Abstract

Kapok (Ceiba pentandra Gaertn.) is one of the plants which has many benefits for the community. This plant is used as a retaining soil from erosion, plant shade, mattress filler material and animal feed. Kapok seed is currently being developed as a herbal contraceptive medicine. The aim of this research is to determine the activity of kapok seed extracts (Ceiba pentandra Gaertn.) on the mating behavior of male rats(Rattus norvegicus).The design used was completely randomized design (CRD) with 4 groups and 8 replications were P0 (distilled water and tween 80), P1 (0,07 mg/kg body weight kapok seed extract), P2 (0,12 mg/kg body weight kapok seed extract), P3 (0,24 mg/kg body weight kapok seed extract). Kapok seed extract treatment is done every day for 33 days, whereas  mating behavior was observed once in 3 days. The parameters observed to assess their mating behavior were the quantity of kissing the vagina, mounting and intromission. The results showed that the kapok seed extract significantly provide a real impact on the mating behavior of male rats which is indicated by a decrease quantity of the kissing vagina, the quantity of mounting andthe quantity of intromission of male rats treated group.

Abstract

This study was conducted to investigate the effect of monosodium glutamate (MSG) on spermatogenesis of male experimental mice.using 32 twelve-week old mice. They were divided randomly into 4 groups, each group consisted of 8 mice. One group was used as control, three groups were used as treatment, which received 1.5 mg/g body weight/day, 3 mg/g body weight/day and 4.5 mg/g body weight/day of MSG solution in destilled water for 35 days. The result showed that MSG treatment led to disturbance on spermatogenesis by reduction in some of the parameter studied. There was a significant (p < 0.05) decrease in the number of pachytene spermatocyte and spermatid 15, where as the decrease in spermatogonia was insignificant (p>0.05).

Abstract

Fish cultures in rice fields or areas that use water from a pesticide-polluted irrigation or river, are particularly vulnerable to the negative impact of pesticides. This study aims to assess the effect of organophosphates on growth, ovarian histopathology and fecundity of red tilapia (Oreochromis niloticus). After acclimatization, 160 fishes (fingerlink stage) were kept in four ponds sized 2x1,5x0,8 m each. This study used a complete randomized block design and organophosphate administration through the feed. The study used four groups: control, and variations in dose of 2 ml (D1), 6 ml (D2), and 10 ml (D3) of pesticide per 100 grams of pellet, which is given in the morning (treatment) and in the evening (regular feed). Data were taken at week 3, 6, and 9, then the organophosphate treatments were stopped, and the last data were taken at week-18. Statistical analysis was performed by Anova and Duncan's Multiple Range Test. The results showed that the organophosphate pesticide inhibits the growth by reducing fish length and weight, reducing ovarian weight, increasing the number of oocyte atresia, damaging structure of the ovarian histology, and decreasing fecundity by reducing the number of egg production in the ovarian of red tilapia.

Abstract

The aimed of this study was to determine the effect of feed containing various concentrations of Calliandra calothyrsus Meissn. leaf meal mixed with pineapple peels and concentrate feed on histopathology and the activity of liver of local rabbit (Lepus sp). This study used a Post-Test Only Control Group Design, using 24 local male rabbits divided into 4 groups of feed treatment namely P0 = concentrate feed as control; P1 = mixture of 15% of calliandra leaf meal (CLM), 30% of pineapple peels (PP) and 55% of commercial feed (CF); P2 = 30% of CLM, 30% of PP and 40% of CF; and P3 = 45% of CLM, 30% of PP and 25% of CF). The treatments were given for 60 days and used 6 replicates for each group. The data were analyzed statistically by SPSS program version 22. The research results showed the increase of calliandra leaf meal levels in concentrated feed which were added 30% of pineapple peels still caused liver damage namely fatty degeneration, necrosis and congestion. It increased the ALT level, but did not affect the AST level in local rabbit serum.

Abstract

Breadfruit (Artocarpus communis Forst.) leaves have a high antioxidant content. This research aims to know the effect of breadfruit leaf extracts on blood sugar levels and the amount of sperm in rat. This study used a Completely Randomized Design (CRD) using 30 rats aged 2-3 months and weighing 150-200 grams. Male rats were divided into six groups, i.e. negative control group (K1) without treatment, groups K2, P1, P2, P3 and P4 are given then NaCl 0.9%, breadfruit leaf extracts 50 mg/kg, 100 mg/kg, 200 mg/kg body weight, and glibenclamide 1 mg/kg body weight respectively. The treatments were given every day for 21 days. Blood sugar levels were measured before and after aloksan administration, day 7, 14, and 21 after treatment as well as counting the number of sperm after 21 days. The data obtained were analyzed with ANOVA and Duncan Multiple Range Test (DMRT), using the program of SPSS For Windows version 22. Results of the study showed that breadfruit (Artocarpus communis Forst.) leaf extracts could lowered blood glucose levels and the most effective concentration was 100 mg/kg body weight of breadfruit leaf extracts given for 21 days with the effectiveness of 66,77%. Breadfruit leaf extracts did not inhibit spermatogenesis. So the amount of rat sperm did not different significantly among treatment. Keywords: blood sugar levels, breadfruit, rat, sperm count

Abstract

This research was conducted to determine the effect of Leucaena leucocephala leaf extract on the development of ovarian follicle cells of mice (Mus musculus). Twenty four female mice, 10 weeks old, were divided into 4 groups. The control group (P0) were given 0.9% NaCl and the treatment group P1, P2 and P3 were given 0.5, 1 and 1.5 g/ kg bw of Leucaena leaf extract, respectively. The treatments were administered daily for 15 days by gavage. After the treatment ended, all mice were dissected to collect the ovaries. Histological preparation of the ovaries used paraffin method and Hematoxylin-Eosin staining. Observations were made on the number of primary, secondary, and tertiary oocytes, and also De Graaf follicles and corpus luteum. Data were statistically analyzed using One Way ANOVA method. The results showed there were no significantly differences (P&gt;0.05) on the number of primary, secondary, and tertiary oocytes, as well as De Graaf follicles and corpus luteum among the control and treatment groups. The mimosine content of Leucaena leaf extract was 0.87% (less than 1%) so it could not inhibit the secretion of Follicle Stimulating hormone by the pituitary gland. The doses of Leucaena leaf extract used in this research did not significantly affect the follicles development of mice ovaries. In this study, the mimosine content of Leucaena leaf extract decreased 91.8%.

Abstract

Leucaena leucocephala provides the highest amount of proteins compared to other greens. However, its use is restricted due to the content of mimosine, an antinutrition non-protein amino acid known to be toxic to animals. This study aimed to observe the effect of administration of more than 10% of processed Leucaena leucocephala leaf meal to the level of T3, T4 and the thyroid histopathology. In this study, Leucaena leucocephala leaves were soaked in distilled water for 12 hours. The processed of Leucaena leucocephala leaf meal was made into pellets containing 0%, 7.5%, 15%, and 22.5% leaf meal of total feeds respectively, and was fed to the rats during pregnancy and lactation. The level of triiodothyronine (T3) and thyroxine (T4), and the thyroid histological features were the parameters observed. The collected data were statistically analyzed by SPSS 20.0 for Windows using One-way ANOVA followed by Duncan Multiple Range Test to observe any significant difference among the 4 treatment groups (?=5%). Meanwhile, the presence of hyperplastic cells and follicle lumens filled with vesicles and colloids was descriptively analyzed by means of available literatures. The analysis of T3 and T4 level revealed that there was no any significant difference between the control and treatment groups. The epithelial cells of thyroid follicles in the treatment groups of P1, P2 and P3 showed hyperplasia and were detached from their respective basal membranes.

Abstract

Lamtoro merupakan salah satu tanaman yang memiliki kandungan protein yang cukup tinggi yaitu berkisar antara 25-35%. Namun, pemanfaatan lamtoro menjadi terbatas karena mengandung zat antinutrisi seperti mimosin. Kandungan mimosin dapat diturunkan melalui beberapa metode salah satunya adalah melalui perendaman dalam air. Penelitian ini bertujuan untuk mengetahui pengaruh dari pemberian tepung daun lamoro hasil perendaman (TDLP) terhadap histologi testis tikus jantan. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) yang terdiri dari empat kelompok dengan masing-masing delapan ulangan. Perlakuan berupa pemberian tepung daun lamtoro hasil perendaman yang dicampur dengan pelet komersial dengan aras 100% pakan komersial (tanpa TDLP) sebagai kontrol (P0), 92,5% pakan komersial + 7,5% TDLP sebagai perlakuan 1 (P1), 85% pakan komersial + 15% TDLP sebagai perlakuan 2 (P2) dan 77,5% pakan komersial + 22,5% TDLP sebagai perlakuan 3 (P3). Perlakuan diberikan pada tikus jantan selama 30 hari. Variabel yang diamati pada penelitian ini adalah jumlah sel spermatogenik. Data hasil penelitian diolah menggunakan program statistik komputer (SPSS 16.0 for Windows) dengan menggunakan uji One Way Anova. Hasil penelitian menunjukkan bahwa penambahan TDLP dalam ransum tidak berpengaruh nyata (P&gt;0,05) terhadap jumlah sel spermatogenik. Hal ini menunjukkan bahwa penambahan TDLP hingga aras 22,5% tidak menurunkan jumlah sel spermatogenik.

Abstract

Indonesian local chicken is one kind of germplasm that has been utilized by the community for various needs, such as consumption, ceremonial, collection, and cockfighting. To maintain its sustainability, an inventory of genetic diversity can be made using molecular markers. This study aimed to determine the genetic diversity and relationship between four Indonesian local chickens: ayam kampung, ayam ketawa, ayam kate lokal, and ayam bekisar kangean, based on RAPD marker. DNA was extracted from blood samples of 12 local chickens consisting of 3 ayam kampung, 3 ayam ketawa, 3 ayam kate lokal, and 3 ayam bekisar kangean using Genetic DNA Mini Kit (Tissue) Geneaid. The PCR-RAPD analysis was performed using 6 primers, OPA 04, OPB 08, OPB 12, OPC 16, OPH 01, and OPH 03. The product of PCR-RAPD was electrophoresed using 1,5% agarose in 1x TAE buffer. The results showed that the similarity value of 12 local chickens using RAPD ranged from 0% to 100%. The relationship between 12 local chickens &nbsp;using RAPD marker indicated that the chickens in the same morphology were not clustered in the same group. This suggests that the same group of chickens phenotypically does not necessarily have the same genotype. &nbsp; Keywords: Local Chicken, PCR-RAPD

Abstract

The research was conducted to identify the effect of rhodamine B on estrous cycle on female mice (Mus musculus L.). Twenty-four female mice were used, and randomly divided into four groups.  Rhodamine B was given orally (gavage) with a dose of  0 ppm (control), 150 ppm, 300 ppm, and 600 ppm in two estrous cycles.  Treatments were started when mice in estrus phases. The time between first and the next estrus phase were observed from vaginal smear preparation. Data obtained were analysed with One Way Anova.  If the differences were appeared between treatments, then the LSD test was performed. The results showed that rhodamine B was significantly increased the time of estrous cycle (P&lt;0.05).
