The Effect of Propofol on Onset and Sedation in Diabetic Mellitus Rat (Rattus norvegicus)

Abstract

This research aimed to examine the time of onset and sedation on diabetic mellitus (DM) rat (Rattus norvegicus) by propofol. This study used 8 female wistar rats 2-3 months old and 150-200 grams of bodyweight, fed with standard feed and water ad libitum. Samples were divided into two groups. Group 1 (KI) as a control group was injected intraperitoneally with aloxan solvent and group 2 (KII) was injected intraperitoneally with 150 mg/kg bodyweight aloxan. Blood glucose was checked 10 days after aloxan injection, DM was categorized if the blood glucose over 150 mg/dl. All rats were injected intravenously with propofol 1% with the dose of 10 mg/kg bodyweight. Onset and sedation time was counted using stopwatch after propofol injection. The result showed that mean (±SD) of onset on KI and KII were 1.39±0.49 and 8.64±1.23 seconds, consecutively and they were significantly different at P<0.01. Whilst mean (±SD) of sedation KI and KII were 12.12±1.47 minutes and 7.62±1.61 minutes, respectively. The onset and sedation time of KI was significantly different from KII P<0.01. The conclusion of this research was DM affects time of onset and sedation when wistar strain rats were anesthetized by 1% propofol.Key words: rats, diabetic mellitus, propofol, onset, sedation

Abstract

The aim of this research was to investigate the effect of red watermelon (Citrullus vulgaris) extract to the value of erythrocytes, hemoglobin, and hematocrit of mice (Mus muscullus) exposed to cigarette smoke. This study followed direct complete randomized design by using 12 males’ mice which divided into 4 groups. The treatment group is divided by the negative control group which given 0,5 ml of aquadest, the positive control group which exposed to cigarette smoke and 0,5 ml of aquadest. The treatment group I was exposed to cigarette smoke and given red watermelon extract dose 22 mg/Bw mice. The treatment group II was exposed to cigarette smoke and given red with watermelon extract dose 44 mg/Bw mice. The exposure to cigarette smoke and the given of red watermelon extract were conducted for 30 days. The blood taking was performed on day 31 in the Plexus retroorbital.  Furthermore, the number of erythrocytes, hemoglobin and hematocrit were calculated. The data were analyzed using ANOVA completely randomized design with SPSS for Windows 16.0. The results of this study showed that the extract of red watermelon for 30 days showed a highly significant effect (P <0.01) to the number of erythrocytes, hemoglobin, and hematocrit of mice. It is concluded that free radicals contained in the cigarette can cause a hemoglobin desaturation, increased blood viscosity, oxidative stress and red watermelon extract can minimize the damage.

Abstract

The aim of this research was to evaluate the effect of chayote (Sechium edule) leaves infusion on hemoglobin and hematocrit level of anemic rat (Rattus norvegicus). Twelve anemic hemorrhagic male rats strain Wistar with body weight ranging from 175-200 g were used as sample in this research. Rats were divided randomly into 3 treatment groups, each group consist of 4 rats. The control group (P0) was not given chayote leaves infusion. Group P1 and P2 were given chayote leaves infusion with the concentration of 25% and 50% respectively, with dose of 1 ml per 200 gram body weight. Rats were fed with standard commercial food (Turbo feed) and aquadest ad libitum. The rats were treated anemic by taking 20% blood from total blood volume prior to administration of chayote laeves infusion. Chayote leaves infusion was given for 10 days consecutively. Blood sampling was carried out on day 0, 5, and 10 via caudal vein. Data were analyzed using anlysis of variance (ANOVA) Split-Plot pattern. The result showed that chayote leaves infusion increase level of hemoglobin and hematocrit in experimental rats. In conclusion 25% concentration of chayote leaves infusion is more effective than 50% concentration.Key words: anemia, chayote leaves, hemoglobin, hematocrit

Abstract

The aim of this research was to find out the effect of water temperature increase on leucocyte count of nile tilapia. The blood sample from 9 tilapia with weigh of 40-50 g were used in this study. The research was conducted using completely randomized design (CRD) with 3 treatments and 3 replications. Treatment P1 as a control with aquarium temperature was maintained at 29±1 C. Water temperature in treatment P2 and P3 was maintained at 32±1 C and 35±1 C respectively. The aquarium size in each treatment group was 80x60x40 cm with height of water 30 cm. The treatment was done for 15 days and on day 16th, the blood were collected from caudal vein. Data were analyzed with analysis of variance (Anova) and followed with Duncans multiple range test. The result showed that total leucocyte count (x 105/mm3) of nile tilapia in group P1, P2, and P3 were 3.08±0.88, 1.70±0.52, and 1.69±0.20 respectively. lts showed that an increase in temperature significantly affect (P<0.05) the leucocyte count of nile tilapia. The conclusion of this research is the increase in water temperature in aquarium decrease the leucocyte count of nile tilapia.

Abstract

This research was aimed to determine the present status of local goats based on the age at UPT experimental animals Faculty of Veterinary Medicine Syiah Kuala University by urine examination. Sample used in this research was urine from 13 local goats (Capra sp.) male and female that collected in the morning. Dipstick urine (Verify TM) was directly dipped into the collected urine and the result were obtained by comparing with a standard color on urine dipstick (Verify TM) with interval 30-120 seconds. The semiquantitatively data obtained using urine dipstick include measurement of leukocyte, nitrite, urobilinogen, protein, pH, blood in the urine, specific gravity, ketones, bilirubin and glucose. From the results obtained age <1 year nitrite 1 goat, blood 1 goat, alkaline pH and density under normal circumstances. At the age of 1-3 years old goats there are leukocytes 4 goats, nitrite 2 goats, proteins 4 goats, bilirubin 1 goat, pH is alkaline and density under normal circumstances. In goats aged 3-4 years there is leukocytes 1 goat, nitrite 2 goats, pH is alkaline and density under normal circumstances.

Abstract

The purpose of this study was to determine the effect of using ketamine-xylazin and propofol on heart rate and breathing in local male dogs (Canis familiaris). Six male dogs with average body weight ±2-3 kg were divided into 2 treatment groups, each group was subcutaneously injected with of atropine sulfate at dose of 0.04 mg/kg as premedication prior to anesthetic administration. The first group (K1) was intravenously injected with anesthetic combination of ketamine-xylazine at dose of 5.5 mg/kg and 1 mg/kg, respectively. The second group (K2) was given propofol at dose of 6.6 mg/kg intravenously. The heart rate and breathing were directly calculated using stethoscope and stopwatch with 10 minutes interval. Data were analyzed statistically using analysis of variance (Anova) with split-splot design and if there is a difference between the two groups followed by least significant difference test (LSD). The administration of ketamine-xylazin (K I) significantly (P<0.05) increase the respiratory frequency compared to propofol administration (K2). The interaction between time and precise observation of K1 and K2 on 30 minute observation (W3) showed significant differences (P<0.05). While on other time observations did not reveal any differences in the interaction between time and observation (P>0.05). It can be concluded that there is no difference effects of given ketamine-xylazin and propofol against heart rate on local male dogs. But there was difference effect of administration ketamine-xylazin and propofol on respiratory frequency on local male dogs.

Abstract

           The purpose of this research was to identified the compounds in ethanol extract of soursop flower and determined the antibacterial activity of the soursop flower extract on Salmonella enteritidis. The phytochemistry screening was done to identify the secondary metabolite compounds of this extract. The concentrations used of ethanol extract of soursop flower were 10%, 20%, 30%, 40%, 50%, with ampicillin 10 μg/disc as the positive control and 10% Dimethyl Sulfoxide (DMSO) as the negative control. The antibacterial activity test of soursop flower ethanol extract against Salmonella enteritidis was held in vitro using the paper disc diffusion method by measuring the diameter of the inhibition zone. The phytochemistry screening showed that the ethanol extract of soursop flower had secondary metabolites such as alkaloids, phenolic, and flavonoid. The result of the antibacterial activity test showed that there was no inhibition zone (bright zone) at various concentrations. So, it can be concluded that soursop flower ethanol extract (Annona muricata L.) contained the secondary metabolites such as alkaloids, phenolic, and flavonoid, and also this extract had no antibacterial activity against Salmonella enteritidis

Abstract

The research aimed to determine the effect of Moringa leaves infuse toward the haemoglobin levels of broiler chickens. This study used 21 broiler chickens which divided into three groups and each group consisting of 7 broiler chickens. The treatment group (P0) were given only mineral water (aquadest), the treatment group (P1) were given moringa leaves infuse with a concentration of 10 % and treatment group (P2) infusion were given moringa leaves with a concentration of 20 %. The given of Moringa leaves infuse was starting at seven days old of chickens and given for 28 days continously as a drinking water.  The chickens were feed by ad libitum during the research. After the treatment completion, a researcher carried out a blood sampling via the axillary vein.  Data were analyzed with ANOVA by Complete Random Design one way and Tukey test to determine a difference between the treatments of each group. The research results showed that ± SD haemoglobin (g / dl) in the group (P0) 7.71 ± 1.13, (P1) of 8.15 ± 1.95 and (P2) 10.8 ± 1.76. Statistical analysis showed the treatment effect (P > 0.05) of the haemoglobin levels of broiler chickens. The Tukey test results of the treatment group (P0) were significantly different (P < 0.05) with treatment group (P2), but not significantly different (P > 0.05) with treatment group (P1). The treatment group (P2) were significantly different (P < 0.05) with treatment group (P1). From the results of this research, it was concluded that infuse Moringa leaves can improve haemoglobin levels of broiler chicken.
