Abstract

The three Kendari local upland rice varieties i.e. Paedai Meeto, Pae Ikulaku and Pae Loio were studied for their ability to produce the embryogenic microspore and microspore callus from anther under high temperature stress pretreatments. The pretreatment was conducted by incubating the anther in a B medium at 33 oC in darkness for 0, 2, 4, and 7 days to induce the growth of embryogenic microspores. The formation of microspore calli was obtained by incubating the embryogenic microspores into the MO-19 embryo induction medium. Scoring was conducted on the percentage of: (1) embryogenic microspores, (2) symmetrical binucleat microspores, (3) multinucleate microspores, and (4) microspore calli or embryos. Each replication consisted of three hundred of individual microspores and every treatment was replicated in three times. The mean values of the data were analyzed by variance analysis, and the Fisher LSD mean separation at 0.05 of significance level were applied to judge the best treatments. The result showed that Pae Loio produced the higher frequency of embryogenic microspores after anther pretreatment in B medium for 4 days at 33 oC in darkness compared to other varieties. Culture of embryogenic microspore of Pae Loio in MO-19 medium over 30 days also produced the symmetrical binucleate and multinucleate microspores, and microspore calli at higher frequencies than those of other local varieties.Keywords: anther, calli, embryogenic microspores, high temperature pretreatment, upland rice

Abstract

Percobaan ini dilakukan di Kebun Praktek dan Laboratorium Agronomi Fakultas Pertanian Universias Haluoleo, sejak April 1998 hingga Februari 1999. Tujuannya adalah ingin mengetahui hubungan antara umur tanaman enam kultivar lokal padi ladang asal Kendari dan panjang pemunculan malai dengan tepungsari berinti satu yang telah mencapai lebih dari 50% dalam populasi. Keenam kultivar tersebut adalah (1) Pae Kuni (PK), (2) Pae Dai Meeto (PD), (3) Pae Hulo (PH), (4) Pae Iku Laku (PL), (5) Pae Mornene (PM), dan (6) Pae Wila (PW). Setiap pengamatan digunakan tiga malai sebagai ulangan, dan masing-masing malai dibagi tiga bagian, yakni: (a)bagian ujung malai, (b)bagian tengah malai, dan (c)bagian pangkal malai. Setiap bagian digunakan tiga bulir sebagai sumber antera. Komposisi tingkat perkembangan tepungsari dalam antera masing-masing malai dicatat sebagai sifat yang diamati. Data yang diperoleh melalui pengukuran dan penghitungan selanjutnya ditabulasi, kemudian dianalisis secara rata rata dan persentase. Hasil penelitian menunjukkan bahwa : (1)Pae Kuni (PK) tercapai pada umur 92 hari sesudah tanam (HST), malai berada dalam bungkusan maksimal kelopak daun bendera dan mengandung 77.55% tepungsari berinti satu, (2)Pae Dai Meeto (PD), tercapai pada 104 HST, malai berada dalam bungkusan maksimal kelopak daun bendera dan mengandung 78.94% tepungsari berinti satu, (3)Pae Hulo (PH) tercapai pada 95 HST, malai telah muncul sepanjang 1.50cm dan mengandung 84.50% tepungsari berinti satu, (4)Pae Iku Laku (PL) tercapai pada umur 118 HST, malai masih berada dalam bungkusan maksimal kelopak daun bendera dan mengandung 76.37% tepungsari berinti satu, (5)Pae Mornene (PM) tercapai pada umur 97 HST, malai telah muncul sepanjang 1.67cm dan mengandung 87.65% tepungsari berinti satu, dan (6)Pae Wila (PW) tercapai pada umur 117 HST, malai masih berada dalam bungkusan maksimal kelopak daun bendera, dan mengandung 81.25% tepungsari berinti satu.
