Abstract

Penicillin G acylase (PGA) which catalyses penicillin G hydrolysis reaction is a key enzyme for the industrial production of penicilin G derivatives used in therapeutics. A new local strain of Bacillus subtilis BAC4 was found capable of producing extracellular PGA. However, characteristics of this extracellular PGA are not known. The goal of this research was to characterize the extracellular PGA produced by B. subtilis BAC4. Enzyme production was carried out by batch fermentation, followed by enzyme purification and characterization of the PGA. The PGA activity was determined by the Kornfeld method, with optimal activity for hydrolysing penicillin G observed at 43 oC and pH 8.5. The activation energy of penicillin G hydrolysis by the PGA of B. subtilis BAC4 was determined as 4.9 kcal.mol-1 and Vmax and Km values were found to be 0.7 µmole.min-1.mg-1 and 3.5 mM respectively. PGA catalytic activity was competitively inhibited by phenylacetic acid with an inhibition constant, Ki(PAA) , of 347.2 mM. Abstract

Background: Knowledge about prevention of wound infection is very important for health care professional and other worker in clinical setting.  The prevention program of nosokomial infection developed in RSUD Dr. Moewardi had not yet show efectiveness of the decreasing. The number of nosokomial infection optimally. Number of nosokomial infection because of operation event still occufy. The aim of the research is knowing difference woundlead infection among antibiotic topical and nacl 0,9% compress in Anggrek 3 room in RSUD Dr. Moewardi Surakarta. Research Method:  This research include an descriptif research with quantitaive approach that aim for giving illustrate of description aboutcondition objectively. This research is carried out in RSUD Dr. Moewardi Surakarta. With takes sample 10 patient in Anggrek 3 room. Data collecting method use observation. Data collected then analyzed with descriptif analysis and t test. Research finding:1) 1) Wounded lead process post operation in patient with antibiotic topical is good (93,3%), and wounded lead process post operation in patient with NaCl 0,9% is good too (88,9%); (2) Process post operation in patient withantibiotic topical founded that is not good for lead connected (40%) but infection is not found. Processpost operation in patient with NaCl 0,9% founded that is not good for dolor (40%) but infection is not found. (3) There are not difference wounded leadquality between antibiotic topical and NaCl 0,9% compress. Conclussion:  Wounded lead process post operation in patient withantibiotic topical is not good enough than NaCl 0,9%. Comparation analysis found tstat= 1,043 with p=0,328 not accepted for 5% significande (p>0,05). Wounded lead process postoperation in patient with antibiotic topical just more faster than NaCl 0,9% compress. Wounded lead process average in patient with antibiotic topical 93,3% more higher than NaCl 0,9% (88,9%).

Abstract

Abstract. This study tried to develop learning material and its implementation using the Cooperative model of  TGT type with ATONG -based. The purpose of this study produce a valid learning device characteristics, practical and effective. These types of devices are developed syllabus, lesson plans, worksheets and learning media. Practicality test done in class V SD Negeri 1 Kendaga, test the effectiveness done in class V SD Negeri 1 Banjarmangu and SD Negeri 2 Kendaga. Data were obtained from: validation, observations, questionnaires and learning achievement test. The data were processed with descriptive analysis. Test validity and practicality mixed with analysis, testing the effectiveness of currently treated with t-test, z test, regression, and test the appeal. The results showed that the learning tools developed declared valid by the validator, with an average score of 94 for the syllabus, 95 for RPP, 100 for LKS, and 92.85 for instructional media. The results of observations obtained 89.57% of teacher activity, student questionnaire results obtained by 93%, this means a device developed practical. Average yield of 84.3 students mastery learning. Student learning activities affect the learning achievement of 85%. The results of experimental class learning achievement better than the control class. This means a device developed effective.Key words: Developing, Learning Material, Social Education, Cooperative of TGT, ATONGAbstrak. Penelitian ini merupakan pengembangan perangkat pembelajaran dengan model Cooperative tipe TGT berbasis ATONG. Tujuan penelitian ini menghasilkan karakteristik perangkat pembelajaran yang valid, praktis dan efektif. Penelitian menggunakan model pengembangan 4-D Thiagarajan. Jenis perangkat yang dikembangkan Silabus, RPP, LKSdanMedia Pembelajaran.Uji kepraktisan dilakukan dikelas V SD Negeri 1 Kendaga, uji keefektifan dilakukan dikelas V SD Negeri 1 Banjarmangu dan SD Negeri 2 Kendaga. Kelas V SD Negeri 1 Banjarmangu sebagai kelas eksperimen dan Kelas V SD Negeri 2 Kendaga sebagai kelas kontrol.Data penelitian diperoleh dari: validasi, pengamatan,angket dan tes prestasi belajar. Data diolah dengan analisis deskriptif. Uji validitas dan uji kepraktisan diolah dengan analisis, sedang uji keefektifan diolah dengan uji t, uji z, regresi, dan uji banding. Hasil penelitian menunjukkanperangkat pembelajaran yang dikembangkan dinyatakan valid oleh validator, dengan skor rata-rata 94 untuk silabus, 95 untuk RPP, 100 untuk LKS, dan 92,85 untuk  media pembelajaran. Hasil observasi aktivitas guru diperoleh 89,57%, hasil angket siswa diperoleh 93%,hal ini berarti perangkat yang dikembangkan praktis.Hasil rata-rata ketuntasan belajar siswa84,3.Aktivitas belajar siswa berpengaruh terhadap prestasi belajar sebesar 85%.Hasil prestasi belajar kelas eksperimenlebih baik daripada prestasi belajar kelas kontrol. Hal ini berarti perangkat yang dikembangkan efektif. Kata Kunci: Pengembangan Perangkat Pembelajaran, IPS, Cooperative tipe TGT, ATONG.

Abstract

Daun sirsak dapat digunakan sebagai zat antibakteri. Salah satunya menghambat bakteri Bacillus subtilis dan Escherichia coli. Penelitian dilakukan untuk mengetahui aktivitas antibakteri ekstrak daun sirsak terhadap bakteri Bacillus subtilis dan Escherichia coli serta penerapannya padalotion. Karakteristik ekstrak menggunakan spektrofotometer Ultra Violet-Visible (UV-Vis), Forier Transform Infrared (FT-IR), dan High Performance Liquid Chromatography (HPLC). Hasil penelitian menunjukkan ekstrak metanol daun sirsak menghambat pertumbuhan bakteri Bacillus subtilis dengan konsentrasi 25, 50, 100% sebesar 2,3; 3;3,9 mm dan kontrol negatif 2 mm, selain itu dapat menghambat pertumbuhan bakteri Escherichia coli sebesar 3,3; 3,3; 3,1 mm dan kontrol negatif  2 mm. Penambahan ekstrak pada lotion sebanyak 1% menghambat pertumbuhan bakteri  Bacillus subtilis dan Escherichia coli sebesar 0,7 dan 0,8 mm. Hasil UV-Vis serapan paling tinggi pada 292 nm, hasil FT-IR mengandung gugus OH, C-H alifatik, C=C aromatik, C-O alkohol, dan C-H aromatik yang diduga senyawa dihidroflavanol, dan hasil HPLC diduga adanya quercetin.

Abstract

Penelitian ini bertujuan untuk meningkatkan hasil belajar siswa dengan menerapkan metode pembelajaran berbasis proyek berbantuan e-learning. Penelitian ini dilakukan di suatu SMA N di Mranggen. Desain penelitian yang digunakan adalah pretest and postest group design. Sampel yang digunakan sebanyak dua kelas dengan teknik cluster random sampling. Metode pengumpulan data berupa metode tes dan observasi. Data yang diperoleh kemudian dianalisis dengan menggunakan uji t dan uji n gain. Hasil uji perbedaan rata-rata menunjukkan bahwa thitung 5,43 lebih besar dari tkritis 1,99 dengan taraf signifikansi 5%. Uji normalitas gain menunjukkan bahwa rata-rata hasil posttest mengalami peningkatan sebesar 0,57 dan 0,52 dengan kriteria sedang pada kelas eksperimen dan kelas kontrol. Penelitian ini menunjukkan bahwa ketercapaian indicator psikomotorik dan afektif menurut analisis deskriptif rata-rata kelas eksperimen lebih baik daripada kelas kontrol. Menurut analisis koefisien determinasi diperoleh hasil bahwa penelitian ini berkontribusi dalam meningkatkan hasil belajar siswa sebesar 12,60%. Berdasarkan hasil analisis tersebut disimpulkan bahwa penerapan pembelajaran berbasis proyek berbantuan e-learning dapat meningkatkan hasil belajar siswa. This research aims to improve student learning outcomes by applying methods project based learning assisted e-learning. The research was conducted at SMA in Mranggen. The research design was pretest and posttest group design. The sample used as much as two groups with cluster random sampling technique. The method of data collection which was utilized in this study was test and observation. Those data are analyzed using t test and normality gain. Based on the mean difference test showed tcalculated 5.43 greater than tcritical 1,99 with 5% significance level. Gain normality test showed that the posttest average increased by 0.57 and 0.52 with medium criteria at the experimental and control groups.The result of this study showed that achievement indicators psycomotor and affective according to descriptive analysis shows the average value of the experimental group better than the control group. According to the determination coefficient analysis showed that the study contributes to improving student learning outcome by 12.60%. Based on the results of the analysis concluded that the application of project based learning assisted e-learning was able to improve student learning outcomes.

Abstract

Susu sapi yang dihasilkan para peternak sapi di RT 03 RW 02 Kelurahan Nongkosawit Kecamatan Gunungpati hanya dijual ke pasar dalam keadaan susu segar dan selebihnya untuk kebutuhan sendiri sehari-hari tanpa ada upaya untuk membuat produk lain yang lebih bervariasi. Tujuan spesifik kegiatan ini adalah agar masyarakat melalui kelompok ibu-ibu PKK RT 03 RW 02 dapat memanfaatkan potensi yang ada didaerahnya, kecuali itu juga meningkatkan gizi pada balitanya mengingat susu merupakan makanan yang bergizi cukup tinggi dan harganya relatif murah. Metode kegiatan yang digunakan adalah metode ceramah mengenai seluk beluk susu, metode praktek tentang cara pengolahan susu sapi segar menjadi youghurt aneka rasa dan yang berhubungan dengan pasca pengolahan, metode tanya jawab untuk mengetahui sampai sejauh mana peserta mampu menerima atau terlibat dalam kegiatan ini dan evaluasi untuk memperoleh gambaran dalam rangka penafsiran dan analisis untuk memperoleh simpulan dari semua kegiatan yang sudah dilaksanakan. Hasil dari kegiatan ini menunjukkan masyarakat sangat antusias untuk mengolah susu sapi segar menjadi youghurt dengan berbagai macam rasa. Perlu dilakukan inovasi dan kreativitas dalam proses pengolahan susu sapi segar agar produk yang dihasilkan bisa lebih baik danbermutu.

Abstract

Pembelajaran yang masih menggunakan metode konvensional menyebabkan pencapaian kompetensi matapelajaran kimia masih tergolong rendah. Penelitian ini bertujuan untuk mengetahui pengaruh Model Cooperative Class Experiment (CCE) tipe Team Games Tournament (TGT) pada KBK dan Hasil Be/ajar siswa SMA topik reaksi redoks. Penentuan sampel menggunakan teknik cluster random sampling dan menghasikan siswa kelas X8 sebagai kelas eksperimen dan kelas X7 sebagai kelas kontrol. Metode yang digunakan dalam pengambilan data adalah dokumentasi, tes dalam bentuk pilihan ganda dan essay berupa open-ended questions serta observasi. Hasil penelitian menunjukkan hasil analisis data tes KBK yaitu pencapaian kelima subindikator KBK pada kelas eksperimen mencapai rata-rata 73, 75% yang berarti siswa dapat mengembangkan KBK dengan kriteria baik, sedangkan kelas kontrol dengan rata-rata 64,68% dalam kriteria KBK cukup. Dari hasil analisis, diperoleh rb 0,51 dengan besarnya kontribusi 26,29%. Dari hasil penelitian dapat disimpulkan bahwa penerapan pembelajaran Model Cooperative Class Experiment (CCE) Tipe Team Games Tournament (TGT) berpengaruh pada KBK dan hasil be/ajar siswa suatu SMA di Semarang pada pokok materi redoks dengan kontribusi sebesar 26,29%.Learning with the conventional method make competency achievement in chemistry subject is still low. This study aimed to determine the effect of Model Cooperative Class Experiment (CCE) type Team Games Tournament (TGT) on CBC and Learning Outcomes on high school students in the topic of redox reactions. Sample determination was using cluster random sampling technique, class X8 grade students is as the experimental class and X7 class is as control class. The method used in data collection is documentation, in the form of multiple choice test and an essay in the form of open-ended questions and observations. The results showed that analysis of CBC data test achieved fifth subindikator CBC in the experimental class gained an average of 73. 75%, which means that students can develop CBC with good criterion, while the control class with an average of 64.68% in the CBC is sufficient criteria. 7he analysis data obtained rb 0.51 with the contribution of 26.29%. The results of this study concluded that the implementation of Cooperative Learning Model Class Experiment (CCE) Study Team Games Tournament (TGT) type have signiffican effect on CBC and student learning outcomes in high school in Semarang on the redox subject with the contribution of 26.29%.

Abstract

Metode diskusi dalam pembelajaran dapat membangkitkan minat, motivasi, dan karakter siswa sehingga metode diskusi dipandang lebih efektif untuk meningkatkan hasil belajar. Penelitian ini bertujuan untuk mengetahui bagaimana karakter siswa yang muncul pada pembelajaran kimia berbasis teknologi informasi menggunakan metode diskusi kompetensi konsep mol siswa SMK. Penelitian dilakukan di suatu SMK swasta di Pati tahun pelajaran 2013/2014.  Populasi penelitian ini siswa kelas X Teknik Mesin Otomotif. Desain yang digunakan yaitu one group design. Teknik sampling yang digunakan adalah teknik sampling jenuh/ sensus karena semua anggota populasi merupakan sampel dan sampel kurang dari 30 orang. Sampel penelitian ini  terdiri dari 22 orang. Metode pengumpulan data adalah observasi. Berdasarkan hasil penelitian didapatkan hasil rata-rata skor diskusi dan psikomotorik sebesar 18 dan 13 yang termasuk dalam kategori tinggi. Nilai rata-rata karakter sebesar 21 dengan kategori baik. Kesimpulan dari penelitian ini adalah ada delapan karakter siswa muncul selama pembelajaran berbasis teknologi informasi dengan metode diskusi. Enam karakter yang muncul berada dalam kategori baik dan dua karakter dalam kategori cukup. Nilai karakter yang muncul yaitu aktif, bekerjasama, rasa ingin tahu, tanggung jawab, kritis, percaya diri, demokratis, dan terampil.

Abstract

Effects of time and temperature on α-pinene isomerization reaction using catalysts Zr/natural zeolitewas studied. Characterization of the catalysts include: crystallinity, observed using X-Ray Diffraction, count Zr 4+ carried observed using X-Ray Fluorescence, area and porosity catalyst was observed using the Surface Area Analyzer, and acidity catalyst observed through gravimetric method. Isomerization reaction carried out in a batch reactor with temperature variations 90, 120 and 150 C and reaction time variations of 60, 90, 120, 150 and 180 minutes. Best results of isomerisation in this study was obtained at 150 derajat C with a reaction time of 180 minutes. Kindsof isomer obtained was observed using GCMS. Catalyst characterization results indicate that modification of the catalyst by cation Zr increases the acidity from 2.76 to 6.64 mmol/g and does not damage the crystal structure significantly. The highest product conversion in this research is 9.24%, less than the maximum results caused by pre-treatment of the catalyst produces a low area. Thus, temperature and reaction time affect the concentration of α pinene isomerization product in addition to the effect of the catalyst used.
