Abstract

This study was conducted to examine activity of medaka (Oryzias latipes) â-actin promoter (mBP) in common carp(Cyprinus carpio) as the first step towards development of common carp transgenic in country. Gene constructpmBP-hrGFP that consists of mBA promoter and humanized Renilla reniformis green fluorescent protein gene(hrGFP) was injected into cytoplasm of one cell stage of common carp by using microinjector. PmBP-hrGFPconcentration used for microinjection was 50 μg/mL aquabides. Parameters observed were survival rate of embryo(SRe), hatching rate (HR) and expression of hrGFP gene. SRe was calculated before eggs hacthed, while hatchingrate (HR) was after all of eggs hatched. The activity of mBA promoter was analyzed by observation of hrGFP genetransient expression using a fluorescence microscope. The results of experiment showed that SRe (87,5%) andHR (79.2%) of control was respectevily higher than that of injected treatment (75.0% & 61.7%). Expression of hrGFPwas observed firstly at blastula (12 hours after fertilization) to 1-day-old larval stages (24 hours after hatching)with higher gene expression at blastula to late gastrula stages. Percentage of micronjected larvae expressinghrGFP at 6 hours after hatching reached 71.6 ± 6.7%. Conclusion was that mBA promoter could drove hrGFPexpression in common carp, hence it can be used to produce common carp transgenic by changing hrGFP withgenes correlated with important traits in aquaculture.

AKUMULASI LOGAM BERAT DAN PENGARUHNYA TERHADAP SPERMATOGENESIS KERANG HIJAU (Perna viridis)

Abstract

Perairan Teluk Jakarta, Banten, dan Lada telah mengalami pencemaran, terutama tercemar oleh logam berat seperti Pb, Cd, Cr dan Hg. Penelitian ini bertujuan untuk mengetahui kandungan logam tersebut dalam gonad kerang hijau (Perna viridis) dan pengaruh akumulasi logam tersebut pada spermatogenesis kerang hijau. Parameter yang diamati adalah jumlah spermatogenium, spermatosit primer, spermatosit sekunder, spermatozoa, total se-sel kelamin, diameter, luas dan volume lumen folikel gonad kerang hijau, pada tingkat IV spermatogenesis. Hasil penelitian menunjukan bahwa gonad kerang hijau yang berasal dari Teluk Jakarta telah terakumulasi logam berat Pb = 359.75±272.41 ppb; Cd = 36.559±21.90 ppb; Cr = 504.21±448.64 ppb dan Hg = 0.0092± 0.0085 ppb. Logam Cd (13.13 ppb) dan Pb (0.077 ppm) ditemukan dalam gonad kerang hijau yang berasal dari Teluk Banten, tetapi tidak terdeteksi logam Cr dan Hg. Namun tidak ditemukan logam berat tersebut dalam gonad kerang hijau berasal dari Teluk Lada. Di Teluk Jakarta ditemukan korelasi antara kandungan logam Cd terhadap perkembangan jumlah sel spermatozoa (r = 0.64), total sel kelamin (r = 0.60, diameter (r = 0.57), luas (r = 0.71) and volume (r = 0.71) lumen folikel gonad. Logam Hg berpengaruh terhadap perkembangan spermatogenium (r = 0.68) dan spermatosit sekunder (r = 0.61), sedangkan logam Cr mempunyai pengaruh terhadap luas (r = 0.75) dan volume (r = 0.75) lumen folikel gonad.Kata kunci: akumulasi, logam berat, spermatogenesis dan kerang hijau.

Abstract

The most widely cultivated seaweed in aquatic Karimunjawa is Kappaphycus alvarezii, because low venture capital, high market demand, low-cost production technology, production cycle is short, post-harvest handling is easy, and simple as well as market share is still open. This study aims to (1) Evaluate the feasibility of seaweed cultivation; (2) Identify factors that influence internal and external business seaweed cultivation; (3) Develop appropriate strategies in business development efforts to the cultivation of seaweed. Financial data were analyzed according to the criteria value NPV, B/C ratio, IRR, PBP, and BEP. Identification of environmental factors was evaluated by IFE dan EFE matrix, mapped to the IE matrix to view the company´s position. Strategy formulation with SWOT and QSP matrix. The result of feasibility analysis showed that seaweed cultivation efforts with longline method financially profitable and feasible. This was indicated by a positive NPV value of 30.81 million rupiah; B/C ratio (2.69), IRR (47.58%); PBP 1.61 years; BEP 13.23 million rupiah or sales of 1,474 kg of dried seaweed. With a total score value of the internal-external matrix of 2.52 and 2.83. The combination of these two values indicates that the position of the business lies in V cells or growth strategies. The most appropriate strategies are the empowerment of members and business groups to increase their business, and increased cultivation of technical skills for the improvement of product quality. These three strategies can be implemented simultaneously as mutually supporting one another.

Abstract

The purpose of this study was to determine the magnification and technology marketing at UD Sumber Rezeki and get a new breakthrough in marketing efforts and the enlargement of catfish, determine the feasibility and efficiency of marketing efforts and the enlargement of catfish, find the prospect of catfish processed products, know the SWOT in the enlargement and marketing of catfish and determine the strategy of business development and marketing of catfish enlargement on UD Sumber Rezeki. Technology production processes carried out by following the catfish aquaculture activities at UD Sumber Rezeki Parung, West Java, and then analyzed the feasibility of their business. The analisis through qualitative analysis, quantitative analysis of IFE and EFE, Matrix IE and SWOT. Catfish farming in the UD Sumber Rezeki Parung, West Java feasible to be implemented with the value of B/C ratio > 1,26. BEP catfish production. Rp9631.76 per kg, NPV 14% on the value of Rp38,140,956, 17% IRR and PBP 3 years 9 months. Efficiency for catfish enlargement in UD Sumber Rezeki determined by the use of water by gravity, the use of probiotics to save time, use sinking feed and floating feed. Based on the IFE (2.83) and EFE (2.81), then the position at UD Sumber Rezeki phase of growth and stability and are in quadrant 5. Based on the analysis of SWOT matrix and IE matrix, then the strategy for developing UD Sumber Rezeki, Parung, West Java are (1) market penetration by selling directly to end customer, (2) diversification of catfish products and (3) improve the competence of human resources.

Abstract

Tuna Indonesia mostly exported in fresh and frozen (55%), and 45% in processed tuna. It is therefore necessary to study the competitiveness of processed tuna. The method used is the Revealed Comparative Advantage (RCA) and the analysis of the Competitive Profile Matrix (CPM) Analysis, In the year 2006 to 2010, Indonesia has a fresh tuna RCA index between 4.56 to 8.18, tuna frozen at 0.49 to 1,43 and processed tuna fish ranged from 1.25 to 2.68. Based on the analysis of the competitive profile, three factors of production and marketing of highly influential on the competitiveness of tuna fish are (1) the quality of the resulting processed tuna fish with a weight of 0.143, (2) tariff and non tariff barriers with a weight of 0.114, and (3) Development and Promotion Intellegence market with the weight of 0.110. As for the human and institutional factors, factors which have an important role in increasing competitiveness is (1) The Role of Government in the development of the tuna processing industry with a weight of 0.147, (2) Availability of human resources capable of handling quality with weights 0.135, and (3) the role of government in the prevention and handling of illegal fishing by weight of 0.130. Based on the analysis of RCA analysis and matrix analysis of the competitive profiles of priority strategies that can be done to improve the competitiveness of Indonesia´s processed tuna to the factors of production and marketing are as follows (1) Improve the quality of processed tuna Indonesia; (2) Encourage overcome tariff and non tariff barriers; (3) enhance market development and promotion Intellegence, The strategic priorities for the human and institutional factors are (1) Increase the Role of Government in the development of the tuna processing industry, (2) Improving the human resources capable of handling quality, (3) Eradication and control of illegal fishing.

Performance of Nile tilapia juvenile fed diet containing different dosages ofrecombinant common carpgrowth hormone

Abstract

The studywas conductedtodetermine thedose of recombinant common carp growth hormone (rCcGH) supplemented in the commercial diet that generates the best performanceonbodyweight, biomass, andfeed conversion rate of juvenile tilapia.The dose of rCcGH administered was10, 20, and30mg/kg of commercial feed, andnorGH supplementation as control. Eachtreatment was designed in triplicates. Juveniles at average bodylengthof about2cm(average body weight of 0.7 g) were reared in a density of25fish/m2, for three weeks in hapa net at a size of 2×2×1 m3. Fish were fed diet containing rCcGH twicea week with an interval ofthreedays. The juvenile were fed with the rCcGH supplemented diet three times daily at satiation. The results showedthat average body weight andbiomass of fish treated by rCcGH with the doses of 20 and 30 mg/kg feed was higher (p<0.05) than that of 10 mg/kg and control. In addition, feed conversionrate of 20 and 30 mg/kg feed treatments was lower (p<0.05) than that of 10 mg/kg and control. RT-PCR analysisshowedthat expression level of IGF-1 gene in juvenile liver was increased in parallel with the increasing of rCcGH dosages supplemented in diet.This suggestedthatIGF-1 plays an important rolein the induction ofgrowth,andfeed conversionefficiencyof tilapiafed diet containing rCcGH. Growth and survival of giant gourami juvenile immerse indifferent frequencies using recombinant growth hormone

Abstract

This study was aimed to determine the immersion frequency of recombinant growth hormone (rGH) in the growth stimulation of giant gourami juvenile. A total of 50 larvae at second day after first feeding, was immersed in 30 ppt salt water for 2 minutes, and then transferred into 200 mL of 9 ppt salt water containing 30 mg/L rGH in inclusion bodies form. Immersion was performed for 60 minutes and repeated once a week for two, three, four, and five weeks. As the control, fish was immersed with inclusion bodies without rGH (control-1), bovine serum albumin (control-2), 9ppt saline water without rGH (control-3) and in freshwater (control-4). Fish were kept in 7L aquaria during the first four weeks, transferred into 50L aquaria for two weeks, and then in hapa (2×1×1 m3 in size) for two weeks. The results showed that the highest (p<0.05) average body weight of giant gourami juvenile (2.23±0.19 g) was obtained at four times (four weeks) immersion treatment. Similar body weight of fish in two times (1.72±0.29 g), three times (1.68±0.34 g), and five times (1.81±0.17 g) immersion was obtained. Fish body weight of all controls was lower than the rGH-immersed fish. Average body weight of four time rGH-immersed fish (4.01 g) remained higher than other treatment and control (1.96 to 2.36 g) on 8th week of fish rearing. Survival of four times (96.67±1.15%), and five times rGH-immersion fish (95.33±4.16%) was similar, and higher than other treatments and controls (77.33 to 89.33%). Thus, higher growth rate and survival of giant gourami juvenile could be achieved by four times rGH immersion. Keywords: recombinant protein, growth hormone, immersion frequency, giant gourami

Abstract

Pecel Lele Lela Restaurant branches Pinangranti  is a restaurant that first presents the traditional Deep-fried Catfish food menu into a unique concept with modern restaurant management. This Research aims to (1) find out feasibility, (2) understand the marketing mix of services and assessments of consumer againt services marketing mix so far applied,  (3) analysis the  factors of internal and external environment that affect marketing strategies, and (4) establish  the  priority of alternative marketing strategies that can be  applied. Sampling technique in this research is purposive sampling. The method used in this research is descriptive method that focuses on invesment and business development aspect, as well as NPV, B/C, PBP, IRR, SWOT and QSPM analysis. Based on financial criteria feasibility on discount rate of 6% NPV Rp2.535.752.046. Net B/C 4,22. IRR 21,34% (IRR > 6%).The return on investment (PBP) which is based on the value of now with the discount rate of 6% are 1.81. It means that the restaurant pecel lele lela branch Pinangranti, East Jakarta worthy to be implemented.The resulted total scores value of both Internal Factor Evaluation (IFE) and External Factor Evaluation (EFE) were respectively 2,543 and 2,310 which puts the position on quadrant V, which hold and maintain. Abstract

The world market demand for fishery products is increasing even frequently not fulfilled. It required an effort to overcome the problems in fulfilling the demand for fishery products. Types of freshwater fish cultivated for balita fish at UD Suhada is a type of tilapia and goldfish. The purpose of this study is to examine the feasibility of balita fish agribusiness investment in UD Suhada; identify the internal and external factors that influence the development of agribusiness balita fish at UD Suhada; formulate and recommend appropriate alternative development strategies to be applied to agribusiness balita fish at UD Suhada in business development. Methods of data collection using purposive sampling method, is by deliberately selecting the sample to be studied as a respondent. Respondents were selected from the management and the workers UD Suhada. Financial analysis at 14 percent of interest rate and 5 years of project lifetime resulted in feasible decision with Rp1,105,005,110 of NPV; 4.85 of net B/C Ratio; 38.95 percent of IRR, 3 years and 5 month of PBP; and Rp1,522,035,876 or 32.725 kg of BEP. The combination of value IFE EFE values for 2.688 and 2.651 in the IE matrix shows that the position of the business in V (five) cells, that cell growth. Based on the SWOT analysis and QSPM analysis the strategic priorities is to create a diverse variety of products while maintaining the quality of balita fish, extending the range of distribution and marketing, in collaboration with researchers in developing the quality of balta fish products to face competition.Key words: balita fish, business feasibility, agribusiness, business development strategy

Abstract

An ironic thing that Indonesia as maritime country have insufficiency salt problem. Salt is strategic commodities that can be easily produced by evaporation of sea water and policy salts have been issued since the Dutch colonial era. In 2011 the Government of Indonesia make toward self sufficiency salt policy. This research was aimed to analyze the implementation efectivity of Salt Business Empowerment Program (Pemberdayaan Usaha Garam Rakyat or PUGAR). The research design used purposive and snowball sampling to select 70 respondents in Losarang Village at Indramayu. The data were analyzed by using Quantitative Analysis, Qualitative Analysis, SWOT and MAHP. The result showed PUGAR in Losarang, Indramayu implemented with effectiveness. This is indicated by salt production target achieved and increasing the salt farmer welfare. Implementation PUGAR make salt productivity in Losarang 90,43 ton/ha, increasing salt farmer income, empower 17 the People´s Business Group Salt (Kelompok Usaha Garam Rakyat or KUGAR) with the amount of 170 salt farmers, give technological innovation and quality salt production and give job for 778 peoples as salt farmers, farm workers and transport workers. Obtained result IFE of salt bussiness is 2,608 and EFE is 2,673. Moreover the research calculate salt business feasibility that known from B/C ratio > 1, business profit margin, appropriate salt area owned by farmers, business gap analysis of salt and break even analysis. Break even analysis to determine the selling price of the large volume of salt and salt production business people to reach the point of no profit and no loss (break even).
