This study aimed to evaluate the effect of probiotic on growth and survival of tilapia (Oreochromis niloticus) against Streptococcus agalactiae infection. The test materials used were commercial probiotic bacteria and Streptococcus agalactiae. The probiotic bacteria were administered by mixing into the feed with a dose of 0 mL (without probiotics), 10 mL / kg of feed, 15 mL / kg of feed and 20 mL / kg of feed.  Feed was given 3 times a day at 08.00, 12.00 and 16.00 for 21 days. After that, the challenge test was done with Streptococcus  agalactiae bacteria by cohabitation. Bacteria with a density of 107 cells / mL were mixed into the water and allowed for 2 hours. The result showed that B treatment (10 mL / kg of feed) gave the best result with 93,33 percent of  survival rate. Statistical analysis showed that the survival of fish in treatments B, C and D was significantly different from treatment A (P <0.1). However, there was no significant difference between treatments B, C, and D. It was also found that the addition of probiotic in feed did not affect the growth of Nile tilapia (Orechromis niloticus). Keywords : Probiotic bacteria,  nile tilapia,  Streptococcus  agalactiae, growth,  infection

This research aimed to determine the optimal doses of honey addition on the sperm dilution to optimize spermatozoa motility, fertilization and hatchability of Patin Siam  eggs (Pangasius hypophthalmus).  The experimental design used was Complete Random Design (RAL).  Diluent solution consisted of a physiological NaCl 0.9% and honey.  Treatment doses of honey in aqueous diluent sperm tested as treatments were A (0 mL), B (0.2 mL), C (0.4 mL) and D (0.6 mL), each with three replications.  Observations made included spermatozoa motility, fertilization and eggs hatchability.  The research results showed that the addition of honey had a significant influence on motility,  fertilization, and eggs hatchability. This study concluded that the addition of honey in sperm diluent solution with a dose of 0.6 mL of honey in 99.4 mL of NaCl could increase the spermatozoa motility and  fertilization of Patin siam. This study aims to obtain isolates and amplify the associative bacterial 16SrRNA gene in K. alvarezii algae. The K. alvarezii algae was collected from seaweed cultivation area in Belang, Southeast Minahasa, North Sulawesi. Associative bacteria were sampled from K. alvarezii algae, grown in Nutrient Agar and separated based on their morphological characteristics.  Each isolates were extracted their DNA genome and.the16S rRNA gene of each isolate was amplified using PCR.  Eight associative bacterial from K. alvarezii algae were successfully isolated based on morphological characteristics which were dominated by round shape, smooth edges, convex elevation, and white color of the isolates. The results of genomic DNA extraction from each of these isolates were successfully used as templates to amplify the 16s rRNA gene.Key word: Bacteria, Kappaphycus alvarezii, Taxsonomy, 16S rRNAABSTRAKPenelitian ini bertujuan untuk mendapatkan isolat dan mengamplifikasi gen 16SrRNA bakteri asosiatif pada alga K. alvarezii. Metode penelitian diawali dengan pengambilan sampel alga K. alvarezii di lokasi pembudidayaan rumput laut di desa Belang, Minahasa Tenggara, Sulawesi Utara. Selanjutnya, bakteri asosiatif pada alga K. alvarezii tersebut ditumbuhkan dalam media Nutrient Agar, isolat yang tumbuh kemudian dipisahkan berdasarkan karakteristik morfologinya. dan gen 16S rRNA masing-masing isolat diamplifikasi menggunakn PCR.  Delapan isolat bakteri asosiatif pada alga K. alvarezii berhasil diisolasi dengan karakteristik morfologi berbeda yang didominasi dengan bentuk round, tepian smooth, elevasi convex, dan warna putih. Hasil ekstraksi DNA genom dari masing-masing isolat tersebut berhasil digunakan sebagai template untuk mengamplifikasi gen 16s rRNA.Kata Kunci: Bakteri, Kappaphycus alvarezii, Taksonomi, 16S rRNA
