Abstract

ABSTRAK Telah dilakukan penelitian tentang akumulasi logam berat krom dan timbal dalam sedimen estuaria sungai Matangpondo Palu dengan tujuan untuk mendapatkan informasi besarnya konsentrasi logam krom dan timbal yang terdapat dalam sedimen.  Pengambilan contoh dilakukan pada beberapa titik di sekitar muara sungai Matangpondo di Teluk Palu dengan menggunakan corel sample (modifikasi peneliti). Hasil penelitian menunjukkan bahwa keberadaan logam krom dan timbal berasal dari aktivitas alamiah dan aktivitas antropogenik. Secara parsial kandungan logam berat krom pada salah satu titik contoh telah melampaui batas konsentrasi maksimum yang diperbolehkan (51 mg/kg) dengan rerata 39,24±23,67 mg/kg dan secara holistik belum melampuai NAB tersebut, dengan demikian perairan estuaria sungai Matangpondo masih aman dari pencemaran oleh logam krom. Sedangkan konsentrasi logam timbal dalam sedimen belum melampaui NAB-nya (33 mg/kg) dengan rata-rata 15,89± 7.43 mg/kg. Kata Kunci: Akumulasi, logam, estuaria ABSTRACT The research about heavy metals accumulation in estuary sediment of Matangpondo River, Palu have been done to get information of chrome and lead concentration in sediment. The sample is taken on some area around Estuary River by using Corel sample (researcher modification). The Research result shows that chrome and lead come from natural and anthropogenic activity. By the partial, chrome content of one of area samples has exceeded enabled maximum concentration (51 mg / kg) with the average 39.24±23.67 mg / kg but by holistic has not yet. Thereby estuary territorial of river Matangpondo is still peaceful from chrome metal contamination. While, lead metal in sediment has not yet exceeded of enabled maximum concentration (33 mg / kg) with the mean 15.89± 7.43 mg / kg. Key word: Accumulation, metal, estuary

Abstract

The use of epichlorohydrin to improve the stability of chitosan adsorbent has been carried out. The purpose of this study was to determine the stability of the chitosan adsorbent after modified into epichlorohydrin cross-linked chitosan and ionic cobalt imprinted adsorbent chitosan cross-linked epichlorohydrin. Adsorbent prepared by homogeneous gel formation of chitosan in acetic acid medium and then modified into epichlorohydrin cross-linked chitosan beads and ionic cobalt imprinted adsorbent chitosan cross-linked epichlorohydrin. Stability of the adsorbent was determined by calculating the percentage of insoluble material after stirring the adsorbent in nitric acid medium at pH 1, 2, and 3 for 12 hours. The results of FTIR spectra show a shift of the absorption band of chitosan were compared with epichlorohydrin cross-linked chitosan and ionic cobalt imprinted adsorbent chitosan cross-linked epichlorohydrin. The use of epichlorohydrin as cross-linked agent may increase the stability of the adsorbent to maintain the percentage of insoluble material by 89% at pH 3 for epichlorohydrin cross-linked chitosan adsorbent and 96% at pH 3 for ionic cobalt imprinted adsorbent chitosan  cross- linked epichlorohydrin.

Abstract

A research has been done to synthesize hexa-p-tert-butylhexaestercalix[6]arene using p-tert-butylcalix[6]arene as a starting material. The synthesis was carried out by esterifying p-tert-butylcalix[6]arene. Synthesis products were identified and characterized by melting point, TLC, FTIR, 1H-NMR, and 13C-NMR spectrometers. Synthesis products was obtained a white solid powder (68.45% recovery), melting point 349-351 °C, TLC (SiO2, ethanol: ethyl acetate = 1: 1 v/v, Rf = 0.90). The results of the analysis  of synthesis products with FTIR, 1H-NMR, and  13C-NMR spectrometer showed that the synthesis products compound is 5, 11, 17, 23, 29, 35-hexakis (p-tert-butyl)-37, 38, 39, 40, 41, 42-hexakis (ethoxy-carbonylmethoxy) calix[6]arene.

Abstract

This study aimed to 1) determine the degree of saturation of ammonium sulfate right to extract and purify the bioactive protein from shells (Atactodea striata), 2) determine the fraction of active protein from shells (Atactodea striata) as a potential antibacterial. In this study used the Lowry method for determine protein concentration and agar diffusion method for antibacterial activity. Extraction of shells Atactodea striata was conducted by making use of buffer solution (0,1 M Tris-HCl of pH 8.3, 2 M NaCl, 0.01 M CaCl2, 1 % β-mercaptoethanol, and  0.5 % Triton X-100). Purification of proteins by  precipitation using ammonium sulfate at saturation level 30 %, 50 %, 70 %, and 90 %. The results showed that the protein concentration of the crude   extract is 41.6354 mg/mL. At fractionation rate of 0-90% saturation showed the highest concentration of protein found in fractions with 70% saturation level is 56.4184 mg/mL. The testing of antibacterial activity against Staphylococcus aureus showed that crude extracts and protein fractions Atactodea striata is considered effective as an antibacterial. The highest bioactivity during 24-hour incubation in protein fractions obtained by ammonium sulfate saturation level of 50% is 25.17 mm. Whereas the lowest activity was obtained at 90% saturation level is 14.05 mm. Bioactivity against Escherichia coli after incubation for 24 hours has the highest activity in the protein fraction with 30% ammonium sulfate saturation is 15.12 mm. Whereas the lowest activity was showed at 70% saturation level is 10.30 mm. After the observation was continued for 48 hours on both test bacteria, which formed a clear area becomes cloudy. It shows that the crude extract and fractions of protein tend to be bacteriostatic against Staphylococcus aureus and Escherichia coli.

Abstract

This research aims to utilize isolated chitin from shrimp waste to develop histamine biosensors basedon diamine oxidase (DAO) enzyme electrode with cyclic voltammetry. DAO enzyme trapped in the chitin-cellulose acetate membranes with various comparisons were layered on the Pt electrode. Histamine will be oxidized by the DAO enzyme produces aldehydes and H2O2 which acts as an electron transfer mediator. Biosensor performance is influenced by several factors, especially the concentration and composition of electrode  membranei.Comparison of chitin-cellulose acetate used in this study were 1:1, 2:1 and 3:1. Isolated chitin from the shrimp waste is chemically obtainedrendamen of 23.6%, and characterization of electrode membrane by FTIR and cyclic voltammetry showed that the DAO enzymes electrode with chitin-cellulose acetate membrane 2:1 is the best composition.

Abstract

This study aims to determine the design pattern ESI-Pb (II) with ionophores pt-butilkaliks [4] arena. In this study, ion selective electrode is designed for the determination of lead (II) (ESI-Pb (II)) using ionophores pt-butilkaliks [4] arenas and diimmobilasi on a matrix of polyvinyl chloride (PVC) in the membrane ESI-Pb (II). The results showed a design pattern ESI-Pb (II) with ionophores pt-butilKaliks [4] arena is best to ESI with membrane composition (% by weight), namely: (pt-butilKaliks [4] arena: KTCPB: DOS: PVC) is 3 : 2: 60: 35 with the Nernst factor: 27.61 mV / decade

Abstract

The effect of PVC (Polyvinylchloride)-Based Membrane Composition to Ion Selective Electrode (ISE)-Hg(II) Performance using Ionophore DBA218C6 (N,N-Dibenzyl-1,4,10,13-tetraoxa-7,16-diazacyclo octadecane),  Plasticizer NPOE (Nitrophenyl Octhyl Ether), Anionic Site KTCPB (Potassium Tetrakis (4-chloro phenyl) borate) have been performed. Membrane compositions used were:(a) PVC (30 mg), NPOE (60 mg), DBA218C6(6 mg) and KTCPB (4 mg); (b) PVC(30 mg), NPOE (60 mg), DBA218C6(7 mg) and KTCPB (3 mg); (c) PVC (30 mg), NPOE (59 mg), DBA218C6 (8 mg) and KTCPB( 3 mg). The concentration range of interference ions Zn(II), Cd(II) and Pb(II)  were 1.0 x 10-3 - 1.0 x 10-1 M. ISE-Hg(II) performance for membrane composition of 30 : 60 : 6 : 4 was 26.34 mV per decade (Nernstian Slope value) on Hg(II) concentration range of 1.0x10-6 - 1.0 x 10-1 M , membrane composition of 30 : 60 : 7 : 3 was 27.71 mV per decade on Hg(II) concentration range of 1.0 x10-6 - 1.0 x10-1 M, and membrane composition of 30 : 59 : 8 : 3 was 28.52 mV per decade on Hg(II) concentration range of  1.0 x10-6 - 1.0 x10-1 M with activity between pH 1.0-3.0. The concentration of interference ions : Zn(II), Cd(II) and Pb(II) in the range of 1.0 x 10-3 - 1.0 x 10-1 M with the ratio of the primary ion to interference ions of 4 : 1 gave real effect. As results, selectivities and sensitivities between ISE-Hg(II) and Ionophore DBA218C6 could be determined by PVC-Based Membrane Composition and the effect of Zn(II), Cd(II) and Pb(II)  interference ions was observed in the concentration of 1,0 x10-3 - 1,0 x10-1 M.   Keywords: membrane composition effect, ionophore DBA218C6, ISE-Hg(II)
