Abstract

Tujuan penelitian adalah mengetahui ultrastruktur oosit pasca vitrifikasi menggunakan transmisi elektron mikroskop. Ovarium dikumpulkan dari rumah potong hewan dan oosit dikoleksi dengan cara aspirasi dari folikel menggunakan jarum ukuran 18 G. Setelah dilakukan klasifikasi kualitas  oosit,  maka  oosit  yang  berkualitas  A  dimaturasi  secara  in  vitro. Oosit   hasil  maturasi  dipaparkan  ke  dalam  larutan  vitrifikasi  yang mengandung 30%  EG  + 0,5 M  sukrosa  dengan  lama  waktu paparan  3 menit,  oosit  dimasukkan  ke dalam  ministraw  0,25 cc  (French  straw), kemudian dipaparkan pada uap nitrogen selama 10 detik dan dimasukkan dalam kontainer nitrogen cair. Setelah 3 minggu ministraw diambil dan dilakukan penghangatan (warming) di udara selama 10 detik, dimasukkan dalam waterbath suhu 35 C selama 30 detik. Isi ministraw dituangkan ke dalam cawan petri dan oosit dibilas dua kali dengan  sukrosa 0,5 M.  Selanjutnya dilakukan pengamatan ultrastruktur oosit dengan transmisi elektron mikroskop. Hasil penelitian menunjukkan oosit hasil vitrifikasi mempunyai zona pelusida yang abnormal (fraktur), mem bran plasma mengalami lisis, beberapa butir korteks mengalami degenerasi dan perpindahan butir korteks di ruang perivitelin. Kesimpulan dari penelitian ini adalah terdapat perubahan ultrastruktur pada oosit setelah vitrifikasi.

Abstract

The purpose of the study was to determine the influence of the concentration of ethylene glycol (EG) and length of exposure to levels of bovine oocytes in vitro fertility. This research was conducted using a completely randomized design factorial 5x3 with 7 replicates. The first factor was the concentration of cryoprotectants EG 10, 20, 30, 40, and 50%. The second factor was the length of exposure 1, 3, and 5 minutes. The results showed that the concentration of EG and the length of exposure were effect on fertilized oocytes (P<0.05). The level of oocytes in vitro fertility after vitrification in 30% EG and long exposure to 3 minutes did not different (P>0.05) compared to fresh oocytes, while the EG treatment 10, 20, 40, and 50% significantly showed a lower fertility rate than the 30% EG (P<0.05). It was concluded that the concentration of EG and length of exposure were effect on fertilized oocytes. The highest oocytes in vitro fertility level were found in 30% EG concentration with 3 minutes exposure time.

Abstract

The aim of the research was to determine the optimum of centrifugation on the quality of Peranakan Ettawah goats spermatozoa prepared for in vitro fertilization preparation.The research of material was frozen semen ofPeranakan Ettawah goat produced by Artificial Insemination Center in Singosari with minimum post thawing motility in 40%. The method of the research was an experiment method with the four treatment, i.e: 1000 (P1), 1500 (P2), 2000 rpm (P3) of different centrifugation. The variable observes were spermatozoa motility, spermatozoa viability, and abnormal morphology of spermatozoa. Data obtain was analyzed statistically using Completely Randomize Design and continued with Tukey test. The result showed that on the upper layer of each treatment (P1, P2, and P3) obtained 55,72±3,55; 69,55±3,35; and 55,19±2,72% for spermatozoa motility, 62,99±5,87; 73,99±4,36; and 57,39±9,22% for spermatozoa viability, and 7,19±3,64; 8,84±2,65; and 6,40±3,00% for abnormal morphology of spermatozoa, descriptively. While on the lower layer the result showed 55,59±3,99; 68,63±3,88; and57,61±2,20% for spermatozoa motility, 70,05±7,47; 77,44±1,08; and 69,93±11,98% for spermatozoa viability, and 10,36±6,20; 9,55±3,27; and 8,09±2,80% for abnormal morphology of spermatozoa, desciptively. It was concluded that centrifugation in 1500 rpm showed the highest motility and viability on the upper layer and lower layer.

Abstract

ABSTRACT: Antibody against progesgerone was produced from serum of local rabbit following low dose of progesterone injection: While a control group (Control; n=5) was injected with Freunds adjuvant solution in aquadest, the treatment groups were either firstly injected with progesterone conjugated to Freunds Adjuvant (P--CFA, 150 p.l : 150 pl) or progesterone conjugated to Freunds Adjuvant and bovine serum albumin (P-CFA-BSA; 135 p;l : 150 tt1 : 15 gl). Twice boostering injections were adminstered using incomplete Freunds Adjuvant on day 14 and 52 after first immunization. Weekly bleeding for serum collection were done from 1 week following first booster immunization to week 10, Using ELISA technique it was shown that the antibody titer to progesterone after first and second booster immunization in the P-CFA groug was higher than Control- and P-CFA-BSA groups. Abstract

ABSTRACT:  The quality of semen inseminated to the dam might influence on the fer-tilization process and fetal growth during pregnancy. The aim of this research was to analysis the litter size, birth and weaning weight of Boer x PE goat crosses. Forty one parous and multiparous Peranakan Etawah (PE) does were naturally mated with selected Boer buck (NM-group), while the other 24 does were inseminated using artificial tech-nique using semen collected from the same buck as NM-group  and called as AI-group. The result showed no significant differences in litter size, birth weight and weaning weight between kids resulting from the does mated naturally and artificially insemina-tion technique (1.8±0.64 vs. 1.79±0.58; 2.93±0.568 vs. 2.96±0.397 kg; and 11.06 ± 2.001 kg vs. 11.31 ± 1.54 kg, for litter size, birth weight and weaning weight, respec-tively. It was concluded that mating technique in cross mating between PE does x Boer buck did not influence on the litter size and early growth phase of kids. Keywords: PE does, Boer buck, natural mating, artificial insemination

Abstract

This study was done to evaluate quality of Limousin’s semen during chilled preservation. The characteristic of spermatozoa were easily damaged, so that they were  necessary to protect viability of spermatozoa. Addition of extender was conducted to determine quality and viability of spermatozoa. Each extender were added Egg Yolk as diluents cryopreservation, the treatment were devided in to three groups, they were ; P0 (CEP-2 + 10% Egg Yolk), P1 (Tris Aminomethane + 20% EggYolk), P2 (Skim + 10% Egg Yolk). The result showed that Tris Aminomethane + 20% Egg Yolk gave the best effect on maintaining quality of spermatozoa (motility, viability and abnormality). The motility in three extender at 24h was high significant different (P&lt;0,01). Furthemore, the Viability and abnormality showed high significant different (P&lt;0,01) since first time evaluation at 0h until 24h. Tris Aminomethane showed the best result among others due to the addition of 20% Egg Yolk, as we know that Egg Yolk was an extracellular cryoprotectan to protect and nourish spermatozoa during chilling preservation.   Keyword : Egg Yolk, CEP-2, Tris Aminomethane, Skim,

Abstract

The objective of this research is to know the influence of vitrification method toward thecondition of bovine oocytes cytoskeleton based on microtubules using the immunohistochemistrytechnique. Oocytes classified in two treatments such as fresh oocytes (control) and oocytes withvitrification treatments of 10, 20, 30, 40, and 50% of ethylene glycol (EG). Data were analyzed bydescriptive analyzing. The results of this research indicated that vitrification treatments were affected tomicrotubules organization of oocytes. Concentration of EG 10, 20, 40, and 50% of EG could not repaircytoskeleton after vitrification, but cytoskeleton structure could be protected by EG 30%.

Abstract

Penelitian ini dilaksanakan pada bulan Mei sampai Juni. Tujuannya adalah untuk mengetahui pengaruh bangsa pejantan Simmental dan Limousin terhadap pertumbuhan pedet hasil Inseminasi Buatan (IB) di Kecamatan Bantur, Kabupaten Malang. Materi yang digunakan dalam penelitian ini adalah pedet hasil IB dengan indukan Limpo yang disilangkan dengan pejantan Simmental 50 ekor dan Limousin 50 ekor pada umur 3−3,5 bulan. Metode yang digunakan dalam penelitian ini adalah survey secara purposive sampling. Data dianalisis dengan menggunakan analisis deskriptif dan uji beda. Hasil penelitian menunjukkan bahwa bangsa pejantan berpengaruh terhadap pertumbuhan pedet yang dihasilkan. Berbeda nyata pada bobot lahir, bobot badan 3-3,5 bulan, dan lingkar dada dari pejantan Simmental x Limpo berturut-turut adalah 35,46+2,86 kg; 134,42+54,61 kg; 120,74+10,64 cm, dan dari pejantan Limousin x Limpo  34,16+2,10 kg; 116,01+14,52 kg; 115+12,06 cm. Tidak berbeda nyata pada panjang badan dan tinggi gumba dari pejantan Simmental x Limpo berturut-turut adalah 99,02+8,81 cm; 105,38+10,13 cm, dan dari pejantan Limousin x Limpo 97,08+7,41 cm; 105,38+8,31 cm. Pedet hasil persilangan Simmental x Limpo memberikan performans produksi yang lebih baik dari pada Limousin x Limpo. Simmental merupakan salah satu pejantan yang berpengaruh cukup tinggi terhadap pertumbuhan pedet yang dihasilkan. Kata kunci: Pedet, Limousin, Simmental, Limpo, Performans Produksi

Abstract

ABSTRAK Tujuan dari penelitian  untuk mengetahui  penampilan reproduksi sapi betina Peranakan Ongole dan Peranakan Limousin yang meliputi Service per Conception, Days Open, Calving Interval, dan Conception Rate. Materi yang digunakan dalam penelitian ini adalah Sapi Peranakan Ongole   50 ekor dan Sapi Peranakan Limousin  50 ekor. Metode yang digunakan dalam penelitian ini adalah studi kasus, pengumpulan data dilakukan primer dan sekunder.  Variabel yang diamati adalah Service per conception (S/C), Days open (DO), Calving interval (CI), Conception rate (CR),  Hasil penelitian menunjukkan S/C sapi Peranakan Ongole 1,28 dan sapi Peranakan Limousin 1,34. Days Open (DO) sapi Peranakan Ongole 130,27±20,99  hari, sedangkan pada sapi Peranakan Limousin sebesar 149,32±24,19 hari. Conception Rate sebesar 75,34% pada sapi Peranakan Ongole dan 66% pada sapi Peranakan Limousin dan untuk CI pada sapi Peranakan Ongole sebesar 414,97±25,53  hari sedangkan pada sapi Peranakan Limousin sebesar 433,67±24,39 hari. Disimpulkan bahwa terdapat perbedaan penampilan reproduksi sapi Peranakan Ongole dan Peranakan Limousin.   Kata Kunci: Service per Conception, Days Open, Calving Interval and Conception Rate ABSTRACT The purpose of this study was to know the difference of reproductive performans Filial Ongole and Filial Limousine covering Service per Conception, Days Open, Calving Interval, and Conception Rate. The materials used in this study is to Filial Ongole by the number 50 objects and Filial Limousin Cattle with the number of 50 objects. The method used in this research is the gathering of primary and secondary data. The observed variables are Service per conception (S / C), Days open (DO), Calving interval (CI), Conception rate (CR), The experiment showed that  S / C , DO, CR and CI   of Filial Ongole  were 34 1.28;130.27 ± 20.99 days, 75.34%;  414.97 ± 25.53 days and for Filial Limousin were  1,34; 149,32±24,19 days; 66 %;433,67±24,39days respectively.  It was concluded  that there was   difference of  reproductive performans between Filial Ongole and Filial Limousin. Abstract

The objective of this research was to determine the effect of the simple modified freezing method, 1°C/minute freezing rate with different diluter ration on a post-thawing quality of local goat sperm namely Peranakan Etawah (PE). This work is aimed to study the quality of post-thawing sperm and to characterize the calcium intensity profile of both fresh and post thawing goat sperm. The method used is the experimental design of a laboratory. Freezing semen was performed in 2 main temperatures of -45°C then -196°C respectively using Mr. Frosty (®) System. Early Sperm characters of Ca+2 intensity was performed by Confocal Laser Scanning Microscope (CLSM) through Fluo-3 staining and Ca++ intensity was analysis descriptively. The result showed that post-thawing qualities are considered as good as standard qualities, at least, more than 40% based on Indonesian National Standard (SNI, 2014). The different level diluents commercial of Andromeda used were influenced highly significant (P&lt;0.01). The best diluents ration is 1:4 (v/v) for final sperms stocked at -196°C. However freezing sperm conserved in -196°C is better than in -45°C. Meanwhile, the sperm characters of two condition showed the important variation of Ca+2 intensity, with the length of region measurement of 39.06±4.595 and 32.696±9.011 µm each.  It was concluded that the calcium intensity pattern was varied more and higher in fresh sperm than in freezing sperms. This simple modified method of a freezing system was considered as a feasible alternative method for goat semen in a reason for both for sperm post-thawing quality and practical purposes.
