Abstract

The objectives of this study was to observe antibacterial activities of water lily seed, against diarrhea-causing pathogenic bacteria as well as lactic acid bacteria; phytochemistry components in water lily seed and to evaluate each component’s activities against pathogen bacteria. Extraction of antibacterial components in the seed was done by fractional extraction methods using solvent based on its polar level, i.e. hexane, ethyl acetate and ethanol. The activities of each extract was tested by using diarrhea-causing bacteria, Enteropathogenic Escherichia coli K.1.1 (EPEC K1.1) and S. typhimurium with agar well diffusion method. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values were calculated with plate counting. Qualitative phytochemical tests were performed on all extracts. Fractionation was performed on extract with the largest antimicrobial activity by using thin-layer chromatography. The activities of each fraction were tested qualitatively by bio-autography method on thin layer chromatographic plates. The water lily seed had an antibacterial activity against EPEC K.1.1 and Salmonella typhimurium, especially in ethyl acetate extract. Ethanol extract had the same, yet lower activity. Ethyl acetate and ethanol extract of the seed did not show inhibition against the growth of lactic acid bacteria (Lactobacillus sp) and Bifidobacterium bifidum. The MIC and MBC values of the ethyl acetate extract on EPEC K1.1 were 0.89 (mg/mL) and 1.33 (mg/mL), respectively, while similar values of that on S. Typhimurium were 1.11 (mg/mL) and 1.33 (mg/mL¬), respectively. Phytochemistry components within ethyl acetate extract were alkaloids, flavonoids, tannins, glycosides, saponins, and triterpenoids. All fractions in the ethyl acetate extract had antimicrobial activities against EPEC K.1.1 and S. Typhimurium. These fractions were thought to inhibit the growth of the test-microbes by synergic action of each component.Key words: water lily seed, antibacterial, ethyl acetate extract

Abstract

The  aims  of  this  experiment  were  to    identify  and  characterize of sediment  product from cattle rumen liquor as a source of amino acids, minerals and vitamins. The sediment were obtained as pellets upon centrifugation of rumen liquor at 10,000 g for 10 minutes for collection of supernatant. The sediment were evaluated for amino  acids, minerals and vitamins composition and chararacterized for the pH,  solubility of dry  matter,  specific  density,  bulk  and  compacted  bulk  densities  and  angle  of  response.  Result  of  the experiment showed that sediment contained higher minerals:  Na, K and Fe compared with the commercial premix, but  lower in B-vitamins and  amino acids. The composition of mineral Na,  K, and Fe from rumen liquor of local cattle was 13.47%, 7.73 % and 14.52 %,  while Na, K, and Fe from  rumen liquor of  imported cattle was 18.40%, 10.25%, and 14.52% respectively. The sediment had pH range from 10.01-10.03, the dry matter solubility   was 35.5%  up to 39.1%. The  sediment from imported cattle had  higher specific density, bulk  and  compacted  bulk  densities  and  angle  of  response  than  that  of  local  cattle. It  is  concluded  that sediment from cattle rumen liquor contained high  Na, K  and Fe, low amino acids and B-vitamin,  high pH and low solubility.

Abstract

In vitro analysis of oligosaccharide from extract rumbia fruit (Metroxylon sago Rottb.) as prebioticABSTRACT. Despite a range of commercially available oligosaccharides there is plenty of room to develop new, functionally enhanced prebiotics. current generation of oligosaccharides was not rationally developed. better understanding of factors determining the prebiotic activity of a particular oligosaccharide. Despite the range of commercially available oligosaccharides mixtures (mainly fructo and galacto-oligosaccharides), very few studies are focused on the mechanisms behind the prebiotic activity of particular oligosaccharides. Probably this lack is due to the unavailability of well characterized oligosaccharide fractions for prebiotic function assessment. The objectives of this research were to asses the ability of lactic acid bacteria in fermentation of oligosaccharide and as prebiotic (in vitro). Material used was oligosaccharide of purified rumbia fruit extract. Analysis of oligosaccharide as prebiotic was conducted in vitro using lactic acid bacteria. The lactic acid bacteria consisted Bifidobacterium bifidum, Bifidobacterium animalis, Lactobacillus bulgaricus and Lactobacillus casei Rhamnosus. The growth media for bacteria was a liquid MRS basic medium where glucose was substituted by oligosaccharide of purified rumbia fruit extract. Incubation was in aerob for Lactobacillus and anaerob for Bifidobacterium in incubator 37oC. The lactic acid bacteria was calculated 24-48 hours during incubation periode. The variables observed were: oligosaccharide component, ability of lactic acid bacteria in fermentation of oligosaccharide, and growth of lactic acid bacteria (Lactobacillus and Bifidobacterium). The result showed that the oligosaccharide component from extract rumbia fruit consisted of: sucrose, stacchiose, and raffinose. The result showed that the oligosaccharide extract rumbia fruit was significantly (P<0.05) the growth of lactic acid bacteria (Lactobacillus and Bifidobacterium) and fermentation of oligosaccharide. It is concluded that oligosaccharide of rumbia fruit extract could be used as prebiotic.

Abstract

This article is a dynamics synthesis of milk production data of dairy goat fed diet containingconcentrate supplemented with PUFA (polyunsaturated fatty acid), yeast and curcuma. Twenty (20) dairy goats of Ettawa crossbred in late lactation phage were grouped into fiveof four goats to receive five treatments in in Randomized Complete Block design. Thetreatments were no supplement (PD0), 3 tablets of Asifit (PDA), 5g/d yeast (PDY), 20g/dcurcuma (PDC), and a mix of 5g/d yeast and 20g/d curcuma (PDM). The concentratecontained of PUFA sources (roasted ground corn, roasted soy bean meal, and corn oil),soybean by-product; roughage provided was King Grass. Milk yield was recorded daily, pre,treatment, and post then calculated all the way from pre to post treatment. Milk yield werehigher significantly (P&lt;0.05) in PDA (538.7 g/d) and PDY (522.3g/d) compared to PD0(336.3g/d), PDC (322.3g/d) and PDM (392.0g/d). Average temperature and relativehumidity in the farm were 26.94 ? 2.280 C and 78.83 ? 7.34 %. Yeast supplementation eithersolely or in combination with curcuma is considered better in milk production dynamics inpre, treatment, post.

Abstract

To study the effect of utilization of passion fruit hulls (Passiflora edulis Sims f. edulis Deg) as feed component on consumption, digestion and nitrogen retention in kacang goats, a trial was conducted using 20 young kacang goats (average initial body weight 23.73±2.16 kg). The experiment was arranged in completely randomized design consisting of 4 diets and 5 replications. Animal were randomly allocated into 4 diets (0, 15, 30, 45% of passion fruit hulls). Each diet contains 2550 Kcal/kg metabolism energy and 14% crude protein. The feeding level was set at 3.8% of body weight based on dry matter. The results of the experiment showed that average dry matter intake, nutrient digestibility, and nitrogen retention were not affected by level of passion fruit hulls (P&gt;0.05), although nutrient digestibility and nitrogen retention tended to decrease with the increasing level of passion fruit hulls. Abstract

Lactic acid bacteria (LAB) regarded as safe microorganisms; they can naturally live in gastrointestinal tract, so appropriately used as a probiotic for chicken. This study aimed to select six isolates of LAB (E1223, E3, E4, E5, E7, and E8) to obtain the isolates potentially as probiotic candidate for chicken. The six isolates were derived from spontaneous fermented corn obtained from Laboratory of Animal Biotechnology and Biomedical, PPSHB, Bogor Agricultural University, Indonesia. LAB isolates were tested their susceptibility to antibiotics (bambermycin, erythromycin, chloramphenicol, and tetracycline) then were examined in vitro for their tolerance to gastrointestinal pH (2, 3, 4, and 7.2) and 0.5% bile salt condition, antimicrobial activity against Salmonella enteritidis and Enterococcus casseliflavus, and ability to adhere to chicken ileal cells. The results showed the isolates E5, E7, and E8 were sensitive to tetracycline and chloramphenicol, they could survive at pH 2, 3, 4, and 7.2, could survive at 0.5% bile salts, produced antimicrobial activity, and able to adhere to ileal cells (9.40±0.00 Log CFU/cm2 of E8) and were significantly (P&lt;0.05) higher than those of control (5.30±0.14 Log CFU/cm2). In conclusion, this study showed that isolate E8 had better potential compared to isolates E5 and E7 in most in vitro assays as a probiotic candidate for chicken. E5, E7, and E8 were closely related with Pediococcus pentosaceus based on 16S rRNA gene. Key words: LAB, probiotic, chicken, in vitro

Abstract

The purpose of this study was to evaluate the effect of a complete ration silage made of cassava biomass  nitrogen retention, and metabolizable energy male ducks. Control diet consisted of corn, rice bran, coconut meal, soybean meal, vegetable oil, fish meal and premix. While the treatment of silage ration of cassava based (BBS) consisted of leaves, peel, and tubers of cassava, as well as a mix of fish meal, vegetable oil, premix, DL-methionine and L-lysine. The experimental design used was completely randomized design with 5 treatments and 4 replicates ie S0 (100% control diet), S25 (75% control diet + 25% BBS silage), S50 (50% control diet + 50% BBS silage), S75 (25% control diet + 75% BBS silage), and S100 (100% BBS silage ration). Measurement of metabolizable energy and nitrogen retention was conducted using 25 male ducks aged 10 weeks maintained in metabolic cages and adapted to experimental diets for seven days. Ducks were fasted for 24 hours before the experimental diets were offered. Metabolizable energy and nitrogen retention were measured by the method of Sibbald (1989). The results showed that a decline in nitrogen retention and, metabolizable energy along with the increasing level of BBS silage in the rations. The conclusion of this study was the use of up to 75% BBS silage generated metabolizable energy and nitrogen retention that do not differ from the control ration.Key words: cassava silage, complete ration, male duck, metabolizable energy, nitrogen retention

Abstract

Effect of prebiotic oligosaccharide extract rumbia fruit (Metroxylon sago Rottb.) in the ration on broiler performanceABSTRACT. Prebiotic oligosaccharides are thought to provide beneficial effects in the gastrointestinal tract of humans and animals by stimulating growth of selected members of the intestinal microflora. Prebiotic oligosaccharides are defined as nondigestible food ingredients that provide beneficial effects to the host by stimulating the growth of selected microbial members of the gastrointestinal tract. Among the colonic bacteria capable of metabolizing prebiotic oligosaccharides and whose growth is stimulated are species of Lactobacillus and Bifidobacterium. Prebiotic oligosaccharides can be produced in transgli-cosylation reactions catalyzed by glycosidases. Glycosidases from different biological sources have specific ability to catalyze the formation of oligosaccharides with particular chain lengths (usually DP < 7) and predominant glycosidic linkages. Oligosaccharide used this research was purified rumbia fruit extract as prebiotic for feed additive in the ration on broiler. The objectives of this research were to study the performance of broiler given of prebiotic oligosaccharide extract rumbia fruit (metroxylon sago Rottb.) in the ration. Two hundred day-old chicks of broiler were divided into three dietary treatments and four replications. Ration used was consisted of: R1 = basal ration (control), R2 = basal ration + 0,4% oligosaccharide extract rumbia fruit, and R3 = basal ration + 0,4% frukto-oligosaccharide (FOS). The variables observed were: feed consumption, body weight, feed conversion ratio, mortality, and production index. The result showed that the performances of the broiler supplemented prebiotic oligosaccharide extract rumbia fruit (R2) was significantly (P < 0.05) differences between of feed consumption, body weight, and mortality. It is concluded that of prebiotic oligosaccharide extract rumbia fruit were able to decrease the mortality and feed consumption at six week of age.
