Abstract

Background: Overweight and obesity can cause high body fat total and lipid profile in the blood that brings risk for diseases to the bearer. Some studies on green tea extract supplementation have been undertaken to find out its effect on the fat reduction in children, men, and mice and the result showed a reduction in body fat, weight, appetite, and triglyceride level. This study uses green tea extract supplemented to overweight and obese women. Objective: To find out the reduction of body fat total and lipid profile (total cholesterol, triglyceride, LDL, HDL) in the blood of overweight and obese women personnel of Yogyakarta Municipal Health Office supplemented with green tea extract.Method: The study was randomized double-blinded control trial. The subject of the study was overweight and obese women of 35-55 years old personnel of Yogyakarta Municipal Health Office. Samples were 86 women randomly taken, comprising experiment group and control group, each of which consisting of 43 women. Each group had diet according to individual appetite. Both groups were given capsules of similar form and color but different ingredients: placebo and green tea extract. The experiment took three months. Blood was taken before and after the experiment to identify lipid profile (total cholesterol, triglyceride, LDL, HDL). Body fat total was examined using body fat analyzer. Data analysis used paired t-test.Results: Green tea extract supplementation could significantly reduce body fat total and insignificantly reduce cholesterol level, triglyceride, and LDL, significantly increase HDL in overweight and obese women.Conclusion: Green tea extract supplementation could reduce body fat total and increase HDL (p<0.005).

Abstract

Background: Province of Yogyakarta Special Territory is a province of “elderly” structure population, as viewed from the number of elderly which is 15% of total population. Anemia is one of nutrition problems of elderly which reaches as high as 50%. This is caused by low intake of iron and other substances related to iron metabolism process in forming hemoglobin. Low hemoglobin concentration can be one of causes of elderly declining physical fitness. Efforts to overcome anemic problems can be done through the supply of iron and zinc for the elderly twice a week.Objective: To know effect of iron and zinc supply to hemoglobin concentration and physical fitness for anemic elderly at Bantul District, Province of Yogyakarta Special Territory.Methods: The study was a randomized double blind controlled trial design. Subject of the study were anemic elderly that fulfilled inclusion criteria. The supply of iron, combination of iron and zinc to trial group was given twice a week on different days, supply of iron was given on Monday and Thursday, while supply of zinc was given on Tuesday and Saturday. Laboratory and physical fitness examination to both group were done prior and after treatment. Nutrition intake data were achieved through food record. Next, to know effect of iron, iron and zinc supply to elderly hemoglobin concentration and physical fitness, covariant analysis was done.Results: The result of the study showed that after 3 months’ treatment, there was significant difference of hemoglobin concentration caused by different treatment of group with iron supply, iron and zinc combination supply and control group (p=0.012) although iron and combined iron and zinc supply had equal effectiveness in increasing hemoglobin concentration (p=0.404). There was no significant relationship between elderly hemoglobin concentration and physical fitness (p=0.448). This showed that treatment given affected increase of hemoglobin concentration but the supply and  increase of  hemoglobin concentration did not affect increase of elderly physical fitness.Conclusion: The supply of iron and combined iron and zinc increase hemoglobin concentration, but the supply and in- crease of hemoglobin concentration did not increase the elderly physical fitness.

Abstract

Latar belakang Stres oksidatif telah diketahui berperan dalam patogenesis diabetes melitus (DM), termasuk penyakit jantung koroner (PJK) sebagai salah satu komplikasi tersering. Kuersetin adalah salah satu kelompok flavonoid alami yang banyak tersebar dalam tumbuhan dan dapat bekerja sebagai antioksidan. Penelitian sebelumnya menunjukkan bahwa kuersetin dapat mencegah komplikasi dan mengontrol kolesterol LDL pada DM tipe 2. Tujuan Tujuan penelitian ini adalah untuk mengetahui pengaruh kombinasi kuersetin dan glibenklamid terhadap kadar kolesterol LDL pada tikus yang mengalami DM tipe 2. Metode Penelitian ini bersifat eksperimental murni dengan desain kelompok kontrol pre dan post- test. Terdapat 16 tikus diabetes sebagai subjek penelitian yang akan dibagi menjadi 4 kelompok perlakuan; kelompok 1 menerima plasebo, kelompok 2 glibenklamid 5 mg/kgBB/peroral, kelompok 3 kuersetin 20 mg/kgBB/peroral dan kelompok 4 kombinasi keduanya. Perlakuan diberikan selama 4 minggu dan diperiksa perubahan kadar kolesterol LDL yang terjadi. Abstract

The tyrosine kinase receptor RET, which is the protein product of the RET gene, is involved in the development of the mammalian nervous system that causes Hirschsprung’s disease (HSCR). RETs are cell surface molecules that are expressed in cells derived from the neural crest. The purpose of this study was to investigate the polymorphism of the RET gene in HSCR in the Yogyakarta population. Genomic DNA was extracted from surgically removed bowel tissues of 54 unrelated HSCR patients. Exon 2 of the RET gene was amplified by polymerase chain reaction (PCR) and analyzed by restriction fragment length polymorphism (RFLP). Molecular results were compared with clinical performance of Hirschsprung patients. RET polymorphism was detected in exon 2 in all of the 54 Indonesian HSCR patients. The allelic distribution of the c135GàA polymorphism in the RET exon 2 indicated that the A allele was more frequent in patients than in control individuals (chi-square test, p= 0.001). Thus the RET variant allele A is over-represented in patients affected with the HSCR phenotype. Polymorphism of exon 2 of the RET gene was found in sporadic Hirschsprung’s disease in the Yogyakarta population, which suggests that the RET gene plays important roles in the pathogenesis of HSCR.

Abstract

Background: Thyroxin plays important role in the metabolism of carbohydrate, protein and cholesterol, and in the process of growth. Iodine deficiency disorder not only can be caused by lack of iodine substance but also another competition factor, which is a pollutant substance which is goitergenic (pesticide). This goitergenic substance interferes hormonogenesis of thyroid causing enlargement of thyroid gland known as goiter.Objective: This study was aimed at examining the relationship between status of pesticide and status of iodine deficiency disorders (urinary iodine excretion) of elementary school children and the difference between them based on level of their endemic in Dairi District.Methods: This was an observational study using a cross-sectional design. Palpation of thyroid gland was made to determine the level of endemic; concentration of blood cholinesterase was determined using tinto meter kit; urinary iodine excretion was analyzed with the ammonium persulfate digestion method. Data was analyzed with chi square and anova.Results: Chi square test indicated that the relationship between the concentration of blood cholinesterase and urinary iodine excretion was significant (p<0.05) with OR 11.1 and the relationship between concentration of cholinesterase and iodine deficiency disorders (palpation) was not significant (p>0.05) with OR 1.3. Anova test, based on endemic, indicated that there was a significant difference between the concentration of blood cholinesterase and urinary iodine excretion (p<0.05).Conclusion: There was significant relationship between status of pesticide (concentration of blood cholinesterase) and status of iodine deficiency disorder based on urinary iodine excretion and there was no significant relationship between concentration of cholinesterase and iodine deficiency disorders based on the result of palpation. There was a difference in the average status of pesticide and iodine deficiency disorders status among the elementary school children based on their endemic.

Abstract

Physical exercise is important in preventing and adjunctive therapy certain diseases. However,over physical exercise for beginners may cause oxidative stress and muscle injury leads to generatechemoattractants whic enables to attract neutrophil and monocyte towards the injury and stimulatesleucocytes activation. The use of antioxidant-rich fruits to mitigate exercise induced oxidative stresshas been applied. This study was conducted to evaluate the effect of guava juice (Psidium guajavaL) consumption during aerobic exercise on leucocytes, neutrophils and superoxydes dismutase(SOD) level of beginners. Sixteen students of National Land College (Sekolah Tinggi PertanahanNasional/STPN), Yogyakarta who met the inclusion and exclusion criteria were involved in thisstudy. Subjects were divided into two groups i.e. a group given mineral water as control andanother group given 240 mL guava juice 0.67 g/mL daily for 27 days as treatment group. Bothgroups then underwent aerobic exercise for 30 minutes every day for 27 days. Blood samples weretaken before and after exercise on day 1, 7 and 14 for leucocyte and nutrophil count as well as SODanalysis. The leucocyte count and SOD level before and after exercise in both Guava juice andMineral water groups were not significantly different (p>0.05). However, the percentage of neutrophilon day 1 and 7 were significantly higher than that in Mineral water group (p<0.05). Moreover,during the exercise, the percentage of neutrophils in Guava juice group significantly increased(p<0.05), whereas in Mineral water group was not significantly different (p>0.05). It can beconcluded that consuming guava juice during exercise for the beginners does not influence leucocytecount and SOD levels. However, it can increase percentage of neutrophil.

Abstract

Kelelahan otot merupakan suatu kondisi dimana otot kehilangan kemampuan berkontraksi karena stimuli terus menerus dan hal ini dikaitkan dengan penurunan cadangan glikogen. Obat derivat methylxanthine (kafein dan teofilin) bermanfaat untuk menghemat penggunaan glikogen. Penelitian ini bertujuan untuk menggali efektivitas kafein dan teofilin dalam menunda kelelahan otot sebagaimana yang diukur dalam lama struggling. Sebanyak 18 (delapan belas) ekor tikus berusia ± 3 bulan, berat 200-210 gram, kadar Hb 11,5-16 mg/dL dan kadar Hmt 35-51%, dibagi secara acak ke dalam 3 kelompok perlakuan: kontrol (akuades 1 mL/kgBB), kafein 3,78% (1 mL/kgBB) dan teofilin 3,15% (1 mL/kgBB). Kelelahan otot diukur dengan lama struggling dalam detik. Data rerata lama struggling antar kelompok dianalisis dengan one way ANOVA. Hasil menunjukkan bahwa lama struggling untuk kelompok kafein (286.83+7.11) detik dan teofilin (312.33+15.92) detik, lebih lama daripada kelompok kontrol (140.33+23.24) (p&lt;0,05), sedangkan lama struggling kelompok kafein dengan teofilin tidak berbeda bermakna (p&gt;0,05). Disimpulkan bahwa kafein dan teofilin mampu menunda kelelahan otot pada tikus dan efektifitasnya sebanding.

Abstract

Latar belakang. Tuberkulosis (TB) merupakan masalah kesehatan dunia yang masih perlu mendapat perhatian. Indonesia sampai saat ini masih menempati urutan ke 3 di dunia untuk jumlah kasus TB setelah India dan Efektifitas Pemberian Ekstra ... (Agus Purwanto dkk.) 11 China dengan prevalensi 415 kasus per 100.000 penduduk pada tahun 2010. Pasien TB mengalami penurunan kadar albumin secara bermakna, dan penyebab penurunannya diduga adalah faktor gizi (asupan makan rendah, anoreksia, peningkatan katabolisme), enteropati dan reaksi protein fase akut. Albumin yang rendah berhubungan dengan peningkatan IL-6 dan sampai saat ini masih sedikit diketahui hubungan albumin dengan sitokin inflamasi terutama IL-6. Tujuan. Penelitian ini bertujuan untuk mengkaji keefektifanputih telur terhadap peningkatan albumin dan penurunan aktivasi IL-6. Metode. Penelitian yang dilakukan dengan rancangan single blind randomized controlled trial. Teknik pengambilan sampel dengan simple random sampling sebanyak 75 pasien terdiri 37 perlakuan dan 38 kontrol. Kelompok perlakuan diberikan terapi diit Tinggi Energi Tinggi Protein standar rumahsakit dan ekstra putih telur dikukus, sesuai dengan kebutuhan albumin pasien berdasarkan perhitungan formula Baxter, diberikan selama 14 hari. Kelompok kontrol diberikan terapi diit Tinggi Energi Tinggi Protein standar rumah sakit dan ekstra makanan terdiri tahu putih dan tepung hunkwe. Kadar albumin dan IL-6 diperiksa sebelum dan setelah perlakuan. Analisis data menggunakan uji Mann Whitney untuk mengetahui perbedaan kenaikan kadar albumin kedua kelompok. Hasil. Hasil penelitian menunjukkan rerata kadar albumin awal kelompok perlakuan adalah 2,82 g/dL dan kelompok kontrol adalah 2,85 g/dL. Kadar rerata albumin akhir pada kelompok perlakuan adalah 3,47 g/dL dan pada kelompok kontrol adalah 2,81 g/dL. Hasil analisa bivariat menunjukkan terdapat perbedaan signifikan terhadap rerata peningkatan kadar albumin pada kelompok perlakuan dan kelompok kontrol dengan p=0,001 (p<0.05). Hasil penelitian menunjukkan rerata IL-6 sebelum penelitian pada kelompokperlakuan 5,07 pg/mL dan kelompok kontrol 3,77 pg/mL. Rerata IL-6 setelah penelitian pada kelompok perlakuan 0.0006 pg/mL dan pada kelompok control 2,57 pg/mL. Uji statistik menunjukkan perbedaan signifikan antara kelompok perlakuan dan kontrol dengan p = 0.001 (p < 0.05). Kesimpulan. Putih telur berdaya guna meningkatkan kadar albumin dan meghambat reaktivasi IL-6 pada pasien TB.

Abstract

Background Hirschsprung disease (HSCR) is a heterogenouscongenital disorder and the current research show that the RETgene is a major locus involved in its pathogenesis. However,whether these genes take a part in sporadically Indonesian HSCRhave not been fully understood.Objective The aim of this study was to analyze the association ofRET gene c2307T&gt;G polymorphism among HSCR patient inYogyakarta population.Methods Genomic DNA was extracted from bowel tissues of 34patients with sporadic HSCR which were removed by surgery ascase group and blood DNA from 46 healthy persons as controlgroup without history of genetic disorder. Exon 13 of RET genewas amplified by polymerase chain reaction (PCR) and wasanalyzed by restriction fragment length polymorphism (RFLP).Results Of 34 patients, 22 were males and 12 were females, givingmale to female ratio of 1.83:1. The c2307T&gt;G polymorphism inRET exon 13 was not significantly difference between patientand control group (chi-square test P=0.17). However, there wasa significant difference in female patient compare with control(chi-square test P=0,04).Conclusion The RET gene c2307T&gt;G polymorphism was foundamong HSCR patient in Yogyakarta population. This poly-morphism can be used as predictor for development of HSCRamong female individual.
