Abstract

The research aimed to evaluate the effect of baker’s yeast as immunostimulant on the growth of carp.  Fish was obtained from Board of Freshwater Aquaculture at Tatelu, North Sulawesi Province.  Before running the experiment, the fish was acclimatized for one week in five concrete tanks measuring 2x1x1 m3 each. During acclimatization, fish was fed commercial pellet at 5%/body weight/day, twice a day.  After acclimatizationm, the fish was distributed in 15 aquaria with a density of 25 individuals per aquarium.  The research used Complete Randomized Design with five treatments namely 0, 5, 10, 15, and 20 g yeast/kg of pellet, each with three replications.  Fish was fed with treatment pellets for three weeks consecutively at a dose of 5%/body weight/day, twice a day at 08.00 am and 16.00 pm. Growth of fish was measured at the end of feeding period.  The result showed that the addition of baker’s yeast cells into fish feed significantly influenced fish growth. The best growth was achieved in fish fed pellet supplemented with 5 g yeast/kg of pellet.Keywords : Cyprinus carpio, Saccharomyces cerevisiae, immunostimulant, growth

Abstract

This study aimed to evaluate the effect of probiotic on growth and survival of tilapia (Oreochromis niloticus) against Streptococcus agalactiae infection. The test materials used were commercial probiotic bacteria and Streptococcus agalactiae. The probiotic bacteria were administered by mixing into the feed with a dose of 0 mL (without probiotics), 10 mL / kg of feed, 15 mL / kg of feed and 20 mL / kg of feed.  Feed was given 3 times a day at 08.00, 12.00 and 16.00 for 21 days. After that, the challenge test was done with Streptococcus  agalactiae bacteria by cohabitation. Bacteria with a density of 107 cells / mL were mixed into the water and allowed for 2 hours. The result showed that B treatment (10 mL / kg of feed) gave the best result with 93,33 percent of  survival rate. Statistical analysis showed that the survival of fish in treatments B, C and D was significantly different from treatment A (P <0.1). However, there was no significant difference between treatments B, C, and D. It was also found that the addition of probiotic in feed did not affect the growth of Nile tilapia (Orechromis niloticus). Keywords : Probiotic bacteria,  nile tilapia,  Streptococcus  agalactiae, growth,  infection

Abstract

The objective of this study was to determine the minimum concentration of extract in inhibiting motile motile aeromonad septicaemia. The material used was the stems and leaves of the plant P. grandiflora as much as 10 g. The crushed simplicia was then macerated with aquades with a ratio of 1:10,  then filtered with filter paper and, put in an Erlenmeyer, and evaporated in an incubator at temperature of 30°C. The sensitivity test was performed by placing a paper disc containing extract on the agar medium that had been scratched with bacterial colonies. Clear zone formed around the paper disc was compared with standard classification table for clarifying the inhibition level.  The results showed that stem and leaf extract had inhibition zone against Aeromonas hydrophila bacteria that caused motile aeromonad septicaemia. The best result was obtain at concentration of extract 3.25 mg / L) with an average diameter of 22.67 mm and for leaf extract had a moderate to strong zone for all treatments. These proved that stem and leaf extract of P. grandiflora plant had antibacterial activity and can be used as control of MAS disease caused by A. hydrophila bacteria. Abstract

The aim of this research was to study the effect of Binahong extract on the growth of Nile tilapia. A number of 225 fish measuring 10-12 cm in length with an average weight of 15 g was obtained from Fishery Development Center and Training, Tateli. The fish were put  in oxygenated plastic bags and transported to Laboratory of Aquaculture Technology. The fish was kept in aquarium equipped with an aerator with a density of 15 fish per aquaria. The test material used was the stem of Binahong plant stem, macerated with alcohol 70% for 3 days. The extract was then filtered with whatman filter paper and extract was concentrated using a Rotary Evaporator. The extract was further diluted to obtain  concentrations of 12.5, 25, 50, and 100 mg / mL Binahong  extract as treatment was given to the fish after mixed first into the pellet. The fish was fed with treatment feed for 30 days. Control feed was not supplemented with binahong extract. Growth data was measured at day 30 after treatment. The results showed that the extract of binahong had significant effect on fish growth (p <0.01) with the best result achieved at concentration of 100 mg/mL. In conclusion, supplementation of  binahong extract into fish feed could improve growth of  fish in which the best growth was obtained in fish fed diet containing 100 mg/mL of binahong extract.Keywords: Medicinal plants, tilapia, Anredera cordifolia, growth, maceration

Abstract

This study aims to analyze the carrageenan quality of seaweed Kappaphycus alvarezii. The test covered rendement, gel strength, viscosity, and ash content. The rendement was 63%. Gel strength was 372.8 g and met the FAO standard, 20 – 500 g/cm2. Viscosity and ash content, 20 cps and 22.68%, respectively, also met the FAO standard, 15-40%. As conclusion, carrageenan of Kappaphycus alvarezii in NSMEC, east Likupang, had very good quality.Key words : NSMEC, Kappaphycus alvarezii, rendement, gel strength, viscosity. ABSTRAKPenelitian ini bertujuan menganalisis kualitas karageenan rumput laut Kappaphycus alverezii.  Pengujian yang dilakukan ialah Uji Rendemen, Uji Kekuatan Gel, Uji Viscositas, dan Kadar abu. Hasil penelitian diperoleh nilai rendemen sebesar 63%.  Nilai ini tergolong sangat tinggi dibanding dengan beberapa hasil penelitian lainnya. Kekuatan gel pada penelitian ini sebesar 372,8 g.  Nilai tersebut telah memenuhi standard FAO (2017) 20 – 500 g/cm2. Nilai viskositas dan kadar abu, masing-masing 20 cps dan 22.68%, memenuhui standar FAO sebesar 15-40%. Kesimpulan bahwa karaginan rumput laut Kappaphycus alvarezii di NSMEC Likupang Timur berkualitas sangat baik.Kata kunci : NSMEC, Kappaphycus alvarezii, rendemen, kekuatan gel, viskositas.

Abstract

This study aims to examine the effect of the use of crude marine sponge extract on the resistance of streptococcus agalactiae infection in tilapia, Oreochromis niloticus, and to establish the effective dose of crude sponge extract in improving the immune system and the growth of the fish. The sponge used in the study was Cribrochalina sp. taken from Malalayang waters, Manado. The fish were taken from Freshwater Aquaculture Center, Tatelu. The fish were acclimatized for a week. After being acclimatized the fish were given feed added with sponge crude extract as a treatment with different concentrations of 20 g, 40 g and 60 g / Kg of feed for 14 days as much as 5% / body weight / day with the frequency of feeding twice a day at 10:00 am and at 5:00 p.m. After being treated, the fish was challenged with S. agalactiae. The data collected consisted of tilapia resistance, Total Leukocyte Count (TLC) as immune parameters and absolute growth. The results showed that the addition of crude extracts of Cribrochalina sp. into feed can increase TLC and growth of tilapia (p <0.05). The best results were achieved in fish fed with the addition of sponge crude extract of 40 g/kg feed. The survival rate of tilapia fed with treatment diet then challenged with pathogenic bacteria S. agalactiae showed the best results (100% survival rate) compared to controls (75%). Spons yang digunakan dalam penelitian adalah spons Cribrochalina sp. yang diambil dari perairan Malalayang. Ikan uji diambil dari Balai Budidaya Air Tawar Tatelu, Provinsi Sulawesi Utara. Ikan diaklimatisasi selama seminggu. Setelah diaklimatisasi ikan diberi pakan yang ditambahkan dengan ekstrak kasar spons sebagai perlakuan dengan konsentrasi berbeda yaitu 0 g, 20 g, 40 g dan 60 g/kg pakan selama 14 hari sebanyak 5%/berat tubuh/hari dengan frekuensi pemberian pakan dua kali sehari yaitu jam 10.00 pagi dan jam 17.00 sore. Setelah diberi perlakuan, ikan diuji tantang dengan bakteri S. agalactiae. Data yang dikumpulkan terdiri dari kelangsunganhidup ikan nila. Total leukosit sebagai parameter imun dan pertumbuhan mutlak. Hasil penelitian mendapatkan bahwa penambahan ekstrak kasar spons Cribrochalina sp. ke dalam pakan mampu meningkatkan total leukosit dan pertumbuhan ikan nila (p<0.05). Dimana hasil terbaik dicapai pada ikan yang diberi pakan dengan penambahan ekstrak kasar spons sebanyak 40 g/kg pakan. Kelangsungan hidup ikan nila yang diberi pakan perlakuan yang diuji tantang dengan bakteri patogen menunjukkan hasil yang paling baik (tingkat kelangsungan hidup 100%) dibandingkan dengan kontrol (75%). Sebagai kesimpulan bahwa pemberian pakan dengan ekstrak kasar spons Cribrochalina sp. berpotensi untuk meningkatkan sistem kekebalan tubuh dan pertumbuhan pada ikan nila.Kata Kunci: Ekstrak kasar, Spons laut, Cribrochalina sp., Tilapia, Resistensi, Streptococcus agalactiae

Bahan uji berupa Daun Ketapang  (Terminalia catappa) dengan dosis A= 0, B= 5 g/kg, C= 10 g/kg, D= 20 g/kg dan E= 40 g/kg  diberikan kepada ikan selama 21 hari. Ikan diberi pakan perlakuan dua kali sehari yakni pukul 09.00 am dan 16.00 pm dengan dosis pemberian 5%/berat tubuh/hari.
