ABSTRACTFifth generations of Nile tilapia from several strains have been produced by using selective breeding program in Main Centre for Freshwater Aquaculture Development (MCFAD) Sukabumi, West Java. This research was aimed to evaluate the impact of family selection program of some highly economic traits on its genetic variability using microsatellite DNA markers. The total of 180 specimens have been collected from fifth generation of nine reciprocal mating between three families selected from fourth generation of Nile tilapia and were screened for genetic variability at three microsatellite loci (UNH 123*, UNH 172*, UNH 216*). The results showed that the amount of genetic variability on fifth generations of Nile tilapia from three strains was ranged between 33 to 100% and the highest genetic distance relationship between families was 0.3875. This research approved that females and males issued from the family which have more amount of genetic variability and higher distance to others could be considered as genetic materials to produce the next generation. Keywords: microsatellite DNA, genotype, genetic variability, genetic distance, Oreochromis nilotiocus ABSTRAKBeberapa strain ikan nila generasi kelima telah dihasilkan dalam program pemuliaan di Main Centre untuk Freshwater Aquaculture Development (MCFAD) Sukabumi, Barat Jawa.

Mangrove leaf litter production is essential matter for organic recycling of productivity and baseline matter for living of mangrove organisms. Mangrove forest in Conservation Area for Mangrove and Bekantan (KKMB) Tarakan is one of Mangrove forest area which is not only for recreation but also ecological function of mangrove living organism’s productivity. The aim of this research was to know the rate of mangrove leaf litter production and Nitrogen organic matter contribution in KKMB Tarakan. This research was conducted from October 2015 to January 2016. The average of mangrove leaf litter productivity were Avicennia alba 3.31 g/m2/day, Rhizopora apiculata 4.65 g/m2/day, Bruguiera parviflora 3.02 g/m2/day and Sonneratia alba 6.20 g/m2/day. The contribution of Nitrogen organic matter from mangrove leaf litter showed that were Avicennia alba 0.0987 g/m2/day, Rhizopora apiculata 0.0755 g/m2/day, Bruguiera parviflora 0.0977 g/m2/day and Sonneratia alba 0.3722 g/m2/day.Key words: litter productivity, litter decomposition, Nitrogen, KKMB

Abstrak Traditional products derived from the local wisdom of the Tarakan City coastal communities, namely in the form of cold protective skin powder from the sun which is made from mangrove raw Sonneratia alba. This product has potential to be developed to improve the economic value of local communities. They can compete with products other sunscreen products produced by industry (commercial) through cream preparation technology (sunscreen). This research was carried out for seven months (February-August 2018) in the Environmental Laboratory of FPIK UBT, Samarinda Pharmacy laboratory (AKFARSAM), and the UNMUL Mathematics and Biochemistry laboratory. The research procedure used maserasi method with 70% ethanol solvent followed by concentrating the extract using a rotary evaporator to obtain extracts of active ingredients and carried out qualitative phytochemical testing. The obtained active ingredient was is then formulated into a sunscreen cream preparation. The level of protection of mixed sunscreen cold powder and commercial cream were tested by using UV-VIS spectrophotometer to see the value of Sun Protection Factor (SPF) at absorbance of 290-320 nm. Qualitative phytochemical test showed that on S.alba contains of positive alkaloids, phenols, flavonoids, steroids, and tannins, and negative saponins. ANOVA statistical analysis (95%) based on the calculated F value <F table so that H1 is rejected and H0 is accepted, ie there is no difference in SPF values among sunscreen products tested in this study. Based on the category of sunscreen ability, it is known that the SPF value of the three different products tested in this study ranged from maximum to ultra categories with pH values ranging from 6.5-7.5.

The aims of this experiment are to know the effect of different medium to seaweed (G. verrucosa) cultured in vitro expressed total number of thallus. The experiment was conducted in the Biotechnology Laboratory of Research Institute for Coastal Aquaculture (RICA) Maros, South Sulawesi. The experiment was arranged in Completely Randomized Design (CRD) with 10 replications and 4 treatments of medium, those are: Sterile Sea Water / SSW 25 ppt, Prevasoli ES Medium / PES, Agrodyke / AG, and Conwy / CW. The last analyze was Least Significant Difference (LSD) to know the best medium to support total number of thallus. The result of the experiment shows that the highest total number of thallus was achieved by SWS (3,75). Keywords: Gracilaria verrucosa, tissue culture, thallus, medium
