Abstract

Tujuan: Mengukur ketebalan serat elastin pada dinding vagina penderita POP anterior dan menganalisis korelasi antara ketebalan serat elastin dengan derajat POP anterior.Bahan dan Metode: Penelitian ini merupakan penelitian analitik observasional secara cross sectional yang dilakukan pada 28 blok paraffin dinding vagina dari penderita POP anterior yang telah dilakukan operasi di RSUD dr. Soetomo Surabaya. Dibagi menjadi 4 kelompok: derajat 0/I, II, III dan IV dengan masing-masing (n=7). Dilakukan pewarnaan imunohistokimia dengan antibodi elastin dilanjutkan pengukuran ketebalan serat elastin menggunakan mikroskop dengan perangkat lunak Leica Application Suite (LAS).Hasil: Didapatkan perbedaan bermakna rerata ketebalan serat elastin kelompok penderita POP anterior derajat 0/I, II, III dan IV yaitu masing-masing sebesar 0,81 + 0,14 µm; 1,63 + 0,19 µm; 2,47 + 0,26µm dan 3,19 + 0,36 µm (p<0,0001). Berdasarkan hasil uji statistik korelasi pearson didapatkan koefisien korelasi r = 0,965 (P<0,0001).Simpulan: Ketebalan serat elastin pada dinding vagina bertambah dengan meningkatnya derajat POP anterior. Peningkatan ketebalan serat elastin merupakan akibat proses remodeling matriks ekstraseluler pada dinding vagina penderita POP anterior.

Abstract

Objective: This study aims to prove the correlation between Prostate Specific Antigen (PSA) blood level and Matrix Metalloproteinase-2 (MMP-2) expression in patients with prostate adenocarcinoma. Material &amp; method: Prostate cancer patients’ data from January 2009 to May 2012 were collected at the Department of Pathology, Soetomo General Hospital Surabaya. Data collected included patient medical documents, PSA blood examination, and histopathological examination. Histopathology slides and paraffin blocks of needle biopsies, Transurethral Resection of Prostate (TURP) and radical prostatectomy of prostate cancer patients werere-read, then the samples that met the inclusion criteria were stained by immunohistochemistry using antibodies MMP-2. Results: Data collection was done to obtain data samples of prostate cancer patients in 2009 to 2012 comprising as many as 22 patients between the ages of 52-91 years. Prostate adenocarcinoma in age of 70-79 was found in 8 patients, with a mean age of 68 years. PSA values obtained from medical documents were between 8.6-594.41 ng/ml. Spearmans test performed in this study showed a positive correlation (one-tailed) (correlation coefficient (r) 0431, p &lt; 0.05) between blood PSA level and MMP-2 expression in patients with prostate adenocarcinoma. Conclusion: Blood PSA level correlates positively with MMP-2 expression in prostate adenocarcinoma.

Abstract

Objectives: To identify the variants of genotype HPV that most often found in ICC at Dr Soetomo Hospital Surabaya.Materials and Methods: This was an explorative study with cross sectional approach. Specimens used were 30 formalin fix parrafin embedded from squamous cell carcinoma (SCC), adenocarcinoma (AD) dan adenosquamous carcinoma (AS) cervival cancer parients at Dr Soetomo Hospital Surabaya around Januari-Desember 2013, then used for DNA virus extraction and continued for PCR and HPV genotyping.Result : The result of HPV genotyping showed 10 positives from SCC, 6 positives and 4 negatives from AD, and 9 positives and 1 negative from AS. HPV infections in SCC were single infection by HPV 16, 18, 45, 56, and multiple infection by HPV 16+45, in AD were single infection by HPV 16,18, anad multiple infection by HPVhr+hr 68b+72 and HPVlr+hr  6+18+45, and in AS were single infection by HPVhr 16, 18, 45 and mutiple infection by HPV lr+hr 6+59.Conclusion : Infections HPV in ICC were single infection by HPV16, 18, 45, 56 and mutiple infection by HPV16+45, 68b+72, 6+18+45, 6+59. HPV 18 has highest prevalence in ICC patient from Dr Soetomo Hospital, followed by HPV16, 6, 45, 56, 59.

Abstract

Colorectal carcinoma has high morbidity and mortality rate worldwide, in which over 90% are adenocarcinoma. Colorectal carcinoma is one of the most cancer that metastasize to the lymph node. Angiogenesis have an important role in tumor growth and metastasis. Hypoxia is the trigger factor for angiogenesis. Hypoxia induced factor-1 (HIF-1) is one critical protein directly reacting to hypoxia. HIF-1alpha, a HIF-1 subunit, is an important regulator of angiogenesis. This study analyzed HIF-1a expression in colorectal adenocarcinoma with and without regional lymph node metastasis. This study was to prove that HIF-1a has a role in regional lymph node metastasis in colorectal adenocarcinoma. An analytical observational study was conducted on thirty formalin fixed paraffin-embedded colorectal adenocarcinoma tissues from Anatomic Pathology Laboratory of Dr. Soetomo Hospital. Detection of HIF-1alpha expressions were performed with immunohistochemistry method, using HIF-1alpha antibody. It were statistically analyzed using Mann-Whitney and Kruskal- Wallis methods. There were no significant differences in the expression of HIF-1alpha in colorectal adenocarcinoma with or without lymph node (LN) metastasis (p>0.05). As a conclusion, this study showed that HIF-1alpha has no role in LN metastasis in colorectal adenocarcinoma.

Abstract

Tujuan: mencari peningkatan ekspresi cleaved caspase-3 pada pasien kanker serviks IIB tipe sel skuamosa sesudah pemberian kemoterapi neoadjuvan cisplatin dan mencari kapan waktu yang tepat untuk mendeteksi apoptosis menggunakan cleaved caspase-3. Bahan dan Metode: Jenis penelitian analitik observasional berpasangan. Dilakukan pada penderita kanker serviks IIB tipe sel skuamosa berdasarkan kriteria FIGO yang berobat di POSA RSUD Dr. Soetomo Surabaya. Penelitian dilakukan mulai bulan Mei 2014. Sebelum dilakukan terapi semua penderita diambil biopsi serviks. Kemudian penderita diberikan cisplatin 50mg/m2/minggu Diambil biopsi serviks setelah diberikan kemoterapi ke 1 dan ke 4 (maksimal 8 jam setelah kemoterapi, sampel kedua). Sampel diperiksakan imunohistokimia cleaved caspase 3. Dilakukan penghitungan ekspresi cleaved caspase 3 di bawah mikroskop dengan perbesaran 400x pada 10 lapang pandang. Pengujian statistik dilakukan dengan nilai kemaknaan p<0,05.Hasil: Rata-rata ekspresi cleaved caspase 3 sebelum kemoterapi adalah 1,46± 1,854. Setelah kemoterapi ke 1 didapatkan rata- rata 10,77± 3,655.Setelah kemoterapi ke 4 didapatkan rata- rata 12,77± 5,703. Pada pemeriksaan didapatkan ekspresi cleaved caspase 3 setelah pemberian kemoterapi lebih meningkat dibandingkan sebelum pemberian kemoterapi (p<0,01), sedangkan ekspresi cleaved caspase 3 setelah kemoterapi ke 1 dan ke 4 tidak didapat-kan perbedaan bermakna (p=0,882).Simpulan: Pemberian kemoterapi terbukti memberikan efek peningkatan ekspresi cleaved caspase-3. Sehingga cleaved caaspase-3 dapat dijadikan sebagai salah satu uji apoptosis pada efek kemoterapi terhadap sel kanker.

Abstract

Background: Human Papillomavirus (HPV) infection can be associated with a variety of cutaneous as well as mucosal manifestations. Some types of HPV are associated with increased risk of epithelial malignancies; these have been divided into low-risk and high-risk types based on their oncogenic potential. Bowenoid papulosis (BP) strongly associated with high risk type. Purpose: To provide information to health workers, in order to be more concern about BP.Review: The term BP refers to multifocal papular lesions on the genitalia with histological features similar to squamous cell carcinoma (SCC) in situ or Bowen’s Disease (BD). Its clinical manifestation is characterized by multiple brownish or erythematous papules located in the anogenital region, affecting mostly young adults with an active sex life. Clinically, it must be differentiated from seborrheic keratosis, and melanocytic nevus. Bowenoid papulosis is strongly associated with HPV 16.Conclusion: The natural course of BP is unpredictable, the lesions may increase, decrease, and even disappear spontaneously. Progression to an invasive SCC has been estimated in 2.6% of cases, HPV type 16 is the most frequent causative agent of BP.

Abstract

HPV infection is known to cause cervical cancer. This study aimed to identify the variant of HPV genotypes of cervical precancerous lesions from low grade squamous intraepithelial lesion  (LSIL) and high grade squamous intraepithelial lesion (HSIL). This was an explorative study using formalin fix paraffin embedded (FFPE) from cervical precancerous lesions at Dr. Soetomo Hospital, Surabaya. DNA was extracted from FFPE and hybridized for HPV genotyping using Ampliquality HPV Type Express kit (AB ANALITICA) by reverse line blot techniques. The results showed that there were variants of HPV genotype in LSIL. The variants were HPV16 (8/15), HPV18 (3/15), HPV52 (1/15), HPV6+31 (1/15), HPV6+18 (1/15), and HPV72+68 (1/15), and in HSIL which were HPV16 (4/10), HPV18 (2/10), HPV59 (1/10), HPV6+45 (1/10), HPV61+26 (1/10), and HPV16+31 (1/10). The characteristics of infection in LSIL were single infection of high-risk (hr) HPV and multiple infection of  low-risk (lr)+hr HPV, and in HSIL were single infection of HPVhr, multiple infection of HPVhr+hr and HPVlr+hr. In conclusion, HPV prevalence in cervical precancerous lesions is single infection by HPV16 (48%), HPV18 (20%), HPV52 (4%), HPV59 (4%), and multiple infection by HPV6+31, HPV6+18, HPV6+45, HPV16+31, HPV61+26, HPV72+68 is 4%.

Abstract

Objectives: To measure the thickness of elastin fibres on vaginal wall of patients with POP anterior and then analyse the correlation of this measurement with the degree of POP anterior.Materials and Methods: This research is an observational analytic study. Data collection was conducted with a cross sectional approach on 28 blocks paraffin of vaginal wall taken from POP anterior patients of whom went through an operation in Dr. Soetomo Hospital of Surabaya. Data was divided into four groups: degree 0/I , II, III and IV respectively (n = 7). Immunohistochemical staining was performed with antibodies elastin. Afterward, the thickness of the elastin fibres was measured by utilizing a microscope with a Leica Application Suite (LAS) software.Results: We found significant differences in the mean of thickness of elastin fibres in each group of patients with degrees of anterior POP 0/I , II , III and IV respectively 0,81 + 0,14 µm; 1,63 + 0,19 µm; 2,47 + 0,26µm and 3,19 + 0,36 µm (p < 0,0001 ). Based on Pearson correlation test, the results shows a correlation coefficient r = 0.965 (p < 0,0001).Conclusion: The thickness of elastin fibre on vaginal wall increased as the degree of POP anterior arise. Increased in the thickness of elastin fibers on the vaginal walls was the result from extracellular matriks remodelling on vaginal wall of anterior POP patient.

Abstract

Background: Breast carcinoma, the most common malignancy in women, are often accompanied by tumor infiltrating lymphocytes (TIL) which has controversial clinical relevance. TIL is thought to reflect the host’s immune response to malignant tumors. FOXP3, specific biomarker of Treg, is an important transcription factors that develops and functions in the maintenance of self tolerance, including inhibition of CD8+ cytotoxic T cell function. Aim: To analyze the differences and the correlation between FOXP3+ and CD8+ TIL in breast carcinoma with different T staging. Methods: An analytical observational research, performed on 44 paraffin block of breast carcinoma of various stages T (AJCC 7th ed) in anatomical pathology installation of RSUD Dr. Soetomo, used FOXP3+ and CD8+ antibodies. The immunoexpression are evaluated on stromal area, then analyzed statistically, period January 1, 2014 – December 31, 2016. Result: Showed significant differences in FOXP3+ expression between T1-T4, T2-T3, T2-T4, T3-T4. There were significant differences in CD8+ expression between T2-T3, T2-T4. There is a correlation between the expression of FOXP3+ and CD8+ in T1 and all T (p < 0.05). Conclusion: There was significant difference in FOXP3+ and CD8+ TIL of breast carcinoma with increasing T stage. There was correlation between FOXP3+ and CD8+ TIL expression of breast carcinoma at all T and T1 stage. ABSTRAKPendahuluan: Karsinoma payudara adalah keganasan terbanyak wanita dan sering didapatkan adanya tumor infiltrating lymphocytes (TIL) dengan relevansi klinis yang masih kontroversial. TIL sering dianggap mencerminkan respon imun inang terhadap tumor ganas. FOXP3, biomarker spesifik Treg, merupakan faktor transkripsi yang penting dalam perkembangan dan berfungsi dalam pemeliharaan self tolerance, termasuk penghambatan fungsi sel T sitotoksik CD8+. Metode: Penelitian observasional analitik terhadap 44 sampel blok parafin karsinoma payudara berbagai stadium T (AJCC edisi ketujuh) di instalasi Patologi Anatomi RSUD Dr. Soetomo Surabaya menggunakan antibodi FOXP3 dan CD8, dihitung pada area stroma tumor, kemudian dilakukan uji statistik periode 1 Januari 2014 – 31 Desember 2016. Hasil: Terdapat perbedaan bermakna ekspresi FOXP3+ antara T1 dan T4, T2 dan T3, T2 dan T4, T3 dan T4. Terdapat perbedaan bermakna ekspresi CD8+ antara T2 dan T3, T2 dan T4. Terdapat korelasi antara tingginya ekspresi FOXP3+ dengan tinginya ekspresi CD8+ pada T1 dan semua T (p < 0,05). Kesimpulan: Terdapat perbedaan bermakna ekspresi FOXP3+ dan CD8+ TIL karsinoma payudara dengan meningkatnya stadium T. Terdapat korelasi antara ekspresi FOXP3+ dan CD8+ TIL karsinoma payudara pada semua stadium T dan T1.

Abstract

Osteosarcoma is the most common malignant bone tumor in children and young adults. The process of metastasis and recurrence involves several proteins, including ezrin, and CD44 that are shown to be involved in tumor growth,metastasis and recurrence. To analyse the corelation of ezrin and CD44 expression with chemotherapy responsse in osteosarcoma patient. Cross sectional method on paraffin block of Osteosarcoma in Anatomic Pathology Laboratory of RSUD dr Soetomo, (January 1, 2010 - December 31, 2015). There were 17 out of 26 cases of amputated osteosarcoma which are met the inclusion criteria were performed immunohistochemical staining with ezrin and CD44 antibodies. The corellation of ezrin and CD44 expression with chemotherapy responsse was analyzed using Spearman’s rho test. The coeficient correlation in this experiment p<0.05, there was no corellation of ezrin expression with chemotherapy responsse of osteosarcoma. There was no correlation of CD44 expression with chemotherapy responsse in osteosarcoma. There was no correlation of ezrin and CD44 expression with chemotherapy responsse of osteosarcoma. There was no corellation of ezrin and CD44 expression with chemotherapy responsse of osteosarcoma.ABSTRAKOsteosarkoma merupakan tumor ganas tulang, sering terjadi pada anak-anak dan dewasa muda. Proses metastasis dan rekurensi melibatkan beberapa protein, di antaranya ezrin dan CD44 yang terbukti ikut serta dalam pertumbuhan tumor, metastasis, dan rekurensi. Penelitian ini bertujuan menganalisis hubungan antara ekspresi ezrin dan CD44 dengan respons kemoterapi. Penelitian ini menggunakan metode cross sectional blok parafin osteosarkoma di Laboratorium Patologi Anatomi RSUD Dr. Soetomo (1 Januari 2010–31 Desember 2015). Sebanyak 17 kasus sesuai kriteria inklusi dari 26 kasus osteosarkoma yang telah diamputasi dilakukan seleksi serta pemeriksaan imunohistokimia dengan antibodi ezrin danCD44. Hubungan ekspresi antara ezrin dan CD44 dengan respons kemoterapi dianalisis menggunakan uji Spearman’srho. Pada penelitian ini, nilai koefisien korelasi p<0,05 sehingga tidak terdapat hubungan antara ekspresi ezrin denganrespons kemoterapi pada osteosarkoma, di mana nilai p=0,868 (p>0,05). Tidak terdapat hubungan antara ekspresi CD44 dengan respons kemoterapi pada osteosarkoma di mana nilai p = 0,740 (p> 0,05).Tidak terdapat korelasi antaraekspresi ezrin dengan CD44 dengan respons kemoterapi osteosarkoma, nilai p=0,113 (p>0,05). Tidak terdapat hubungan antara ekspresi ezrin dan CD44 dengan respons kemoterapi pada pasien osteosarkoma.
