Abstract

The aims of this study were: 1) to develop a biology practical guidance based on TGT cooperative models equipped with practical performance assessment instrument; 2) to determine the characteristics and suitability of products than have been developed; 3) to know it’s effectiveness in critical  thinking abilities on biology subject of students in class XI. Product development process in this study by following Borg and Gall’s Development. The result of the expert validation by 3 validators showed that each of  the average score in biology practical guidance based on TGT and practical performance assessment instrument were 4,41 and 4,48 and categorized as “very good”. Limited testing conducted on 10 students with high, medium, and low ability. The result of limited testing showed that each of percentage score average in biology practical guidance based on TGT and practical performance assessment instrument were 82,67% and 83,33% and categorized as “very good”. Field trial testing using a pre-test post-test control group design. The data of critical  thinking abilities are tested in this study consist of 5 indicator skills:  (1) formulate the problem; (2) recognize the existence of a logical relationship between issues; (3) collect and collate information; (4) asses and evaluate the statement of facts; (5) Make some conclude were needed. Data analysis were performed  using two independent sample test formula by SPSS 20 for windows. The analysis showed that the critical  thinking abilities for indicator 1, 2, 3, and 5 of experimental group different from the control group, while the fourth indicator not different. Data analysis equipped with calculation of the value of n-gain to determine  the effectiveness of learning by using product that have been developed. The result of the data analysis showed that the value of n-gain in experimental groupis equal to 0,71 and categorized as “high”.

Abstract

This study aims to determine the concentration of growth regulators BAP and 2,4-D which have the highest effect in stimulating the formation of shoots and roots melon plants (Cucumis melo) Mai 119 variety. The callus leaves of seedlings were used as a source explant. Completely randomized design (CRD) was used in this research. Media used on shoot induction was MS medium added with several concentration of BAP (0 mg/L, 1 mg/L, 2 mg/L, 3 mg/L) and 2,4-D (0 mg/L, 1 mg/L, 2 mg/L, 3 mg/L) either alone or in a combination of both. Media used in the formation of roots were MS medium added with 2,4-D at a several concentration of 0 mg/L, 1 mg/L, 2 mg/L, and 3 mg/L. Parameters measured were the time appearing of shoots, number of shoots, time appearing of roots, and the number of roots. Parameters measured were the time appearing of shoots and number of shoots using descriptive test. Anova followed by Tukey's test was used in the analyse time appearing of roots. In the parameter number of roots using Kruskal Wallis test followed by Mann-Whitney test. The results showed that there were differences in the effect of growth regulators on the shoots and roots formation. The fastest shoot induction was obtained in the media 3 mg/L BAP+1 mg/L 2,4-D with the highest number of shoots contained in the medium at a concentration of 3 mg/L BAP + 1 mg/L 2,4-D and 2 mg/L BAP+1 mg/L 2,4-D. The fastest root induction was obtained in media 0 mg/L 2,4-D, 1 mg/L 2,4-D, and 2 mg/L 2,4-D. The highest number of roots were obtained in media with 0 mg/L 2,4-D and 1 mg/L 2,4-D.

Abstract

Callus induction determines the successfulness of micropropagation through somatic embryogenesis. This research was conducted to determine the effect of 2,4-D and BAP on callus induction of peanut (Kelinci strain). Callus induction was done on MS medium, with the observed variables were of initiation time, diameter, texture and color callus. Statistical analysis showed that 2,4-D and BAP affect the initiation time, diameter, texture and color callus. The earliest initiation of callus formation was obtained from MS medium with the addition of 4 mg/L 2,4-D. The diameter of the largest callus obtained from MS medium with the addition of 3 mg/L 2,4-D + 1 mg/L BAP. Callus color were affected by the treatment of 2,4-D and BAP, while callus texture was not affected by these treatmen, in which compact callus texture was not affected by these treatment, in which compact callus texture was observed in all treatments.Keywords: Induction of callus, 2,4-D, BAP, peanuts

Abstract

Abstract : Strychnos ligustrina stem has been empirically used by the people of West Nusa Tenggara and Bali in the treatment of malaria, tooth ache and diarrhea, but there is no scientific data that supports it. This study aims to determine and prove the antibacterial activity of Strychnos ligustrina methanol extract to pathogenic bacteria in vitro and in vivo. This research is an experimental study conducted by measuring the inhibition zone (mm) growth of pathogenic bacteria, determining minimum inhibitory concentration (MIC) and minimum killing concentration (MKC) in vitro, and determining the percentage of antibacterial activity of methanol extract of S. ligustrina stem in vivo. The experiment was conducted using 4 groups of concentrations of S. ligustrina stem methanol extract in an in vitro study of 25, 50, 75, and 100% with ciprofloxacin as a positive control and aquadest as a negative control. In vivo studies experiments were carried out using 6 treatment groups of test animals male mice Balb / c (Mus musculus). The in vitro test results showed that methanol extract of S. ligustrina stems was able to inhibit the growth of pathogenic bacteria with medium categories of clinical isolates of Staphylococcus aureus and categories of weaks to Klebsiella pneumonia and Escherichia coli isolates. The minimum inhibitory concentration (MIC) for S. aureus and K. pneumonia bacteria isolates was at a concentration of 25% while for E. coli isolates at a concentration of 30%. The methanol extract of the S. ligustrina stem has no killing power against the pathogenic bacteria tested. Antibacterial activity in vivo was able to inhibit the growth of S. aureus pathogenic bacteria by 6.60% (at 25% concentration), 8.62% (at 50% concentration), and 17.31% (at 100% concentration), against K. pneumonia was 11.85% (at 25% concentration), 51.21% (at 50% concentration), and 65.92% (at 100% concentration), against E. coliat 19.18% (at concentration 25%), 29.98% (at 50% concentration), and 40.88% (at 100% concentration). Methanol extract of S. ligustrina stem proved to have antibacterial activity in vitro and in vivo. Key words: Srychnos ligustrina, pathogenic bacteria, antibacterial, in vitro, in Vivo. Abstrak : Strychnos ligustrina secara empiris  telah digunakan oleh masyarakat Nusa Tenggara Barat dan Bali dalam pengobatan penyakit malaria, sakit gigi, dan diare, tetapi belum ada data ilmiah yang mendukung. Penelitian ini bertujuan untuk menentukan dan membuktikan aktivitas antibakteri ekstrak metanol batang bidara laut terhadap bakteri patogen secara in vitrodan in vivo. Penelitian ini merupakan penelitian eksperimental yang dilakukan dengan mengukur zona hambat (mm) pertumbuhan bakteri patogen, menentukan konsentrasi hambat minimum (KHM) dan konsentrasi bunuh minimum (KBM) secara in vitro, serta menentukan persentase aktivitas antibakteri ekstrak metanol batang bidara laut secara in vivo. Percobaan dilakukan menggunakan 4 kelompok konsentrasi ekstrak metanol batang bidara laut pada penelitian in vitro yaitu 25, 50, 75, dan 100% dengan ciprofloxacin sebagai kontrol positif serta aquadest sebagai kontrol negatif. Pada penelitian in vivo percobaan dilakukan menggunakan 6 kelompok perlakuan hewan uji mencit jantan galur Balb/c (Mus musculus). Hasil uji in vitro menunjukkan ekstrak metanol batang bidara laut mampu menghambat pertumbuhan bakteri patogen dengan kategori sedang terhadap Staphylococcus aureus isolat klinis dan kategori lemah terhadap Klebsiella pneumonia dan Escherichia coli isolat klinis. Nilai konsentrasi hambat minimum (KHM) untuk isolat bakteri S. aureus dan K. pneumoniae adalah pada konsentrasi 25% sedangkan untuk isolat E. coli pada konsentrasi 30%. Ekstrak metanol batang bidara laut tidak memiliki daya bunuh terhadap bakteri patogen yang diuji. Aktivitas antibakteri secara in vivo mampu menghambat pertumbuhan bakteri patogen S. aureus sebesar 6,60% (pada konsentrasi 25%), 8,62% (pada konsentrasi 50%), dan 17,31% (pada konsentrasi 100%), terhadap K. pneumonia sebesar 11,85% (pada konsentrasi 25%), 51,21% (pada konsentrasi 50%), dan 65,92% (pada konsentrasi 100%),   terhadap E. coli sebesar 19,18% (pada konsentrasi 25%), 29,98% (pada konsentrasi 50%), dan 40,88% (pada konsentrasi 100%). Ekstrak metanol batang bidara laut terbukti memiliki aktivitas antibakteri secara in vitro dan in vivo. Kata kunci: Srychnos ligustrina, bakteri patogen, antibakteri, in vitro, in vivo

Abstract

The objective of this study was to isolate endophytic bacteria from bark of srikaya, analyzing antibacterial activity of endophytic bacteria in S. aureus, B. cereus and E. coli pathogenic bacteria, characterizing endophytic bacteria capable of inhibiting the growth of pathogenic bacteria and identification of endophytic bacteria able to inhibit the growth of pathogenic bacteria. Isolation of endophytic bacteria using TSA and NA media, bioassay on pathogen bacteria with concentration of 106 cells / ml with using Ø 6 mm wells and entering supernatant of 100 μl. Supernatant was obtained by growing endophytic bacteria in NB media shaken with shaker 150 cycles / min for 48 h at 32ᵒC then culture centrifuge at 5000 g for 30 min. Positive control using cyprofloxacin. Characterization is based on the nature of the colony, Gram paint, spore formation, and biochemical tests. The results of the study yielded 13 endophytic bacterial isolates and 4 endophytic isolates capable of inhibiting the growth of 8 pathogenic bacteria with sensitive criteria, 2 pathogenic bacteria with resistant criteria and 1 pathogen bacteria can not be inhibited its growth. Gram's paint results show that 4 endophytic isolates belong to Gram-positive, rod-shaped and spore forming cells. From the character possessed by the bacteria can be identified that the 4 bacteria endofit capable of inhibiting pathogenic bacteria are Bacillus brevis, Bacillus latesporus, Virgibacillus pantothenticus, and Bacillus circulansKeywords: Characterization, Endophytic Bacteria, Bark of Srikaya, Antibacterial

Abstract

This study aims to identify the types of endophytic bacteria on the bark of terap (Artocarpus elasticus) capable of inhibiting the growth of clinical isolate bacteria of Staphylococcus aureus, Escherichia coli, Salmonella typhi, and Pseudomonas aeruginosa. Stages of this study were (1) isolating the endophytic bacteria from the bark of terap plants (2) antibacterial test (inhibition test) against clinical isolate bacteria, (3) biochemical test upon endophytic isolate bacteria showing strong  inhibition effect, (4) moleculer identifcation upon endophytic bacteria isolate showing strong  inhibition effect. From this study, it was obtained 34 barks of terap isolates, and 12 isolates have inhibitory effect on S. aureus. Based on 16S rRNA   molecular identification, the isolate of endophytic bacteria of the barks were identified as Bacillus cereus, Bacillus subtilis and Bacillus amyloliquefaciens. Those  endophytic bacteria of Terap plants  are potentially  to be  developed as a the producer of antibacterial compounds.Keywords: Terap bark, Endophytic bacteria, Identification, Antibacteria

Abstract

This study aims to know the abilities of the teachers in applying authentical assessment in biology at grade X IPA Senior High School of Lombok Timur district. This study using descriptive method which getting data in the verbal only not in numbers. Getting samples at 8 Senior High Schools of Lombok Timur district which aplicate kurikulum 2013 they are SMAN 1 Masbagik, SMAN 2 Aikmel, SMAN 1 Sikur, SMAN 1 Terara, SMAN 2 Selong, SMAN 1 Keruak, SMAN 1 Montong Gading and SMAN 1 Sakra. To these teachers of biology were given instruments which asked how the authentical assessment reseach done in the schools and the result were presented in this study. The ability of teachers in applying authentical assessment were different from every questions there are attitude, knowledge and skill. Attitude they are observations, self assessment, assessment among students, journal and interview were answered by teachers with yes 65,62% , knowledge were answered by teachers by yes 88,75% and skill were answered yes 87,75%. Ability of teachers in applying authentical assessment were answered yes as many as 78,75% and the next verbally answered by teachers because of the problems in applying authentical assessments. Abstract

Abstrak: Kegiatan pengabdian pada masyarakat ini secara umum bertujuan memberi keterampilan kepada guru-guru Biologi SMA dalam memelihara hewan percobaan Drosophila, serta teknik-teknik dasar yang diperlukan dalam menggunakan hewan ini sebagai bahan percobaan pada pokok-pokok bahasan yang terkait dengan mekanisme pewarisan sifat. Kegiatan pelatihan ini dilaksanakan di SMAN 2 Labu Api dan diikuti oleh 23 orang peserta yang merupakan guru-guru Biologi SMA yang tergabung dalam MGMP Biologi Lombok Barat. Materi pelatihan meliputi Teknik Kultur Drosophila, Teknik Isolasi Betina Virgin, Teknik Menyilangkan mutan Drosophila, dan Teknik Mengatasi Kontaminasi Kultur. Hasil observasi menunjukkan bahwa dari 23 orang peserta pelatihan, 96% peserta belum pernah menggunakan Drosophila dalam praktikum, dan 100% peserta tidak memiliki keterampilan memelihara dan menggunakan Drosophila sebagai bahan praktikum di sekolah tempat mereka bertugas. Monitoring dan diskusi selama 10 hari pasca pelatihan menunjukkan bahwa hampir 74% peserta telah mampu melakukan dengan baik teknik-teknik dasar yang diperlukan dalam kultur Drosophila. Materi pelatihan tersulit yang dialami peserta adalah Teknik Mengatasi Kontaminasi Medium Kultur. Hingga 10 hari pasca pelatihan tidak seorang pun peserta melaporkan keberhasilan merekan dalam mengerjakan teknik ini. Meskipun demikian pelatihan ini berhasil karena sebagian besar materi pelatihan mampu dikerjakan dengan baik oleh mayoritas peserta pelatihan. Kata Kunci: pelatihan, kultur Drosophila

Abstract

Abstrak: Kurikulum Mata Pelajaran IPA SMP tahun 2017 di Kota Mataram  menuntut para guru IPA untuk bisa meningkatkan keterampilan dalam mengembangkan perencanaan  dan mengimplementasikannya secara baik. Salah satunya adalah guru IPA SMP bisa mengembangkan perangkat pembelajaran. Untuk dapat mengembangkan perangkat pembelajaran IPA sesuai dengan standar,  para guru IPA harus memiliki keterampilan dalam mengembangkan perangkat pembelajaran, Para guru masih terpaku dengan meodel yang disarankan kemnetrian terkait, sementara  banyak model pembelajaran berbasisi siswa aktif salah satunya model 5E (Engage, Explore, Explain, Extend dan Evaluate).  Oleh karena itu perlu dilakukan kegiatan untuk mengmbangkan keterampilan  bapak/ibu guru IPA anggota MGMP IPA dalam perangkat pembelajaran model 5E. Desain yang digunakan adalah post tes no control group design. Populasi adalah semua guru IPA anggota MGMP Kota Mataram, sampel yang digunakan sebanyak 40 guru. Pengumpulan data dilakukan dengan mengevaluasi produk berupa perangkat pembelajaran hasil kegiatan workshop. Data dianalisis deskriptif kuantitatif. Hasil menunjukan bahwa perangkat pembelajaran yang disusun oleh guru peserta (90%) sudah baik dalam merencanakan langkah-langkah 5E. Tetapi dalam kaitannya dengan tagihan dan pengetahuan sebagian besar perangkat pembelajaran kurang mencakup keluasan dan kedalaman materi yang ditagih oleh kompetensi dasar pengetahuan dan keterampilan. Dengan demikian walaupun langkah-langkah pembelajaran berbasis siswa aktif dan memenuhi tagihan kurikulum tetapi tingkat asesmen belum memenuhi tagihan. Berdasarkan hasil penjelasan di atas dapat disimpulkan bahwa  90% bapak ibu guru IPA dapat menyusun perangkat pembelajaran cukup baik khususnya terkait sintaks  5E. Kata Kunci : pembelajaran IPA, model 5E, kajian kritis
