Abstract

This experiment was designed to study the impact of mammary carcinogenesis induced by 7,12-dimetilbenz (?) antracene (DMBA) on histopathological features of the gastric in Sprague Dawley rat.Eight female Sprague Dawley (SD) rats, of four week-old, were used in this study. The rats were randomlydivided into 2 groups of four. Group I were treated orally with 20 mg/kg BW of 7,12-dimetilbenz (?)antracene (DMBA) treatment, twice a week for 5 weeks. Group II were used as negative control withoutDMBA treatment. At 5 week after the end DMBA of, the rats of the both groups were sacrificed and thegastric were fixed in 10% buffer formalin. Subsequently, tissue sections were stained with hematoxylinand eosin (H&E) for microscopic examination. The results of the experiment show that oral administrationof DMBA (20 mg/kg BW) twice a week, for 5 weeks result in epithelial hyperplasia of cardia. Hemorrhagicand inflammation reactions are also found on the mucous membrane of fundic. However, macroscopicexamination shows no specific changes of the gastric. Histopathological changes were suspected as a precancer lesion of the gastric due to chronic exposure of DMBA.

Abstract

The sponge Aaptos suberitoides can produce high secondary metabolite with some farmocological activities as antimicrobial, antiviral, antiinflamatory, and anticancer agents. The purpose of this research is to find out correlations between activities of the etanol extracts of marine sponges Aaptos suberitoides on the cancer growth of subcutaneous mice (Mus musculus) injected by Benzo(a)piren. For the purpose the mice were divided into six groups, i.e. I, II, III, IV, V, and VI. Each group was treated with carcinogenic inductions by intravenously injecting the Benzo(a)piren concentration of 0.3g/0.2 ml oleum olivarum. After the cancer was appeared at the fifteenth day, the mice were treated by the anticancer extracts of marine sponges Aaptos suberitoides with concentrations of 500 mg/kg BB (Group IV), 1000 mg/kg BB (Group IV), and 1500 mg/kg BB (Group IV). The treatments were orally done each day for two weeks. At the twentieth week, subcutaneous cancer tissues were taken to make histological preparates using a parafin method. Result of the histological observation indicates that cancer in the mice was fibrosarcoma characterized by the thickening dermis layers, necrosis, mitosis, and nuclear polymorphsm. Necrosis, mitosis, and nuclear polymorphism occurred in Groups II, IV, V, and VI, and did not in Groups I and II. Presentation of necrosis was 20-60%, mitosis was in 3-4 cells, and nuclear polymorphism was 100%. Result of the statistical analyses by using the Kruskal-Wallis method and the Pair Comparison test indicates that the anticancer extracts of marine sponges with the concentrations of 500 mg/kg BB, 1000 mg/kg BB, and 1500 mg/kg BB had no activity inducing mice skin cancer.

Abstract

The studies have reported that ginger have many activities such as antiemesis, anti-inflammatory,anti-bacterial and anti-parasites. Therefore, this study was conducted to evaluate antiviral effect of essentialred ginger oil againts Avian Influenza (AI) in ovo using hemagglutination test (HA). Avian Influenzaviruses were treated with 0,01%, 0,1% and 1% of essential red ginger oil, and then inoculated in chickenembryonated egg via allantoic sac. Allantoic fluid was harvested using for HA test . Result of this studyshows that application of 1% of essential red ginger oil results in the reduction of titer HA . Interestingly,essential oil shows antiviral activity revealed HA titre 20 whereas the titre HA AI which AI virus treatedwith 0,01% and 0,1% essential red ginger oil, the HA titer was 25. The conclution of this study proved thatessensial oil 1% of the red gingger is the best concentration as antiviral activity .

Abstract

Leptin merupakan hormon yang disintesis oleh sel lemak dan berperan dalam proses kesembuhan luka. Tujuan penelitian ini adalah mengidentifikasi leptin dan leukosit setelah luka irisan pada tikus yang diberi pakan lemak tinggi dan aplikasi topikal zinc. Penelitian ini menggunakan 16 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masingmasing terdiri dari empat ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup kembali dengan jahitan. Pada kelompok A dan C, luka jahitan diberi aplikasi topikal vaselin (tanpa zinc), sedangkan pada kelompok B dan D, luka jahitan diberi aplikasi topikal zinc 10%. Tiga hari setelah operasi, tikus diambil darahnya untuk analisis kadar leptin dan leukosit total. Data leptin dan total leukosit dianalisa dengan ANOVA untuk faktorial eksperimen (2x2). Hasil penelitian menunjukkan kadar leptin secara signifikan lebih tinggi pada tikus yang diberi pakan lemak tinggi dibandingkan tikus yang diberipakan normal (P<0.05), sedangkan leukosit total tidak menunjukkan adanya perbedaan yang signifikan. Dengan demikian, kadar leptin pada hari ketiga setelah luka irisan dipengaruhi oleh pakan, yaitu pakan lemak tinggi dapat meningkatkan kadar leptin. Leukosit total tidak dipengaruhi oleh pakan dan aplikasi topikal, meskipun demikian, leukosit total cenderung lebih rendah pada tikus yang diberi aplikasi topikal zinc.Kata kunci: pakan lemak tinggi, leptin, zinc, leukosit total, tikus Sprague Dawley

Abstract

The  objective of this study was to determine the pathological lesions of the broiler chickens having the clinical signs that appeared to be the swollen head syndrome (SHS),  such as simetrically and bilaterally swollen periorbital areas. Eighteen broiler chickens were collected from three poultry farms of the three diffrerent areas, e.g. Kulonprogo, Sleman and Bantul at Special Province of Yogyakarta, respectively. Six of those broiler chickens were selected from each poultry farm consisted of three SHS- broiler chickens and three non SHS-broiler chickens. All of broiler chickens were necropsied, observed anatomic patologic lesions, and then the organs of interest were processed and stained routinely with hematoxylin and eosin for histopathologic examination. Subcutaneus edema and cellulitis were found in the frontal and occipital skin. Based on the histopathological examination, purulent and granulomatous cellulitis and lymphocytic perivasculitis, and subcutaneus edema were found in the frontal and occipital skin. Other lesions, such as: necrotic, lymphocytic sinusitis, tracheitis and pneumonia accompanied by atrophy of mucosal glands were also present. It is concluded that the pathological lesions of SHS in the broiler chickens were found not only in the frontal and occipatal skin, but also in both lower and upper respiratory tracts.

Abstract

Gangguan metabolisme karbohidrat dapat menyebabkan Diabetes mellitus. Karbohidrat dalam saluran cerna mengalami metabolisme menjadi glukosa yang sederhana kemudian diabsorbsi masuk kedalam peredaran darah serta mempengaruhi kadar glukosa dalam darah. Proses penyerapan ini dikatalisis enzim pemecah ikatan α-1,4-glikosida yaitu α-amilase dan enzim pemecah ikatan α-1,6-glikosida yaitu α-glukosidase yang terdapat pada sel usus. Telah dilakukan penelitian sebagai upaya mengembangkan pengobatan alternatif diabetes mellitus dengan menguji kemampuan isolat andrografolid dalam menghambat aktivitas α-amilase dan α-glukosidase secara in vitro. Isolat andrografolid memperlihatkan aktivitas yang cukup baik dalam menghambat α-amilase (IC50= 12,49 mg/mL) dan lemah dalam menghambat α-glukosidase (IC50= 38,86 mg/mL). Penghambatan aktivitas α-amilase ini menjadi bukti salah satu mekanisme andrografolid dalam mengurangi metabolisme karbohidrat yang dapat mempengaruhi kadar glukosa dalam darah dan mengindikasikan andrografolid sebagai obat alternatif yang cukup potensial dalam mengatasi penyakit diabetes mellitus.

Abstract

This research was conducted to study the histopathological effect of ethanol extract of Gynuraprocumbens (Lour) Merr leaves in male Sprague Dawley rats. Ten rats were divided into an experimental group and a control group, each consisting of five rats. The test extract was administered to the experimental group for 36 weeks, twice weekly. The control group was untreated up to the end of the experiment. The rats were necropsized at the end of the 18th and 36th week. The gastrium was excised to be fixed in 10% buffered formalin for 24 hours. The H&E sections were then prepared for histological examination. The histological appearance in 4 of 5 experimental rats shows extensive mucus secretion and a mild mucosal epithelial erosion of gastrium. No histological changes were detected in the control group. It was concluded that the administered extract of G. procumbens for 36 weeks twice weekly did not result in significant pathological changes.

Abstract

This study was carried out to investigate the effect of 4NQO oral induction in oesophagus of male rats. Sixteen male Sprague Dawley rats were divided into three experimental groups and one untreated group as control. The experimental groups were applied with 0.5% 4-nitroquinoline 1-oxide on the dorsal mucosa of tongue thrice weekly for 8,16 or 24 weeks, one brush stroke per application. At the end of the 36th week, all rats were sacrificed and the tongue and oesophagus were excised and fixed in 10% buffered formalin for 24 hours. The H&E sections were prepared for histological examination. The microscopical assessment showed that all rat tongues whether applied with 4NQO for 8, 16 or 24 weeks were identified having Squamous Cell Carcinoma (SCC). Microscopical examination of oesophagus indicated that 75% of the rats applied with 4NQO for 16 weeks showed hyperkeratosis, and 20% of the rats applied with 4NQO for 24 weeks showed malignancy changes and hyperkeratosis, respectively. No histological changes were detected either in the tongue or the oesophagus of the control rats. It was concluded that the effect of carcinogenic induction in oral mucosa caused malignant changes in oesophagus.

Abstract

Infectious Bovine Rhinotracheitis (IBR) is caused by Bovine Herpes virus-1 in the cattle. The clinicalsigns demonstrate depression, anorexia, swelling of the vulva, redness of the vestibule, pustule and ulceron the vaginal mucosal. Based on previous research, IBR virus from the nasal swab could be grown inchorio-allantoic membrane of embryonated chicken eggs. This study aim was to confirm whether IBR virusin cattle could be grown in embryonated chicken eggs as a substitute for cell culture. A total of five nasalswab samples from the cows that were positive for IBR infection (diagnosed by Polymerase Chain Reactionand cell culture) were inoculated on the chorio-allantois membrane of embryonated chicken eggs.Observation of lesions performed at 3-5 days after inoculation. Re-inoculation (passage) was done threetimes. Pock characteristic lesions were observed on the corioallantoic membrane with the size of 5-7 mm,rounded shape, opaque edge, with necrosis in the central area. Furthermore, pock lesions were processedfor hematoxylin and eosin staining and immuno-histochemistry. The result of hematoxylin and eosinstaining showed that the formation of intranuclear inclusion bodies and vacuolization of the epithelial cellof membrane was observed. Immuno-histochemistry staining showed positive reaction for antibodiesagainst BHV-1 in the epithelial cells membrane. In conclusion, embryonated chicken eggs could be usedas a medium for detection of IBR.

Abstract

The cytochrome P-450 (CYP) and glutathione S-transferase (GST) enzyme systems may influence the biological effects of carcinogens. As such, these enzymes may predict the developmental risk of breast cancer, as well as be potential targets for chemoprevention. The purpose of this study was to compare the expression of CYP1A1 and GST* between treatment groups. Expression of CYP1A1 and GST* was quantified in liver tissue from 18 female Sprague Dawley rats aged 40 days, which randomly divided into six treatment groups. Those are base line group (without DMBA and ethanolic extract treatment), DMBA induced cancer group, the other two groups was administrated by DMBA after treated by ethanolic extract in two different doses, 300 mg /kg BW and 750 mg/kg BW. The last two groups were given two doses of extract ethanolic only, without initiated of DMBA. The ingestion of the extract was carried for three weeks and the ingestion of DMBA was performed twice in the third week. The expression CYP1A1 and GST was quantified by immunohistochemistry. CYP1A1 expression was significantly higher (P < 0.05) in cancer group (DMBA) as compared with other groups. On the other hand, GST expression was lower in cancer group (P < 0.05). Result of this study demonstrated that ethanolic extract leaves of G. procumbens in 300 mg/kg BW dose could inhibit CYP1A1 stronger than are other and induced GST* level. Has effect on ethanolic extract leaves of G. procumbens has ability in played role of as blocking agent in preventing initiation stage of carsinogenesis, therefore it should be taken into account for chemopreventing agent in mammary carsinogenesis.Key words : Gynura procumbens, breast cancer, Chemopreventive, CYP1A1, GSTμ
