Abstract

Psychological stress is often suffered by people with body rhythm disorders, one of whom is a person with sleep disorders upside down. Psychological stress can trigger oxidative stress. Oxidative stress can be overcome by increasing antioxidants, beans as antioxidants are considered to reduce oxodative stress. The objective of the study was to examine the effect of green bean juice on the levels of Malondialdehyde (MDA) in male white wistar strains exposed to psychological stress. The design used in this research is Randomized Post Test Only Group Design. There were 5 groups tested, 2 normal and stress control groups, and 3 treatment groups. Exposure to psychological stress with reverse sleep guard pattern, so that the condition of experimental animals experiencing oxidative stress, then given bean juice with a dose of 4.5 gr / 1 ml in the treatment group. The results showed MDA levels dropped in the first hour and remained down until the 24th hour after the provision of bean juice. The effect of antioxidant flavonoids on beans decreases the amount of free radicals by scavenging mechanisms in free radicals and increases levels of endogenous antioxidants and increases insulin levels. The role of arginine in beans acts as a precursor of the formation of Nitric Oxide (NO) as vasodilator of blood vessels so as to decrease oxidative stress. A single dose of bean juice in experimental animals has been shown to overcome oxidative stress due to exposure to psychological stress since the first hour of green bean juice and its antioxidant activity persisted until the 24th hour.

Abstract

Stres oksidatif dapat dipicu oleh paparan stres psikologis. Stres oksidatif dapat dikurangi dengan konsumsi zat antioksidan eksogen, teh hijau dianggap mampu menurunkan stres oksidatif. Tujuan penelitian ini untuk menguji pengaruh pemberian seduhan teh hijau terhadap glukosa darah, MDA dan aktivitas SOD pada tikus putih jantan Strain Wistar yang dipapar stres psikologis. Rancang bangun yang digunakan pada penelitian ini adalah Randomized Post Test Only Group Design. Terdapat 5 kelompok tikus, 2 adalah kelompok kontrol normal dan kelompok kontrol stres, serta 3 kelompok perlakuan. Sebelumnya tikus diberikan stres psikologis 1 x 24 jam berupa gangguan pola tidur jaga, sehingga didapatkan kondisi hewan coba yang mengalami stres oksidatif, kemudian dilakukan pemberian seduhan teh hijau pada kelompok perlakuan dengan dosis tunggal 3,6 ml/ekor pada jam ke-1 pada kelompok perlakuan 1, jam ke-6 pada kelompok perlakuan 2 dan jam ke-24 pada kelompok perlakuan 3. Analisis statistik menggunakan uji Manova. Hasil Berdasarkan analisis statistik menunjukkan bahwa terdapat perbedaan glukosa darah, dimana kelompok perlakuan menurun setelah jam ke-1 pemberian seduhan teh hijau, dan tetap bertahan di jam ke-6 dan ke-24, begitu pula dengan MDA, kelompok perlakuan menurun setelah jam ke-1 pemberian seduhan teh hijau, dan tetap bertahan di jam ke-6 dan ke-24. Sedangkan aktivitas SOD meningkat pada jam ke-1, kembali meningkat pada jam ke-6 dan meningkat maksimal pada jam ke-24 setelah pemberian teh hijau. Dosis tunggal seduhan teh hijau pada hewan coba yang mengalami stres oksidatif terbukti dapat menetralisir stres oksidatif pada jam ke-1 setelah pemberian teh hijau dan aktivitas antioksidannya masih bertahan sampai 24 jam.

Abstract

Hyperlipidemia is a disease whichthe effect in increased fat levels in the blood. In hyperlipidemia, the condition of high oxidative stress is obtained due to the oxidation process of LDL (Low Density Lipoprotein) in the body, this condition showed by increased of MDA (Malondialdehyde) levels in serum. Bitter melon (Momordica Charantia L.) contains a lot of active ingredients, one of the ingredients of bitter melon that is thought to have antilipidemic effects is flavonoid compounds. This study was experimental study using a randomized posttest only control group design study with a sample size of 30 male white rats (Rattus norvegicus) wistar strain which was divided into 3 groups, namely K1, K2, and K3. In each group, different treatments are given. Negative control group (K1), given standard feed, water ad libitum; Positive control group (K2), given standard feed, high fat feed with a dose of 2.5 ml water ad libitum; The treatment group (K3) was given a standard diet, high-fat feed with a dose of 2.5 ml water ad libitum and extract of bitter melonat a dose of 250 mg / kg BB by orally every morning for 3 weeks. Data analysis using One Way Anova Test. From the results of the test shows the significance of p-value = 0.000 which is <α (0.05). The results showed that there was an effect of  administration ethanol extract of bitter melon (Momordica charantia L.) to decrease in serum MDA (malondialdehyde) levels in male white wistar rats (Rattus norvegicus) given a high-fat diet.

Water Clover Extract in Liprotein Lipase enzyme Activity of The Rats with High Cholesterol Diet

Abstract

Hypertriglyceridemia or high blood triglyceride levels is one of the lipid fraction disorders associated with an increased risk of cardiovascular disease. Flavonoids has an important role as cardioprotection by lowering triglyceride levels. The flavonoids in water clover contain 10,71 mg of extract. This research aims to prove that flavonoids can increase Lipoprotein Lipase enzymes activity. True experimental study used design pre-post test control group design in rats. The sample was selected based on by completed randomized design sampling to be devided into five groups, the control positive (n=5) were only given normal diet, control negative (n=5) were only given high cholesterol diet and treatment groups with three doses (n=15) were given cholesterol diet during the 7 days and water clover extract that intervention during the 14 days. The results of this research can increase activity of Lipoprotein Lipase enzymes as an influence of water clover extract based on Paired T-Test, before and after treatment there was a significant different (p<0.05). Activity of Lipoprotein Lipase enzymes in the negative control group and third treatment showed a significant difference (p.< 0.05) based on MANOVA (Multivariate Analyses of Variance). Conclusions of this research is water clover extract affecting increase Lipoprotein Lipase enzymes activity significantly.

Abstract

Mekanisme kadmium menginduksi toksisitas pada hepar yaitu dengan menimbulkan stres oksidatif. Stres oksidatif menghambat enzim-enzim yang berperan dalam proses metabolisme glukosa. Penelitian ini bertujuan untuk menganalisis perbedaan kadar glukosa pada homogenat hepar. Penelitian ini merupakan penelitian eksperimental laboratorik yang dilakukan pada 4 kelompok: kelompok P0 yang tidak dipajankan Cd, kelompok P1 dipajankan Cd 0,03 mg/L, kelompok P2 dipajankan Cd 0,3 mg/L, dan  kelompok P3 dipajankan Cd 3 mg/L. Hasil penelitian didapatkan rata-rata kadar glukosa pada kelompok P0 sebesar 21.667 μM, kelompok P1 sebesar 33.278 μM, kelompok P2 sebesar 69.889 μM, dankelompok P3 sebesar 150.667 μM. Melalui uji statistik Kruskal-Wallis didapatkan perbedaan yang bermakna p=0,000 (p<0,05) dan uji pos hoc Mann-Whitney juga menunjukkan perbedaan yang bermakna pada semua kelompok. Dapat disimpulkan bahwa semakin tinggi konsentrasi pemajanan Cd terhadap homogenat hepar maka semakin tinggi pula peningkatan kadar glukosa pada hepar tikus.

Effects of Solo Garlic Extract in Decreasing Malondialdehyde (MDA) Levels due to E-Cigarette Exposure

Abstract

The popularity of E-Cigarette has increased significantly over the last decade, and has potential to replace tobacco cigarettes. Aerosol which is produced by E-Cigarette, contains reactive carbonyl and free radicals, inducing oxidative stress and causing inflammation, proliferation, and cell damage. This oxidative stress can be seen through the Malondialdehyde (MDA) levels in the body, and reduced by antioxidant activity in Solo Garlic extract. The aim of this study is analyzing the differences of MDA levels in four treatment groups. 28 white rats were divided into four groups and three of them were exposured by E-Cigarette. This study showed that the highest MDA levels was found in the group which was exposured by E-Cigarette without solo garlic extract (7,179 nmol/ml). MDA levels can be reduced by antioxidant activity shown in the group which was exposured by E-Cigarette and solo garlic extract dose 0,05 gram/day (the smallest dose), amount 3,715 nmol/ml. The average of MDA levels in group which was given by the smallest dose of solo garlic extract, is lower than the control group. So, it can be concluded that using solo garlic extract less than 0,05 gram/day can restore MDA levels to a normal state (4,210 nmol/ml).

Abstract

Hiperkolesterolemia merupakan suatu keadaan dimana kadar kolesterol didalam darah mengalami peningkatan diatas angka rata-rata nilai normal yang telah di tetapkan secara standar. Terapi herbal tradisional merupakan terapi alternatif yang dapat digunakan untuk mencegah dan menurunkan kadar trigliserida di dalam darah. Salah satu pilihan terapi yang digunakan adalah bawang putih siung tunggal (Allium Sativum Linn.). Bawang putih siung tunggal memiliki kandungan antioksidan yang kuat yaitu flavonoid, apabila dengan rutin mengkonsumsinya dapat menurunkan kadar trigliserida didalam darah sehingga resiko penyakit degeneratif dapat dikurangi. Flavonoid adalah senyawa golongan polifenol. flavonoid memiliki kemampuan sebagai antikolesterol. Flavonoid bekerja pada banyak enzim dalam pembentukan kolesterol atau trigliserida, salah satunya adalah enzim lipoprotein lipase. Sehingga, apabila aktivitas enzim lipoprotein lipase meningkat, maka proses pembentukan trigliserida melalui mekanisme jalur eksogen di pembuluh darah terhambat sehingga kadar trigliserida dapat menurun. Penelitian ini menggunakan jenis penelitian eksperimental laboratorium dengan design Pre test- Post test Controlled. Menggunakan 5 kelompok dengan masing-masing kelompok menggunakan 5 ekor tikus. uji Paired T-Test  menunjukkan bahwa terdapat perbedaan aktivitas enzim lipoprotein lipase dengan nilai signifikansi (p < 0.05) sedangkan pada uji Manova, nilai signifikansi yang didapatkan adalah (p. < 0.05). Berdasarkan hasil penelitian, dapat disimpulkan bahwa pemberian ekstrak bawang putih siung tunggal dapat meningkatkan aktivitas dari enzim lipoprotein lipase.

Abstract

Trauma menjadi masalah kesehatan masyarakat yang utama di seluruh dunia karena dikaitkan dengan morbiditas dan mortalitas yang tinggi baik di negara maju dan berkembang. Trauma toraks terjadi hampir pada 50% dari semua kecelakaan dan menjadi penyebab penting kematian. Penelitian ini bertujuan untuk mendapatkan data tentang trauma toraks di ruang rawat inap bedah RSUD Gambiran Kediri periode Maret 2017 ?? Maret 2018. Jenis penelitian ini adalah deskriptif retrospektif dengan menggunakan semua jumlah kasus trauma torak sebanyak 31 pasien dari 545 pasien trauma bedah yang membutuhkan rawat inap. Hasil penelitian mendapatkan kasus trauma toraks paling banyak ialah fraktur iga (29%) yang terjadi pada jenis kelamin laki-laki (93,5%) dan kelompok usia 46-60 tahun (35,5%). Penyebab tersering adalah kecelakaan lalu lintas (80,6%) sedangkan penanganan paling banyak dilakukan ialah penanganan konservatif (100%). Durasi rawat inap pasien trauma toraks paling sering sekitar 1-3 hari (58,1%) dengan hasil akhir trauma toraks yang didapat umumnya sembuh (93,6%).

The Effect of Melinjo Peel Extract in Activity of Lipoprotein Lipase Enzym of The Rats Fed a Hypercholesterolemia Diet

Abstract

The intake of saturated fat and cholesterol that comes from food digested in the intestine resulting in free fatty acids, triglycerides, phospholipids, and cholesterol. Triglyceride levels increased can be caused by presence of impaired lipoprotein lipase (LPL) enzyme activity. LPL activity can be increased by flavonoid in plant, like Melinjo peel. The purpose of this study is to measure LPL enzyme activity before and after being given extract of Melinjo peel treatment. This research use true experimental, with study design of pretest posttest control group design. Hypercholesterolemic diet given to rats by oral gavage as much as 1,8 grams for 14 days. Melinjo peel extract were given by oral gavage for 14 days after the hypercholesterolemic diet is given. Statistical analysis used Paired Sample T-Test to compare lipoprotein lipase activity before and after treatment. Then, used MANOVA (Multivariate Analyses of Variance) to see the difference of lipoprotein lipase activity in each group after treatment.Based on statistical analysis showed that there were differences of lipoprotein lipase activity in the hypercholesterolemic diet group, HD+54.15, HD+108.30, and HD+216.60 before and after treatment significant (p. < 0.05). The activity of lipoprotein lipase in the normal diet group compared with the hypercholesterolemic and HD+216.60 group showed a significant difference (p. < 0.05). Melinjo peel extract can increased activity of lipoprotein lipase enzym after treatment, those can be due to the flavonoid in Melinjo peel extract.

Efek Abelmoschus esculentus terhadap Glukosa Darah, Superoksida Dismutase, dan Malondialdehid Rattus norvegicus Diabetes Melitus dengan Induksi Streptozotocin

Abstract

In diabetes mellitus there is an increase in oxidative stress in which oxidants exceed the antioxidant system resulting from the production of oxidant-antioxidant imbalance and/or limited antioxidant defenses. Abelmoschus esculentus (??okra?) is known as lowering blood glucose vegetable by Indonesian. A. esculentus has a high antioxidant activity that can: 1) lower H2O2 level; 2) decrease the amount of ROS; 3) lower ?OH level. This study tried to prove that administration of A. esculentus may decrease fasting blood glucose, serum malondialdehyde, and increase superoxide dismutase activity of diabetic male white rats. This research is a pure experimental research, with randomized post test only control group designs, held in 28 days. Thirty one rats were divided into 4 groups randomly: KN = normal control group given 0.1% Na-CMC solution; KDM = DM control group injected with STZ, followed by 0.1% Na-CMC solution; P1 = treatment group 1 injected with STZ, followed by administration of A. esculentus at a dose of 775 mg/kgBW/day; P2 = treatment group 2 injected with STZ, followed by administration of A. esculentus at a dose of 1550 mg/kgBW/day. The result: administration of A. esculentus at doses of 775 mg/kgBW/day and 1550 mg/kgBW/day can not decrease fasting blood glucose and MDA serum level of diabetic male white rats. Giving A. esculentus at a dose of 1550 mg/kgBW/day may increase the activity of SOD erythrocytes of diabetic male white rats.. Conclusions: A. esculentus has the potential to lower blood glucose and MDA serum levels, also increase erythrocyte SOD activity.
