Abstract

ABSTRAK Tujuan penelitian ini adalah untuk mengevaluasi dinamika folikel pada ovarium domba pascatransplantasi secara intrauterin pada kelinci pseudopregnansi. Transplantasi dilakukan pada kelinci pseudopregnant hari ke 1 atau ke 7. Ovarium kembali diambil pada hari ke 5, 7, atau 9 setelah transplantasi. Untuk menentukan dinamika folikel pada ovarium domba pascatransplantasi dan menghitung jumlah folikel pada berbagai tahap perkembangan, ovarium domba pascatransplantasi dijadikan preparat histologis dengan metode parafin dan pewarnaan HE. Hasilnya masih ditemukan semua tahapan perkembangan folikel (folikel primordial, primer, preantral, dan antral) pada semua kelompok perlakuan. Jumlah folikel pada 5, 7 atau 9 hari pascatransplantasi menurun nyata (p<0,05) kecuali jumlah folikel primordial pada kelompok 5 hari pascatransplantasi (634,7±56,88) tidak berbeda nyata dengan kontrol (683,7±61,55). Dapat disimpulkan bahwa dinamika folikel ovarium domba pascatransplantasi pada kelinci pseudopregnansi masih dapat ditemukan pada semua kelompok perlakuan. THE FOLLICLE DYNAMICS OF EWE OVARIAN POST-INTRAUTERINE TRANSPLANTATION TO PSEUDOPREGNANCY RABBIT ABSTRACT The objective of this study was to evaluate the follicle dynamics of ewe ovarium post-intrauterine transplantation to pseudopregnanty rabbit. The experiment was concerned with the 1st or 7th days of pseudopregnancy to receive the ewe ovarium. After 5, 7, and 9 days transplantation the ewe ovarium were recollected. In order to determine the follicle dynamics of ewe ovari post-intrauterin transplantation and to count the number of each stage, the ewe ovari was prepared using the paraffin methods and staining with HE. The results showed all stages of the follicle dynamics (Primordial, Primary, Preantral and Antral follicle stages) were still found in all groups of treatment. The number of follicles decreased significantly (p<0.05) except the number of Primordial follicles of the 5 days post transplantation (634.7±56.88) was not significantly different (p<0.05) compared to the control group (683.7±61.55). It could be concluded that the follicle dynamics of ewe ovarium post-intrauterine transplantation in pseudopregnancy rabbit still found in all groups of the treatment.

Abstract

ABSTRAK Telah dilakukan penelitian untuk mengetahui viabilitas oosit domba yang dikoleksi dari ovarium domba pascatransplantasi intrauterus pada kelinci pseudopregnant. Ovarium domba ditransplantasikan dalam uterus kelinci pseudopregnant pada hari pertama bunting semu dan kemudian diambil kembali setelah lima (P5) atau tujuh (P7) hari transplantasi. Sebagai kontrol digunakan ovarium segar. Oosit dikoleksi dari ovarium pascatransplantasi dengan metode slicing (pencacahan) di dalam media phosphate buffer saline (PBS) yang disuplementasi dengan 5% fetal bovine serum (FBS) dan 100 IU penicillin-streptomisin/ml. Oosit hasil koleksi selanjutnya dimaturasi dalam Tissue Culture Medium (TCM)-199 yang disuplementasi dengan 10% FBS, 10 IU follicle stimulating hormone (FSH)/ml dan 100 IU penicillin-streptomisin/ml. Oosit selanjutnya diinkubasi dalam inkubator CO2, dengan kandungan CO2 5% dan suhu 38ºC, selama 24 jam. Setelah dimaturasi, oosit diwarnai dengan aceto-orcein 2% untuk menentukan status inti oosit. Hasilnya memperlihatkan bahwa oosit yang mampu mencapai tahapan perkembangan dengan status inti M-II setelah maturasi pada P5 (35,05%) dan P7 (35,24%) nyata lebih sedikit (p<0,05) dibandingkan dengan kontrol (56,65%). Dapat disimpulkan bahwa viabilitas oosit domba pascatransplantasi di dalam uterus kelinci pseudopregnant masih ditemukan meskipun persentasenya lebih rendah. EWE OOCYTE VIABILITY FROM EWE OVARIAN AFTER INTRAUTERINE TRANSPLANTATION TO PSEUDOPREGNANT RABBIT ABSTRACT The aim of the present study was to investigate the ewe oocyte viability from ewe ovary after intrauterine transplantation to pseudopregnant rabbit. The ewe ovary was transplanted intrauterine on day 1 to pseudopregnant rabbit and oocytes recollected on five (P5) or seven (P7) days posttransplantation. The fresh ovary was used as the control. The oocytes were collected from the ovaries by slicing method in Phosphate Buffer Saline (PBS) supplemented with 5% of Fetal Bovine Serum (FBS), and 100 IU/ml of penicillin-streptomycin. Oocytes were matured in Tissue Culture Medium (TCM)-199 supplemented with 10% FBS, 10 IU/ml of Follicle Stimulating Hormone (FSH), and 100 IU/ml of penicillin-streptomycin. Oocytes were incubated in CO2 incubator with 5% CO2, 38°C for 24 h. After maturation, the oocytes were stained with 2% aceto-orcein to determine the nuclear oocytes status. The result showed that the oocytes could reach the M-II phase from P5 (35.05%) and P7 (35.24%) decreased significantly (p<0.05) compared to the control (56.65%). However it can be concluded that the oocytes viability still preserved intrauterine in pseudopregnant rabbit.

Abstract

The objective of this study was to evaluate the development of ewe follicles and oocytes viabilityfollowing an intrauterine transplantation of the ovaries into pseudopregnant rabbit. The experiment wasconducted in day 1 of pseudopregnant rabbit. Following transplantation for 5, 7, and 9 days, the ovarieswere recollected. The development of follicles determined by counting the number of follicles in paraffinembedded ovaries after staining with haematoxylin-eosin (HE). The viability of oocytes was determined byslicing the ovaries. Oocytes were incubated in CO2 incubator with 5% CO2, 38°C for 24 hours. After maturation,the oocytes were stained with 2% aceto-orcein to determine the nuclear oocytes status. The result showedthat follicles were detected in all stages of their development (primordial, primary, preantral, and antralfollicle stages), but their number decreased significantly (P<0.05) 5, 7 or 9 days after transplantation,except for those at primordial stage which at day 5 post-transplantation (634.7±56.88) were not significantlydifferent to the control (683.7±61.55). After maturation, the oocytes that were able to reach the M-II phaseat day 5 and day 7 post-transplantation were 35.05% and 35.24% respectively. They were significantly(P<0.05) lower than the control (56.65%). In conclusion, the development of follicles and oocytes viability inthe ewe ovaries in pseudopregnant rabbits was still preserved during intrauterine transplantation.

Abstract

The balenggek chickens has been raised by rural communities for many generations in Payung Sakaki, Tigo Lurah, and Pinang Awan, Regency of Solok, Province of West Su matera.The Balenggek chicken had low sperm concentration per ejaculated. This study was carried out to identify the influence of PBS as suspender to the sperm quality of ba lenggek chickens for Artificial Insemination (AI). Sperm collected by massage technique and suspended in physiologic solution (NaCl 0,9%) or PBS (Phosphate Buffer Saline) with sperm concentration 200 million sperm/ ml suspender. Suspended sperm were places in Cool Case (-4°C), and Room Temperature (26-28°C) for 2 or 4 hours before AI. The egg that resulted in AI was incubated to analysis the egg fertility and hatchability. The results shows there were no significantly higher (p<0,05) to all treatments. The eggs fertility 85-90% and the eggs hatchability was 65-70% in both treatments for all temperature condi tions. It can conclude that PBS can suspender and preserve the Balenggek chicken sperm quality for AI.   Keywords: balenggek chickens, PBS, sperm quality, artificial insemination

Abstract

Hen’s egg is one of the source of protein that contains very high quality of nutrition, beside the hen’s egg is easy to find. Avian influenza which is very popular is lately, especially in Indonesia has been the causes of decreasing of egg’s producing more over, this case also brings some bad effect for hen’s population that die because of the ‘stamping out”. It cause hen’s population becomes decreasing and the repopulation going slowly, however to know more about the most productive chicken is still less. This research in order to know the fertility and the lay egg force of chicken (Gallus domestica) that came from the regions all around Padang. This research is descriptive the sample is take with purposive sampling way. One hundred egg from all around Padang is incubated at 370C degree and at the 7th day, the incubation is observed bout the fertility and candling. However at the 23rd day the lay egg force of the incubation is observed. The data for this research with percentage way. The result is proving that the lay egg force in Padang is high. Abstract

Linear Alkylbenzene Sulphonate (LAS) is a detergent surfactant that impact most of water included the ponds. The aim of this research to evaluate the effects of Linear Alkyl benzene Sulphonate to opperculum movement and coughing frequency of carpy fish. This research was conducted in Zoology Laboratory, Biology Department of Negeri Padang University. Experimental design was completely Randomized Design (CRD), 5 time repli cated per group treatment. The treatment were A. control 0 ppm LAS; B. 0.1 ppm LAS; C. 0.2 ppm LAS; D. 0.3 ppm LAS, and E. 0.4 ppm LAS. The data of the number of operculum movement and coughing frequency was counted for a minute per a carpy fish, for 5 time. The data were analyzed using one tail ANOVA and Scheffe Test. The results showed that the operculum movement was increased significantly higher (p<0.05) in the all group treatment compare to the control, as the LAS concentration treatment increased too. The LAS treatments were not significant to coughing frequency in all treatment. It can conclude that LAS coused increased the opperculum movement in carpy fish (Cyprinus carpio L.). Abstract

The aimed of this research was to identify the diversity of diatomae in the river of Negeri Padang University. This research was done in February – March 2011. Sampling technique was used Stratified Random Sampling with four stations. The samples got from every station five times in 10.00-14.00 pm.The Paralon pipe (15 cm in diameter) used to sampling vertically in the bottom of the river. One liter water was used as sample for diatomae identification. Diatomae samples drooped ten droop by 4% formaldehide prior identified in Zoology Laboratory, Biology Department. Before, microscope analyze, the diatomae was prepare with dried technique prepared slide. All the species of diatomae then documented by photomicrography technique. The result shows threa are 42 genus of diatomae contained 9 genus from class of Coscinodiscophyceae, 7 genus from class of Fragilariophyceae, and  26 genus from class of Bacillariophyceae. Abstract

The aimed of this research to know the coused of the Hawksbill Turtle (Eretmochelys imbricata L.) population decreased. The World Conservation Union lists them into endangered species. The hatchability of Hawksbill Turtle eggs is 50%. One of the influent factors of it was the nest pattern including the nest-depth and eggs layer. The research conducted factorial CRD with 35, 55, and 75 cm depth nest factor and 1, 2, or 3 egg layers factor. The natural nest became a control group. The data analyzed by ANOVA and DNMRT. The result shows percentage of eggs hatchability based on depth-nest in control, 35, 55, and 75 cm are 91.2%, 83.6%, 23.3% and78.7%. Based on the eggs layer, the eggs hatchability shows in 1, 2, or 3 egg layers and control are 79.12%, 85%, 91.6% and 82.23%. It can conclude that the depth-nest and eggs layer influence the eggs hatchability of Hawksbill Turtle (Eretmochelys imbricata L.) Keywords: Hawksbill Turtle, nest, eggs, hatchability

Abstract

Abstract:Scylla paramamosain E. is one type of crab that lives in mud substrat at mangroveareas Buo Gulf. Scylla Paramamosain E. is favored by the people in the Buo Gulf. Now Scylla paramamosain E. population has begun to decrease and sizes caught are relatively small . Based on at this research take plan to identification gonad maturity of Scylla paramamosain E. at mangrove forest in the Buo Gulf Bungus of Padang. This study was conducted in May until July 2013 used the descriptive method. The results showed there were different gonad maturity of Scylla paramamosain was caught in the Buo Gulf. In May, Scylla paramamosain E. had immature gonads 33.33 % while the percentage of matured at a rate of66.67 % . In June, the percentage of immature gonads and matured amount 30.0% and 70 % . In July, there were 41 , 66 % in the immature stage and 58.32 % matured. Among of the three months, gonad maturity Scylla paramamosain E in July at the mostly mature gonad condition and ready for spawning. In addition biogeochemical cycles can influence and bring sufficient nutrients for survival Scylla paramamosain E. In general the level gonad maturity in Scylla paramamosain E. in the Gulf Buo mangrove forest in May to July 2013 TKG I5.68 % , 29.32 % TKG II , III TKG 21.27 % , 11.15 % IV TKG , TKG V 32.59 %. Environmental factors such as temperature measured during the study ranged from 280C , salinity ranges from 31 % o - 32 % o , pH around 7 ( Neutral ) and an average water depth of 58.5 cm .
