Abstract

Tujuan penelitian ini adalah untuk mengetahui  suplementasi ampas kecap terhadap intensitas libido dan produksi sperma kambing peranakan etawa. Penelitian dirancang dalam pre-test dan post-test design, dimana pengambilan data dilakukan sebelum dan setelah perlakuan. Tiga ekor kambing diberi perlakuan masing-masing 10 hari pada  pre-test dengan pakan rumput lapangan tanpa suplementasi, post-test-1 dengan pakan rumput lapangan disuplementasi 5% dan post-test-2 dengan rumput lapangan disuplementasi 10% ampas kecap. Intensitas libido dan produsi sperma secara makroskopis dan mikroskopis diobservasi setiap tiga hari sebanyak 3 kali. Data yang diperoleh dianalisis secara statistik. Hasil penelitian menunjukkan, nilai intensitas libido meningkat sangat signifikan dari 3.11 ± 0.78 tanpa suplementasi menjadi 4.67 ± 0.50 setelah suplementasi 10% ampas kecap. Rataan volume sperma meningkat dari 0.71 ± 0.13ml menjadi 0.87 ± 0.17 ml, nilai pH rata-rata mendekati 7.0, konsistensi sperma menjadi kental dan motilitas massa spermatozoa dari ++ menjadi +++. Motilitas progresif dan viabilitas spermatozoa meningkat signifikan dari 72.78 ± 2.64% menjadi 82.22 ± 2.64% dan dari 79.98 ± 1.25% menjadi 81.68 ± 1.36%. Persentase abnormalitas spermatozoa terendah terdapat pada suplementasi 10% dan konsentrasi tertinggi pada suplementasi 5% ampas kecap. Kesimpulan, suplementasi 10% ampas kecap di dalam pakan rumput lapangan mampu meningkatkan intensitas libido dan produksi sperma kambing peranakan etawa.Kata kunci : Suplementasi, viabilitas, spermatozoa This study was conducted to observed the supplementation of soy sauce dregs in wild-grass ration on libido intensity and semen production of peranakan etawa (PE) buck. The experiment was done in pre-test and post-test design. Three bucks were treated each 10 days at pre-test by given of wild-grass without supplementation, post-test-1 with wild-grass supplemented by 5% and post-test-2 with wild-grass supplemented by 10% of soy sauce dregs. The libido intensity and semen production were observed every three days for three times. The data was analyzed statistically. The results showed, the libido intensity score was increased very significantly from 3.11 ± 0.78 to 4.67 ± 0.50 after supplementation of 10% soy sauce dregs. The average of semen volume, consistency and  mass motility of spermatozoa were increased from 0.71 ± 0.13ml to 0.87 ± 0.17ml, rather thick to thick and ++ to +++.  Progressive motility and viability of spermatozoa were increased significantly after supplementation of 10% soy sauce dregs. The lowest percentage of spermatozoa abnormality was found on supplementation of 10% and the highest concentration found on supplementation of 5% soy sauce dregs. Abstract

This study aimed to determine the role of guava fruit filtrate (GFF) on maintaining the pre-freezing and post-thawed quality of Bali bullspermatozoa in tris-egg yolk (TEY) extender. The bovine semen was collected using artificial vagina twice a week (n= 10). Ejaculate sampleswere divided into four tubes and each tube was respectively added the following diluents: TEY, TEY + 5% GFF, TEY + 10% GFF, and TEY +15% GFF. The diluted semen was filled into the straws (0.25 mL) and cooled in refrigerator at 4° C for 2.5 hours. The samples were kept onvapor phase of liquid nitrogen (N2) (-120° C) for 10 minutes prior to be stored in liquid phase (-196° C). The quality of pre-freezing and postthawedspermatozoa in straw sample (i.e. motility, viability, and abnormality) was evaluated under light microscopy at 400x magnification. Datawere analysed statistically. The percentage of motility and viability both on pre-freezing and post-thawed process were significantly higher insemen diluted with TEY + 10% GFF compared to control (TEY), TEY + 5% GFF, and TEY + 15% GFF. Addition of 10% GFF into tris-egg yolkextender play role for maintaining the quality of pre-freezing and post-thawed Bali bull spermatozoa.
