Raya Bandung-Sumedang Jatinangor, Sumedang 45363   Diterima  1 Juli  2009 / Disetujui  7 Desember 2009   ABSTRACT   The aim of this study was to determine the content of total monomeric anthocyanins and to identify the anthocyanin composition of jambolan fruit (Syzygium cumini) growing in Indonesia. Anthocyanins were extracted with methanol containing 0.1% HCl, removed from non-polar impurities by partition with chloroform and purified in a C18 solid phase cartridge. Anthocyanins were identified by high-performance liquid chromatography with diode array detection (HPLC-DAD), UV-visible spectral analysis, and physico-chemical reactions. Using pH-differential method, it was known that the content of total monomeric anthocyanin in the ripe fruit was 161 mg/100 g fresh fruit (731 mg/100 g skin on wet basis or 3430 mg/100 g skin on dry basis), based on cyanidin-3-glucoside. Furthermore, five anthocyanins without acylating groups were identified as 3,5-diglucoside derivatives of delphinidin (41.29%), petunidin (27.79%), malvidin (25.60%), cyanidin (4.19%), and peonidin (1.13%). These results demonstrate that the skin of jambolan fruit especially the ripe one is rich in anthocyanin, with wide range in anthocyanidin backbone (five aglycons).   Key words: jambolan fruit, Syzygium cumini, anthocyanin, HPLC-DAD

Abstract

The research of the bioactivity effect of the leaves extract Kalanchoe daigremontiana Hammet & Perrier (Crassulaceae) against the fourth instar larvae of Epilachna vigintioctopunctata Fabricius (Coleoptera) the harm of Solanaceae were conducted at Laboratory of Animal Taxonomy, Department of Biology, FMIPA UNPAD Jatinangor. The experiment of bioassay test was conducted with observasion in 1 x 24 hours and continue to antifeedant test with no choice test. The data were analysed by the Mann Whitney U test to compare between the control with the treatments. The result showed that the leaves extract of Kalanchoe daigremontiana have antifeedant bioactivity against to early fourth instar larvae of E. vigintioctopunctata which significantly reduce the feeding activity of larvae at  78,125 ppm, 156,25 ppm, 312,5 ppm, 625 ppm, 1250 ppm, 2500 ppm, dan 5000 ppm concentrations.

Abstract

Dalam penelitian berkelanjutan untuk pencarian senyawa-senyawa yang bersifat insektisida daritanaman Kalanchoe (Cocor bebek) Indonesia, diperoleh hasil bahwa ekstrak metanol daun Cocorbebek (Kalanchoe daigremontiana) memberikan aktivitas insektisida yang kuat terhadap instar ke tigalarva ulat sutera (Bombyx mori). Ekstrak metanol yang diperoleh selanjutnya dipisahkan senyawasenyawanyamenggunakan partisi dengan pelarut organik dan kombinasi kolom kromatografi padasilika gel dan ODS sehingga dihasilkan satu senyawa aktif insektisida. Struktur kimia senyawa yangbersifat insektisida tersebut ditentukan dengan metoda spektroskopi dan perbandingan data spektrasenyawa analog yang telah diperoleh dari penelitian sebelumnya sehingga senyawa aktif insektisidatersebut diidentifikasikan sebagai senyawa turunan bufadienolida, daigremontianin. Daegremontianinmenunjukkan aktivitas insektisidal yang kuat terhadap instar ke tiga ulat sutera (Bombyx mori) denganLD50 0,9 μg/g diet.

Potensi Tumbuhan Erythrina (Leguminosae) sebagai Antifertilitas

Abstract

Erythrina plants (Leguminosae) were used as traditional antifertility agents. In the continuing research for novel plant with antifertility capacity from Indonesian plants, the methanol extract of Erythrina plants showed significant antifertility activity in an in vitro assay of spermatozoa of white rat (R. norvegicus). The methanol extract was separated using bioassay-guide n-hexane, ethylacetate, and n-buthanol fractionation. The ethylacetate fraction was tested in vitro on spermatozoa of white rat (R. norvegicus) at a concentration of 0.25 x 10-3 μg/μL. It showed a decrease of their motilities and viabilities, as well as an increasing abnormality of the spermatozoa.

Abstract

Traditional medicines originating from plants have been widely used as an alternative therapy and extensive research have identified several bioactive compounds of the plants. In Indonesia, Erythrina variegata (Leguminosae) plants have been used for medication to many diseases including anthelmintic, anticancer,antimalaria, and antifertility.Objective: In the course of our continuing search for novel bioactive compounds from Indonesian Erythrina plants, we isolated and described several bioactive compounds such as anticancer, antimalaria, and antifertility from E. variegata. Method: Activity assay in vitro against breast cancer cell T47D using the Sulforhodamine B (SRB) method, activity assay in vitro against antiplasmodium using lactatedehydrogenase (LDH) method, and antifertility activity on spermatozoa Rattus novergicus. Their chemical structures were determined based on spectroscopic evidences and comparison with related compound previously reported. Result: Extract and compounds of E. variegata showed anticancer in vitro against breast cancer cell T47D, antiplasmodial in vitro K1 and 3D7 strain parasites, and antifertility on spermatozoa R. norvegicus with midle and high activity. Besides that, the toxicity assay of it was also conducted using white mice to show the safety grade of the plant. Conclusion: This results strongly suggested that E. variegata is a promising sources as herbal medicine of anticancer, antimalaria, and antifertility agents.

Triterpenoid Pentacyclic Antimalarial Activity from the Leaves of Erythrina variegata

Abstract

In the course of our continuing research for novel antimalarial agent from Indonesian plants, the methanol extract of the Erythrina variegata leaves showed significant antimalarial activity against Plasmodium falciparum strain K1 in vitro with IC50 6.8 μg/mL. The methanol extract was separated by using variety of chromatography techniques. The chemical structure of an antimalarial compound was determined on the basis of spectroscopic evidence and compared to previous data then this compound is identified as a pentacyclic triterpenoid oleane derivative, namely 3,22,23-trihydroxy-oleane-12-ene. The pentacyclic triterpenoid, 3,22,23-trihydroxy-oleane- 12-ene showed antimalarial activity against 3D7 and K1 strains with IC50 4.3 μg/mL and 24 μg/mL, respectively. Abstract

Kalanchoe is the biggest genera of Crassulaceae family and distributed in tropical and subtropical regions. This genera is found to be a rich source of biologically active natural products such as triterpenes, flavonoids and steroids. As a part of our continuing search for novel insecticidal compounds from Indonesian Kalanchoe plants, we examined Kalanchoe daigremontiana collected from Bandung region, West Java, Indonesia. The methanolic extract of the dried leaves of K. daigremontiana was concentrated and extracted with methylene chloride The methylene chloride extract exhibited an insecticidal activity toward silkworms. The methylene chloride extract was partitioned between n-hexane and methanol containing 10% water. The active lower layer was extracted with ethyl acetate. By using the insecticidal activity to follow the separations, the ethyl acetate fraction was separated by combination of column chromatography on Kieselgel 60 and ODS to afford two insecticidal bufadienolides 1 and 2. The structures of these compounds were elucidated based on spectroscopic analysis (UV, IR, NMR, 2D-NMR) and comparison with those related data previously reported. In this paper, the isolation, structural elucidation, and insecticidal activities against the third instar larvae of silkworm will be described. Abstract

In the course of our continuing search for novel paralytic compound from Indonesian plants, the methanol extract of the stem bark of Erythrina variegata (Leguminosae) showed significant paralytic activity against the third instar larvae of silkworm (Bombyx mori). The purposes of this research were isolation and structural elucidation of paralytic compound from the stem bark of E. variegata. Using the paralytic activity following the separation, the methanol extract was separated by combination of column chromatography to yield prenylisoflavone, warangalone. The chemical structure of warangalone was identified based on spectroscopic evidence and comparison with the previous reported. The paralytic activity of warangalone showed weak activity against the third instar larvae of silkworm (B. mori).Keywords: Erythrina variegata, Leguminosae, isoflavonoid, warangalone, paralytic activity

Abstract

Peptida P25 merupakan epitop yang dapat dengan mudah mengaktivasisel T penolong. Sel T penolong teraktivasi berperan dalam aktivasi sel Buntuk membentuk antibodi. Peptida bahkan protein dapat dibuat denganmetode sintesis peptida fasa padat Fmoc/tBu menggunakan aditif HOBt.Namun sayangnya HOBt memiliki karakter yang mudah meledak. Padapenelitian ini telah berhasil disintesis bagian epitop P25 (peptida P251-9bergugus pelindung) yang memiliki tingkat kemurnian tinggi dan perolehan36,4% dengan metode sintesis peptida fasa padat Fmoc/tBu menggunakanaditif oksima pengganti HOBt.Kata kunci: Epitop, P251-9, sintesis peptida fasa padat Fmoc/tBu, oksima.

Poriferasta-5,22E,25-trien-3β-ol,22-dehidrokolesterol dari daun Kalanchoe serrata (Crassulaceae)

Abstract

Dalam penelitian berkelanjutan untuk pencarian senyawa metabolit sekunder baru dari tumbuhan Kalanchoe Indonesia, telah dilakukan kajian fitokimia terhadap Kalanchoe serrata. Daun segar K. serrata sebanyak 14,8 kg diekstraksi dengan metanol pada temperatur kamar. Ekstrak metanol (292 g) dipartisi berturut-turut dengan n-heksana dan metilenklorida. Ekstrak metilenklorida (0,80 g) selanjutnya dipisahkan pada kromatografi cair vakum pada silika gel G60 dengan eluen kloroform-aseton yang meningkat kepolarannya sehingga dihasilkan 10 fraksi yang dikelompokan berdasarkan analisis KLT. Padatan yang diperoleh pada fraksi yang terelusi dengan 20% aseton selanjutnya dipisahkan pada kromatografi kolom pada silika gel (230-400 mesh) dengan eluen kloroform dan dimurnikan lebih lanjut dengan kristalisasi pada aseton sehingga dihasilkan isolat berbentuk kristal jarum tak-berwarna sebanyak 23 mg. Isolat menunjukkan titik leleh 115-118oC dan memberikan warna hijau-kebiruan pada uji Liebermann-Burchard menunjukkan adanya kerangka steroid.  Hasil analisis spektroskopi yang meliputi UV, IR, 1D-NMR dan 2D-NMR menunjukan bahwa isolat merupakan turunan sterol dan diidentifikasi sebagai poriferasta-5,22E,25-trien-3β-ol,22-dehidrokolesterol.
