Abstract

A Research About Activity of Ethanol of Pearl Grass (Hedyotis corymbosa) On The Imunoglobulin M (IgM) by Hemaglutination Method Had Been Conducted of Male Mice. This research purpose to established of activity imunoglobulin M (IgM) from ethanol extract of pearl grass (Hedyotis corymbosa). Mice is animal that used for this research,The mice was divided into 4 treatment groups, where each group consisted of 3 mice. The first group as negative control, and each group was given Sheel Red Blood Cell (SRBC) 2 % as many 0,1 ml intraperitonially, after that negative control group was administrated with air sulingt, and other group was administrated with ethanol extract  dose 26 mg, 78 mg, dan 130 mg for five days. The blood from all group was collected by intracardial after five days. The activity of imunoglobulin M (IgM) was measured by hemaglutination method based on titer of the imunoglobulin M (IgM) there was highest dilution of mice blood serum in which aglutination was showed. Research result showed   Ethanol extract of Pearl Grass (Hedyotis corymbosa) at 26 mg, 78 mg, and 130 mg dose, can increased imunoglobulin M (IgM) activity.Key words : Imunoglobulin M, Hedyotis corymbosa, Hemaglutination Method

Abstract

The research concerning antimitotic assay of “Sisik Naga” (Drymoglossum piloselloides [L] Presl) ethanol extract  using Tripneustes gratilla Linn eggs cleavage inhibition method had been conducted. The aim of this study was to evaluate which the extract of that provide higher effect in to inhibit cell division of fertilized sea urchin eggs. Quantivication of division was performed visually by light microscopy by counting the number of cleaveaged and non cleaveaged embryos after an incubation time of  ± 2 hours. If 80 – 100% inhibition of cleavage (IC80-100) occurs at ≤ 16 mg/ml, the compound is considered to be active as antimitotic assay.Key word : Antimitotic, Drymoglossum piloselloides, vinkristin

Abstract

Inflammatory medication which is commonly used is the treatment synthetically or traditionally from the natural ingredients, the green grass jelly leaves (Cyclea barbata Miers). This research aimed to determine the effects and the most effective dose of the ethanol extract of green grass jelly leaves as an anti-inflammatory in white rats induced by carrageenan. This research used 15 mice divide into 5 group. Group I (negative control) was given Na.CMC; group II (positive control) was given Na.Diclofenac; group III, IV and V were given ethanol extract of green grass jelly leaves (EEDCH) at the dose of 1.875 mg/kgBW, 3.75 mg/kgBW, and 7.5 mg/kgBW. Pretreatment on animals induced by carrageenan 1% amounted to 0.1 mL in sub plantar on the soles of the mice. The measurements were made every 1 hour for 5 hour by using the pletismometer tool. The result showed that the ethanol extract of green grass jelly leaves has a potential as anti-inflammatory. Based on the result  of statistical analysis of the one way ANOVA followed by Post Hoc Test LSD showed that substance EEDCH at the dose of 7.5 mg/kgBW has anti-inflammatory effect were not significantly different from the positive control (Na.Diclofenac).Keywords: Green grass jelly leaves (Cyclea barbata Miers), anti-inflammatory, Na.Diclofenac.

Abstract

The research of toxicity assay of Ethanol Extracts of Taro tubers (Colocasia esculenta L. Schott) have been conducted with the aim of determining the effects of toxicity and anticancer potential of ethanol extract of tubers of taro (Colocasia esculenta L. Schott) by Brine Shrimp Lethality Test method against Artemia salina Leach. Ethanol extract, made with concentrations of 0.1, 1, 10, 100, 1000, and 10000 µg/ml and for comparison with the drugs vincristine concentrations of 0.01, 0.1, 1, and 10 µg/ml. Negative control using sea water made with the same concentrations. Furthermore, each concentration 10 ml of sea water and then put 10 shrimp larvae. The treatment is done by 10 replication at each concentration. Observations were made after 24 hours of treatment by counting the number of dead larva. The observations calculated using probit analysis program, showed LC50 values with 95% confidence level is 33,997 µg/ml (7,970 µg/ml-151,231 µg/ml). And the results of a comparison of data from drug vincristine showed LC50 value is 0,398 µg/ml (0,259 µg/ml-0,624 µg/ml).

Abstract

The research was about Tuberculosis Drugs Using System in the  Balai Besar Kesehatan Paru Masyarakat Makassar (BBKPM) Period January 2011 – June 2011. The aim of this research was to describe about using system so obtained TBC drugs which used in the Tuberculosis Drugs Using-System in the  Balai Besar Kesehatan Paru Masyarakat Makassar (BBKPM) Period January 2011 – June 2011. The data taking prospectly via all the tuberculosis patient medical record in the Tuberculosis Drugs Using System in the  Balai Besar Kesehatan Paru Masyarakat Makassar (BBKPM) Period January 2011 – June 2011 obtained 30 patients. The result of this research can be concluded that from the regiment side, about 96,7% Tuberculosis patients which treatment in the  Balai Besar Kesehatan Paru Masyarakat Makassar (BBKPM) Period January 2011 – June 2011 were category 1 and the residual was category 3. While the side of combination with the other drug, giving combination dominated about 96,7%. While if the give single drug about 3,3%. The frequency of  the other drug usage was dextromethorphan about 90% and the smaller was pyridoxin about 10%.Key Word : Using System, Tuberculosis Drugs, BBKPM

Abstract

Hypolipidemics a drug used to lower plasma lipid levels in the blood. This study aimed to examine the hypolipidemic activity of ethanol extract of purslane herb (Portulaca oleracea L.) against the triglyceride level reduction in obese mice. The purslane herb contains omega-3, flavonoids and hight ascorbic acid as a lowering of triglyceride levels in the blood. This study used 15 malerats divided into 5 groups. Group I (negative control) was given Na.CMC 1%, group II (positive control) was given Gemfibrozil, group III, IV, and V were given ethanol extract of purslane herb with dose of 100 mg/kgBW, 200mg/kgBW, and 300 mg/kgBW as therapy group. All the administration were done orally once a day for 16 days. The measurement of triglyceride level was performed on early triglycerides before induction, after induction and after therapy. Before therapy, the rats were given hight-calorie diet. The results showed that the ethanol extract of purslanes herb has activity as hypolipidemic in lowering trygliseride level in obese mice. From the statistic result of One-Way ANOVA followed by LSD test, it can be concluded that the ethanol extract of purslane herb of the dose 100 mg/kgBW provide the most effective hypolipidemic activity that triglyceride level is not significantly different from positive control group (p>0,05).

Cytotoxic activity of simvastatin in T47D breast cancer cell lines and its effect on cyclin D1 expression and apoptosis

Abstract

Background: Statins (HMG-CoA Inhibitors) is a drug used for decreasing plasma cholesterol levels and used in therapy to prevent coronary artery disease. Research in animals and epidemiological studies showed that statin therapy can decrease risk against cancer associated with cholesterol. Based on that result then research of cytotoxic activity against simvastatin knowing cultur T47D breast cancer cells and his influence in decreasing expression of cyclin D1 and induction of apoptosis has been done.Research objectives: The aims of this research is to prove activities of simvastatin against T47D breast cancer cell culture, especially to examine cytotoxic activity, cyclin D1 expression, and simvastatin effect in apoptotic induction.Research method: The type of this research is quasi experiment with using posttest with non-equivalent control group design. Cytotoxicity test performed on T47D breast cancer cell cultures using MTT assay to determine IC50 values after given simvastatine. Expression of cyclin D1 and apoptosis induction test detected using flow cytometry with antibody monoclonal anti-cyclin D1 and Annexin V-Pi, then analyzed by FACS-Calibur program.Results: Simvastatin has cytotoxic effect against T47D breast cancer cells with IC50 values 25.25 µg/mL. Simvastatin with concentrations of 6.31; 12.62; 25.25 and 50.5 µg/mL was able to decrease the cyclin D1 expression. Furthermore, simvastatin can induce apoptosis with EC50 values 26.96 µg/mL in T47D breast cancer cells.Conclusion: Simvastatin has cytotoxic activity of in T47D breast cancer cells and decreasing cyclin D1 expression and inducing apoptosis activity in T47D breast cancer cells.Keywords: simvastatin, cytotoxic, cyclin D1, apoptotic, T47D.
