Abstract

Madura cattle is one of the Indonesian local cattle breeds derived from crossing between Zebu cattle (Bos indicus) and banteng (Bos javanicus). Branched-chain α-ketoacid dehydrogenase (BCKDH) is one of the main enzyme complexes in the inner mitochondrial membrane that metabolizes branched chain amino acid (BCAA), ie valine, leucine, and isoleucine. The diversity of the nucleotide sequences of the genes largely determine the efficiency of enzyme encoded. This paper aimed to determine the nucleotide variation contained in section  intron 7, exon 8, and intron 8 genes BCKDHA on Madura cattle. This study was conducted on three Madura cattle that used as bull race (karapan), beauty contest (sonok), and beef cattle. The analysis showed that the variation in intron higher than occurred in the exon. Simultaneous indel found at base position 34 and 68 in sonok cattle. In addition, the C266T variant found in beef cattle. These variants do not cause significant changes in amino acids. There was no specific mutation in intron 7, exon 8, and intron 8 were found in Madura cattle designation. This indicated the absence of differentiation Madura cattle designation of selection pressure of BCKDHA gene.

Abstract

Local beef cattle is still the major source of domestic beef consumption, yet their productivity and quality is relatively low. This study was aimed to investigate productivity and meat quality traits of local beef cattle through fattening using cereals based concentrate containing different energy level. Nine Ongole cattle with average initial liveweight of 254.67 ± 20.7 kg and age between I1-I2 permanent incisor teeth were used in the study. They were fattened for four months and alloted into three different energy rations; they were low (ER) 57.88% TDN, medium (ES) 63.72% TDN, and high (ET) 69.67% TDN. The three rations had simmilar protein contents, between 12.42-12.96%. Observed parameters included cattle performance (ration and nutrition dry mater intake, daily gain, and feed conversion), carcass traits (carcass weight and percentage, fat thickness, and loin eye area at the 12th rib), and meat quality traits (pH, water holding capacity, tenderness, cooking loss, marbling score, and meat color). The results showed that cattle fed with high energy (ET) ration had the highest body weight and marbling score, and more efficient in feed conversion than medium (ES) and low energy (ER) rations. Local beef cattle productivity and there meat quality could be improve through fattening using high energy cereals based concentrate.

Abstract

This study were conducted to determine the presence of toxocariosis in beef cattle in Kasiman Subdistrict, the Regency of Bojonegoro kept under the Sekolah Peternakan Rakyat (SPR) programme. The experiment were done to identify several risk factors that may play certain roles on the degree of prevalence and infection intensity. A total of 263 head of local beef cattle with various ages were taken for faecal samples and 80 farmers were interviewed. Results of the experiment indicated the prevalence of toxocariosis was 5.32% (CI 95% = 2.61-8.04%) influenced by age and gender which showed an effect on the risk of infection. The prevalence tends to decline by age group (p <0.001): calves under 6 months of age (23.68%), and between 6-12 months of age (7.41%), 1-2 years old calves (2.04%) and the above 2 years adults (1.34%). The prevalence is higher in the males group (12.7%) compared to the females group (3.0%), (p=0.003). Logistic regression analysis concluded that age was the only affecting variable to the rate of infection with mathematical models for toxocariosis. Logit toxocariosis = -0.058 -1.098 age.

Abstract

A group of components called as 3-monochloropropane-1,2-diol (3-MCPD) esters has been found in some vegetable oils, particularly palm oil. Analysis of these components needs GC-MS instrument to reach a low level detection, parts per billion or ppb level. An effort to validate the 3-MCPD esters analysis by GC-MS method (Weiβhaar method) using phenyl boronic acid (PBA) as a derivatizing agent and isotopic internal standard (IS) 3-MCPD-D5 followed by the determination of 3-MCPD in commercial palm oils has been conducted. The analytical results showed the instrumental performance as follows: instrument linearity for the analysis of 3-MCPD standard solutions containing IS 2.0 μg/mL was obtained at a concentration range of 0.25-7.50 μg/mL test solution with R2 higher than 0.990, precision of 3-MCPD as well as IS retention time was less than 2.0% which was acceptable according to JECFA guidance. Instrument detection limit (LOD) and limit of quantitation (LOQ) were read from GC-MS based on the linearity experiment, i.e. 0.17 and 0.59 μg/mL solution, respectively. Method linearity by using palm oil as a matrix also showed R2 higher than 0.990. Recovery test result at 40 μg/g gave an accuracy at 83.29 ± 7.12% which is in the range of AOAC standard (80-110%) and precision at 5.38% (less than RSDHORWITZ). Analysis of 14 palm oils from local market showed 100% of the sampels were positive to contain 3-MCPD esters. 3-MCPD esters in palm oil were found at a concentration range of 8.15-58.14 µg 3-MCPD/g sample.
