The Effect of Tender Coconut Water on Free Radical Due to Mercury Exposure

Abstract

Mercury pollution can results from gold mining. Furthermore, a continuous mercury exposure can trigger the formation of Reactive Oxygen Species (ROS) and interfere with the body's antioxidant metabolism. A high level of free radicals in the body can be characterized by a low level of an antioxidant enzyme. Prevention to avoid the impacts of mercury poisoning is better than treatment. Tender coconut water can eliminate ROS and improve antioxidant status. This study aimed to was to confirm that tender coconut water is able to decrease the free radical which is characterized by increased antioxidant enzyme GPx status. This study was designed as randomized pre- and posttest control group design, 40 traditional gold mining workers devided into 2 groups: the control and treatment groups. After the pre test of GPx level, the workers were either given tender coconut water at the dose of 450 m L or water for 4 weeks every day. Four weeks after the pre-test, the level of GPx was assessed in the laboratory PAU-Gajahmada University. The post test results of the control group and treatments group were compared and analyzed using Man Whitney test. The mean levels of GPx post-test in the control group and treated group were 63.5±1.1 U/mL and 79.9±8.8 U/mL respectively. The analysis result obtained p-value 0.000 <0.05. Tender coconut water has an effect on free radicals due to to mercury exposure. Tender coconut water can be consumed daily to prevent degenerative diseases caused by mercury exposure.

Abstract

Background: Low birth weight (<2500 grams) is representation of a nutritional deficiency during pregnancy, thiscondition is associated with increased incidence of degenerative diseases. Tempe as traditional Indonesian foodhas the potential to improve of DNA damage and to inhibit the progression of degenerative diseases. This studyaimed to determine the effect of tempe flour supplementation during pregnancy on blood sugar and blood lipidprofiles of offsringMethods: An experimental study with a sample of 20 female Wistar rats were pregnant, were divided into 4groups. Treatment during pregnancy: the first group received a diet 50% of the normal diet (10 g / day), group IIget a normal diet and 50% tempe flour 1 g / day, group III received normal diet and 50% tempe flour 2 g / day,group IV, 50% received a normal diet and 1 g egg chicken / day. Provision of drinking with distilled water adlibitum. During lactation are given a normal diet, after weaning are given a diet high in saturated fat for 4 weeksin each group.Results: The mean number of children in group 1: 9.33 with a mean BB: 4.93 g, group II: with a mean of 6.17 BB:5.32, group III: 5.17 with a mean BB: 5.33, group IV : BB with a mean of 6.33: 5.01. There are significant differencesin the levels of blood sugar, total cholesterol, triglycerides, LDL, and HDL in each treatment group (p= 0,00).Conclusion: Supplementation of tempe flour effect on blood sugar levels and lipid profiles rat pups that receiveda restricted diet during pregnancy (Sains Medika, 4(2):182-194).Latar belakang: Berat badan lahir rendah (< 2500 gram) merupakan gambaran dari defisiensi nutrisi selamakehamilan. Keadaan ini berhubungan dengan peningkatan insiden penyakit degeneratif. Tempe sebagai makananasli Indonesia mempunyai kandungan nutrisi yang berpotensi memperbaiki kerusakan DNA maupun untukmenghambat progresifitas penyakit degeneratif. Penelitian ini bertujuan untuk mengetahui pengaruhsuplementasi tepung tempe selama kehamilan terhadap kadar gula dan profil lipid darah anak yang dilahirkan.Metode penelitian: merupakan penelitian eksperimental dengan sampel 20 ekor tikus betina galur Wistar hamil,dibagi menjadi 4 kelompok masing-masing 5 ekor. Perlakuan selama hamil: kelompok I mendapat diet 50% daridiet normal (10 g/ekor/hari), kelompok II mendapatkan 50% diet normal dan tepung tempe 1 g/ekor/ hari,kelompok III mendapat 50% diet normal dan tepung tempe 2 g/ekor/hari, kelompok IV, mendapat 50% dietnormal dan 1 g telor ayam /ekor/ hari. Pemberian minum dengan aquades ad libitum. Selama masa menyusuidiberikan diet normal, setelah disapih diberikan diet tinggi lemak jenuh selama 4 minggu pada masing-masingkelompok.Hasil: Rerata jumlah anak kelompok 1: 9,33 dengan rerata BB: 4,93 g, kelompok II:6,17 dengan rerata BB: 5,32,kelompok III: 5,17 dengan rerata BB : 5,33, kelompok IV: 6,33 dengan rerata BB: 5,01. terdapat perbedaan yangsignifikan pada kadar gula darah, kolesterol total, trigliserid, LDL, dan HDL pada masing-masing kelompok perlakuan(p= 0,00).Kesimpulan: Suplementasi tepung tempe berpengaruh terhadap kadar gula darah dan profil lipid anak tikusyang mendapat pembatasan diet selama hamil (Sains Medika, 4(2):182-194).

Abstract

Low birth weight, a nutritional deficiency is related to the increased in the coronary heart disease insidence.Low birth weight is correlated with the hipotalamus-pituitary-adrenal responsible for the the concentrationof cortisol in sirculation, increased in homosistein, insuline resistence and increased C reactive proteinplaying role on the aterosclerosis process predispose the corronary disease. This paper will discuss therelationship between the low birth weight and the ateroclesoris process leading to coronary heart disesase(Sains Medika, 3(2):185-200).Key words : low birth weight, aterosklerosisBerat badan lahir rendah (BBRL), yaitu kondisi berat badan lahir bayi < 2500 gram merupakan gambarandari defisiensi nutrisi selama kehamilan berhubungan dengan peningkatan insiden penyakit jantungkoroner. BBLR dihubungkan dengan kelainan aksis hipotalamus-pituitary-adrenal yang bertanggung jawabterhadap kadar kortisol darah, peningkatan kadar homosistein darah, resistensi insulin, peningkatankadar C-reactif protein yang berperan pada peningkatan proses aterosklerosis sebagai awal dari penyakitjantung koroner. Abstract

Radikal bebas selalu terbentuk dalam tubuh sebagai hasil samping respirasi aerobik. Pada bunting terjadi peningkatan produksi radikalbebas yang berpotensi untuk menimbulkan kerusakan struktural maupun fungsional pada janin. Tempe sebagai makanan asli Indonesiamempunyai kandungan vitamin, mineral, maupun isoflavon yang bersifat antioksidan, berpotensi mengurangi kerusakan akibat radikalbebas dan memperbaiki kerusakan DNA yang mungkin terjadi selama bunting. Penelitian ini bertujuan untuk mengetahui pengaruhsuplementasi tepung tempe selama bunting terhadap kadar Glutathione peroksidase (GPx) dan kadar Malondealdehyde (MDA) sebagaiindikator terjadinya peroksidasi lipid pada anak yang dilahirkan. Penelitian eksperimental dengan sampel 20 ekor tikus betina galurWistar bunting, dibagi menjadi 4 kelompok masing-masing 5 ekor. Perlakuan selama bunting: kelompok I mendapat pakan standar10 g/ekor/hari, kelompok II mendapatkan 10g/ekor/hari pakan standar dan tepung tempe 1g/ekor/ hari, kelompok III mendapatpakan standar 10 g/ekor/hari dan tepung tempe 2 g/ekor/hari, kelompok IV mendapat pakan standar 10 g/ekor/hari dan 1 g telorayam /ekor/ hari. Pemberian minum dengan aquades ad libitum. Selama masa menyusui (sampai umur 1 bulan) anak tikus diberi dietnormal 10g/100gBB/hari, setelah disapih diberi tambahan diet tinggi lemak jenuh selama 4 minggu pada masing-masing kelompok.Rerata jumlah anak kelompok 1: 9,33 dengan rerata BB: 4,93 g, kelompok II:6,17 dengan rerata BB: 5,32, kelompok III: 5,17 denganrerata BB : 5,33, kelompok IV: 6,33 dengan rerata BB: 5,01. Rerata kadar GPx pada kelompok I : 22,38 U/mg, kelompok II: 28,94 U/mg,kelompokII: 38,59 U/mg, dan kelompok IV: 16,98 U/mg. sedangkan rerata kadar MDA kelompok I: 11,07 nmol/mg, kelompok II: 9,89nmol/mg, kelompok III: 10,06 nmol/mg, dan kelompok IV: 12,25 nmol/mg. Terdapat perbedaan signifikan antara kelompok kontroldengan perlakuan tepung tempe 1g/ ekor/hari maupun 2g/ekor/hari baik kadar GPx maupun MDA (p;0,00). Means of GPx for the four groups were 22.38 U/mg, 28.94 U/mg, 38.59 U/mg, 16.98 U/mg respectively. The meanof MDA for the four groups were 11.07 nmol/mg, 9.89 nmol/mg, 10.06 nmol/mg and 12.25 nmol/mg respectively. There was a significant difference inGPx and MDA (p=0.00).
