Abstract

The main objective of this research was to obtain the effect of sheep serum (SS) and estrus sheep serum (ESS) on in vitro maturation oocyte and ovine fertilization. This study was carried out in experimental laboratory in animal reproduction laboratory, Faculty of Animal Husbandry, Padjadjaran University. Results showed that treatments significantly (P<0,05) influenced on maturation rate for germinal vesicle (GV), germinal vesicle breakdown (GVBD), metaphase-I (M-I) and metaphase-II (M-II), but no significantly (P>0.05) results observed on ovine in vitro fertilization (1, 2 and >2 pronuclei). Concentration of 10-20% ESS in CR1aa media were significantly (P<0,05) better than that of SS on maturation rate of ovine oocyte.       Key Words: Sheep Serum, Estrus Sheep Serum, Maturation, Fertilization

Abstract

The aim of this research was to know the effect of incubation time on capacitation and acrosome reaction of in vitro ovine spermatozoa. Twelve ejaculates from two Garut sheep were collected using artificial vagina and then evaluated macro and microscopically. After semen washing (centrifugation method), semen was diluted in Brackett and Oliphant (BO) medium and then incubated during 6 hours in incubator CO2, temperature 38.5ºC. Evaluation of capacitation and acrosome reaction was conducted on 0 hour, 0.5 hour, 1 hour, 2 hours, 4 hours and 6 hours incubation time. The result showed that incubation time had significant (P<0.05) effect on no capacitation sperm (pattern F), capacitation (pattern B) nad acrosome reaction (pattern AR). Incubation time had significantly (P<0.01) effect on subjective motility, life sperm and membrane integrity. Key Words: Ovine spermatozoa, incubation, capacitation, reaction, in vitro

Abstract

The objective of the research was to know the effect of serum type and concentration on in vitro maturation ovine oocyte. The result was expected to give information about the effect of serum type and concentration on in vitro maturation ovine oocyte, in order to find an alternative serum to reduce the use of serum from pharmacy industry. The result of research shows that concentration and type of serum significantly (P<0,05) influenced the maturation rate germinal vesicle (GV), germinal vesicle breakdown (GVBD), metaphase-I (M-I) and metaphase-II (M-II) phase. The result of research shows that concentration and tyoe of serum has no significant (P>0,05) influence on cumulus expansion rate grade 0, 1 and 2. Abstract

The objectives of the research were to know the effect of various hormone on in vitro maturation ovine oocyte. The result was expected to give information about the effect of various hormone on in vitro maturation ovine oocyte. The result of research show that treatment no significant (P>0.05) influenced on germinal vesicle (GV), germinal vesicle breakdown (GVBD) and metaphase-I (M-I) phase. The treatment P3 (CR1aa+FSH+ Estradiol+hCG) significant (P<0.05) increased maturation rate then P1 (CR1aa+FSH) dan P2 (CR1aa+FSH+ Estradiol).
