Abstract

A method for identification of the causal bacteria of citrus vein phloem degeneration (CVPD) based on polymerase chain reaction (PCR) technique using template DNA extracted from leaf midrib of citrus has been implemented routinely. The method has not been validated, therefore it is necessary to validate the method to confirm that the method fit for its intended use. Leaf samples showing chlorotic symptom was obtained from Bogor and Bekasi, West Java and used for test samples. These samples was differentiated into 2 groups, i.e. leaf midrib and leaf mesophyll. Validation test involved homogenicity, and reproducibility test; each test was replicated 2 times. The test showed that using leaf midrib gave better result for detection of bacteria causing CVPD disease than using leaf mesophyll. Therefore, this method is recommended as routine detection method for bacteria causing CVPD disease.

Abstract

This study aims to analyze the perceptions and expectations of premarital counseling. As the study sample was 70 couple. The results showed that of the indicators that are part of the five dimensions of the societal expectations of counseling before marriage, the highest expectations on religious dimensions.        By 99.5% what is expected by respondents occurred in his life after experiencing the developed relationship counseling and a balanced attitude toward the family on both sides of husband and wife. Indicator with the smallest percentage value is the dimension of integrity with a ratio of 83.25%, namely: Maintaining unity both in relationships with individuals and institutions. This means that the respondent would hope the preservation of unity in both individual and institutional relationships that exist after counseling, only 83.25% according to their perception becomes reality. The value of spearman rank correlation (rs) between perception (X) and hope (Y) is at 99., 91%. This indicated that the relationship between perceptions as independent variables is very strong in the hope (Y) as the dependent variable. Based on t test, t values obtained count equal 114.207, this value is very far when compared with the t table at ∞ = 0.05%, thus it can be concluded that the perception variables (X) is very significant influence expectations variable (Y) as the dependent variable.

Abstract

Tabat Barito (Ficus deltoidea) is known as a plant that has potency as an antioxidant because of containing a significant phenolic compound. In this study, we performed metabolite profiling on F. deltoidea leaves by UPLC-QTOF-MS/MS to identify its phenolic compounds. Extraction by maceration and ultrasonication techniques with methanol (MM and UM) and ethanol (ME and UE) were used to extract the F. deltoidea metabolites. About 70 metabolites were identified by using UPLC-QTOF-MS/MS in negative ion mode. The amounts of metabolites found in each extract were different, i.e., 45 metabolites in MM, 64 metabolites in UM, 42 metabolites in UE and 41 metabolites in ME. MS/MS could further tentatively identify 16 metabolites. The identified compounds belonged to the class of flavonoids and phenolic acid. Also, we conducted an antioxidant activity by using DPPH method on each extract to determine its potency as an antioxidant. The highest antioxidant activity was exhibited by UM extract (IC50 71.93 ppm) may be due to the number of metabolites in UM extract which was higher than the other extract based on the detected metabolites.
