In Vitro Selection of Peanut Somatic Embryos on Medium Containing Culture Filtrate of Sclerotium rolfsii and Plantlet Regeneration

Abstract

Attempts to identify somaclonal variants of peanut with resistance to Sclerotium stem rot disease due to infection of S. rolfsii were conducted. The objectives of this study were to develop in vitro selection method using culture filtrates of S. rolfsii, identify culture filtrate-insensitive somatic embryo (SE) of peanut after in vitro selection and regenerate peanut R0 lines originated from culture filtrate-insensitive SE. To achieve these objectives, peanut embryogenic tissues were cultured on selective medium containing various concentrations of S. rolfsii culture filtrates and sublethal concentration of the filtrates. Medium containing sublethal level of S. rolfsii culture filtrates was used to identify culture filtrate-insensitive SE of peanut. Subsequently, the selected SEs were germinated, plantlets were regenerated and preliminary tested against S. rolfsii. Results of the experiments showed that addition of S. rolfsii culture filtrates into medium for inducing peanut somatic embryos drastically reduced their growth and proliferation. S. rolfsii culture filtrates at 10% concentration has significantly reduced the number of proliferated SE per explant. However, sublethal level was achieved at 30% of culture filtrates concentration. Responses of five peanut cultivars against 30% of culture filtrates were similar, indicating they were similar in their susceptibility against S. rolfsii. A number of culture filtrate-insensitive SE were identified after culturing 1500 clumps of embryogenic tissue of peanut cv. Kelinci for three consecutive passages on medium containing 30% of culture filtrates. Germination of selected SE and regeneration of plantlet from culture filtrate-insensitive SE resulted in 50 peanut R0 lines. These lines have been grown in the plastic house and produced normal seeds for further evaluation. Results of S. rolfsii inoculation indicated the existence of chimera for insensitivity against S. rolfsii.

Abstract

Penelitian ini dimaksudkan untuk memberikan suata cara sederhana dalam mengendalikan arus start motor induksi 3-fasa. Penelitian ini dibantu oleh program komputer Matlab untuk menganalisa kinerja motor selama proses start dan operasi. Motor yang digunakan pada penelitian ini adalah motoro induksi 3-fasa, 1500 W, 380 V, 4 kutup, 50 Hz and 1400 rpm. Dari hasil penelitian yang telah dilakukan menunjukkan bahwa untuk mengendalikan arus start sebaiknya pada awal start motor diberi tegangan 26.32% dari tegangan nominalnya,kemudian dinaikan secara bertahap menjadi 69% and 100% dari tegangan nominalnya.

Abstract

ABSTRAK Penelitian ini bertujuan untuk menemukan sikap siswa terhadap penilaian yang diberikan oleh dosen dalam mata kuliah translation. Penelitian ini merupakan penelitian kualitatif, responden yang dipilih dalam penelitian ini adalah mahasiswa tahun ketiga pada tahun akademik 2011/2012 yang terdiri dari 12 kelas. Teknik pengambilan responden yang digunakan adalah Purposive Random Sampling. Jumlah responden yang diambil pada penelitian ini berjumlah 120 mahasiswa dan beberapa dosen yang sudah pernah mengajar mata kuliah translation. Instrumen dari penelitian ini adalah wawancara dan angket. Berdasarkan data yang didapat selama peneliti melakukan wawancara dengan beberapa dosen yang sudah pernah mengajar mata kuliah translation dapat diketahui bahwa tipe-tipe penilaian yang mereka berikan meliputi Free Response Items, True False Items, Matching,  Portfolio, Translation Production Test (Essay) dan Multiple Choice. Kemudian setelah peneliti menganalisa hasil jawaban mahasiswa terhadap angket, peneliti menemukan bahwa persentase mahasiswa yang mempunyai sikap positif terhadap penilaian yang diberikan dosen dalam mata kuliah translation adalah 36,80% dan persentase mahasiswa yang mempunyai sikap negatif adalah 63,20%, sehingga disimpulkan bahwa kebanyakan  mahasiswa masih salah dalam menyikapi penilaian dalam mata kuliah translation. Hal ini dikarenakan mereka sering merasa takut, cemas, khawatir, tidak percaya diri, bingung serta kurang termotivasi dalam melaksanakan  latihan yang diberikan dosen.

Abstract

Strategi global pemberantasan vektor DBD salah satunya adalah pengendalian nyamuk secara selektif dan terpadu yang melibatkan partisipasi masyarakat. Laporan cakupan Angka Bebas Jentik (ABJ) yang menjadi indikator pengendalian DBD merupakan output peran serta aktif kader jumantik. Tujuan kegiatan adalah penyuluhan pada ibu-ibu rumah tangga di Serdang daerah Kemayoran tentang jumantik mandiri sebagai upaya meningkatkan pengetahuan warga dan berikutnya diharapkan dapat menjadi jumantik mandiri minimal di rumah tangganya masing-masing.Jumlah sasaran kegiatan sebanyak 28 orang ibu rumah tangga di kelurahan Serdang, Kemayoran. Target kegiatan setelah penyuluhan adalah pengetahuan peserta meningkat di atas 70%. Bahan presentasi disusun menyesuaikan sasaran kegiatan penyuluhan yaitu Ibu Rumah Tangga. Lebih banyak materi disampaikan dalam bentuk gambar. Kegiatan penyuluhan diselingi dengan ice breaking untuk menyegarkan suasana penyuluhan. Sosialisasi dan advokasi dilaksanakan pada tanggal 12 Januari 2018 pelaksanaan penyuluhan pada hari Rabu, 17 Januari 2018 di Sekretariat RPTRA RW 1 kelurahan Serdang. Hasil kegiatan menunjukkan bahwa nilai benar untuk jawaban soal post test penyuluhan yang dibawah 70% ada lima soal dari total 16 soal. Hasil uji T berpasangan terhadap nilai pre dan post tes didapatkan nilai p = 0,087 (> 0,05) artinya tidak terdapat perbendaan signifikan antara nilai sebelum dan sesudah penyuluhan. Saran yang bisa diberikan untuk soal post test masih dibawah 70% yaitu perlu dilakukan evaluasi materi, soal pre post test dan cara menyampaikan materinya.

Abstract

Effectiveness of the application of organic matter and Trichoderma viride from suppresive soil to control fusarium wilt on banana plant. Fusarium wilt disease caused by Fusarium oxysporum f.sp. cubense is one of the problems in banana production. This research was aimed to evaluate the effect of  Trichoderma viride isolated from suppresive soil and organic matter on controlling fusarium wilt on banana plant. The efficcacy trial consisted of 12 treatments and four replications within each treatment. Each experimental unit consisted of 10 plants. Organic matters used were rice straw, cassava peel, and compost. Treatments were arranged in a randomized complete block design. Data obtained were analyzed by analysis of variance followed by orthogonal analysis (P≤0.05). Application of T. viride suppressed disease incident up to 65%. The highest disease incident occurred in plants treated without T. viride and organic matter and in plants treated with organic matter only (78.33%). Disease incidence in plants treated with T.viride was lower than those treated with the combination of T. viride and organic matter. Different application times (in nursery or soil medium) did not significantly affect fusarium wilt incidence. The type of organic matter did not significantly affect the incidence and severity of fusarium wilt.

Abstract

Tuberculosis (TB) is a contagious disease caused by the group of Mycobacterium bacteria, Mycobacterium tuberculosis. There are six countries with the highest number of TB incidents in the world, including Indonesia with a prevalence of 1,020 per 100,000 inhabitants located second after India. Based on the results of monitoring and evaluation of Tuberculosis External Monitoring Mission Team in 2008, the success of treatment is still low. Many factors that influence the success of tuberculosis treatment one of them is the level of knowledge. This study aims to see and prove whether there is a relationship of knowledge level to the success of treatment in patients with tuberculosis and done cross-sectional. The result of the research shows that there are 45 respondents (80,4%) and the knowledge level of the respondent has good knowledge, 22 people (39,3%) from 56 samples. From the results of statistical tests using Chi-Square test obtained P value> 0.05 is 0.069.

Abstract

Optimization of planlets growth during acclimatization can be done by using plant growth regulator. This research was aimed to know the effect of two foliar fertilizer and benzyladenin on the growth of Phalaenopsis hybrid orchid planlets during acclimatization stage. The treatment was Growmore and Bayfolan as the foliar fertilizer with the various concentrate of benzyladenin (BA) at BA 0 mg/l, BA 15 mg/l and BA 30 mg/l. The results showed that Growmore foliar fertilizer had the better response of plant height and fresh weight plant compared to Bayfolan foliar fertilizer. BA 30 mg/l was increased the leaf number, and BA 15 mg/l and 30 mg/l was decreased root length and fresh weight plant. There was no interaction between foliar fertilizer and benzyladenin on the growth of Phalaenopsis hybrid orchid planlets

Abstract

Forest area of Balik Bukit Resort allocated as regions of natural preservation.  This area hasmany potential/natural resources that could be developed. This area is located at BukitBarisan Selatan National Park which have diverse types of flora, including various types oforchids. Approximately there were 154 typeof orchids in this area(Indonesia Institute ofSciences, 2011). This research aims were to know the population and distribution patterns ofEria spp orchids.This research was conducted in August 2015. The method used is the doubleplots systematically method with sampling intensity (SI) 0.1%. The size of each swath of 20 mx 20 m and the total area of the Balik Bukit Resort was 3,560 ha, so that the total number ofsample plots were 90 units. The observed variables were types of Orchid, Orchid populationsand patterns of spread of Eria spp. Based on the results of the study, there were 10 different types of Eria spp.. Eria iridifolia found most of 22.50 individuals/ha compared to other typesof orchids Eria. Followed by the type of E. erecta of 14.72 individuals/ha E. flavescens andtype of 8.33 individuals/ha. Whereas other types is E. discolor, E. javanica, E. junghunii, E.mucronata, E. oblitterata, E. retusa, E. robusta has a density of 1 — 4 individuals/ha.Distribution pattern of the orchids E. discolor, E. erecta, E. flavescens, E. iridifolia, E.javanica, E. mucronata, E. obletterata, and E. retusa enter into the category of a huddle.While the orchids E. junghunii belonging to the category of uniform and the orchids E.robusta into random categories.Keywords: distribution pattern, Eria spp. orchids, population

Abstract

 Dendrobium (Orchidaceae) is one of the most popular orchid genera consisting more than 2000 species, which is widely known for its long vaselife, various, shapes and colors, high frequency of flowering per year and long and flexible stalks, so it is easily assembled for flower arrangements. The objectives of this research was to study the effects of different host trees and position of plant attachment at the tree on growth of Dendrobium seedlings. This experiment was conducted with treatments arranged factorally (3 X 2) in a completely randomized design with three replicates. The first factor was the species of host three consisted of Akasia (Acacia auriculiformis), Cemara Bundel (Cupressus retusa) and Kerai Payung (Filicium decipiens) and the second factor was position of plant attachment on the tree, which consisted of attachment at the bottom ( 0,5 m above ground ) and the upper was ( 1,5 m above ground ). Each experimental unit consisted of two Dendrobium seedlings. The results showed that in general both host trees and position of attachment did not affect number of shoots, number of leaves, plant height and the longest root length. however, different host trees and its interaction with the position of attachment affected the number of roots, resulted in the highest number of roots obtained in Kerai Payung (Fillicium decipiens) at 1,5 m above ground.Keywords: Acacia auriculiformis, Cupressus retusa, host tree, Filicium decipiens, orchid.
