Abstract

Background: stichopus hermanii (golden sea cucumber) is one of the many types of marine organisms containing glycosaminoglycans (GAGs), a polysaccharide that promote wound healing. The content of this GAGs, mainly dermatan sulfate, chondroitin sulfate and heparan sulfate has the ability to modulate FGF2. FGF2 many found in the oral mucosa to activate fibroblast proliferation. Purpose: The study was aimed to determine the modulation of FGF2after topical application of stichopus hemanii gel on traumatic ulcers in wistar rat. Methods: The sample was 36 male Wistar rats which were divided into 6 groups. C1 and C2 group was placebo gel, sC1 and sC2 group was stichopus hermanii gel, HA1 and HA2 was hyaluronic acid A gel. The gel was given shortly after the traumatic ulcer (TU) formed and 24 hour later. Then all rats were sacrificed lips mucosa were taken and ELIsA examination was done. Results: All data were analyzed by ANOVA test followed by Tukey HsD. Test results show significant differences between sC1-sC2 with C1- C2 group, while the HA1-HA2 with C1-C2 group showed no significant difference. Conclusion: The study showed that modulation of FGF2 increased after topical application of stichopus hermanii gel on traumatic ulcers in wistar rats.Latar belakang: stichopus hermanii (teripang emas) merupakan salah satu jenis biota laut yang banyak mengandung GAG, suatu polisakarida yang sangat bermanfaat dalam proses penyembuhan luka. Kandungan GAG ini, terutama dermatan sulfat, chondroitin sulfat dan heparan sulfat memiliki kemampuan untuk memodulasi FGF2. FGF2 banyak didapatkan pada mukosa rongga mulut untuk mengaktifkan proliferasi fibroblas. Tujuan: studi ini bertujuan meneliti modulasi FGF2setelah pemberian aplikasi topikal gel stichopus hermanii pada ulkus traumatikus pada tikus wistar. Metode: sampel penelitian ini adalah 36 ekor tikus wistar jantan yang dibagi dalam 6 kelompok. Kelompok C1 dan C2 merupakan kelompok kontrol negatif (gel plasebo), Kelompok sC1 dan sC2 merupakan kelompok pemberian gel stichopus hermanii (sC), serta kelompok asam hyaluronat HA1 dan HA2 merupakan kelompok pemberian gel asam hialuronat (AH). Pemberian gel dilakukan sesaat setelah pembuatan ulkus traumatikus dan 24 jam kemudian. Kemudian semua tikus dikorbankan untuk diambil mukosa bibirnya dan dilakukan pemeriksaan ELIsA. Hasil: semua data dilakukan analisa dengan uji Anova dan dilanjutkan dengan uji Tukey HsD yang hasilnya menunjukkan adanya perbedaan bermakna antara kelompok sC1-sC2 dengan kelompok C1-C2, sedangkan kelompok HA1-HA2 dengan kelompok C1-C2 tidak menunjukkan perbedaan yang bermakna. Simpulan: Modulasi FGF2 meningkat setelah pemberian aplikasi topikal gel stichopus hermanii pada ulkus traumatikus tikus wistar.

Abstract

Background: Bone morphogenetic protein-2 (BMP-2) is bone stimulator which capable of inducing differentiation of mesenchymal cells into osteoblast, stimulating bone formation in wound healing process of dental extraction. Chitosan is polymer composed N-acetylD-glucosamine unit that has been used in various applications in wound healing process and bone tissue engineering. Purpose: The objective of this research was to analyzed expressions of BMP-2 for 7,14 and 21 days after using chitosan gel with different molecular weight on wound healing process of dental extraction. Method: The research was an experimental laboratory study. Rattus nornegicus strain wistar male, aged 8-16 weeks, divided into 3 treatment groups namely group 1 and II which given chitosan gel 1 % with high and low molecular weight and group III as control which were not given chitosan gel. Chitosan gel were applied into the socket of dental extraction. Rat was decaputated 7,14 and 21 days after chitosan gel application and the jaw in the treated regions and control group were cut for immunohistochemical examination to observe BMP-2. Data were analyzed using ANOVA test. Result: The result of this research showed significant differences on BMP-2 for 7,14 and 21 days observation (p<0,05). The increasing of BMP-2 were found in the group which given chitosan gel with high molecular weight. Conclusion: It may be concluded that chitosan gel with high molecular weight can enhance the expresion of BMP-2 on wound healing process ofdental extraction.

Abstract

Intraoral periapical radiograph examination is the additional examination which is the most widely used in Dentistry. This radiograph examination using an x-ray ionizing radiation with low LET (Linear Energy Transfer), and may affect submandibular salivary gland. Ionizing radiation exposure can cause damage by inducing a series of changes at the molecular and cellular level. This study aimed to prove the effects of x-ray ionizing radiation with low LET towards the catalase activity of Rattus norvegicus strain Wistar’s submandibular gland. The subjects were 28 male Wistar rats and divided into 4 groups (n=7). Three groups were exposed 4, 8 and 14 times to radiation with 0.002 µSv for each exposure. The catalase activity of each rat was examined by a spectrophotometer. Data were analyzed using one-way ANOVA followed by Bonferroni test. The results showed the average of catalase activity on Wistar rat’s submandibular gland, respectively for: 0.150±0.0895 (KK), 0.1405±0.0607 (K1), 0.1228±0.0290 (K2), 0.1227±0.0556 (K3). Data showed significant differences of catalase activity between test groups, but showed not significant differences of catalase activity between each groups of Rattus norvegicus strain Wistar’s submandibular gland. In this study concluded decreased catalase activity of Rattus norvegicus strain Wistar’s submandibular gland resulting from x-rays ionizing radiation by 4 times, 8 times and 14 times exposures.

Abstract

Background: The traumatic ulcer is the most common oral soft tissue lesion that cause physiologic dissorder (pain) in patients, so it need a medicine to improving healing processs.Stichopus hermanii contains glycosaminoglycans, omega-3 and other agents that accelerate wound healing. Their influence againts signaling cell for proliferation and angiogenic for healing procce. Endoglin plays in a number of cellu- lar processes and the elevated expression during wound healing. Purpose: Knowing the effectivity of ethanol extract of Stichopus hermanii towards the expression on traumatic ulcer maturation phase in oral mucosa. Materials and Methods: The research subjects were forty eight wistar rats divided into 6 groups. Traumatic ulcer was made in the center-rat?s-labial-mucosa. The X1 group was smeared with sterilized aquades as the control group. The X2? wassmeared with 0,2% hialuronic acid,? X3 with 60% ethanol extract, and? X4 with 80% ethanol extract. They were smeared once a day during 4 days. On the sixth day, the rat?s labial mucosa was biopsied to make histological preparation in order to study the amount of fibroblast. Results: Kruskal-Wallis test showed not significant difference (p=0,000) after applying? Abstract

Latar Belakang: Resiko osteoporosis dapat terdeteksi melalui parameter lebar kortikal mandibula dan porositas yang dapat dilihat pada radiografi panoramik. Pengukuran dengan menggunakan radiografi panoramik dalam menilai kualitas tulang dan mengobservasi tanda dari resorpsi dan osteoporosis salah satunya dengan menggunakan Mental Index. Mental Index merupakan perhitungan rata-rata lebar korteks dibawah foramen mentale karena korteks mandibula yang tipis dikorelasikan dengan penurunan densitas tulang. Tujuan: Untuk membandingkan hasil penilaian ketebalan korteks dengan menggunakan Mental Index pada pasien wanita berdasarkan kelompok umur 30 ? 70 tahun di RSGM FKG Universitas Hang Tuah Surabaya. Bahan dan Metode: Penelitian ini menggunakan? radiografi panoramik pasien wanita umur 30-70 tahun dengan menggunakan radiomorfometri Mental Index untuk menilai ketebalan korteks mandibula secara manual dengan menggunakan kaliper digital (ketelitian 0,01mm). Hasil: Berdasarkan uji One Way ANOVA diperoleh nilai p=0.000 yang berarti terdapat perbedaan yang bermakna dari hasil penilaian ketebalan korteks antar kelompok umur menggunakan Mental Index berdasarkan radiografi panoramik pasien wanita berumur 30-70 tahun di RSGM FKG Universitas Hang Tuah Surabaya. Kesimpulan: Penilaian ketebalan korteks dengan menggunakan Mental Index pada pasien wanita mengalami penurunan pada kelompok umur 56-70 tahun yang dibandingkan dengan tiga kelompok umur sebelumnya yaitu kelompok umur 30-35 tahun, 36-45 tahun dan 46-55 tahun dan sebanyak 11 radiografi panoramik atau 12% dari total 93 radiografi panoramik yang terdeteksi osteoporosis berdasarkan Mental Index

Abstract

Dentists have an important role in detecting osteoporosis that can be found incidentally on radiographic panoramic examination. Radiomorfometri mandible on panoramic radiography is a simple technique for screening osteoporosis and provides additional benefits for patients with radiographic examination for oral care. This study used a sample aged 35 years and over because the theory is based on the influence of estrogen hormone responsible for the regulation of calcium levels in the blood which begin to decline levels in the body since the age of 35 years. Radiomorfometri used were mandibular cortical index (MCI) and panoramic mandibular index (PMI). Each of these has a different way of identifying osteoporosis by panoramic. The sample of this cross-sectional study, with analytic observational study design is the total sampling, ie all data panoramic radiographs of patients over the age of 35 years and over who come to the Hang Tuah University Hospital Surabaya and required panoramic radiographs for dental and oral care needs, and in accordance with the criteria. The results of the Mann Whitney test showed no significant difference with p = 0.624 (p&gt; 0.05), which means that there is no significant difference between the results of os teoporosis based on MCI and PMI. It is concluded that there was no significant difference between the results of osteoporosis based on radiomorfometri MCI and PMI on Hang Tuah University Hospital patients.
