Abstract

Kumbang badak (Oryctes rhinoceros L.) dapat hidup dengan optimal dengan dukungan dari faktor ekologis yang cocok. Faktor ekologis meliputi habitat limbah material organik di vegetasi tersebut, musuh alami, musim, dan faktor cuaca (suhu, curah hujan dan kelembaban) diteliti pengaruhnya terhadap struktur populasi kumbang badak pada Oktober-Desember 2015. Penelitian ini merupakan penelitian survei dengan pengambilan sampel dilakukan sebanyak empat kali, dua kali pada musim kemarau dan dua kali pada musim penghujan. Data dianalisis dengan analisis ANOVA dan untuk mengetahui faktor ekologis yang paling berpengaruh dilakukan uji lanjut dengan Duncan. Berdasarkan ANOVA, ada perbedaan antar habitat F=0,012 untuk perbandingan antara jerami dan serasah daun, 0,002 antara jerami dan tanpa limbah, 0,000 antara serasah daun dan tanpa limbah, serta 0,001 untuk batang pohon dan tanpa limbah (P<0,05). Untuk stadium pupa dan imago hasil analisis ANOVA menunjukkan hasil tidak berbeda nyata. Faktor musim memberikan hasil bahwa terjadi peningkatan di ketiga stadium yang ditemukan dengan jumlah yang berbeda. Stadium larva mengalami peningkatan sebanyak 381 larva, stadium pupa sebanyak 25 pupa, dan peningkatan sebanyak 166 ekor kumbang badak stadium imago. Setelah dilakukan uji statistik dengan uji t, jumlah populasi kumbang badak pada musim kemarau dan penghujan berbeda nyata dengan nilai uji t =0,000 untuk stadium larva, sedangkan pada pupa, t = 0,038 (P<0,05). Setelah dilakukan uji lanjut diperoleh hasil habitat limbah material organik merupakan faktor ekologi yang paling berpengaruh. Habitat limbah material organik yang paling berpengaruh adalah batang pohon, disusul dengan habitat serasah dan jerami.

Abstract

Javanese Gibbon (Hylobates moloch) is the endemic primate species in Indonesia. IUCN - World Concervation has given this species an endangered status. The habitat Hylobates moloch’s were found at Slamet Mountain in Central Java. The study entitled "Study of Javanese Gibbon (Hylobates moloch) Population in Slopes of Slamet Mountain, Central Java" aimed to determine the number of population in slopes of Mount Slamet. This research was conducted in the exact line these July -November 2013, the survey method with stratified sampling base on line transect method was used in this research. Transects pathway in this study used existing pathways wich the passed and cut the contour of research sites. Javanese Gibbon (Hylobates moloch)  obtained along the transect was calculated and analyzed using perpendiculars distance approach (DISTANCE 5.0’s software). Analysis by DISTANCE 5.0 showed that the density of Javanese Gibbon (Hylobates moloch) at Slamet Mountain in 170 km2 area was 0.26 individual/km2, coefficient of variation was 34.88 % , lower limit of 95 % confidence interval was 0.13365 to 0.51981. Based on the analysis, total number of javanese gibbon population at Slamet Mountain was 45 individuals of the density 0.26 individuals/km2.

Abstract

Darah merupakan komponen penting karena menunjukkan kondisi fisiologis individu. Oleh  karena  itu  darah  menjadi salah  satu  parameter  pokok  dalam penelitian praklinik/ biomedik. Hematologi merupakan ilmu yang mempelajari kondisi sel-sel darah perifer dalam kondisi normal maupun patologis. Parameter pemeriksaan hematologis yang rutin dilakukan antara lain profil eritrosit dan leukosit. Sampel darah yang diterima kadangkala tidak langsung diperiksa karena berbagai alasan. Untuk menjaga supaya kondisinya tidak rusak, maka sampel darah ditambah antikoagulan dan disimpan di dalam lemari pendingin selama beberapa jam hingga beberapa hari. Penelitian ini bertujuan untuk mempelajari profil eritrosit dan leukosit pada sampel darah tikus (Rattus norvegicus Berkenhout, 1769) Galur Wistar yang sehat/normal dengan antikoagulan EDTA atau Heparin dan variasi waktu penyimpanan (0, 6, 18, 24, dan 48 jam). Untuk pembahasan lebih lanjut, data dianalisis secara statistik berdasarkan ANOVA two-factor (P<0,05). Hasil menunjukkan bahwa antikoagulan berpengaruh secara signifikan terhadap profil eritrosit dan leukosit (P<0,05). Nilai profil eritrosit pada sampel darah-EDTA lebih rendah daripada sampel darah-Heparin. Sebaliknya pada pemeriksaan profil leukosit, sampel darah-EDTA memiliki kecenderungan nilai lebih tinggi daripada sampel darah-Heparin. Seiring bertambahnya waktu penyimpanan, jumlah eritrosit, total leukosit, neutrofil, dan limfosit berkurang secara signifikan (P<0,05). Waktu penyimpanan tidak berpengaruh secara signifikan terhadap nilai hematokrit, kadar hemoglobin, jumlah monosit, eosinofil, dan basofil (P>0,05). Disimpulkan bahwa pemeriksaan profil hematologis yang terbaik adalah menggunakan darah tanpa antikoagulan namun harus langsung dilakukan segera setelah sampel diperoleh (sebelum darah mengalami koagulasi). Apabila tidak memungkinkan, maka dapat digunakan EDTA atau Heparin, dan jenis antikoagulan harus dijelaskan dalam pelaporannya. Pemeriksaan darah dengan antikoagulan hendaknya juga tetap dilakukan segera setelah sampel diterima (tidak ditunda).

Abstract

This research was aimed to observe the anatomical character of mangrove plants leave and examine the differences from leaves anatomical character of mangroves in Donan river and Tritih forest in Cilacap regency as an self-adaptation indicator. The method which is used was survey method by collecting leaves sample of each mangrove plants. The sample was preparated using embedding method, and stained with safranin 1% disolved in alcohol 70%. The parameter of variable which is observed in leaves anatomical character are thickness of leaves,  leaves mesophyll thickness, cuticle thickness, stomata length size, and amount of stomata and trichome per leaves unit area. The data were analyzed with t-test for examining anatomical character difference. The results showed that four usual types of mangrove plants such as Achantus ilicifolius, Rhizospora mucronata, Aegiceros corniculatum, and Bruguiera gymnorrhiza have similar leaves anatomical structure. The anatomical character from polluted habitat were different from unpolluted habitat. This differences was showed in leaves mesophyll thickness, cuticle thickness, stomata length size, and amount of stomata and trichome per mm2 leaves unit area.

Abstract

This research aimed to find out the interaction effect between mixed Vesicular Arbuscular Mycorrhiza (VAM) and phosphate fertilizer to the growth of red chili (C. annuum) in acid soil, and to fnd out the best combination of mixed VAM and phosphate fertilizer to the growth of red chili (C. annuum) in acid soil. This research used an experimental method with Completely Randomized Design (CRD) in a factorial pattern with two factors. The first factor was mixed VAM dosages consisted of four levels: 0; 10; 15; 20 g/plant. The second factor was phosphate fertilizer dosages consisted of four levels:  0; 0,2; 0,4; 0,6 g/plant. Each combination treatment had three replication. The parameters were observed in the form of plant height, stem diameter, plant top dry weight, degree of VAM infection, and P content of plant tissue. Data obtained from the observation was analyzed with Analysis of Variance (ANOVA) at an error rate of 5% and 1%, treatment that showed significant or very significant result, then followed with Honestly Significant Difference (HSD) test. The result showed that interaction between mixed Vesicular Arbuscular Mycorrhiza (VAM) and phosphate fertilizer did not increase the plant height, stem diameter, and plant top dry weight, but each factor increased the plant height, stem diameter, and plant top dry weight. VAM dosage inoculation of 20 g/plant without phosphate fertilizer is the most effective combination in increasing the degree of VAM infection.

Abstract

Bakteri asam laktat (BAL) yang digunakan dalam pembuatan soyghurt di antaranya     Lactobacillus bulgaricus, Streptococcus thermophillus, dan Bifidobacterium sp. dengan suhu optimal pertumbuhan 36°-37°C dan lama inkubasi berkisar 1-2 hari. BAL diketahui mampu menghasilkan bakteriosin yang dapat menghambat bakteri patogen dalam saluran pencernaan salah satunya ialah Bacillus cereus. Tujuan penelitian ini ialah mengetahui perbandingan BAL pada soyghurt terhadap daya hambat B. cereus, mengetahui lama inkubasi soyghurt dengan penambahan Bifidobacterium sp terhadap pertumbuhan B. cereus, dan mengetahui pengaruh konsentrasi BAL dan lama inkubasi pada soyghurt terhadap pertumbuhan B. cereus. Hasil penelitian menunjukkan perbandingan BAL (L. bulgaricus : S. thermophillus : Bifidobacterium sp.) 1:1:1 (K1) pada soyghurt mampu menghambat B. cereus. Lama inkubasi soyghurt dengan penambahan Bifidobacterium sp. optimal pada 24 jam terhadap pertumbuhan B. cereus. Konsentrasi BAL dengan lama inkubasi pada soyghurt tidak mampu meningkatkan daya hambat pertumbuhan B.cereus.

Abstract

Genetic diversity of winged-bean [Psophocarpus tetragonolobus (L.) DC] can be produced by induced mutation with EMS (ethyl-methane sulphonate). To identify the genetic diversity after EMS application on winged-bean, molecular analysis using RAPD (Randomly Amplified Polymorphic DNA) technique was carried out. This study aims to identify RAPD profile of winged-bean after EMS application. Winged-bean seeds of both long and short pod types were soaked in wet paper towels for 6 hours followed by soaking in EMS of 0.3% and 0.5% for 4 hours. Winged-bean seeds of both pod types without EMS served as control. Ten of the 30 primers operon technologies USA and 2 primers bangalore genei showed sufficiently clear bands (OPA 1, OPA 3, OPA 10, OPA 15, OPA 19, OPB 1, OPB 6, OPB 7, ES10C27, and ES10C28). All the ten primers produced polymorphic fragments with a total number of 320 and size ranging from 100 bp to 2,000 bp. As many as 66 loci were observed. The polymorphism values obtained ranged from 16.67% to 46.97%. This low polymorphism values indicate that EMS concentrations applied have not resulted in sufficiently high genetic variation. At the same time, the genetic distance observed were only 0.11 to 0.41.
