Abstract

Highly pathogenic avian influenza (HPAI) HSN1 virus is a known pathogen in birds. Recently, the virus has been reported to cause sporadic fatal disease in tigers, leopards, and other exotic felids as well as domestic cats in Thailand. The present study was carried out to investigate the presence of AI HSN1 virus infection in stray catsroaming around residential, traditional and chicken farms in Bogor, West Java. Ninety serum samples were tested using HI test to screened for the presence of antibody to AI HSN1. Virus isolation was done in SPF embrionated chicken eggs and identify using HI, AGP and RT -PCR.

Abstract

Reactive Oxygen Species (ROS) merupakan salah satu lethal chemical yang dapatmembunuh dan mengeliminasi bakteri pada sel fagosit. Lidah Buaya (Aloevera) banyak dipakai sebagai pengobatan tradisional, tetapi belum ada buktiilmiah sampai tingkat seluler apalagi subseluler dalam hal efek imunostimulanpada penyakit infeksi. Tujuan penelitian ini adalah untuk mengetahui aktivitasimunostimulan dari gel lidah buaya yang ditunjukkan oleh aktivitas ROS makrofagsecara in vivo terhadap infeksi bakteri patogen Salmonella typhimurium. Sebanyak24 ekor mencit BABL/c betina umur 8-10 minggu berat 20-30 gram dikelompokkansecara acak menjadi empat kelompok, masing-masing kelompok enam ekor.Kelompok kontrol tidak diberi gel Aloe vera, sementara kelompok P1, P2, dan P3berturut-turut diberi gel Aloe vera 0,5 ml/ekor/hari; 1,0 ml/ekor/hari, dan 1,5ml/ekor/hari. Pemberian gel Aloe vera dilakukan selama sembilan hari. Pada harike-6, mencit diinfeksi bakteri patogen Salmonella typhimurium intraperitoneal105 CFU. Selanjutnya pada hari ke-10 mencit didislokasi dan dibedah, diambilmakrofag dari peritoneum untuk dianalisis produksi ROS-nya. Hasil penelitianmenunjukkan bahwa pemberian gel Aloe vera berpengaruh signi..ikan terhadappeningkatan produksi ROS makrofag mencit BALB/c yang diinfeksi Salmonellatyphimurium. Terdapat perbedaan secara signi..ikan antara kelompok kontroldengan kelompok P1, P2, dan P3, tetapi tidak terdapat perbedaan signi..ikan antarkelompok P1, P2, dan P3. Pemberian gel Aloe vera dosis 0,5 ml/ekor/hari sudahmampu meningkatkan produksi ROS makrofag. Reactive Oxygen Species (ROS) is one of lethal chemicals that can kill and eliminatebacteria in phagocytic cells. Aloe vera is widely used as traditional medicine, but thereis no scienti..ic evidence to prove the effect of immunostimulatory of the Aloe vera gel oninfectious disease in the cellular or subcellular level. This research aims to determinethe immunostimulatory activity of Aloe vera gel showed by ROS macrophage activityin vivo on the infection of bacterial pathogen Salmonella typhimurium. This researchused 24 female mice BABL/c age 8-10 weeks weight 20-30 Gram. They were groupedrandomly in four groups consisting of six mice each. The mice in the control groupwere not given the Aloe vera gel, while group P1 was given the gel of 0,5ml/mice/day,group P2 was 1,0ml/mice/day, and group P3 was 1,5ml/mice/day. The gel was givenin nine days, and in 6th day the mice were infected by bacterial pathogen Salmonellatyphimurium then in 10th day the mice were dissected to take their macrophage fromperitoneum for being analyzed its ROS production. The result showed that there wasa signi..icant difference between control group and P1, P2, and P3 group, but there isno signi..icant difference between P1, P2, and P3 group. In conclusion, the dose of 0,5ml/mice/day was able to increase ROS macrophage production.

Abstract

Tujuan penelitian untuk meningkatkan hasil belajar IPA melalui pembelajaran model examples non examples dengan berbantuan powerpoint. Model pembelajaran examples non examples adalah suatu model pembelajaran yang menggunakan gambar yang sesuai dengan kompetensi dasar. Hasil penelitian diperoleh bahwa terdapat kenaikan rata-rata nilai dan prosentase ketuntasan belajar. Pada siklus 3 siswa yang tuntas 80,4 %, sesuai dengan ketercapaian indikator keberhasilan yang ditetapkan. Berdasarkan hasil yang telah dicapai maka dapat disimpulkan pembelajaran dengan menggunakan model examples non examples dapat meningkatkan hasil belajar IPA. The objective of the research is to increase the result of learning Science through learning model of ‘examples non examples’ with power point. learning model of ‘examples non examples’ is a learning model which uses pictures related to basic competence. The result of the research obtains the increase score and the percentage of learning exhaustiveness. On the third cycle, the student learning exhaustiveness obtains 80,4 %. It refers to the success indicator established. The conclusion is that through learning model of ‘examples non examples’ can increase the result of Science learning.

Abstract

Recently, the virus has been reported to cause sporadic fatal disease in tigers, leopards, and other exotic felids as well as domestic cats  in Thailand. The present study was carried out to investigate the presence of AI H5N1 virus infection in stray cats roaming around residential, traditional and chicken farms in Bogor, West Java. Ninety serum samples were tested using HI test to screened for the presence of antibody to AI H5N1. Virus isolation was done in SPF embrionated chicken eggs and identify using HI, AGP and RT-PCR. The results showed that 18,9% of stray cats developed antibodies against H5 with geometric mean titre 23,1 . Stray cats lived in traditional markets 18–40% developed antibodies in the titre ranging from 22,8  to 24,5. Only two out of nine stray cats which lived in chicken farm developed low antibody titres again H5 (21). None of the stray cats lived in residencial area have developed antibodies against H5. This study revealed that stray cats have been contact with AI H5. Avian influenza H5 viruses were isolated in eight out of 33 pooled of rectal swab samples. The viral cleavage site sequences are CCTCAAAGAGAGAGC AGAAGAAAGAAGAGAGGT which represent amino acid sequences of PQRESRRKKRG. Based on the cleavage site sequence, the isolates are similar with the AI H5 virus subtype isolated from human in Indonesia during 2005–2007.

Abstract

Glutation merupakan antioksidan yang berperan dalam fungsi imun, dan diekspresikan secara genetik oleh urutan gen yang membentuk protein enzim Glutation Peroxidase (GPx1). Bila ekspresi gen berubah maka terjadi perubahan fungsi glutation dan kerentanan terhadap stress oksidatif. Metode yang digunakan adalah Kasus-kontrol. Sampel yang digunakan adalah sampel darah. Kelompok kasus adalah sampel darah pasien tuberkulosis paru sedangkan kelompok kontrol adalah sampel darah orang sehat. Pemeriksaan gen Glutation peroxidase (GPx1) menggunakan metode Polymerase Chain Reaction (PCR) untuk melihat pita DNA pada pasien tuberkulosis par serta elektroforesis produk PCR-RFLP gen GPx1 kelompok sampel tuberkulosis. Hasil penelitian menunjukkan bahwa tidak terdapat hubungan yang bermakna antara polimorfisme gen GPx1 (p=0,365) pasein tuberkulois dengan individu sehat, sehingga tidak dapat digunakan sebagai alat deteksi kerentanan terhadap stress oksidatif pada pasien tuberkulosis. Perlu penelitian lanjutan yang menggunakan sampel lebih besar dan populasi etnik yang berbeda.

Abstract

Penelitian ini bertujuan untuk menganalisis kadar protein dan profil protein pada beberapa susu (susu kedelai, susu kambing) dan olahannya (yogurt, tofu). Kadar protein diukur dengan metode Lowry, sedangkan profil protein dianalisis menggunakan SDS PAGE. Data yang diperoleh dianalisis secara deskriptif. Kadar protein tertinggi pada sampel yang dianalisis terdapat pada produk yogurt A (579,5 mg/ml), disusul susu kedelai (289,99 mg/ml) dan susu kambing (133,1 mg/ml). Analisis profil protein terlihat pita protein dengan mobilitas terendah sampai tertinggi terletak pada berat molekul 14-150 KDa. Pita protein khas yang hanya dimiliki susu kambing adalah pita 150kDa. Sementara pita protein khas yang hanya dimiliki susu kedelai adalah pita 44 kDa dan 55kDa. Pita protein yang khas hanya dimiliki yogurt A (dengan bakteri Lactobacillus bulgaricus dan Streptococcus thermophillus) adalah pita 65Da. Semua jenis susu dan olahannya memiliki pita 70kDa, kecuali susu kedelai. Profil protein susu kedelai dan tofu menunjukkan profil protein yang sangat berbeda, namun keduanya memiliki pita 18kDa.This study aimed to observe protein level and profiles on some milks (soy milk, goats milk) and dairy (yogurt, tofu) product. Protein content was observed by Lowry method, whereas the protein profiles were analyzed by polyacrylamide gel electrophoresis. Data were analyzed descriptively. The highest protein content of the observed sample was in yogurt A products (579,5 mg/ml), followed by soy milk (289,99 mg/ml) and goats milk (133,1 mg/ml). Analysis of protein profiles showed protein bands with lowest to highest mobility lies in the molecular weight of 14-150 KDa. Typical protein band of goats milk was a 150kDa band. While the typical protein bands of soy milk were 44 kDa and 55kDa band. The typical protein band of yogurt A (with Lactobacillus bulgaricus and Streptococcus thermophillus bacterium) was 65Da. All types of milks and dairy had 70kDa band, except for soy milk. Protein profile of soy milk and tofu was very different, but both had 18kDa band.

Abstract

Agrowisata Sidomuncul berpotensi sebagai sumber belajar Kingdom Plantae dan manfaatnya bagi kehidupan. Namun perizinan yang cukup lama, biaya tranportasi, tenaga, dan kemungkinan terganggunya mata pelajaran lain menjadi pertimbangan dalam melakukan kunjungan ke agrowisata Sidomuncul. Tujuan penelitian ini adalah menguji kelayakan dalam pengembangan Ensiklopedia Tanaman Obat koleksi PT. Sidomuncul sebagai media pembelajaran materi plantae kelas X SMA. Penelitian ini merupakan penelitian Research and Development (R&D). Langkah penelitian ini meliputi penelitian pendahuluan observasi jenis spesies tanaman obat koleksi PT. Sidomuncul (melihat, mendokumentasikan, dan mengidentifikasi untuk kemudian dibuat dalam bentuk ensiklopedia), desain produk, dan validasi produk. Penelitian pendahuluan dilakukan di Agrowisata Sidomuncul milik PT. Sidomuncul Klepu, Kecamatan Bergas, Kabupaten Semarang. Validasi Ensiklopedia Tanaman Obat koleksi PT. Sidomuncul melibatkan pakar media, pakar materi, dan pakar perangkat pembelajaran untuk penilaian keterterapan Ensiklopedia Tanaman Obat koleksi PT. Sidomuncul sebagai media pembelajaran. Hasil dari penelitian ini teridentifikasi sebanyak 210 spesies tanaman obat yang kemudian disusun ke dalam ensiklopedia. Validasi Ensiklopedia Tanaman Obat koleksi PT. Sidomuncul dinyatakan dalam kategori sangat layak dengan persentase 97,5% oleh validator media, layak dengan persentase 77,5% oleh validator materi, dan sangat layak dengan persentase 87,5% oleh validator perangkat pembelajaran sebagai media pembelajaran materi Plantae kelas X SMA Sidomuncul Agrotourism have potential as a source of learning Plantae Kingdom and benefits for life. Licensing process long enough, the cost of transportation, energy, and possible disruptions of other subjects into consideration in making a visit to Sidomuncul Agrotourism. The purpose of this study was to test the feasibility of the Encyclopedia of Medicinal Plants PT. Sidomuncul collection development, as a learning medium of Plantae materials class X SMA. This study is a Research and Development (R & D). Step of this study include observational preliminary study of medicinal plants species PT. Sidomuncul collection (see, documenting, identifying and then made in the form of an encyclopedia), product design and product validation. Preliminary research conducted at Agrotourism of PT. Sidomuncul Klepu, District Bergas, Semarang Regency. Validation Encyclopedia of Medicinal Plants PT. Sidomuncul collection involving media experts, materials experts and experts of learning devic for the assessment of implementation Encyclopedia of Medicinal Plants PT. Sidomuncul collection as a learning medium. The results of this study identified as many as 210 species of medicinal plants which are then compiled into the encyclopedia. Validation Encyclopedia of Medicinal Plants collection PT. Sidomuncul expressed in the category of very decent with a percentage of 97.5% by the media validator, feasible with the percentage of 77.5% by validator material, and very decent with a percentage of 87.5% by validator learning device as a medium of learning materials Plantae class X SMA.

Abstract

Website can be made as source and learning medium because of its application in helping students to understand the unprecise material. Web educative is a learning model of distance learning system with the use of IT-based interactive and virtual.The objective of the study as the learning resource was to find out the feasibility and the effectiveness in learning process. The type of the study was Research and Development (R&D). The data of this study were detemined from the feasibility validation of web educative by validator, student’s and teacher’s responses, and the last was student’s achievement. The result of web educative feasibility which was determined from the expert could be said very suitable with the average score 91%. Student’s completeness achievement with the score ≥78 in product trial reached 100% and 93% in application trial. The score 84% showed positive student’s response about this web educative. Based on this study, it can be concluded that the developing of web educative was very suitable to be applied in learning process especially in the material of self defenses; it also proved the effectiveness of this method in student’s achievement.Website dapat dijadikan media dan sumber belajar karena menyediakan banyak aplikasi yang membantu siswa memahami konsep materi yang abstrak. Web educative merupakan sebuah model pembelajaran sistem pembelajaran jarak jauh dengan pemanfaatan TI yang bersifat interaktif dan berbasis virtual. Tujuan pengembangan web educative sebagai sumber belajar materi sistem pertahanan tubuh adalah untuk mengetahui kelayakan dan keefektifan. Penelitian ini merupakan penelitian Research and Development (R&D) melalui tahapan identifikasi potensi dan masalah, pengumpulan data, desain produk, validasi desain, revisi desain, uji coba produk, revisi produk, uji coba pemakaian, revisi produk dan menghasilkan produk final.. Data yang diambil dalam penelitian ini adalah kelayakan web educative yang divalidasi validator, tanggapan siswa dan guru mengenai kelayakan web educative dan hasil belajar siswa. Hasil penelitian dapat diketahui bahwa hasil penilaian kelayakan web educative dari pakar media dan materi memperoleh skor rata-rata 91% dengan kriteria sangat layak. Ketuntasan hasil belajar siswa dengan nilai ≥78 pada uji coba produk mencapai 100% dan pada uji coba pemakaian 93%. Siswa memberikan tanggapan yang baik dengan ditunjukkan perolehan skor rata-rata sebesar 84%. Berdasarkan hasil penelitian dapat disimpulkan bahwa web educative sangat layak digunakan sebagai sumber belajar materi sistem pertahanan tubuh dan efektif terhadap hasil belajar siswa.
