Abstract

Heavy metal Zn comes into waters from paint and textile factories that have not adequately treated their waste. Small amount of Zn is necessary for metabolic activity of aquatic organisms, but it may cause toxicity when presents in abundance. This study was conducted to know differences between Zn effects on microanatomy structures of intestine and those on liver of Cyprinus carpio. The treatments arranged in a Completely Randomized Design (CRD) consisted of 0 ppm (control), 2 ppm, 4 ppm, and 6 ppm of Zn, which were applied respectively for 96 hours and replicated five times. The observed parameters were height of columnar epithellial cells of intestine and hepatocyte diameter. Data were analysed with F test followed by Least Significant Difference (LSD). The results showed that there was significant difference among treatments indicating that Zn affected microanatomy structures of intestine and liver of the fish.  Increase in height of columnar epithellial cells of intestine was recorded to begin at level of 4 ppm, while that in hepatocyte diameter was found to begin at level of 6 ppm.

Abstract

The aims of this study were to investigate the effects of the mixture of tapioca waste, rice bran, starch (BOC), and humic substance (BH) on Aspergillus niger growth and its phosphate solubilizing capability. The experiment was conducted in Erlenmeyer flask containing 200 ml Pikovskaya liquid media and incubated on shaker with 100 rotaries per minute for nine days.  The Christmas Island phosphate rock was used as phosphate source.  Aspergillus niger YD 17 was taken from the Laboratory of Microbiology, Faculty of Agriculture, Gadjah Mada University. The experiment was arranged using factorial completely randomized design with three-replications (3x4x3). The level of BOC (i.e. 0, 2, and 4 g.l-1 media)   and BH   (i.e. 0, 1, 2, and 3 ml.l-1) were applied as experimental factors. Dry weight of mycelia and the soluble phosphate in media were parameters of Aspergillus niger growth. The results showed that BOC and BH increased the Aspergillus niger growth and its phosphate solubilizing capability (P£0.05). Four-gram of BOC and 3 ml of BH per litter of media were the best level for Aspergillus niger growth, while 4 g of BOC and 2 ml of BH was the best level for phosphorus solubilization capability.

Abstract

The objective of this study was to know some factors related to microbial contamination in herbal medicine products at Semarang City. This study was conducted by descriptive method employing cross sectional design in which samples were taken by purposive random sampling. The results showed that Escherichia coli, Salmonella, Staphylococcus aureus, Pseudomonas aeruginosa were identified. The number of fungi exceeded the standard indicating that 62.5% of the herbal medicines surveyed did not meet the required standard of Kepmenkes no. 661/Menkes/SK/VII/1994. The relationship between the quality of herbal row material, processing, and serving to the microbial contamination in herbal medicine products was observed.

Abstract

Study was conducted to know forest structure and regeneration of mangrove Cikiperan Cilacap. Transect method was used for this study, the data of seedling were collected by quadrat method, sapling and tree were collected by quarter method. The result of this research showed there were 7 species seedling, 11 species sapling and 18 species trees. Dominant species was Rhizophora mucronata, Rhizophora apiculata, Bruguiera gymnorhiza, Avicenia alba, Aegiceros corniculatum, Nypa fruticans, and Ficus retusa. The forest of mangrove regeneration was generally very good. Seven species, however, were recorded in critical condition, i.e. Ceriops tagal, Sonneratia alba, Intsia bijuga, Cynometra ramniflora, Cerbera adolam, Leucaena leucodendrom, and Lumnitzera littorea.

Abstract

Dicksonia is one of tree ferns species that can be found in Indonesian highlands. Due to their wide distribution this species was suggested to have genetic variation among their population. Samples from Bali, West Java (Cibodas) and East Nusa Tenggara were compared using the PCR-SSCP methods to detect genetic variation of trnL-trnF intergenic spacer from the cpDNA plastid genome. Sample from Bali and West Java (Cibodas) did not morphologically and genetically variated, but they were variated to samples from East Nusa Tenggara.

Abstract

A study on the distribution and zonation of mangrove crustacean in Mahakam Delta, East Kalimantan was conducted in July 2004. The three research locations were selected i.e. Muara Ilu, Muara Kaeli, Muara Bayor and Muara Beji. Crustaceans were collected by applying quandran methods (1 x 1 m) in transect lines. The diversity index (H’) of crustacean in Delta Mahakam: 1.382 - 2.028 and the Eveness  index (E) : 0.5877 - 0.7101. The highest diversity index was recorded from muara Kaeli and the highest Evenes index  was in Muara Ilu.

Abstract

The controlling of fascioliasis has not been satisfying so far, since the effort of breaking off the fluke life cycle often leads to resistance. The resistance is due to the synchronization between the parasites and their snail host and high infection ability of the parasites enabling them to finish their life cycle. Ultraviolet irradiation provides alternative approach possibly applied to cope with the resistance. The objectives of this research were to know the ability of F. gigantica miracidium in infecting Lymnaea sp. at various ages and  to determine time exposure of ultraviolet (254 nm) effectively reducing the ability of miracidium infection. The results indicated that susceptible age of Lymnea javanica is five weeks old, while the most effective time exposure of ultraviolet (254 nm) in weakening  the ability of Fasciola gigantica miracidium infection to L. javanica is 60 seconds (P < 0,05).  At the moment the study on reinfection effect of F. gigantica miracidium on the immunity of L. javanica is still in progress.

Abstract

An experiment was conducted to determine the activity of pearl grass (rumput mutiara, Hedyotis corymbosa) in inhibiting growth of bacterial and fungi causing diseases in poultry.The current study used 3000 gram of sorted and cleaned pearl grass from dust and other weeds. This study employed sample collection, extraction, fractionation, preparation of medium of Agar and Mueller Hinton Broth, culturing bacterial, antimicrobial test and measuring minimum inhibitory concentrations. Extraction was conducted by maceration of pearl grass with methanol for five days followed by evaporating in vacuo to get methanol extract. The extract was then fractionated by adding hexane or acetic acid and ethyl acetic or sodium bicarbonate or n-buthanol to get fraction of hexsane, acid ethyl acetic, base ethyl acetic, n-buthanol. Each fraction was then tested with microbial test that were Salmonella sp, Escherichia coli, and Candida albicans. The results showed that each fraction at 2 % of concentration could inhibit bacterial growth except for Candida albicans that was not inhibited by any fraction of pearl grass. The minimum inhibitory concentrations was 0.4 – 0.8% (4 mg/ml – 8 mg/ml) indicating that pearl grass fractions were highly active on inhibiting bacterial growth that cause poultry diseases and the inhibiting activity on E. coli was found higher than that on Salmonella sp.
