Abstract

Sandy coastal area is a poor land, with minimal nutrients and low ability to store water and nutrition. Therefore, plant is difficult to grow well, consequently the treatments for improving sandy coastal is required before planting to support their growth. Application of Trichoderma as a plant-growth promoting and controlling pathogens had been known. Shallot seed treatment by Trichoderma agents was intended to improve and support plant growth and yield in the sandy coastal land. The aim of experiment was to determine the most effective treatment to improve growth and yield of shallot on sandy coastal land. The experiment had been carried out in Yogyakarta during August - November 2015. The factorial treatments of cultivar and Trichoderma applications were arranged in Completely Randomized Design with three replications. The shallot cultivars consisted of Tiron, Crok, and Biru, while Trichoderma application consisted of control (no treatment), sprayed with Trichoderma, soaked in Trichoderma, matricontioning media i.e. rice husk charcoal and brick in combination with Trichoderma. The collected data related to plant growth and yield. The result showed that seed treatment methods with Trichoderma increased the plant height, the leaf area index, extensive root, net assimilation rate (NAR), plant growth rate (PGR), Nitrate Reductase Activities (NRA), total chlorophyll, and fresh bulb weight. The best technical treatment for cultivar Tiron was soaked with Trichoderma, and Crok was sprayed with Trichoderma, while Biru cultivar was using brick in combination with Trichoderma.

Abstract

Fusarium wilt caused by Fusarium oxysporum f.sp.cepae (Foc) is one of the most important diseases of onion in Indonesia. Induced resistance was one of the effective techniques to acquire resistance to fusarium wilt in shallot. Salicylic acid (SA) had been inferred to be in endogenous signal in the systemic acquired resistance response of plants. This research aimed to study the effect of exogenously added SA to resistance in shallot callus cv ‘Bima Brebes’ to fusarium wilt disease in vitro; and to determine the effective concentration of SA to induce resistance. A group of shallot callus was grown in MS medium containing varying SA concentration (0, 15, 20 and 25 ppm) in vitro for 2 weeks. Then, the callus were treated with toxin of Fusarium oxysporum, namely fusaric acid, to have observe the resistance response. In vitro selection was done twice in different fusaric acid concentration. Application of exogenous SA at all concentration did not suppres time of appearance of disease symptom yet. Toxic symptom in the callus was shown by the browning or blackening (off) of callus. Salicylic acid at concentration of 20 ppm effectively reduced the toxic symptom up to 16.66% and supported callus regeneration better than the concentration of 15 ppm and 25 ppm. The number of resistant callus regenerated was 66.67% at pretreatment of 20 ppm of SA.

Abstract

Corn is one of strategic commodities in Indonesia. Corn productivity from year to year continues to increase, but it is lower than the world productivity. This research analyzes the comparative and competitive advantages of corn farm management in Grobogan district, Central Java province. Corn is financially and economically profitable, besides that, it also has comparative and competitive advantages. Government policy on corn commodity has not been running effectively. The most sensitive component of the profit earned in corn commodity in Grobogan is a component of the output price.

Abstract

Molecular and biochemical characteristics of Fusarium oxysporum f.sp. cubense (Foc) were detected. Six of Indonesian Foc isolates were artificially inoculated on “Ambon Kuning” banana. DNA of one-week culture isolates was extracted by three methods prior to PCR assay using Foc TR4 (tropical race 4) specific primer. Activity of extracellular enzyme was determined with reduction sugar, agar diffusion and SDS-PAGE assays. Statistical analysis revealed that all isolates insignificantly caused Fusarium wilt symptoms on tested banana with disease severity index ranging from 3 to 3.6. Maximum DNA concentration was obtained by CTAB method (766.25 µg mL-1), followed by SDS and alkaline lysis methods, i.e. 553.75 and 211.25 µg mL-1, respectively. PCR analysis showed that Bnt2 and Kjg1 isolates positively reacted to TR4 of Foc primer (DNA size of 1400 bp approximately). Reduction sugar and agar diffusion assays demonstrated that Kjg1 isolate significantly produced more extracellular enzyme, with 6.53 × 10-2 mg mL-1 in concentration and 20 mm in halo diameter. Meanwhile, SDS-PAGE assay viewed diverse bands of tested fungi (20.6 to 80 kDa), representing four extracellular enzymes. Positive PCR results highlighted the presence of Foc TR4 infecting banana in Indonesia. Various activities of extracellular enzymes did not influence the pathogenicity of Foc.   Key words: pathogenicity, DNA concentration, extracellular enzyme

Abstract

Abstract: Effect of Banking Service Quality (BSQ) Dimensions towards Bank Customer Satisfaction. This study aims to determine the effect of Banking Service Quality (BSQ) to the customer satisfaction of BPD Bank in DIY. This research is a survey research with a sample of 1.536 respondents. The sample was taken by purposive sampling technique. Data collection instrument was questionnaires and analyzed using multiple regression. The results showed that six dimensions of BSQ simultaneously gave positive and significant impact on customer satisfaction. If seen from each dimension individually; the highest influence came from the reliability dimension and the lowest dimensions were from Access and Services Portfolio dimensions. For the national servants (PNS) customers, the BSQ six dimensions simultaneously gave a positive and significant impact on customer satisfaction; the highest influence came from the reliability dimension and the lowest was from portfolio services dimension. For the non national servants, the BSQ six dimensions simultaneously gave a positive and significant impact on customer satisfaction; the highest influence came from the reliability dimension and the lowest was from portfolio services dimension. Penelitian ini bertujuan untuk mengetahui pengaruh Banking Service Quality (BSQ) terhadap kepuasan nasabah Bank BPD di DIY. Penelitian ini merupakan penelitian survey dengan sampel secara keseluruhan berjumlah 1.536 responden. Sampel diambil dengan teknik purposive sampling. Instrumen pengumpulan data berupa kuesioner dan dianalisis  dengan regresi ganda. Hasil penelitian menunjukkan keenam dimensi BSQ secara simultan berpengaruh positif dan signifikan terhadap kepuasan nasabah, jika dilihat tiap dimensi maka pengaruh tertinggi berasal dari dimensi keterhandalan dan terendah dari dimensi Akses serta Portofolio Jasa. Untuk nasabah PNS, keenam dimensi BSQ secara simultan berpengaruh positif dan signifikan terhadap kepuasan nasabah, pengaruh tertinggi berasal dari dimensi keterhandalan dan terendah berasal dari dimensi portofolio jasa, dan untuk nasabah non PNS, keenam dimensi BSQ secara simultan berpengaruh positif dan signifikan terhadap kepuasan nasabah, pengaruh tertinggi berasal dari dimensi keterhandalan dan terendah berasal dari dimensi portofolio jasa. Kata Kunci: Banking Service Quality, Kepuasan Nasabah

Abstract

Tulisan ini bertujuan untuk melakukan analisis  atas kemungkinan terjalinnya kerjasama yang  saling menguntungkan antara sektor UMKM (Usaha Mikro Kecil Menengah) dengan Perguruan Tinggi.   Sebuah bentuk kerjasama  dengan skema Joint Venture Profit Sharing (JVPS) merupakan sebuah alternatif desain yang akan dianalisis dalam tulisan ini. UMKM hingga saat ini menghadapi berbagai permasalahan klasik, seperti permodalan, pemasaran, produksi dan teknologi, pengelolaan Sumber Daya Manusia, dan permasalahan akses informasi dan jaringan.  Sementara itu, Perguruan Tinggi (PT) di satu sisi adalah tempat bertemunya para ahli dari berbagai bidang dan tempat dikembangannya bermacam ilmu  dan teknologi yang sebenarnya sangat dibutuhkan oleh UMKM.  JVPS yang merupakan bentuk penyertaan modal dengan hak keterlibatan dalam pengelolaan usaha adalah bentuk kerjasama yang sangat dibutuhkan oleh UMKM sekaligus bisa dipenuhi leh PT. Diperlukan sebuah Unit Pengelola Kerjasama JVPS di Perguruan Tinggi yang bertugas mengelola penarikan dana-dana PT untuk tujuan pemberdayaan UMKM.  Dana yang terkumpul untuk kemudian dialokasikan kepada UMKM yang potensial, dengan sebuah perjanjian kerjasama yang mengatur hak dan kewajiban masing-masing pihak.  Perjanjian ini menjadi syarat mutlak sebelum kerjasama dilakukan, untuk menjamin bahwa hak semua pihak yang terlibat akan terlindungi dengan baik. Abstract

ABSTRACT : Along with the development of rapid information technology and the increasing use of information systems, requires a company or agency to promote information services faster and more efficiently. In the Store Joice group, author noticed many problems in the data processing sales and purchases, as it is still done conventionally so the impact on the delay in the delivery of an information data, both to managers and to consumers. With this author system provides a solution of making the sale and purchase of information. With a system of this information, it can help in the process of selling, buying pemriksaan accuracy in inventory and sales reports.In this study describes what are the things that factor into making the sale and purchase of information system and what is needed in the making. Resulting from the research and design of information systems sales and purchases are provided at the Joice Store group in the data processing and provide information funiture sales inventories, sales reports and purchase at the store Joice group. Key Word : Development of Information Systems and Electronic Sales Funiture ABSTRAKSI : Seiring dengan perkembangan teknologi informasi yang semakin pesat dan semakin bertambahnya penggunaan sistem informasi, menuntut suatu perusahaan atau instansi untuk mengedepankan layanan informasi yang lebih cepat dan lebih efisien. Pada Toko Joice group, penulis memperhatikan masih banyak permasalahan pada proses pengolahan data penjualan dan pembelian , karena  masih dilakukan secara konvensional sehingga berdampak terhadap keterlambatan di dalam penyampaian suatu data informasi, baik kepada manajer maupun kepada konsumen. Dengan ini penulis memberikan solusi pembuatan sistem informasi penjualan dan pembelian. Dengan adanya sistem informasi ini, maka dapat membantu dalam proses penjualan, pembelian  ketepatan dalam pemriksaan persediaan dan laporan penjualan. Dalam penelitian ini menguraikan tentang hal-hal apa saja yang menjadi faktor pembuatan sitem informasi penjualan dan pembelian dan apa saja yang dibutuhkan dalam pembuatannya. Yang dihasilkan dari penelitian dan perancangan sistem informasi penjualan dan pembelian ini adalah memberikan kemudahan pihak Toko Joice group dalam proses pengolahan data penjualan funiture dan memberikan informasi persedian barang, laporan penjualan dan pembelian pada pihak toko Joice group. Abstract

Fusarium oxysporum f. sp. cubense tropic race 4 (Foc TR4) strain which belong to Vegetative Compatibility Group (VCG) 01213 is the most devastating disease in global banana production. Validation of specific primer sets using the positive control (Foc TR4). In total, 50 isolates of Foc are collected from several banana production regions in Indonesia represent the group of VCG, races, genotype, cultivars, which are confirmed as Foc based on the tested using FocEf3 primer set, except Cjr-2 and Lmp-4 isolates. Foc-1/Foc-2 could amplify 34 Foc isolates included in Foc race 4. Three specific primer sets i.e. TR4-F/TR4-R, Six-1c, and TR4-F2/TR4-R1 are used to classify Foc isolates into Foc tropic race 4. TR4-F/TR4-R is known have the highest specificity as it could amplify 35 Foc isolates including positive controls (Foc TR4) compared to the other primer sets (Six-1c and TR4-F2/TR4-R1). This research indicates that there are a large number of diversity strains found in Foc isolates to be studied for further research. Race 4 of Foc (STR4 or TR4) is known to be widespread in several regions in Indonesia. Therefore, specific primer set development needs to be done to detect Foc TR4 and the most damaging strains on Foc TR4 based on molecular data.IntisariFusarium oxysporum f. sp. cubense ras 4 tropika (Foc TR4) yang termasuk ke dalam kelompok VCG 01213 merupakan patogen yang paling merusak dalam produksi tanaman pisang secara global. Validasi primer spesifik berbasis PCR menggunakan kontrol positif (Foc TR4). Total, 50 isolat Foc dikoleksi dari  beberapa daerah produksi pisang di Indonesia mewakili VCG, ras, genotipe dan kultivar yang dikonfirmasi sebagai isolat Foc berdasarkan pengujian menggunakan primer FocEf3, kecuali isolat Cjr-2 dan Lmp-4. Foc-1/Foc-2 dapat mengamplifikasi 34 isolat Foc yang termasuk ke dalam Foc ras 4. Selanjutnya tiga pasang primer spesifik yaitu TR4-F/TR4-R, Six-1c, dan TR4-F2/TR4-R1 digunakan untuk mengelompokkan isolat-isolat tersebut ke dalam isolat Foc ras 4 tropika. TR4-F/TR4-R diketahui memiliki spesifitas tertinggi karena dapat mengamplifikasi sebanyak 35 isolat Foc termasuk kontrol positif (Foc TR4) dibandingkan dengan primer lainnya (Six-1c dan TR4-F2/TR4-R1). Penelitian ini menunjukkan bahwa terdapat sejumlah besar keragaman strain yang terlihat pada isolat-isolat Foc tersebut untuk dapat dipelajari lebih lanjut. Ras 4 dari Foc (STR4 atau TR4) diketahui tersebar luas pada beberapa daerah di Indonesia. Oleh karena itu, perlu dilakukan pengembangan primer spesifik untuk mendeteksi Foc TR4 dan strain yang paling merusak pada Foc TR4 berdasarkan data molekuler.

Abstract

Isolation method of Pyricularia oryzae the causal agent of rice blast disease. Rice blast disease is a disease that reduces rice productivity and threatens global food reserves. The study of diversity and distribution of race fungal causing rice blast disease required a fungal isolates collection from different places and times. One of the challenges in collecting these fungi is the difficulty of isolation process. The purpose of this research was to study the proper isolation method of rice blast pathogen. The most appropriate isolation method of Pyricularia oryzae was to moisten the infected panicle, place on moist filter paper in a petri dish, and incubate plate for 2 days at room temperature under fluorescent lamp. Afterward, conidium was picked using sterile needle and transfered to potato dextrose agar without lactic acid.

Abstract

Banana is one of the important fruit crop in Village of Sidomulyo, Bantul, Yogyakarta. One of important diseases which become the constraint in development of banana is Fusarium wilt caused by Fusarium oxysporum f. sp. cubense (Foc). This fungus has high race diversity and  virulence, so that it required early detection for prevention and control of disease. This experiment was aimed to figure out race and virulence of Foc isolates from Village of Sidomulyo, Bantul, Yogyakarta. The 13 tested isolates were isolates of PR11, PKJ20, RU20, PR30, AH40, PKJ40, A41, RB42, PR43, RU51, A60, RP60, and A80. Race was molecularly detected using two types of primers, i.e. General Foc primer FocEf3 and specific primer for race 4 (Foc-1/Foc-2). Virulence test was performed on banana seedlings of Ambon Kuning cultivar using Completely Randomized Design (CRD) with 14 treatments and 4 repetitons. The observed parameters were external and internal symptoms, calculation of disease severity index and disease intensity. Data were analyzed using variance and further test of Duncan Multiple Range Test (DMRT) at 5 % level. The results showed that all isolates were Foc and 9 of 13 isolates were grouped into race 4, i.e. A80, RP60, PR11, A41, AH40, PKJ40, PR30, RB42, and PR43. The highest and lowest virulences were consecutively expressed by PR30, RB42, RU51, RP60, PR43, PKJ40, PR11, A41, AH40, RU20, PKJ20, A60, and A80, with severity index on leaves and rhizomes ranging 1.61-2.91 and 2.25-7, respectively. IntisariPisang merupakan tanaman buah unggulan di Desa Sidomulyo Kecamatan Bambanglipuro, Kabupaten Bantul. Salah satu penyakit penting yang menjadi kendala dalam pengembangan pisang adalah layu fusarium yang disebabkan oleh jamur Fusarium oxysporum f. sp. cubense (Foc). Jamur ini memiliki keragaman ras dan virulensi yang tinggi, sehingga deteksi dini diperlukan untuk pencegahan dan pengendalian penyakit. Penelitian ini bertujuan untuk mengetahui ras dan virulensi isolat Foc asal Desa Sidomulyo, Kecamatan Bambanglipuro, Kabupaten Bantul. Isolat yang diuji sebanyak 13 isolat, yakni isolat PR11, PKJ20, RU20, PR30, AH40, PKJ40, A41, RB42, PR43, RU51, A60, RP60, dan A80. Pengujian ras secara molekuler dengan menggunakan dua jenis primer yakni primer Foc in general FocEf3 dan primer spesifik ras 4 Foc-1/Foc-2. Uji virulensi pada bibit kultivar ambon kuning dengan menggunakan Racangan Acak Lengkap (RAL) yang terdiri dari 14 perlakuan dan 4 ulangan. Parameter yang diamati berupa pengamatan gejala luar dan gejala dalam, penghitungan indeks keparahan penyakit dan intensitas penyakit. Analisis data menggunakan sidik ragam dan uji lanjut Duncan Multiple Range Test (DMRT) pada taraf 5 %. Hasil pengujian menunjukkan bahwa semua isolat merupakan isolat Foc dan dari 13 isolat yang digunakan terdapat 9 isolat yang merupakan ras 4 yakni isolat A80, RP60, PR11, A41, AH40, PKJ40, PR30, RB42, dan PR43. Isolat yang memiliki virulensi tertinggi sampai terendah berturut-turut adalah PR30, RB42, RU51, RP60, PR43, PKJ40, PR11, A41, AH40, RU20, PKJ20, A60, dan A80, dengan indeks keparahan pada daun berkisar 1,61−2,91 dan indeks keparahan pada bonggol 2,25−7.
