Abstract

Background: Enterococcus faecalis and Fusobacterium nucleatum are the most common bacteria found in infected root canal teeth and most of them often caused failure in endodontic treatments. These bacteria can form biofilm which makes them more resistant against antibacterial agents. Biofilm formation also causes a decrease in antibiotics and antimicrobials sensitivity. Pluchea indica Less leaves is a species of plants that has several chemical properties. It consists of flavonoids and polyphenols which have benefits to inhibit biofilm formation. Because of its benefits, the extract of Pluchea indica Less leaves can be potentially developed as one of ?sterilization dressing in root canal teeth. Purpose: The aim of this study was to determine biofilm formation inhibition of Pluchea indica Less leaves extract against Enterococcus faecalis and Fusobacterium nucleatum. Materials and Methods: The dilution method was done first to show the Minimum Inhibitory Concentration (MIC) of the extract. The inhibition biolfilm formation was tested using microtitter plate assay by measuring the bacterial biofilm Optical Density (OD) from ELISA reader?s results and using autoagregation assay to show the inhibition of adherance bacteria.? The Pluchea indica Less leaves extract concentration used for inhibition of biofilm formation were 100%, 50%, 25%, 12,5%, and 6,25%. Results: The result of biofilm formation inhibition showed that Pluchea indica Less leaves extract were able to inhibit Enterococcus faecalis and Fusobacterium nucleatum? biofilm formation with strong moderate effect. The autoagregation assay showed a decrease in autoagregation percentation of Enterococcus faecalis and Fusobacterium nucleatum. Abstract

Background: Tooth loss can be replaced by the use of dentures. Cleaning of dentures by brushing using toothpaste can cause surface roughness because of abrasive materials. The use of toothpaste can be replaced with paste of extract Cymbopogon citratus because it has antibacterial and antifungal effects. Purpose: to determine the effect of brushing using paste of extract Cymbopogon citratus on the surface roughness of heat cured acrylic resin. Methods: Sample were heat cured acrylic resin (62.5 x 10 x 2.5) mm, divided into 4 groups consist of negative control group was brushed with basic paste, positive control group was brushed with herb paste, and two groups were brushed with paste of Cymbopogon citratus extract with 30% and 60% concentrations. Brushing was done for 3 minutes with 60 brushing times and then measured with surface roughness tester. The data was processed by ANOVA test. Result: The average surface roughness in K-, K+, P1, and P2 were 0,1217, 0,3433, 0,1633, and 0,27 ?m. There was significant difference between negative control group and positive control group; negative control group with P1 and P2 group; positive control group with? P1 and P2 group; P1 group with P2 group (P <0.05). Conclusion: The paste of Cymbopogon citratus extract concentration of 30% was effectively used because the level of surface roughness below the maximum limit of recommended surface roughness of dentistry material which is 0,2 ?m. The 60% concentration paste of Cymbopogon citratus extract was less effective because the level of surface roughness above the maximum limit of surface roughness is recommended.

Abstract

Latar belakang: Salah satu bahan pengisi suatu defek tulang dapat berupa hidroksiapatit. Hidroksiapatit dapat diperoleh dari ekstrak cangkang kerang darah Anadara granosa yang memiliki kandungan kalsium yang tinggi. Kandungan mineral tersebut dapat dijadikan sebagai bahan pengisi tulang untuk bone grafting. Namun, bahan tersebut belum diketahui bioviabilitasnya terhadap jaringan periodontal sehingga perlu dijui bioviabilitasnya. Pengujian bioviabilitas ini menggunakan sel punca mesenkimal karena sel punca mesenkimal dapat berdiferensiasi menjadi jaringan periodontal. Tujuan: Untuk mengetahui bioviabilitas ekstrak cangkang kerang darah Anadara granosa terhadap sel punca mesenkimal. Bahan dan Metode: Penelitian ini dilakukan menggunakan rancangan post only control group design. Stem sel mesenkim dalam 96 sumuran dibagi menjadi kelompok kontrol sel (n=7), kontrol media (n=7), dan perlakuan (n=7). Kelompok perlakuan diberi berbagai dosis ekstrak cangkang kerang darah Anadara granosa dengan konsentrasi 54 mg/ml, 27 mg/ml, 13,5 mg/ml dan 6,75 mg/ml. Sel punca mesenkimal tersebut diinkubasi selama 24 jam sebelum dan sesudah perlakuan. Setelah diberi MTT, optical density dibaca dengan ELISA reader dan dihitung persentase viabilitasnya. Data viabilitas sel tersebut dianalisa dengan uji statistik Kruskall-Wallis, Mann-Whitney. Hasil: Dari menunjukan hasil bahwa terjadi peningkatan viabilitas sel terhadap ekstrak cangkang kerang darah Anadara granosa. Viabilitas sel meningkat dimulai dari kelompok perlakuan konsentrasi 54 mg/ml (23,67%), 27 mg/ml (57,43%), 13,5 mg/ml (68,87%), 6,75 mg/ml (81,92%). Viabilitas sel tertinggi pada konsentrasi 6,75 mg/ml (81,92%). Kesimpulan: Ekstrak cangkang kerang darah Anadara granosa memiliki bioviabilitas sel paling tinggi pada konsentrasi 6,75 mg/ml dan bioviabilitas sel yang paling rendah pada konsentrasi 54 mg/ml.?

Abstract

Latar belakang: ?Perawatan ortodonti membutuhkan waktu yang lama sehingga perlu diusahakan? cara untuk mempersingkatnya. Ekstrak propolis memiliki berbagai kandungan, antara lain flavonoid yang mungkin dapat meningkatkan jumlah fibroblas di daerah tarikan pada perawatan ortodonti. Tujuan: Penelitian ini bertujuan untuk membuktikan pengaruh gel ekstrak propolis terhadap jumlah fibroblas di daerah tarikan untuk mempercepat pergerakan gigi pada perawatan ortodonti. Bahan dan Metode: Subyek penelitian adalah tiga puluh Cavia cobaya jantan yang dibagi dalam tiga kelompok. Kelompok pertama, (P) diberi perlakuan dan pemberian gel ekstrak propolis. Kelompok kedua K(+) diberi perlakuan dan pemberian gel NaCMC. Kelompok ketiga, K(-) sebagai kontrol negatif. Setelah 14 hari Cavia cobaya dikorbankan. Fibroblas diperiksa menggunakan mikroskop cahaya dengan menggunakan pembesaran 400X. Data yang diperoleh dianalisis dengan uji ANOVA dan dilanjutkan dengan uji LSD (p=0,05). Hasil: Rerata dan SD pada kelompok K(-):100 ? 13,2; K(+):80 ? 13,05; (P):176 ? 17,9. Uji ANOVA menunjukkan adanya perbedaan bermakna pada jumlah fibrolas. Uji beda memperlihatkan perbedaan bermakna pada semua kelompok dengan hasil signifikansi lebih kecil dari 0,05. Simpulan: Melalui penelitian ini dapat disimpulkan bahwa gel ekstrak propolis efektif meningkatkan? jumlah fibroblas di daerah tarikan pada pergerakan gigi ortodonti.

Abstract

Introduction:The Dental Public Health Warrior (DPHW) project is a social activity which provides a creative, fun, and interactive dental health education throughout?? elementary schools in Keputih Area-Surabaya,Indonesia. Purpose: The aim of this study was to determine the effectivity of those projects in increasing the Performance Treatment Index of elementary school?s student. PTI score used to measure the willingness to treat their teeth. Materials and Methods:In this pre experimental study (pretest and posttest only design), subjects were 54 elementary students (4th grade) . All subjects were educated by the DPHW team and their DMF-t score and PTI score were collected by calibrated dentists before the education and in 3 months after the education. Results: The mean of DMF-t score increase? from 6,7 to 6,9. The mean of PTI score increase from 3,7 to 9,5 . Wilcoxon analysis showed there is significant different between before and after education (p= 0,001). Conclusion:. The DPHW project can be effectively used to increase the Performance Treatment Index of elementary school?s student. The increase of PTI indekx means there is increase in willingness to treat their teeth.?Keywords: Dental Public Health Warrior Project,? Abstract

Background: Channa striata contains important compounds in the process of tissue synthesis and important role in wound healing, such as albumin, zinc (Zn), copper (Cu), iron (Fe) and unsaturated fat acids. Purpose : To determine the effect of Channa striata extract to the amount of neutrophil in healing process of traumatic ulcer. Material and Method : The design of this study used randomized post test only control group design. 18 wistar rats were divided into 6 groups, i.e: K-1 (no treatment for 1 day), K-3 (no treatment for 3 days), K+1 (giving hyaluronic acid 0.2% for 1 day), K+3 (giving hyaluronic acis 0.2% for 3 days), P1 (giving Channa striata extract 100% for 1 day), P3 (giving Channa striata extract 100% for 3 days). Results : There was a decrease in the number of neutrophil in the treatment group. The mean and standart deviation of the number of neutrophil in the K-1 group (75,96?3,65), K-3 (50,76?2,04), K+1(72,96?2,97),K+3(45,06?2,83),P1(54,20?1,31),P3(32,50?,85). Conclusion : The using of Channa striata extract 100% for 3 days are the most effective in decreasing the amount of neutrophil in healing process of traumatic ulcer of Rattus Novergicus Strain Wistar.

Abstract

Latar belakang: Pergerakan gigi ortodonti tergantung pada remodeling tulang yang melibatkan osteoklas dan osteoblas. RANKL memiliki peran penting pada osteoklastogenesis dan meningkat selama pergerakan gigi ortodonti. Kafein yang terkandung dalam minuman kopi teh, dan minuman berkarbonasi dapat meningkatkan osteoklastogenesis. Tujuan: untuk menganalisis efek pemberian kafein terhadap ekspresi RANKL dan jumlah osteoklas pada pergerakan gigi ortodonti. Bahan dan Metode: Metode penelitian yang digunakan adalah eksperimental laboratoris, 16 tikus wistar dibagi menjadi 2 kelompok, yaitu kelompok K: tikus diberikan gaya mekanis ortodonti (GMO) dan kelompok P: tikus diberikan GMO dan kafein sebesar 1,37mg / 100g BB dua kali sehari pagi dan sore. GMO pada tikus dilakukan dengan cara pada gigi molar-1( M-1) rahang atas (RA) kanan dan pada kedua gigi insivus RA diberi kawat ligatur dengan diameter 0,20 mm. Kemudian M-1 RA kanan digerakkan ke mesial dengan Niti closed coil spring. Pengamatan dilakukan pada hari ke 15 dengan pemeriksaan histologi untuk menghitung jumlah osteoklas dan pemeriksaan imunohistokimia untuk menentukan ekspresi RANKL. Hasil: Pemberian kafein meningkatkan jumlah osteoklas pada daerah tekanan dan daerah tarikan (p <0,05). Jumlah osteoklas pada daerah tekanan lebih besar daripada daerah tarikan (p <0,05). Pemberian kafein meningkatkan ekspresi? RANKL pada daerah tekanan dan daerah tarikan (p <0,05). Ekspresi RANKL pada daerah tekanan lebih besar daripada daerah tarikan (p <0,05). Simpulan: Pemberian kafein meningkatkan ekspresi RANKL dan jumlah osteoklas pada pergerakan? gigi ortodonti, sehingga dapat menjadi alternatif mempercepat perawatan ortodonti.

Abstract

Background: Candidiasis is one of oral infectious disease caused by Candida albicans.The role of C. albicans as pathogen opportunistic in oral infection can affected by immune system. Phagocytosis has contributed in immune system against C. albicans infection, which is the role of monocytes. Grapthophyllum L. Griff has been proved to increase macrophage?s phagocytic activity, but the effect of violet leaves ethanol extract (EEDU) to increase monocyte?s phagocytic activity on C. albicans is unknown. Objectives: To determine the effect of violet leaves extract on monocyte?s phagocytic activity on C. albicans. Methods: This research was acted in vitro on human monocyte cell culture. There was 5 groups: negative control group, positive control group (incubated in Isoprinosine) and treatment groups was incubated in EEDU 2,5%, 5%, 10%. The percentage of activated monocytes was counted. Result: The result showed that incubation of EEDU 2,5%, 5% and 10% could increase monocyte?s phagocytic activity significantly (p<0,05) compared to control groups. The number of monocyte?s phagocytosis on C. albicans incubated with EEDU 2,5%, 5% and 10% is 65%, 60,5%, and 58,75%. The optimal concentration of EEDU to increase monocyte?s phagocytic activity was 2,5%. Conclusion: The study showed that EEDU can increase monocyte?s phagocytic activity on C. albicans.??????????? Abstract

Latar belakang: Penyembuhan pada kerusakan tulang membutuhkan sel-sel untuk membentuk tulang baru. Fibroblast growth factor-2 termasuk sinyaling kontrol pada diferensiasi fibroblas menjadi sel osteoblas. Aplikasi hidroksiapatit pada proses penyembuhan tulang dapat membantu proses regenerasi tulang sebagai kerangka. Pemberian minyak ikan lemuru yang mengandung EPA dan DHA sebagai immunodulator yang dapat mempercepat dan mengoptimalkan fase inflamasi pada fase penyembuhan tulang. Tujuan: Penelitian ini bertujuan untuk mengetahui efektivitas kombinasi minyak ikan lemuru (Sardinella longiceps) dan aplikasi hidroksiapatit terhadap ekspresi fibroblast growth factor-2 (FGF-2) pada proses bone healing. Bahan dan Metode: Unit eksperimen (tikus wistar jantan, n=20) dibagi menjadi 2 kelompok, kelompok kontrol dan kelompok perlakuan. Kelompok perlakuan diberikan minyak ikan lemuru secara per oral selama 7 hari sebelum dilakukan defek, dan dilanjutkan 7 hari setelah dilakukan defek. Kelompok kontrol tidak diberikan minyak ikan lemuru. Semua tikus wistar yang telah diberikan 2 defek difemur kanan, ada yang diaplikasikan hidroksiapatit (K+ dan P2) dan tidak diaplikasikan hidroksiapatit (K- dan P1). Defek dibuat sebesar setengah diameter round bur di femur. Tikus dikorbankan 7 hari setelah pembedahan dan femur di potong secara transversal dan dilakukan pengecatan IHC menggunakan Anti-bFGF Polyclonal antibody (Bioss?). Pengamatan dilakukan menggunakan mikroskop cahaya. Data dianalisis menggunakan one-way Anova. Hasil: Berdasarkan pada hasil uji ANOVA dan uji LSD, terjadi peningkatan rata-rata ekspresi FGF-2 pada kelompok P2 : 15,29?5,251, dibandingkan dengan K- : 5,57?2,070 , K+ : 12,14?3,976? dan P1 : 8,14?1,676 (P<0.05). Simpulan: Kombinasi minyak ikan lemuru (Sardinella longiceps) dan aplikasi hidroksiapatit efektiv secara signifikan terhadap ekspresi fibroblast growth factor-2? FGF-2 pada proses bone healing pada hari ke-7.

Abstract

Background: Dental caries is a major oral health problem which is most commonly found in Indonesia. One of the main factors in role of causing dental caries is Streptococcus mutans which is the primary target of the prevention of caries. Leaf extract of Sonneratia alba hasbeen proven to have an antimicrobial effect towards Streptococcus mutans, but its cytotoxic activity on normal cells is still unknown. The initial cell viability in this study was conducted using cell line BHK-21. Purpose: The aim of this study is to determine the cell viability offibroblast cells line BHK-21 by given Sonneratia alba?s leaf extract.?Materials and Methods: The experiment was conducted using post test only control group design. BHK-21 fibroblast cell culture on 96 wells were divided into cell control group (n=9), media control group (n=9) and test groups (n=9). The test groups were given various concentration of Sonneratia alba?s leaf extract, they were: 5%; 10%; and 20%. The cell cultures were incubated for 24 hours before and after treatment using Sonneratia alba?s leaf extract. After dripping MTT into the microplates, OD were read using ELISA reader and the viability were counted. The data that was acquired from the viability calculation was statistically analyzed using One Way ANOVA and LSD test. Results: Data showed the increase of viability cell in all the test groups. The highest viability cell showed by the test group with 10% concentration of Sonneratia alba (153,7122%?7,42458), and the lowest found in the test group with 20% concentration of leaf extract (141,4682%?7,48752).
