Abstract

Study of glycoconjugates in the mandibular gland of swine was performed. The aim of thisstudy was to identify the composition of mainly terminal carbohydrate residues ofglycoconjugates in the mandibular gland of swine using Alcian Blue (AB), Periodic AcidSchiff (PAS) and lectin histochemistry staining methods. Results showed that neutralcarbohydrates were detected in the mandibular gland ie. mucous cells, serous cells, epithelductus and lumen ductus cells. Acidic carbohydrates were detected in sereus cells of thegland. Whereas in the mucous cells, epitel ductus and lumen ductus cells there were noacidic carbohydrates detected. Using six types of lectins ie: PNA, RCA, WGA, LCA, ConAand UEA the terminal carbohydrate residues detected in the mucous cells of the gland weregalactose, GlcNAc, GalNAc, sialic acid and fructose. In the epitel ductus cellscarbohydrates detected were galactose, GalNAc, sialic acid, fucose and manose,respectivelly.

Abstract

In recent years, groups of antimicrobial peptides, mainly produced from lactic acidbacteria in yoghurt has been used extensively in the health field as an antibacterial andfood biopreservative. This aims of this study to determine the physicochemicalcharacterization and antimicrobial properties of bacteriocin which extracted from theyoghurt. We used Saphylococcus aureus and Eschericia coli bacteria for this trial. Weexamined physicochemical characters of the bacteriocin such as pH, thermal stability, andproteolytic effect. Also, we conducted the antimicrobial activties to S. aureus and E. coli.The results showed that the bacteriocin which extracted from the yoghurt could inhibit thegrowth of bacteria S.aureus, and E. coli. In order to, bacteriocin havephysicochemical properties, such as optimum active at pH 5, is stable against heating upto 121°C for 15 minutes, and sensitive to proteolytic enzymes

Abstract

ABSTRAK Kitosan adalah polimer polisakarida merupakan suplemen diet yang telah luas digunakan di bidang kedokteran untuk penurun kolesterol dan mengurangi berat badan. Penelitian ini bertujuan untuk mengetahui pengaruh konsumsi kitosan terhadap kadar kolesterol, trigliserida, kadar glukosa darah serta terhadap kadar mineral kalsium dan fosfat dalam serum. Sebanyak 12 ekor kelinci jantan ras New Zealand white berumur 5 bulan dengan bobot badan antara 1,5 – 1,6 kg telah digunakan dalam penelitian ini. Hewan percobaan dibagi menjadi empat kelompok perlakuan, yaitu kelompok I (kontrol negatif), kelompok II-IV, yaitu kelompok perlakuan yang diberi kitosan masing-masing 1, 2, dan 4%. Kadar total kolesterol dan trigliserida serum di analisis menggunakan metode kalorometri enzimatis, kadar glukosa darah dianalisis dengan metode biosensor glukose oksidase, kadar kalsium metode spektrophotometri serapan atom (AAS), dan fosfat dengan metode spektrophotometri. Hasil penelitian menunjukkan bahwa pemberian kitosan 2% dan 4% dapat menurunkan kadar total kolesterol, trigliserida, kalsium, dan fosfat serum serta berbeda nyata (P<0,05) dibanding dengan kontrol negatif, dan tidak berpengaruh terhadap kadar glukosa darah. THE EFFECT OF ADMINISTRATION CHITOSAN ON MINERAL AND CHOLESTEROL CONCENTRATION SERUM IN RABBITS ABSTRACT Chitosan is a polysaccharide polymer is a dietary supplement that has been widely used in medicine for lowering cholesterol and reducing weight. This study aims to determine the effect of chitosan intake on cholesterol, triglycerides, blood glucose levels as well as the minerals calcium and phosphate levels in serum. A total of 12 male rabbits of New Zealand white 5 months old with body weight between 1.5-1.6 kg were used in this study. The animals were randomly divided into four treatment groups: group I (negative control), group II-IV, namely chitosan treatment group were given respectively 1%, 2%, and 4%. Levels of total serum cholesterol and triglycerides in analysis kalorometri enzymatic method, blood glucose analysis by glucose oxidase biosensor method, calcium by atomic absorption spectrophotometri method (AAS), and phosphate spectrophotometri method. The results showed that administration of chitosan 2% and 4% can reduce total cholesterol, triglycerides, calcium, and phosphate and serum significantly different (P<0.05) compared with negative control, and no effect on blood glucose levels.

Abstract

Isoflavone compounds have been known to have biological effects in improving health. The purpose ofthis study was to determine the bioavailability of isoflavone in plasma and its role as lowering bloodglucose levels in hyperglycemia rats. Bioavailability of isoflavones in plasma was performed in 18 ratsstrains Spraque Dawley age 3 months with 200 g of body weight on average and were given 300 ppm oralof isoflavone single dose. The analysis of isoflavon as done at 0, 1, 6, 12, 18 and 24 hours using HPLCmethod. Study effect on blood biochemical values used 25 male rats, which was divided into 5 treatmentgroups. One group as a negative control (K1) was given aquades; positive group hyperglycemia (K2), and 3other groups (K3, K4 and K5) were a positive groups of hyperglycemia and given isoflavone different doses,i.e. 1.0, 1.5 and 2.0 mg/200g bw per oral. Blood glucose were analyzed by using glucose oxidase biosensor,while cholesterol, HDL (Hight-density lipoprotein) and triglycerides were analyzed enzymatically using aspectrophotometer. The results showed the analyzes isoflavone genistein was more efficiently absorbedthan daidzein and glisitein. Isoflavone as found at highest concentration in plasma at 6 hours after oraladministration i.e: genistein of 11,17 ppm, daidzein of 2,86 ppm and glisitein of 2,47 ppm. Theadministration isoflavones at dose 2 mg/200g bw could decrease blood glucose levels in hyperglycemiarats, and also has hipocholesterolemia effect.

Abstract

The present study was carried out to investigate the effects of tapak dara (Catharanthus roseus) onblood glucose level and insulin profile in hyperglicemic rabbits. Fifeteen local male rabbits were used forthis study. The rabbits were randomly divided into five groups. Group 1 (K-), a control negative group;group 2 (K+), a control positive hipergliccemia; group 3 (KT1) and group 4 (KT2), were groups hiperglicemiaand treated with water extract of tapak dara doses 1 and 2 g/kg bw, respectively; and group 5 (KO), a grouphiperglicemia that treated with glibenclamide 2 mg/kg bw. The result showed water extract of tapak daradose 1 g/kgbw could not decrease the blood glucose level in hyperglycemic rabbits, while dose 2 g/kg bwcould decline blood glucose level in rabbits. This decline had no significantly difference compared withglibenclamide treatment (P> 0.05). Immunohistchemistry result indicated that water extract of tapakdara could stimulate beta cells pancreas to produce insulin.

Abstract

A study was conducted to isolate and identify of lactic acid bacteria originated from gastric fluid of bali cattle, and to determine their potential as the candidates of biopreservative. Lactic acid bacteria were isolated by culturing the gastric fluid of bali cattle in de Mann, Rogosa, Sharpe (MRS) medium; screening the bacteria, and identification of bacteria species by Analytical Profile Index (API) 50 CHL Kit. The results showed that, the new species of lactic acid bacteria were isolated and identified as Lactococcus lactis spp lactis 1 (SR21 isolate) and Lactobacillus brevis 1 (SR54 isolate) that have broad spectrum antimicrobial activities. It is clear from this study that a potential lactic acid bacteria producing antimicrobial agent can be isolated from the gastric fluid of bali cattle.

Abstract

This research was carried out to evaluate the effect of chitosan supplementation in the chicken food with high cholesterol diet on total cholesterol level and serum lipoprotein profile of chicken. The aims of this study were to evaluate the effects of chitosan supplementation on lipid level, total cholesterol, triglyceride, High density lipoprotein (HDL), Very low density lipoprotein (VLDL) and Low density lipoprotein (LDL) concentration on serum chicken. The experimental design used in this study was completely randomized design with 4 treatment groups. Group K (as a controlled without cholesterol er and chitosan), group A (treated with cholesterol 1% without chitosan), group B (treated with cholesterol 1% and chitosan 15%) and group C (treatment with cholesterol 1% and chitosan 30%). The results showed that treatment with egg yolk powder (group A) could cause hyperlipydemia on chickens and cholesterol total was significantly different (p<0,05) compared to group K (control). Treatment with 30% chitosan were reducing lipid concentration, cholesterol total, triglyceride, VLDL and LDL serum of chicken, respectively (p<0,05) compared to group A (egg yolk powder without chitosan), total HDL concentration was not significantly different (P>0,05). It was concluded that the suplementation of 30% chitosan in the chicken food could reduce total serum cholesterol level in hyperlipydemia chicken.

Abstract

Research to confirm the abnormalities of blood corpuscles in Bali ducks based on blood smear examination has been conducted. The research samples consisted of 105 ducks from various regencies in Bali. The blood smear examination was conducted by the method described in this literature and the data was collected and tabulated by means of the descriptive method. Erythrocyte abnormalities were: polychromasia (61.9%), anisocytosis (17.1%), poikilocytosis (21.0%). Abnormalities associated with leukocyte composition were: Lymphopenic (46.7%); Heterophylic (44.8%); Heteropenic (3.8%), also Thrombocyto penia (2.9%) was present in 105 ducks. In conclusion 61,9% ducks examined deal with regenerative anaemia. They also have some blood abnormalities such as basophylia, eosinophylia, heterophylia, heteropenia, lymphopenia, and monocytosis.

Abstract

Glycogen is found at all of body tissue, especially mostly in liver and muscle. The objectives of thisresearch was to evaluate the content of glycogen in liver and muscle of diabetic rats that were treated withextract of tempe. A total of twenty male Spraque Dawley rats of 2 months old were used in this study. Therats were divided into four groups: (1) negative control group (K-), that were not treated extract of tempeand nondiabetic, (2) positif extract of tempe group (ET), that were treated with extract of tempe andnondiabetic (3) positif diabetic group (DM), and (4) diabetic and extract of tempe group (DM+ET). Extractof tempe was orally administered with dose 300 mg/kg bw/day. The treatment was conducted for 28 days.Effect of extract of tempe on body weight of all rats was determined at various time interval at 0, 4, 7, 14,21, and 28 days. At the end of the experiment, all rats were sacrificed by cervical dislocation. Liver andmuscle gastrocnemius were collected for analysis of glycogen level. The result of this study showed thatadministration methanol extract of tempe of 300 mg/kg bw/day can increase body weight, glycogen synthesisin the liver and muscle in normal rats (rat of ET group) and also diabetic rats (rat of DM+ET group). At theend of research, diabetic rats (rat of DM group) were decrease of body weight up to 5.7%. On the rat ofDM+ET group, rat of K(-) group and rat of ET group were increase of body weight of 5.7%, 19.3% and 20.3%,respectively. Glycogen level both liver, and muscle at rat of ET group and rat of DM+ET group wereincrease each of 9.29%, 2.2% in liver and 18.27% and 4.02% in muscle. Glycogen level at rat of DM groupwere decrease up to 42.5% in liver and 31.6% in muscle.
