1.8, 2.0 and 4.5 AU/Ha/year, respectively. About 70,91% of the farmers are free from poverty, and 46.54% are in food security status, which means that cattle raising will not be disturbed by the basic requirement of life but it needs an initial investment to support cattle raising. Based on the technical feasibility of cattle raising as much as 64,39% having no knowledge of breeding, therefore training and livestock education should be done. The business feasibility result shows that cattle raising on a breeding scale of 60 heads were feasible with positive NPV value of IDR 2.536.610,211, IRR of 23.09% and B/C of 1.22. Based on the overall indicators, it was concluded that the development of beef cattle in Bintuni regency was feasible to be done with requisite that improvement on land aspects and community knowledge about raising cattle.

Abstract

This research aims to determine the performance and income of a closed-house broiler farm with partnership business model. The research was conducted in a closed broiler house owned by Faculty of Animal and Agricultural Sciences, Diponegoro University, Semarang, and in a partnership with Cemerlang Unggas Lestari Ltd. A total of 11,000 broilers were raised for each raising period in the closed broiler house. A case study method was used for this research. The data were collected using survey method; the primary data were collected directly through interviews with the farm operators and field observation. Secondary data were collected through data recording which includes production cost and obtained revenue in each raising period. The collected data were then analyzed through quantitative-descriptive study. A total 7 raising periods of production cost, revenue, and income in a year were analyzed. The results of this study showed that the average performances of closed-house broiler farm yield 0.37% depletion, 1.49 FCR, and 398.46 PI. The economic indicators showed that the average production cost was Rp292,668,800.00; which generated Rp327,300,779.00 revenue, Rp34,631,978.00 income, Rp183.055.535.00 NPV, and 1.24 BCR. The study concluded that the closed-house broiler farm generates higher income compared with the open house system.

Abstract

Tanning process using free chromed material is needed to reduce toxic content in leather. The aims of this study is to increase byproducts of livestock by goat skin tanning with free chrome tanning materials. This study used glutaraldehyde as tanning materials. Goat skins were tanned become upholstery leather, and then physical quality was determined. The materials of this study were pickle goat skin. Physical testing conducted in Balai Besar Kulit, Karet dan Plastik (BBKKP) Yogyakarta. The results were analyzed descriptively using SPSS version 17.0 for Windows. Statistical analysis showed that the value of tensile strength, elongation, tear strength, sewing strength, scrub resistance of paint to dry and wet were 166.025 ± 72.315 kg/cm²; 69.910 ± 9.107%; 26.785 ± 6.031 N/cm; 115.120 ± 18.681 kg/cm; 0.775 ± 0.353; 0.775 ± 0.353 respectively. This study showed that the physical quality of upholstery leather which tanned using free chrome tanning materials have the physical qualities that met with SNI standard for leather upholstery.

Abstract

This research aims to understand the effect of metal ions bioabsorption which belong on different elemental groups to the methanol dehydrogenase (MDH) enzymatic activity in nitrogen-fixing bacteria Bradyhizobium japonicum USDA 110. Ten metal ions with each have 30μM concentration were added to grow Bradyhizobium japonicum USDA 110 in 10-1 diluted nutrient medium. The MDH activity test showed a similar result between the bacteria grown in medium without metal ions addition (control) and the bacteria were grown in a calcium ion/Ca2+ added media. The highest MDH enzymatic activity was shown on the bacteria grown in a magnesium/Mg2+ added medium, which showed 0.08 (U/mg) enzymatic activities. The addition of magnesium/Mg2+ metal ion accelerates the bacterial growth by 2.6 times and MDH activity by 1.28 times compared to control. The MDH enzyme is essential, especially for bacteria which exist in the soil environment, to adapt to high methanol concentration and to support bacterial anaerobic growth capacity along with plant symbiotic process. Moreover, the MDH activity staining method could also act as pollutant indicators like metal ions and hydrocarbon derivates. This research concluded that metal ions biosorption (calcium/Ca2+ and magnesium/Mg2+) are required for bacterial cells reproduction and oxidation of single carbon chain compounds like methanol. The nitrogen-fixing symbiotic bacteria, Bradyhizobium japonicum USDA 110 showed high MDH activity after the two metal ions absorption. However, contrary results were shown on vanadium/V3+, manganese/Mn2+, iron/Fe3+, copper/Cu2+, zinc/Zn2+, and aluminum/Al3+ absorption, which showed low MDH activity and cells biomass.
