Abstract

Percobaan ini dilaksanakan di Laboratorium Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Brawijaya Malang dan Balai Inseminasi Buatan Singosari pada bulan Agustus sampai Desember 1998. Tujuan penelitian ini adalah untuk mencari metode pembekuan spermatozoa hasil seleksi jenis kelamin dengan menggunakan filtrasi sephadex G-200. Percobaan ini terdiri atas 10 kali ulangan dengan dua perlakuan yaitu krioprotektan ekstraseluler TCM- 199+ 10% serum+ 14% kuning telur dan tris aminomethan- kuning telur. Parameter yang diukur meliputi persentase motilitas, hidup, kapasitasi dan reaksi akrosom. Media TCM-199 kuning telur lebih dapat mempertahankan mutu semen dibandingkan dengan tris aminomethan kuning telur, dan proses pembekuan meningkatkan kapasitasi dan reaksi akrosom.

Abstract

Effect of thawing technique to quality frozen semen spermatozoa in bali, madura and PO cattleABSTRACT. The experiment was conducted from November 2011 to January 2012 in BBIB Singosari. The study aims was to determine the effect of thawing techniques on the quality frozen bovine sperm Bali, Madura and PO cattle. This material used 90 frozen straw of three cattle from the three breed. The method is an experiment with three treatments and 10 replications for frozen to thawing in temperature water is 5 0C of duration 60 seconds, 15 0C duration 30 seconds and 37 0C duration 15 seconds. Variables observed in this study were sperm quality are motility, viability and abnormality. Data in percentage transformed in to arcus sinus were analyzed using Completely Randomized Block Design (CRBD) with replication as blocks of 10x. The results showed the thawing at 37 0C duration 15 seconds to highly significant effect (P<0,01) viability and motility and significant effect (P<0.05). The abnormalities, no significant effect (P>0,05). Breed cattle not significant effect to quality. It can be concluded that thawing techniques impact the motility and viability in frozen bovine semen spermatozoa Bali, Madura and PO cattle. Thawing using 37 0C temperature water with duration of 15 seconds is the best method to obtain the quality of spermatozoa post thawing frozen sperm.

Abstract

The objective of this study was to determine sperm membrane stability after sexing with gradientalbumin (egg white) using andromed and CEP-2 supplemented with 10% egg yolk. The observations ofsperm membrane included membrane integrity, uncapacitated, capacitated  and acrosome reacted. Albumingradient was made by mixing egg white with andromed or CEP-2 supplemented with 10% egg yolk resultingin a concentration of egg whites 10%, 30% and 50%. Membrane integrity was observed using HOST(Hypoosmotic Swelling Test), while uncapacitated, capacitated and acrosome reacted sperm  were observedusing CTC (Chlortetracycline) staining. The results showed that both andromed and CEP-2 supplementedwith 10% egg yolk could protect membrane integrity, uncapacitated, capacitated and acrosome reactedsperm. Andromed and CEP-2 supplement with 10% egg yolk could reduce the demage of membrane integrityand uncapacitated sperm, whereas capacitated and acrosome reacted sperm could be kept low.

Abstract

This study aimed to determine the effect of different methods of saving the storage by using a waterjacket and a free water jacket to quality semen Limousin cattle after being stored at a temperature of 5°C.Semen used the criteria motility e” + + collected two times a week. Semen evaluated macroscopically andmicroscopically and diluted in the ratio 1: 1 using the Cauda Epididymal Plasma (CEP-2) diluent added10% egg yolk. Stored in the refrigerator temperature 5°C and semen quality  was observed immediatelyafter dilution, 1 hour, 2 hours, 3 hours, 24 hours, 48 hours and 72 hours. The results showed that themethod of free storage waterjacket better in maintaining the percentage of motility, viability, abnormalityand plasma membrane intact spermatozoa

Abstract

The aim of this study was to observe the best extender in protecting the membrane of bovine spermatozoafollowing sexing by percoll density gradient centrifugation. Freshly collected semen were obtained fromBalai Besar Inseminasi Buatan Singosari-Malang. The semen were diluted in andromed and CaudalEpididymal Plasma-2 (CEP-2) added with 10% egg yolk extenders.The sperm membrane integrity wasobserved using Hypo-osmotic Swelling Test (HOST). Sperm capacitation and acrososome reaction wereassessed using Chlortetracycline Fluorescence Assay.The results showed that andromed and CEP-2 addedwith 10% egg yolk were able to retain the sperm membrane integrity, whereas sperm capacitation andacrosome reaction were kept low. Caudal Epididymal Plasma-2 (CEP-2) added with 10% egg yolk seemedto give better protection towards the sperm membrane intact in comparison to andromed extender.

Abstract

The use of appropriate diluent is intended to ensure the physical and chemical requirements ofspermatozoa so that the quality, particularly capacitation can be maintained. This study aimed todetermine the quality of Limousin cattle sperm after sexing with density percoll centrifugation methodusing Andromed diluent and Cauda epididymal Plasma 2 (CEP-2) plus 10% egg yolk. Observations ofspermatozoa quality including : motility, viability, percentage of abnormal, concentrations, membraneintegrity and state of capacitation. The semen used were obtained from Limousin cattle at the Center forArtificial Insemination(BBIB) Singosari Malang, with motility criteria e” 70%. Data were analysed usinga paired T-test in order to determine differences in the use of thinning treatments at each layer. Theresults showed that both diluent, andromed and CEP-2 diluent plus 10% egg yolk were able to maintainthe quality of spermatozoa either in the upper and lower layers

Abstract

The purpose of this research was to determine the success of AI using sexed semen results based on NRR, S/C, CR, and rectal palpation were also determine.  The total number of 81 acceptors divided into 3 treatment groups, such as P0 (non- sexed semen) ; P1 (Frozen semen Sexed using White Yolk Sedimentation),  P2 (Frozen semen sexed using Percoll Gradient Centrifugation) . The results showed the NRR percentage of P0, P1 and P2 were between the treatments are P0 of 74.07 ± 12:00%; 65.43 ± 5.66% and 64.19 ±  8:56% respectively. Furthermore the highest of CR was in P0 44%,   P2 25.91% and P1 18.51%. Conclution of this research was the highest percentage of pregnancy was 59.25 %(P0) ,  P1 51.85% and the lowest was in 44.44%. The S/C was in (P0) 2.31 follow by 3.00 in P2 and 3.33 in P1.   Key words : artificial insemination , frozen semen sexed, the percentage of Gestation and feed

Abstract

Penelitian ini bertujuan untuk mengetahui perbedaan produksi semen beku pada berbagai bangsa sapi potong dan untuk mengetahui produksi semen pada masing-masing  individu pada bangsa yang sama. Penelitian dilakukan di Balai Inseminasi Buatan Daerah Ungaran Jawa Tengah Indonesia. Materi yang dipergunakan dalam penelitian ini adalah catatan produksi semen yang terdiri kualitas semen segar dan jumlah straw semen beku pada bulan Januari sampai Desember tahun 2014  sebanyak  39 ekor sapi  yang terdiri dari 18 ekor sapi Simental, 8 ekor sapi  Limousin, 8 ekor PO dan 5 ekor sapi Brahman. Pengencer yang dipergunakan adalah skim kuning telur.  Metode yang dipergunakan adalah studi kasus dengan rancangan acak kelompok. Variabel yang diamati adalah Volume semen, Konsentrasi, Motilitas individu, total spermatozoa dan total spermatozoa yang motil. Hasilnya menunjukkan volume sapi Limousin yang terbanyak = 7,2 + 1,3 ml, diikuti oleh  oleh sapi simental = 6,8 + 1,6 ml,  sapi PO = 6,1 + 1,2 ml dan yang paling sedikit adalah sapi Brahman = 4,2 + 1,8 ml. Rata-rata volume semen pada berbagai bangsa terdapat perbedaan yang nyata (P&lt;0,05). Rata-rata persentase motilitas spermatozoa sapi  Simental =  57,8 + 13,05 %, Sapi  Limousin = 61,17 +1.37%, sapi PO = 63,5 + 6,62 %  dan  Brahman = 44,8 + 25,33 %, Rata-rata persentase motilitas pada berbagai bangsa tidak menunjukkan perbedaan yang nyata (P&gt;0,05). Rata-rata Total spermatozoa dan total spermatozoa yang motil pada sapi Simental adalah 9.320,3 + 2.461,2 and 5.532,9 + 2.314,2 ; Sapi Limousin = 11.266,9 + 1.159,6 dan  7.908,9 + 3.851,2; Sapi PO = 8.995,1 + 2.860,8 dan  5.651,6 + 2.418,4 , Sapi  Brahman =  6.189,3 + 22,6 dan  3.053,3 + 4.356,6. Rata-rata kedua parameter ini tidak menunjukkan perbedaan yang nyata pada berbagai bangsa sapi  (P&gt;0,05). Rata-rata produksi semen beku per hari pada sapi  Simental 280,67 + 68,57 straw ; sapi Limousin 315,6 + 44,32 straw ;  sapi PO 275,15 +70,61  straw  dan sapi   Brahman 225,18 + 59,74 straw. Rata-rata produksi semen beku tidak menunjukkan perbedaan yang nyata pada berbagai bangsa sapi  (P&gt;0,05). Volume semen, motilitas individu dan produksi semen beku harian masing-masing  individu pada bangsa yang sama pada sapi Limousin, Simental, PO dan Brahman terdapat perbedaan yang sangat nyata (P&lt;0,05). Keyword: Sapi Limousin , Sapi Simental, Sapi PO dan Sapi Brahman

Abstract

The purpose of this study was to determine the performance of reproduction in dairy cattle, the views of service per conception, days open, and calving interval. The material used in this study is the implementation of recording data or artificial insemination in 100 dairy cows females have at least twice to parturition known calving interval. Samples were selected randomly within the same district in Batu. The method used in this research is a case study. The data is taken primary and secondary data. Primary data retrieval is done by direct observation (observation), which includes several variables, where the variables include: the identity of the breeder, the number of cattle, cages and feeding and drinking. As for the secondary data obtained by recording the card Artificial Insemination (AI) owned by farmers. Sampling was done by purposive sampling and random with the following conditions: sampling was done by purposive sampling with the following conditions: minimal yearling heifer parturition twice and had a complete recording AI, including, the date AI and the date of parturition. Results from this study is the performance of reproduction in the dairy cooperatives Batu showed unfavorable results, average - the average total value of S/C of parity 1 s / d 6 is, 1.93 ± 1.18. As for the average - the average total value of DO and CI parity 1 s / d 5 is, 171.70 ± 132.20 days and 450.92 ± 132.83 days. In addition, the assessment of S / C, DO and CI at different parity using statistical tests showed that the S / C, DO and CI at various parity was not significantly different (P&gt; 0.05).It concluded that reproduction display AI results in the KUD BATU less than the maximum and the difference between parity does not affect the value of the S / C, DO, and CI.

Abstract

The purpose of the study was to determine the correlation between cow weight and birth weight of Brahman Cross calves with different sex. The materials of the study were one hundred Brahman Cross cows which had been purposively selected. The variables of the study were cow body weight, calves birth weight, and sex. Data were analyzed using Pearson Correlation test (Product-Moment), Linear Regression and t Test. The study found that the birth weight diferred significantly (P &lt;0.05) among male and female calves at the first and third parities. Also, there was significant correlation (P &lt;0.05) between cow body weight and calves birth weight at the first parity. However, this cow body weight only influenced 22-25% on birth weight so that the study suggests to considering other factors such as environment and animal feed.
