Abstract

Tujuan penelitian ini adalah untuk mengisolasi dan menguji potensi antibakterial bakteri asam laktat dari ikan Kerapu Macan (Epinephelus fuscoguttatus) terhadap bakteri patogen Vibrio alginolyticus. Bakteri tersebut diisolasi dari usus ikan dengan metode sebarulas pada media agar MRS dan GYP+CaCO3. Koloni yang tumbuh diidentifikasi  berdasarkan  karakter  morfologi,  biokimia  dan  fisiologi.  Aktivitas  antibacterial  terhadap  V. alginolyticus diuji dengan metode difusi kertas cakram pada media Zobell 2216E lapis ganda, diameter zona jernih disekitar kertas cakram diukur dengan kaliper. Pada medium agar GYP+CaCO3 didapatkan 21 isolat yang menampakkan zona jernih disekitar koloninya. Dari karakteristik morfologi, biokimia dan fisiologi dari isolat-isolat tersebut semua isolat dapat dikategorikan kedalam genus Lactobacillus. Tetapi, dari uji aktivitas antivibrio hanya 20 isolat menunjukkan zona hambat yang berkisar dari 14,0-21,5 mm, dimana tiga isolat menunjukkan aktivitas tertinggi, yaitu berturut-turut isolat KSBU 12C, KSBU 13D dan KSBU 5Da.Kata kunci: Isolasi, bakteri asam laktat, antivibrio, Kerapu Macan The objectives of this research were to isolate and examine antibacterial potency of lactic acid bacteria from Tiger Grouper (Epinephelus fuscoguttatus) against pathogen, Vibrio alginolyticus. The bacteria were isolated from the fish intestine by spread plate method on MRS and GYP+CaCO3  agar media. The grown colonies were identified based on morphological, biochemical and physiological characters. Antibacterial activity against V. alginolyticus was examined by the paper disc diffusion method on Zobell 2216E double layer agar, and clear zone diameter around the paper disc was measured by using caliper. Twenty one isolates  with  clear zone around the colonies were obtained from the GYP+CaCO3 agar. Morphological, biochemical and physiological characteristics of the colonies and cells indicated that all isolates might be categorized into Lactobacillus. However, there were only twenty isolates showed inhibition zones from 14.0-21.5 mm in antivibrio activity test, of which the highest activity was indicated by three isolates namely KSBU12C, KSBU 13D and KSBU 5Da, respectively. Abstract

  Tujuan penelitian ini adalah untuk menyeleksi isolat bakteri asam laktat (BAL) yang telah diisolasi dari ikan Kerapu Macan (Epinephelus fuscoguttatus) berdasarkan toleransi terhadap pH dan bile salts, patogenisitas dan kemampuannya menekan pertumbuhan Vibrio alginolyticus pada uji ko-kultur. Hasil penelitian menunjukkan bahwa dua puluh isolat BAL yang telah diuji antagonismenya terhadap V. alginolyticus pada penelitian terdahulu, dapat hidup pada pH rendah 3 dan tumbuh pada konsentrasi bile salts 0,5% kecuali isolat KSBU 13D.  Semua isolat tidak patogen terhadap ikan Kerapu Macan setelah diinfeksi BAL pada konsentrasi 107 cfu/ml secara intraperitoneal (IP). Pada uji ko-kultur, dari 6 isolat BAL yang dipilih, tiga isolat (KSBU 12C, KSBU 5Da, KSBU 9) berturut-turut mampu menekan pertumbuhan V. alginolyticus, dan penambahan konsentrasi BAL terbaik adalah 107 cfu/ml. Kata kunci: bakteri asam laktat, vibriosis,  patogenisitas, ko-kultur, kerapu macan The research objective was to select lactic acid bacteria which previously isolated from Tiger Grouper (Epinephelus fuscoguttatus) based on pH and bile salts tolerances, pathogenicity test and ability to decrease the growth of Vibrio alginolyticus in co-culture test. The result shows that twenty isolates which had also been examined their antagonisms against V. alginolyticus previously, were able to survive relatively low pH  values 3 and able to survive at high bile salts concentrations 0.5% except KSBU 13D isolate.  All isolates were nonpathogenic to Tiger Grouper after being intraperitoneally infected with the lactic acid bacteria at concentration 107 cfu/ml. In co-culture test, V. alginolyticus was reduced significantly by three isolates (KSBU 12C, KSBU 5Da, KSBU 9) from 6 isolates selected, and the best additional bacterial concentration was  107 cfu/ml. Effects Of pH, Temperature And Salinity In Growth And Organic Acid Production Of Lactic Acid Bacteria Isolated From Penaeid Shrimp Intestine

Abstract

Bakteri asam laktat telah lama dikembangkan sebagai probiotik. Penentuan kondisi lingkungan yang optimum untuk pertumbuhan sel serta asam organik memberikan gambaran aktivitas optimum untuk kinerja probiotik baik dalam sistem fisiologi inang maupun dalam sistem bioproses untuk produksi sel dan metabolit. Penelitian ini bertujuan untuk mengetahui pengaruh faktor lingkungan (pH, suhu dan salinitas) terhadap pertumbuhan dan produksi total asam organik tiga isolat bakteri asam laktat yang telah diseleksi dari intestinum udang penaeid. Eksperimen menggunakan  medium deMan, Rogosa and Sharpe (MRS) cair. Perlakuan pH awal meliputi  nilai pH 4, 5 dan 6. Perlakuan suhu meliputi suhu 25, 30 dan 35OC serta perlakuan salinitas  meliputi salinitas 0,75 %, 1,5 % dan 3 %.  Setiap interval 6 jam dilakukan pengambilan sampel kultur bakteri dan penghitungan pertumbuhan berdasarkan perubahan optical density (pada panjang gelombang 600 nm) sedangkan produksi asam laktat dianalisis dengan metode titrimetrik menggunakan NaOH 1 N sebagai larutan titrasinya. Berdasarkan hasil penelitian disimpulkan bahwa suhu, pH awal dan salinitas berpengaruh terhadap pertumbuhan dan produksi asam organik. Nilai kondisi lingkungan terbaik untuk pertumbuhan dapat berbeda dengan nilai terbaik untuk produksi asam organic. Hal ini ditunjukan oleh nilai laju pertumbuhan dan produksi asam laktat tertinggi dari tiga isolat uji terjadi pada suhu, pH awal dan salinitas yang berbeda.  Isolat L12 tumbuh optimum pada suhu 30oC, pH awal 6 dan salinitas 0,75%. Isolat L14 tumbuh optimum pada suhu 30oC, pH awal 6 dan salinitas 1.5%. Isolat L 21 tumbuh optimum pada suhu 30 oC, pH awal 6 dan salinitas 1.5%. Kata kunci: bakteri asam laktat, suhu, pH, salinitas, asamorganik, pertumbuhan, Lactic acid bacteria are widely distributed in intestinal tracts of various animals where they live as normal flora.Strains of lactic acid bacteria are the most common microbes employed as probiotics, The optimum condition for growth are important to mass production and to determined parameters most suitable for growth. The effects of  temperature, pH and salinity on the growth and production of lactic acid from the three shrimp intestinal lactic acid bacteria isolates were conducted using bacth culture in a flask. These variables for growth were determined based on the growth curves and lactic acid production. Data from the flask batch experiment demonstrated that the best initial pH and temperature  for growth of isolat L12 ,L14 and L21 were found to be pH 6 and 30 OC.  Salinity (NaCl concentration) 0,75% were the best for growth of isolat L12. Salnity  1,5 % were best for growth of isolat L14 and L21. Key words : growth, temperature, pH, salinity, lactic aid bacteria

Abstract

Seiring dengan perkembangan zaman dan teknologiyang semakin pesat, sebagian besar masyarakat telah menggunakan komputer diberbagai bidang pekerjaannya. Namun masalah yang dipantau pada sebuah Pusat Perbelanjaan adalah proses pencarian barang yang masih manual dan belum memiliki basis data,sehingga sulit dalam meninjau perubahan data yang telah terjadi. Untuk itulah penelitian dilakukan dan dibuatlah Aplikasi Pencarian Barang  ini, yang kemudian digunakan untukmengefisiensikan waktu dan memudahkan pencarian barang yang diinginkan olehPelanggan, pada gudang penyimpanan barang yang dilakukan secara komputerisasi. Piranti lunak yang digunakan untuk membuatAplikasi Pencarian Barangini adalahMacromedia Dreamweaver 8 dan PHP. dengan metode penelitian yang digunakan terdiridari metode pengumpulandata danmetode pengembangan sistem, yang terdiri dari metode perancangan basis data konseptual, logical, fisikal, dan perancangan aplikasi. Di akhir tahapan, dilakukan implementasi pada Pusat Perbelanjaantersebut dandipantautingkat keberhasilannya.Diharapkanaplikasi inidapat menjawab kebutuhan danmemecahkan permasalahan yang ada.

Abstract

The purpose of this study was to develop and implement model and the learning of Mathematics Statistics I through modification method of learning with concept maps through analogy approach, with expectations of the quality of learning can be improved, In line with the objectives of this research study it is research and development with 4-D models. While in practice, This research uses descriptive quantitative methods investigate the effect of concept map of learning through analogy approach (developed) to the learning achievement of students in the course of Mathematics Statistics I. The populations in this study were all students of study program Mathematics Education, Faculty of Teacher Training and Education, University of Eleven March Surakarta. While the sample used in this study were 70 students randomly from the students who took the eye Mathematical Statistics I lectures the academic year 2010/2011.Of samples 35 students placed in the experimental class and 35 other students placed in the control class. The results of this study were: (1) has developed the learning in the form of teaching materials and lesson plans for Mathematical Statistics I course is refers to a concept map of learning through analogy approach; (2) There is positive influence on our concept of learning through the eyes of the analogy approach Mathematical Statistics I lectures in the Faculty of Mathematics Education Study Program Teacher Training and Education Sebelas Maret University of Surakarta.

Separation of Radiocopper 64/67Cu from the Matrix of Neutron-Irradiated Natural Zinc Applicable for 64Cu Production

Abstract

Radioisotope 64Cu is a promising radiometallic-isotope for molecular-targeted-radiopharmaceuticals. Having a half-life of 12.70 hours and emitting β+-radiation (Eβ+ = 0.6531 MeV) as well as β-ray (Eβ- = 0.5787 MeV), it is widely used in the form of biomedical-substrate-radiopharmaceutical for positron emission tomography (PET) diagnosis and simultaneously for targeted radiotherapy of cancer. The potential needs on the availability of 64Cu-labeled pharmaceuticals for domestic nuclear medicine hospitals lead to a necessity for the local production of carrier-free 64Cu using BATAN’s G.A. Siwabessy reactor because of the technical and economical constraints in the production using BATAN’s cyclotron. The presented work is accordingly to study whether the radioisotope 64Cu can be produced and separated from the matrix of post-neutron-irradiated-natural zinc. This study is expected can be further improved and implemented in production technology of carrier-free 64Cu based on 64Zn (n,p) 64Cu nuclear reaction exploiting the fast neutron fraction among the major thermal fraction due to unavailability of fast-neutron-irradiation facility in the BATAN’s G.A. Siwabessy reactor. The solution of post-neutron-irradiated-natural zinc in 1M acetic acid was loaded into Chelex-100 cation exchanger resin column to pass out the Zn/Zn* fraction whereas the Cu* fraction which remained in the column was then eluted out from the column by using 1.5 M HCl and loaded into the second column containing Dowex-1X8 anion exchanger resin. The second column was then eluted with 0.5 M HCl. The collected eluate was expected to be zinc-free Cu* fraction. It was observed from the half-life and the γ-spectrometric analysis that radioactive copper-64Cu containing 67Cu was produced by neutron activation on the natural Zn-foil target and can be separated from the target matrix by the presented two-steps-column-chromatographic separation technique. The radioactivity measurement showed that wrapping the Zn target with cadmium foil increased the activity of radioactive copper and, thus, the Cu*/Zn*-ratio.Received: 28 June 2011; Revised: 21 February 2012; Accepted: 24 February 2012

Abstract

SIMULATIONS ON NICKEL TARGET PREPARATION AND SEPARATION.OF Ni(II)-Cu(II) MATRIX FORPRODUCTION OF RADIOISOTOPE64Cu64Ni (p,n) 64  and retained on the column while the nickel was kept in the form of Ni2+  2+  2+ and CuCl while the nickel was totally in the form of Ni2+  while the nickel was found as both Ni2+ and NiCl   while the nickel was mostly in the form of Ni2+. The retained CuCl was then changed back into Cu2+     Keywords 64 Cu, Anion exchange chromatography.: Nickel target preparation, Radioisotope Cu-64, Separation of Ni(II)-Cu(II) matrix, Nuclear reaction of 64Ni(p,n) cation form andeluted out the column by using HCl 0.05 M. The 42– 4 2–.The best condition of separation was in HCl 8 M in which the radioactive copper was mostly in the form of CuCl 42– 42– . In the condition ofHCl 9 M, the radioactive copper was mostly in the form of CuCl 42– cation. It was found that the electroplating result from the acidic solution was more satisfied than that from the basic solution. By conditioning the matrix solution at HCl 6 M, the radioactivecopper was found in the forms of Cucation and eluted off from the column. The retained radioactive copper was then eluted out the column in the condition of dilute HCl changingback the copper anion complex into Cu42– Cu. The nickel target preparation was performed by means of electroplating method using acidic solution of nickel chloride - boric acid mixture and basic solution of nickel sulphate – nickel chloridemixture on a silver- surfaced-target holder. The simulated solution of Ni(II) – Cu(II) matrix was considered as thesolution of post-proton-irradiated nickel target containing both irradiated nickel and radioactive copper, but in thepresented work the proton irradiation of nickel target was omitted, while the radioactive copper was originallyobtained from neutron irradiation of CuO target. The separation of radioactive copper from the nickel target matrixwas based on anion exchange column chromatography in which the radiocopper was conditioned to form anioncomplex CuClg-spectrometric analysis showed a single strong peak at 511 keVwhich is in accord to g-annihilation peak coming from positron decay of Cu-64, and a very weak peak at 1346 keVwhich is in accord to g-ray of Cu-64.. Abstract

Pendekatan, strategi, metode, dan teknik pembelajaran pada hakikatnya dapat dibedakan tetapi tidak dapat dipisahkan. Keempat istilah tersebut merupakan satu kesatuan dalam pembelajaran. Pendekatan, strategi, metode, dan teknik pembelajaran yang akan dan/atau sedang digunakan dapat diketahui dari langkah-langkah pembelajaran yang telah tersusun dan/atau sedang terjadi. Pendekatan pembelajaran adalah cara umum dalam memandang pembelajaran. Sedangkan strategi pembelajaran adalah ilmu dan kiat di dalam memanfaatkan segala sumber belajar yang dimiliki dan/atau yang dapat dikerahkan untuk mencapai tujuan pembelajaran yang telah ditetapkan. Metode mengajar adalah berbagai cara kerja yang bersifat relatif umum yang sesuai untuk mencapai tujuan pembelajaran tertentu dan teknik pembelajaran adalah ragam khas penerapan suatu metode sesuai dengan latar penerapan tertentu. Teknik pembelajaran mengambarkan langkah-langkah penggunaan metode mengajar yang sifatnya lebih operasional. Faktor-faktor yang perlu diperhatikan dalam penentuan teknik pembelajaran di antaranya adalah kemampuan dan kebiasaan guru, ketersedian sarana dan waktu, serta kesiapan siswa. Faktor-faktor yang perlu dipertimbangkan dalam memilih strategi pembelajaran ialah tujuan pembelajaran, jenis dan tingkat kesulitan materi pelajaran, sarana, waktu yang tersedia, siswa, dan guru.

Abstract

Abstract: The objective of this research are to investigate: (1) which one that is more effective in teaching geometry, the STAD type of cooperative learning model with Power Point media or that with Geometrical Model media or that with Conventional media, (2) which learning style results in a better learning achievement and (3) which media used in the STAD learning model results in a better learning achievement in each learning style of the students.This research used the quasi-experimental method. The population of this research was the 9th-grade students of the state junior secondary schools in Ngawi regency in the academic year of 2012/2013. The samples of this research consisted of 250 students who were divided into 85 students of the first experiment class, 83 students of the second experiment class, and 82 students of the control class. The samples were taken by using stratified cluster random sampling. The data of this research were gathered through documentation, test, and questionnaire. The hypothesis of this research were tested by using unbalanced Two-way Analysis of Variance. Conclusions drawn are as follows: (1) the STAD learning model with the Power Point media is better than the STAD learning models with the Geometrical Model and the Conventional media whereas the STAD learning model with the Geometrical Model media is equal to the STAD learning model with the Conventional media; (2) the students with the kinesthetic learning style have a better learning achievement than those with the visual and auditory learning styles whereas the students with the visual learning style have an equal learning achievement to those with the auditory learning style; (3) the STAD learning model with the Power Point media results in a better learning achievement than that with the Conventional media whereas the STAD learning model with the Power Point media results in an equal learning achievement to that with the Geometrical Model media and the STAD learning model with the Power Point media results in an equal learning achievement to that with the Conventional media in the visual learning style; (4) the STAD learning model with the all of the three types of media results in an equal learning achievement in the auditory learning style; and (5) the STAD learning model with the Geometrical Model media results in a better learning achievement than that with the Conventional media whereas the STAD learning model with the Power Point media results in an equal learning achievement to that with the Geometrical Model media and the STAD learning model with the Power Point media results in an equal learning achievement to that with the Conventional media in the kinesthetic learning style.Keywords: STAD, Power Point media, Geometrical Model media and learning style.

Abstract: The objectives of this research are to investigate: (1) wich learning model of Think Pair Share (TPS), Team Assisted Individualization (TAI), or conventional learning model results in a better learning achievement in Mathematics, (2) which learning interest of the high learning interest, the moderate learning interest, and the low learning interest results in a better learning achievement in Mathematics,(3) in the students with the high, moderate, and low learning interests, wich learning model of Think Pair Share (TPS), Team Assisted Individualization (TAI), and conventional learning model results in a better learning achievement in Mathematics. This research used the quasi experimental research method with the factorial design of 3x3. The population of the research were the students of Vocational High School (SMK) in Ponorogo regency on Academic Year 2011/2012. The samples of the research were taken by using the stratified cluster random sampling technique. It was conducted at SMK 1 of Ponorogo, SMK Bakti of Ponorogo, and SMK Sore 1 of Ponorogo. The samples included two experimental classes and one control class of each of the schools. The size of the sample was 275 students consisted of 84 students in the first experimental class, 105 sudents in the second experimental class and 86 students in control class. The data of the research were gathered through mathematics achievement tes and quetionary of learning interest. The data was analyzed by using two-way analysis of variance with.The results of the reseach are as follows: (1) both TPS and TAI result in the same good learning achievement in Mathematics, and result in a better learning achievement than the conventional one does; (2) the students with the higher learning interest have a better learning achievement in Mathematics than those with the moderate learning interest and the low learning interest, but the students with the moderat learning interest have the same learning achievement in Mathematics those with the low learning interest; (3) in the students with the high, moderat, and low learning interest, both TPS and TAI result in the same good learning achievement in Mathematics, and result in a better learning achievement than the conventional one does.Key words: Think Pair Share, Team Assisted Individualization, Conventional, Learning Interest
