Abstract

Several isolates of agarwood-forming fungus in Gyrinops versteegii have been isolated from the result of exploration in Lombok and Sumbawa Islands. This study aimed to identify the three fungus associated with the agarwood formation in G. versteegii originated from Lombok Tengah, Alas, and Lombok Barat, Nusa Tenggara Barat. The three fungus cultured in liquid medium PDB (Potato Dextrose Broth) and incubated for 1 month in shaken culture. The mycelium of each fungus was harvested for DNA isolation. Amplification is done by Polymerase Chain Reaction (PCR) using the Primers ITS and TEF with obtained amplicon having base length ranging of 300-600 bs. BLAST analysis showed that the three fungus have similarity with Fusarium solani. These results were confirmed by phylogenetic tree where all fungus has genetic relationship with F. solani.

Abstract

Gaharu telah dikenal secara luas sebagai salah satu produk hasil hutan bukan kayu unggulan karena manfaat dan nilai ekonomisnya yang cukup tinggi. Beberapa isolat cendawan pembentuk gaharu pada jenis  Gyrinops  versteegii  telah  diisolasi  dari  hasil  eksplorasi  di  pulau  Lombok  dan  Sumbawa. Penelitian ini bertujuan untuk mengidentifikasi tiga cendawan yang berasosiasi dengan pembentukan gaharu pada jenis Gyrinops versteegii asal Lombok Tengah, Alas, dan Lombok Barat, Nusa Tenggara Barat. Ketiga cendawan dibiakan dalam medium cair PDB (Potato Dextrose Broth) dan diinkubasikan selama 1 bulan sambil digojok. Miselium dari masing-masing cendawan dipanen untuk isolasi DNA. Amplifikasi dilakukan dengan Polymerase Chain Reaction (PCR) menggunakan primer ITS dan TEF dan diperoleh amplikon yang memiliki panjang berkisar 300-600 pb. Hasil analisis BLAST menunjukkan bahwa ketiga cendawan memiliki kesamaan dengan Fusarium solani. Hasil tersebut dikonfirmasi dengan pohon filogenetik dimana ketiga cendawan uji mempunyai kekerabatan dengan F. solani.

Abstract

Nyawai (Ficus variegata  Blume) is a priority alternative tree species in the establishment of timber plantations. One of the factors determining the success of the development of nyawai forest plantations is the use of high quality seedlings because high quality seedlings would result high productivity stands. Application of fertilizer and mycorrhizal fungi can improve seedling quality. The purpose of this study is to get an optimum combination of fertilizer and mycorrhizal to produce high quality nyawai seedlings. Nyawai fruits were collected from Cibodas Botanical Garden  Cianjur, Ir. H. Djuanda Grand Forest Park (Bandung), and Cikampek Forest Research. The experimental design used was randomized block design (RBD) with factorial pattern. Each replication consisted consisted of 30 seedlings Germination was done by sowing seed on the sprouting tubs contained by sterilized media. Transplanting media used sub soil plus mycorrhizal soil: without mycorrhizal (C0 ), Glomus sp.  (C1 ), Acaulospora sp.  (C2 ) and NPK : 0.0 g  (P0 ), 0.5 g / polybag  (P1 ), 1.0 g / poybag  (P2 ). To produce  nyawai seedling can use a mix media of subsoil + 30% coconut coir dust (coco peat) +10% rice husk (v / v), CMA Glomus  sp and Acaulospora  sp  able to colonize with the roots of nyawai seedlings.

Abstract

Gyrinops versteegii is one of protected species because of the rare existence. One of the efforts to keep the preserved is by cultivation. Cultivation techniques using generative basis has already done a lot. However, this technique has several limitations, due to the recalcitrant character of the seeds and the inequality of the characteristics of the progeny with their parent. Meanwhile, vegetative techniques will produce a new individual that have the same characteristics as the parent. The alternative-promising vegetative techniques of G. versteegii are using media because it is cheap and easy to obtain, and increase the achievement if additional hormones were used. The purpose of this research is to determine the optimum concentration of the NAA hormone for G. versteegii cuttings. The research method is done by cutting the saplings of G. versteegii on the shoots. The medium used are water without hormones (as control), water with hormones of NAA 100, 200, and 300 ppm. Observations 10 times, at the end of the observation carried out measurements on the root, percent of life, buds, dry stem weight, and dry root weight. The result showed that NAA hormone treatment influenced significantly on the growth of the roots, percent of life, dry stem weight and dry weight of roots of G. versteegii. Medium of water with the addition of NAA 300 ppm was the composition that can be used as a medium to trigger initial rooting.

Abstract

Sengon known as forest product with high market demand. Tissue culture is one multiplication method to get seeds in large quantities. One factors that support the success of reproduction is the health of the plant material. The purpose of the study is to determine the influence of pre-sterilization treatments towards the successful initiation of sengon organogenesis. The research method was done by spraying the seedlings with Pyraclostrobin 50 g.kg -1+ Metiram 550 g.kg -1of 2,500 ppm every week for 3 months. Each explant was grown on MS medium.The observation was carried out done every 1 week for 12 weeks, including the amount of contamination (bacteria, fungi), browning and the appearance of buds, roots and callus. Data were analyzed by t-test. The next stage is multiplication by using treatments of 24D 0.1 mM, 24D 0.5 mM, BAP 0.5 mM, BAP 2 mM, K 0.1 mM, K 0.5 mM, TDZ 0.1 mM, TDZ 0, 5 mM, and control. Completely randomized design was used and analyzed by using one way ANOVA, and tested with Duncan for the differences at 0,05 significant level. The results showed that pra-sterilization treatment was capable of supporting the successful initiation of sengon shoots characterized by low levels of contamination, increased number of callus and number of sengon shoots explants. Kinetin 0.01 mM in the multiplication media of sengon shoots was able to increase the number of adventitious shoots.
