Abstract

Sindroma Down merupakan salah satu kejadian kelainan kromosom tipe aneuploidi yang kerap ditemukan.  Kejadian Sindroma Down terkait erat dengan disabilitas intelektual dan stugma sosial yang melekat padanya.  Pandangan orang tua mengenai Sindroma Down yang tercerminkan melalui perilaku amat penting karena dukungan keluarga merupakan salah satu kunci utama untuk mengoptimalkan kualitas hidup penyandang Sindroma Down. Studi ini merupakan studi deskriptif pada orang tua penyandang Sindroma Down yang bersekolah di Sekolah Luar Biasa (SLB) di Palembang.  Perilaku orang tua diukur dengan menggunakan kuesioner HealthStyles©.  Sebagian besar orang tua penyandang Sindroma Down sepakat bahwa anak dengan Sindroma Down harus bersekolah di SLB (82,5%) karena mayoritas meyakini pencampuran penyandang Sindroma Down di sekolah biasa dapat mengganggu murid lain (60%).  Namun orang tua memiliki pandangan optimis bahwa anak penyandang Sindroma Down dapat bekerja jika telah dewasa (72,5%).  Mereka pun percaya bahwa penyandang Sindroma Down tidak lebih rentan mengalami kecelakaan kerja di tempat kerja (37,5%).  Dari studi ini terlihat bahwa orang tua penyandang Sindroma Down memiliki perilaku yang bervariasi tergantung dari aspek mana mereka memandang Sindroma Down.

Abstract

Daun Gaharu (Aquilaria malaccensis) sejak lama telah digunakan masyarakat Kabupaten Bangka Tengah untuk menangkal keletihan akibat gangguan tidur ringan. Minuman yang diramu dari daun Gaharu yang diiris-iris lalu dijemur sebentar kemudian disedu air hangat kuku ternyata berkhasiat untuk menimbulkan rasa kantuk dan setelah bangun dari terlelap sejenak maka tubuh akan menjadi lebih segar. Penelitian ini ingin membuktikan manfaat tersebut di atas. Sejumlah 60 orang sukarelawan sehat terdiri dari 30 orang laki-laki dan 30 orang wanita dengan rentang umur 18 – 60 tahun berpartisipasi untuk membuktikan efek sedativa dan pengaruh kesehatan lainnya. Dengan disain pre-post test serta kerelaan sukarelawan sehat ini (dengan mengisi informed consent) sebagai responden, maka semuanya diberi minum teh celup Gaharu yang berisi 1 g simplisia kering/saset setiap hari selama 3 minggu berturut-turut. Setelah selesai satu sesi minum, semuanya diwawancarai pemeriksaan fisik, mental dengan menggunakan kuesioner model Pittsburgh Insomnia Rating Scale (PIRS) serta dikoleksi darah intravenanya. Hasil menunjukkan bahwa 32 % responden timbul rasa kantuk ringan, 10 % merasa lebih segar walaupun terasa sakit kepala ringan, serta berkurangnya rasa pegal dan nyeri sendi namun nafsu makan bertambah. Profil darah rutin tidak berubah namun kadar hormon seks seperti testosteron, progesteron, estradiol dan FSH meningkat ringan tanpa LH serta penurunan fibrinogen dan hs-CRP. Sedangkan transaminase hepar tidak berubah. Dari temuan ini disimpulkan bahwa konsumsi rutin teh gaharu memperbaiki profil tidur dan kesegaran jasmani namun tidak bersifat hepatotoksik. Perubahan kadar hormon seks, fibrinogen dan hs-CRP perlu dikaji lebih lanjut.

Abstract

Selain berdasarkan temuan klinis, penentuan diagnosis Disorders of sex development (DSD) juga didukung oleh ragam pemeriksaan penunjang seperti analisis kromosom, radiologi, dan pemeriksaan laboratorium yang salah satunya berupa pemeriksaan kadar hormon. Pemeriksaan kadar hormon dapat memberikan gambaran fungsi gonadal dan adrenal, yang bermanfaat baik untuk penapisan dan juga penentuan diagnosis DSD. Studi yang bertujuan untuk mengidentifikasi profil hormon pasien DSD di Indonesia masih sangat minim, dan studi ini merupakan studi pertama yang meninjau profil hormon DSD di RSUP Dr. Mohammad Hoesin Palembang. Studi ini merupakan studi deskriptif menggunakan data sekunder dari rekam medik pasien DSD yang menjalani pemeriksaan hormon selama periode 2013-2017.  Dari total 173 pasien DSD yang datang berobat ke RSUP Dr. Mohammad Hoesin Palembang selama lima tahun terakhir, hanya 22 (12.72%) pasien yang menjalani pemeriksaan hormon. Ada 13 profil hormon yang diperiksa pada pasien DSD di RSUP Dr. Mohammad Hoesin Palembang dengan hormon testosteron (54.54%) sebagai hormon yang paling sering diperiksa. Pemeriksaan profil hormon sebagai salah satu alat bantu penapisan dan penegakkan diagnosa DSD belum menjadi salah satu pemeriksaan penunjang yang umum dilakukan di RSUP Dr. Mohammad Hoesin Palembang.

Abstract

Introduction Candida is a yeast fungus classes most commonly found in the oral cavity, gastrointestinal tract, reproductive tract and skin, especially the species Candida albicans. Alternative medicines such as herbs are easily available and affordable by the community, such as keghenyat leaves (Acmella uliginosa). Aim of StudyTo determine the activity of Keghenyat leaves fraction (Acmella uliginosa) compared to Nystatin against Candida albicans in vitro. MethodsIn vitro study was held in Laboratory of Genetics and Biotechnology Department of Biology, Faculty of Science and Medical Faculty, Sriwijaya University in December 2015 and January 2016. Research stages: extraction, fractionation, sensitivity test using Nystatin and Candida albicans, fractions antifungal activity test, the determination of minimum inhibitory concentration (MIC), bioautografi test and determination of compound classes. One final stages of testing the active fraction Keghenyat leaves (Acmella uliginosa) with Nystatin. Data were analyzed using ANOVA and Post Hoc Duncan test and linear regression using SPSS 20. ResultsHexane faction of Keghenyat leaves (Acmella uliginosa) is active against the fungus Candida albicans, has a Minimum Inhibitory Concentration (MIC) of 0.625% (6250μl) against the fungus Candida albicans antifungal contain active compounds that terpenoids and flavonoids, amounting to 84.68 mg / ml equivalent to 1 mg / ml Nystatin against the fungus Candida albicans. ConclusionThere is a significance different MIC between hexane fraction of Keghenyat leaves (Acmella uliginosa) and Nystatin, Nystatin have more high activity.

Abstract

Abstract Background Aloe vera is a plant that has been used as an alternative drug. This plant contains various compounds, like anthraquinone, saponin, flavonoid, alkaloid, and tannin that has an antibacterial effect against Staphylococcus aureus and Escherichia coli. Both of it were responsible for the infection incident. This study aims to determine the efficacy of Aloe vera sap as an antibacterial against Staphylococcus aureus and Escherichia coli. Methods An experimental study, in vitro using post-test only control group design, has been done at laboratory of Medical Faculty of Sriwijaya University, by examining the antibacterial activity of Aloe vera sap in five different concentration (5%, 10%, 20%, 40%, dan 80%) using well diffusion and solid dillusion method to determine the minimum bactericidal concentration (MBC). And then continued with the phytocemical screening to determine the compound inside the Aloe vera sap. Results Aloe vera sap were able to kill Staphylococcus aureus at 5% and Eshcerichia coli at 80%. Compatibility test showed that Aloe vera sap with concentration of 10%, 20%, 40%, and 80% are compatible with amoxicillin, therefore 80% is compatible with cefotaxime. This ability due to the compound that it contains, which is alkaloid, flavonoid, tannin, quinone, and saponin. Conclusion Aloe vera sap is effective as an antibacterial against Staphylococcus aureus and Escherichia coli.   Keyword: Aloe vera sap, antibacterial, efficacy, Staphylococcus aureus, Escherichia coli.  

Telmisartan Prevents Myocardial Fibrosis via Decreasing Fraction of Colagen Type 1 Volume in Myocardial Tissue in Wistar Rats-Induced High Salt Intake

Abstract

Myocardial fibrosis is a pathological condition that responsible for initiation of heart failure. Neurohormonal endogen, angiotensin II, has a potential role to activate endothelin I, TGF-β1, myocardial fibroblast, extracelullar matrix deposition, structural changes and decreasing of cardiac function. Fibrotic process is also influenced by PPAR γ. Telmisartan has a potential effect to inactivate angiotensinergic system and to activate PPAR γ. It is expected that telmisartan has optimal effect to protect myocardial fibrosis. To know the role of variation dose of telmisartan to decrease collagen type 1 fraction volume in cardiac tissue of Wistar rats. Ten-week-old male Wistar Rat (n = 30) were randomized into five groups, and each group consisted of 6 rats. Group 1 : negative control. Group 2 : rats were induced by intake Nacl 8% doses 2% body weight for eight weeks. Group 3 : rats were induced by intake Nacl 8% doses 2% body weight and telmisartan 3 mg/kgBB for eight weeks. Group 4 : rats were induced by intake Nacl 8% doses 2% body weight and telmisartan 6 mg/kgBB for eight weeks. Group 5 : rats were induced by intake Nacl 8% doses 2% body weight and telmisartan 12 mg/kgBB for eight weeks. Collagen volume fraction was assessed by immunohistochemistry and ImageJ program. ANOVA test followed pos hoc test was used to analyzed each variable. Collagen volume fraction significantly decreased in group 3, 4 and 5 compared in group 2. Telmisartan decreases collagen type 1 volume fraction of myocardial tissue .

The Efficacy of Cinnamomum burmanii Extract on the Protection of Neuronal Cell Death in Haloperidol Induced Male Wistar Rats

Abstract

Background Haloperidol is categorized as the first class antipsychotic drug. Long-term use of haloperidol may convey to increased Reactive Oxygen Species (ROS) that will yield oxidative damage which further leads to cell death. Several studies had identified the effects of cinnamon extract on cell death. This study aimed to determine the efficacy of cinnamon extract (Cinnamomum burmanii) on the protection of neuronal cell death in haloperidol-induced male Wistar white rats. Methods This study was experimental with pre and post-test design. Thirty male Wistar rats were divided into 5 groups, induced with haloperidol and followed by treatment. Caspase-3 and dopamine were assayed by ELISA sandwich method using ELISA kit. Mean difference of caspase expression and dopamine levels before and after induction were shown (p<0.05). Results There were mean differences of caspase-3 expression level in the positive control group, cinnamon extract of 100 and 200mg/kgBW before and after treatment (p<0.05). Whereas for dopamine levels, there were mean differences in positive control group, cinnamon extract of 50, 100 and 200mg/kgBW before and after treatment (p<0.05). With Post Hoc test, it was found that there were no mean differences of caspase-3 expression level between positive group with cinnamon extract group of 100 and 200mg/kgBW (p>0,05) and there were also no mean differences of positive group dopamine level with group of cinnamon extract of 100 and 200mg/kgBW (p>0.05). Conclussion Cinnamomum burmanii extract at dose of 100 and 200mg/kgBW were effective in the protection against neuronal cell death in haloperidol induced male Wistar white rats.

Abstract

Background Phrynium maximum (local name : lerek ) is a plant native to Sumatera, Malaya and Borneo. This plants have a potential as a local wisdom for therapeutics. In Sumatera, Lerek was used as traditional treatment for fever, diarrhea, diabetics and for wound healing.   Aim of Study Aim of this study to explore the efficacy of lerek fruits extract in body weight, lipid profile and leptin level in Wistar Rats-Induced High Fat Diet   Methods This study was an experimental study , pre-post test with control group design. The sample in this study was  30 male rats, 8 weeks old, weight 150-200 gram. Rats were given high fat diet and lerek fruits extract  at dose of 50, 100 and 200 mg/kgBW/day for 2 weeks. Lerek fruits was extracted by infusion methods. The results of this study were assayed by SPSS 18.   Results Lerek fruit extract 100 mg/kgBW  was more potent to reduce body weight gain, triglyseride level and leptin level than lerek fruit extract 50 mg/kgBW, 200 mg.kgBW, negative control and positive control.   Conclusion Lerek fruit extracts had a potention to reduce body weight, triglyceride and leptin level.   Keywords: Lerek fruit extract – body weight- triglyserida - leptin

Abstract

Abstract Background Receptor for advanced glycation endproducts (RAGE) is a transmembrane protein that belongs to the immunoglobulin superfamily. As its name implicates, it can bind to advanced glycation endproducts, the resulting product of non-enzymatic glycosylation, and it also has the ability to interact with multiple ligands having common motifs as a so-called multi-ligand receptor. The ligands include high-mobility group protein (B)1 (HMGB1), S-100 calcium-binding protein, amyloid-β-protein, Mac-1, and phosphatidylserine. Interaction between RAGE and its ligands activates various cellular processes, including inflammation, proliferation, apoptosis, autophagy, and migration. Methods The proccess of isolation protein of RAGE was initiated with extraction protein and purification of RAGE protein. After that, the immunization of Rats was be done to produce Anti-RAGE. The confirmation of Anti RAGE was be done by SDS PAGE and Immunobloting. Results The production of Anti RAGE was enough pure compared by Anti RAGE commercial. Anti RAGE was protein that have molecular weight around 35 kD. Conclusion The methods that used in this study effective to develop production of anti-RAGE. The Efficacy of Temu Putih Fraction (Curcuma Zedoaria (Berg) Roscoe) Related Quality and Quantity of Spermatozoa in Male Wistar Rats

Abstract

Background Male participation in KB is still relatively low when compared to the participationof women. Researchers have to do research to find the contraception drug. Temu putih (Curcuma Zedoaria (Berg) Roscoe) is one of traditional herb that used as antifertility.   Aim of Study Aim of this study to examine change in the amount, motility, morphology, and viability spermatozoa male rats (rattus norvegicus) due to temu putih fraction suplementation.   Methods This study was an experimental study using a completely randomized design (CRD), post test with control group design. The sample in this study was  30 male rats, 10 weeks old, weight 150-200 gram. Rats were given temu putih fraction (n hexan, etylacetate and methanol-water) at dose of 300 mg/kgBB/day for 48 days. Temu putih was extracted by ethanol and did fractionation by liquid-liquid methods. The results of this study were assayed by SPSS 18.   Results The amount, motility, morphology and viability of spermatozoa in the group of metanol fraction of water  decreased  compared with the control group (p= 0,000), motilitas of spermatozoa in the group of metanol water fraction  decreased compared with the control group.   Conclusion Temu putih fraction can reduce the amount, motility, morphology, and viability of spermatozoa in male rats.   Keywords:  Fraction, Temu Putih, Amount of spermatozoa, Motility of spermatozoa, Viability Of Spermatozoa
