Soy Germed Protein Plus Zn as an Inducer Insulin Secretion on Type-2 Diabetes Mellitus

Abstract

Hyperglicemic induces pancreatic cells to produce inadequate insulin. However, previous studies revealed that soy protein induce pancreatic cells to secrete insulin. Hence, this study was aimed to investigate effect of soy germed protein on the insulin and blood glucose level of type-2 diabetes mellitus with Zn enrichment. The research involved twenty four women that characterized with having more blood glucose level than normal, body mass index more than twenty three kg/m2, and age more than fourty years old. They were divided into three groups randomly, eight woman for each group. The first, second and third group were treated respectively with milk containing soy germed protein plus Zn, this product without Zn, and placebo, all for two months. Blood samples were taken at baseline, one and two months after observation. Results showed that two months after observation the insulin level increased from 194.79 to 519.82 pmol/ml (P = 0.033) in group consuming milk containing soy germed protein with or without Zn, supported by significantly reduced blood glucose level. This result might be correlated with the potency of isoflavones in soy germ protein to protect pancreatic beta cellsmembrane from free radicals attack. Therefore, this maintain the cells integrity and to secrete optimal insulin.

Virgin Coconut Oil (VCO) Enriched with Zn as Immunostimulator for Vaginal Candidiasis Patient

Abstract

Disturbance on the immune system and deficiency of Zn is two factors which often trigger vaginal candidiasis patient. The aim of the research was to study the effect of virgin coconut oil (VCO) enriched with Zn to the amount of neutrophil and lymphocyte subset cells, and the level of IL-2 and IgG in vaginal candidiasis patient. Thirty women were grouped into three (ten women in each group): A, B and C, and intervened for two months. Women in A group were intervened with two tablespoon/day; those in B group were intervened with one tablespoon/day; while those in C group served as control (placebo). Blood was sampled at baseline time, one and two months after intervention. Hematological test by Micros-OT was done on a part of blood, and the plasma was used for IL-2 and IgG level tests using ELISA. The virgin coconut oil enriched with Zn maintained the number of neutrophil and NK cells, but increased Tc cells from 521 to 649 cells/mm3, increased Th cells from 1.090 to 1.380 cells/mm3. The enriched VCO also increase level of IL-2 from 0.25313 to 0.27337 pg/ml, while the IgG level changed from equivocal to negative. The recommended dosage was one tablespoon each day. Effect of Zn Supplemented to Immune Status Premenopausal Women Intervented with Isoflavoned Drinking

Abstract

The research was conducted to find out the effect of Zn supplement to immune status of premenopausal women intervented with isoflavoned drinking. Respondents were 22 women, more than 40 year of age. They were divided into two groups, i.e. 11 women intervented with isoflavone, and other 11 women intervented with isoflavone and 8 mg Zn. The activities of SOD, catalase and GPX were determined by spectrophotometer, thymulin levels by ELISA, whereas Zn levels by AAS. Result showed that Zn had significantly increased SOD lymphocyte activities (p=0.002) and thymulin plasma (p=0.011). Zn had increased catalase (p=0.103) and GPX (p=0.322) as well, but Zn plasma had decreased (0.163). It was indicated that Zn had improved the immune status by increasing lymphocyte and thymus cells activities.

Soy Germ Protein With or Without-Zn Improve Plasma Lipid Profile in Metabolic Syndrome Women

Abstract

The aim of this research was to determine the effect of soy germ protein on lipid profile of metabolic syndrome (MetS) patients. Respondents were 30 women with criteria, i.e. blood glucose level > normal, body mass index > 25 kg/m2, hypertriglyceridemia, low cholesterol-HDL level, 40-65 years old, living in Purwokerto, and signed the informed consent. The project was approved by the ethics committee of the Medical Faculty from Gadjah Mada University-Yogyakarta. Respondents were divided into three randomly chosen groups consisting of ten women each. The first, second, and third groups were treated, respectively, with milk enriched soy germ protein plus Zn, milk enriched soy germ protein (without Zn), and placebo for two months. Blood samples were taken at baseline, one and two months after observation. Two months after observation the groups consuming milk enriched with soy germ protein, both with or without Zn, had their level of cholesterol-total decrease from 215.8 to 180.2 mg/dl (P = 0.03), triglyceride from 240.2 to 162.5 mg/dl (P = 0.02), and LDL from 154.01 to 93.85 mg/dl (P = 0.03). In contrast, HDL increased from 38.91 to 49.49 mg/dl (P = 0.0008). In conclusion, soy germ protein can improve lipid profile, thus it can inhibit atherosclerosis incident.

EFEK SUPLEMENTASI EKSTRAK PROTEIN KECAMBAH KEDELAI TERHADAP KADAR IL-1BETA PENDERITA DIABETES TIPE-2 [The Effect of Soy Germ Protein Extract Supplementation on the Level of IL-1 Beta of Type-2 Diabetic Woman] Hery Winarsi1)* dan Agus Purwanto2) 1) Fakultas Biologi, Universitas Soedirman Purwokerto  2) RSUD Margono Soekarjo Purwokerto   Diterima  5 April  2009 / Disetujui  30  Desember 2009 ABSTRACT   The research was conducted to determine the effect of soy germ protein extract supplementation on the IL-1beta level of type-2 diabetic women. Research subjects were 32 type-2 diabetic women, outpatients of Diabetic Clinics of Margono Soekarjo General Hospital-Purwokerto. These women had blood glucose level above  normal, BMI > 23 kg/m2, over 40 years old, lived in Purwokerto, and signed the informed consent. Group A (n=8) was given milk containing soy germ protein extract plus Zinc, Group B (n=8) was given milk containing soy germ protein extract without Zinc, Group C (n=8) was given placebo, and Group D (n=8) was given glibenclamide for 8 weeks. Blood samples ware taken 3 times: 0, 4 and 8 weeks after intervention. The plasma IL-1beta level was analyzed using Elisa. After 8 weeks, the IL-1beta level decreased from 6.01 pg/mL to 2.63 pg/mL (p=0.022) in group A.  However, the IL-1beta level in Group A was not different from the Group B (p=0.51). Soy germ protein extract  decreased cytokine inflammatory production of type-2 dibetic. Soy germ protein extract with or without Zn had similar effects to glibenclamide in reducing the IL-1beta level (p=0.76).   Key words: IL-1beta, soy germ protein, Zn, type-2 diabetic.

Abstract

Premenopause is part of natural aging process in women indicated by the decrease of ovarial estrogen production, and usually followed by development of menopausal syndrome. Susumeno, skim milk fortified with 100 mg of soy isoflavone and 8 mg of Zn was given to premenopausal women for decreasing menopausal syndrome. This research aimed to evaluate the effect of susumeno on menopausal syndrome in premenopausal women. About 33 premenopausal women aged 43-52 years old in Purwokerto were choosen as respondent. Their menopausal syndrome were identified by using questioner. The answers were scored from 1 to 5 ; i.e., 1 = not detected; 2 = less detected; 3 = mildly detected; 4 = detected; 5 = much detected. It was revealed that some of menopausal syndromes had significantly decreased, especially vaginal dryness from less detected to not detected (p=0,01); fatigue from mildly detected to not detected (p=4,07E-05); mindless from mildly detected to not detected (p=0,022); and dyspareunia from less detected to not detected (p=0,036). Abstract

Aging diinisiasi oleh terakumulasinya radikal bebas. Antioksidan yang diketahui dapat mengendalikan reaktivitas radikal bebas diasumsi sebagai anti-aging. Penelitian ini bertujuan untuk mengetahui usia awal terjadi aging pada perempuan berdasarkan status antioksidan. Penelitian ini bersifat observasional analitik yang dirancang secara purposive random sampling dilakukan di Purwokerto pada tahun 2008 dengan melibatkan 34 perempuan usia balita, anak-anak, remaja, dewasa, tua dan lansia, yang sehat, dan tinggal di Purwokerto. Status antioksidan diketahui berdasarkan aktivitas enzim superoksida dismutase (SOD), katalase, dan glutation peroksidase (GSHPX) eritrosit serta kadar malondialdehid (MDA) plasma responden. Data diuji menggunakan analysis of varians (ANOVA). Status antioksidan perempuan sehat dari usia balita hingga lansia adalah prima yang ditunjukkan oleh tingginya aktivitas SOD berkisar 1.469+3,58–2.009+4,12 U/mg; katalase 20,37+0,01–31,45+0.04 UI/mg; GSH-PX 79,03+0,0–225,2+0,04 μmol/g protein eritrosit; didukung oleh kadar MDA yang rendah dengan kisaran 3.134+2,56 –3.185+3,06 pmol/mL plasma. Secara umum, penurunan status antioksidan terjadi pada usia dewasa (24–<45 tahun), sehingga pada usia tersebut para perempuan mulai memerlukan tambahan suplemen antioksidan guna menghambat laju proses aging dalam tubuhnya. Antioxidants which were known to control the reactivity of free radicals can be assumed as an anti aging. This study aimed to determine the age of the onset of aging in female based on antioxidant status. The analytical observational study with purposively random sampling design was conducted in Purwokerto in the year of 2008 with 34 females included toddlers, children, adolescents, adults, old, and elderly who were healthy, and live in Purwokerto. Antioxidant status was known by enzyme activities of superoxide dismutase (SOD), catalase, gluthathion peroxidase (GSH-PX) in the erythrocytes and malondialdehyde (MDA) levels in the plasma. Data were tested by analysis of varians (ANOVA). Antioxidant status in healthy female from toddlers to the elderly were prime as indicated by the high SOD, catalase, and GSH-PX activities that ranged 1,469+3.58–2,009+4.12 U/mg; 20.37+0.01–31.45+0.04 UI/mg; 79.03+0.01–225.2+0.04 μmol/g protein of erythrocytes, respectively; supported by low levels of MDA that ranged from 3,134+2.56–3,185+3.06 pmol/mL plasma. In general, the decrease in antioxidant status occurred in adults, so at that age female began to need additional antioxidant supplements in order to inhibit the rate of aging processes in the body. Abstract

Rendahnya status antioksidan endogenus memicu perkembangan kondisi sindrom metabolik (SM). Penelitian ini bertujuan untuk mengeksplorasi aktivitas enzim antioksidan superoksida dismutase (SOD), katalase, dan glutation peroksidase (GSH-PX) dalam plasma wanita penderita sindrom metabolik. Penelitian dilakukan periode Maret–November 2010, dengan responden penelitian adalah 30 wanita yang memiliki kriteria sebagai berikut: kadar guladarah >normal, body mass index >25 kg/m2, hipertrigliseridemia, high-density lipoprotein (HDL) rendah, usia >40 tahun, dan tinggal di Purwokerto. Seleksi responden dimulai melalui survey di poliklinik internis Rumah Sakit Umum Margono Soekarjo Purwokerto. Kepada calon responden dijelaskan tentang tujuan penelitian, kondisi SM, dan perkembangannya. Calon responden yang bersedia bergabung diminta menandatangani informed consent, diukur berat badan, tinggi badan, dan tekanan darahnya. Diambil sampel darahnya 2 mL, intravena, ditentukan kadar glukosa darah, trigliserida, dan HDL. Bagian plasma, diuji aktivitas enzim SOD, katalase, dan GSH-PX. Data dipresentasikan sebagai rata-rata + standard error. Status antioksidan wanita penderita sindrom metabolik rendah, ditunjukkan oleh aktivitas SOD 379,3 Unit/mg protein, katalase 6,42 UI/mg protein, dan GSH-PX 804,9 nmol/g protein, serta kadar malondialdehid (MDA) sebesar 2.943,4 pmol/mL. Karena itu responden perlu asupan kaya antioksidan. Simpulan, status antioksidan peroksidase sama rendah yang ditunjukkan oleh rendahnya aktivitas enzim SOD, katalase, dan glutation peroksidase. Respondents were thirty women with blood glucose level >normal, body mass index >25 kg/m2, hypertrigliceridemic, low level cholesterol-high-density lipoprotein (HDL), the age >40 years and live in Purwokerto. Respondents’ selection was started by a survey at internist polyclinic of Margono Soekarjo General Hospital Purwokerto. They were motivated and given an explanation about aim of the research, their diseases and development risk who were willing to be respondents were asked to sign the informed consent. Then, their body weight, height and blood pressure were measured. Their blood sample of 2 mL were taken intravenously and tested for blood glucose, triglyceride and HDL levels. Plasma was tested for the activities of SOD, Cat and GSH-PX enzymes. Presented data was mean + standard error. The antioxidant status of MetS women were low, showed on the activities of SOD, catalase, and glutathione peroxidase enzyme were 379.3 Unit/mg protein, 6.42 UI/mL, 804.9 nmol/g protein respectively, and malondialdehyde (MDA) level was 2,943.4 pmol/mL. Therefore, they need food enrich antioxidant. Profil Lipid, Peroksidasi Lipid, dan Status Inflamatif Wanita Penderita Sindrom Metabolik

Abstract

Prevalensi sindrom metabolik (SM) di Indonesia (13,13%) tergolong tinggi dengan kecenderungan terus meningkat. Salah satu akibat SM adalah disfungsi endotel, sebagai awal penyakit kardiovaskuler yang diinduksi oleh stres oksidatif dan inflamatif. Penelitian ini bertujuan untuk mengeksplorasiprofil lipid, peroksidasi lipid, dan marker inflamasi pada wanita penderita SM di Purwokerto. Sebanyak 30 wanita dengan kadar gula darah diatas normal, obesitas body mass index (BMI) > 25 kg/m2, dan berusia 40-65 tahun dilibatkan sebagai responden yang dipilih melalui survei di PoliklinikPenyakit Dalam Rumah Sakit Margono Soekarjo. Kadar kolesterol total, trigliserida, high density lipoprotein, low density lipoprotein, malondialdehid, dan plasma C-reactive protein ditentukan dalam darah responden yang mempunyai kadar gula sewaktu > 200 mg/dL. Ditemukan bahwa wanitadengan SM rata-rata berumur 50,4 tahun; BMI 31,89 kg/m2; kadar gula darah 219,4 mg/dL; kolesterol total 216,73 mg/dL; trigliserida 218,13 mg/dL; HDL 46,59 mg/dL; LDL 146,27 mg/dL; MDA 2943,4 pmol/mL; C-RP 7,62 mg/L; dan tekanan darah 153/103 mmHg. Hasil ini menunjukkan bahwapenderita SM mengalami dislipidemia disertai dengan status antioksidan rendah dan inflamasi.Kata kunci: Wanita sindrom metabolik, profil lipid, lipid peroksida, malondialdehid, C-reactive proteinAbstractPrevalence of metabolic syndrome (MS) in Indonesia (13,13%) is high and tends to increase. One of the consequences of MS is endothelial dysfunction leading to cardiovascular disease which is inducted by oxidative stressand inflammation. The aim of the present research is to explore lipid profile, lipid peroxidation, and inflammatory marker level on metabolic syndrome women in Purwokerto. Thirty women with blood glucose level greater than normal, body mass index (BMI) > 25 kg/m2, 40-65 years of age were recruited as respondent through selection by a survey in Internal Medicine Polyclinic of Margono Soekarjo Hospital in Purwokerto. In respondents with blood glucose level > 200 mg/dL, total blood cholesterol level, high density lipoprotein, low density lipoprotein, malondialdehid, and plasma C-reactive protein were determined. It was found that the MS women were 50,4 years of age; BMI 31,89 kg/m2; blood glucose 219,4 mg/dL; total cholesterol 216,73 mg/dL; triglyceride 218,13 mg/dL; HDL 46,59 mg/dL; LDL 146,27mg/dL; MDA 2943,4 pmol/mL; C-RP 7,62 mg/L; and blood pressure 153/103 mmHg. It indicates that SM women experience dyslipidemia with low antioxidant and inflammation.Key words: Metabolic syndrome women, lipid profile, peroxide lipid, malondialdehid, C-reactive protein
