Abstract

Furosemide is a diuretic having low solubility and low bioavailability thus the diuretic activity is not optimal. The purpose of this study was to determine the effect of furosemide SNEDDS formation to  the diuretic activity. SNEDDS was made with a mixture of 66% tween 80, 26% propilene glycol, 8% oleic acid and furosemide 40 mg/mL. A total of 20 test animals were divided into 4 groups, namely the negative control 1 (CMC 0,5% suspension), negativ control 2 (a mixture of tween 80, propilene glycol and oleic acid), furosemid suspension and SNEDDS furosemid. Tests on the diuretic activity is done by measuring the volume of urine released for 6 hours.Furosemide SNEDDS  could enhance the diuretic activity of furosemide.

Abstract

ABSTRACTResearch of immunostimulatory application as an activator of the immune system are inconclusive and needa new immunostimulatory search from new sources. Red betel leaf is widely used as a traditional medicine totreat a variety of diseases. Many plants used as traditional medicines reported to have immunostimulatory activity. The isolation of Pc-2 compound of methanolic extract of red betel leaf have been done and was testedin vivo on macrophage phagocytic activity. Methanolic extract of red betel leaf is obtained by maceration,fractionated by vacuum liquid chromatography method, and compound code Pc-2 was isolated by preparativethin layer chromatography method. The content of the compounds in the extract, fractions, and isolateswere monitored by thin layer chromatography. Macrophage phagocytic activity assays using BALB/c mice inducedwith Lysteria monocytogenes bacteria. Phagocytic activity expressed in percentage of phagocytosis (PF),phagocytic index (IF), and the effectiveness of phagocytosis (EF). The results showed that mice treated with thecompound code Pc-2 dose of 10 mg/kg both at day3rd, 10th, and 21st after infection with L. monocytogeneshave PF, IF, and EF that significantly greater than the controlgroup mice; macrophage phagocytic activity occurredon day 21 after infection with L. monocytogenes, and there is no correlation between the PF and IF. ABSTRAKPenelitian terkait aplikasi imunostimulan sebagai aktivator sistem imun hasilnya tidak meyakinkan dan perlupencarian imunostimulan baru dari sumber baru. Secara empiris daun sirih merah banyak digunakan sebagaiobat tradisional untuk mengobati berbagai penyakit. Banyak tanaman yang digunakan sebagai obat tradisionaldilaporkan memiliki aktivitas imunostimulan. Telah dilakukan isolasi senyawa kode Pc-2 dari ekstrakmetanolik daun sirih merah dan pengujian aktivitas fagositosis makrofag secara in-vivo. Ekstrak metanolikdaun sirih merah diperoleh secara maserasi, difraksinasi dengan metode kromatografi cair vakum, dan senyawakode Pc-2 diisolasi dengan metode kromatografi lapis tipis preparatif. Kandungan senyawa dalamekstrak, fraksi, dan isolat dimonitor dengan kromatografi lapis tipis. Pengujian aktivitas fagositosis makrofagmenggunakan mencit BALB/c dan induksi dengan bakteri Lysteria monocytogenes. Aktivitas fagositosis dinyatakandalam persen fagositosis (PF), indeks fagositosis (IF), dan efektivitas fagositosis (EF). Hasil penelitianmenunjukkan bahwa mencit yang diberi perlakuan dengan senyawa kode Pc-2 dosis 10 mg/kg bb baik padahari ke-3, ke-10, maupun ke-21 setelah infeksi dengan L. monocytogenes, mempunyai PF, IF, dan EF yang lebihbesar secara bermakna dibandingkan pada mencit kelompok kontrol; aktivitas fagositosis makrofag terbesarterjadi pada hari ke-21 setelah infeksi dengan L.monocytogenes; dan tidak ada korelasi antara PF dengan IF.Kata kunci: fagositosis makrofag,isolat kode Pc-2, Piper crocatum Ruiz & Pav.

Abstract

Ribosome inactivating proteins from Mirabilis jalapa L. (RIP MJ) has shown higher cytotoxic activity when being formulated as a nanoparticle. However, the selectivity of the delivery system is also an important aspect when it comes to cytotoxic cell therapy. Epithelial cell adhesion molecule (EpCAM) is a monomeric glycoprotein which is overexpressed in epithelial cancer cells. This study aim was to develop a model of targeted protein delivery system by formulating the base fraction of RIP MJ (RIP MJ-30) into alginate nanoparticles and conjugating it with anti-EpCAM antibody. RIP MJ-30 was formulated in to nanoparticle using alginate and CaCl2 as cross-linker. Optimization of conjugation reaction condition was done in the pH variation of 4.5, 5.5, and 6.5. The success of conjugation was analyzed qualitatively using native polyacrylamide gel electrophoresis (native-PAGE) method and BCA assay. The optimum formula of RIP MJ-30 nanoparticles was produced using 0.3% alginate and 0.2% CaCl2. Results indicated that optimum conjugation reaction was carried out at pH level of 5.5. The optimum native-PAGE condition was by using 8% polyacrylamide gel in duration of 6h. Characterization of nanoparticle resulted in particle size of 205.0nm, zeta potential of -6.9mV, entrapment efficiency of 71.11±4.84%, and conjugation efficiency of 89.55±6.18%. It was concluded that RIP MJ-30 was successfully formulated into alginate nanoparticle and conjugated to anti-EpCAM antibody through carbodiimide reaction using 1-ethyl-(dimethylprophilamine) carbodiimide (EDAC).
