Abstract

Growth and Production of soybean varieties of Bradyrhizobium inoculation. Demand of soybean as a protein source continues to increase, but the increasing of domestic soybean production is still low so it must import from abroad. This is due to the decline in soil fertility and agricultural land conversion to residential. The solution, soybean planting can be done by optimizing the existing land commonly called agricultural intensification. In this research, agricultural intensification is done by adding Bradyrhizobium as biofertilizer. symbiosis between soybean plants with these bacteria can directly fix the nitrogen (N) from the air and can save in the N fertilizer of inorganic form. The objective of the research was to know the responses of the soybean varieties toward Bradyrhizobium inoculation. The split plot design with three replication was used, Bradyrhizobium as main plot and soybean varieties (Anjasmoro, Burangrang, Mitani, Grobogan, Tanggamus, Detam 1, and Detam 2) as sub plot. The result showed that the Bradyrhizobium were not significant affected on all of parameters. The varieties significantly affected on plant height at 5 weeks after planted, flower initiation, the time of harvest, the seed weights per plant, and 100 seeds weight. Interaction between Bradyrhizobium inoculation and soybean varieties were not significant on all of parameters .   Keyword : growth, soybean, inoculation, bradyrhizobium, variety

Abstract

The objective of this study is to have a reliable variety to extend on an used cultivated land. Thisresearch was carried out on an used cultivated land as fish pond with salty sulfate contained onKecamatan Percut Sei Tuan in a height site ± 2 m asl with its coastal far distance ± 1 km, has DHL6.5 - 6.8 and pH 4.1 – 4.6. The research adopted group random design with 18 varieties of rice asplant to test. The parameter to analyze such as height of plant, number of saplings, number ofproductive saplings, length of grains, numbers of grains, number of decomposed grains, grainsproduction and wet weight and dried weight of roots.The result of research indicated amongst different of varieties as tested showed a significantdifferent on all parameter as analyzed. Out of 18 varieties that has been tested were found 12varieties survivable, whereas the varieties prospective to extend on the used cultivated land locationwith salty sulfate such as Banyuasin, Dendang, Siak Raya, Punggur and Lambur regions.Keywords : tolerated test, rice, salty sulfate

Abstract

Centella asiatica is still categorized as a wild plant that has not undergone domestication. Sustainability of medicinal and aromatic plants for sustainable harvesting of medicinal plants so that it is not only in nature but also the harvest of cultivated so obtained uniform quality. Chemical constituents arealready known: contain some saponin compounds, including asiaticoside, madeccasoside and asiatic acid. This study aims to determine the bioactive content; asiaticoside, madeccasoside and asiatic acid on the top(leaves and petiol) and bottom (roots and tendrils) with HPLC method. The experiment was conducted in growth chamber Department of Biological Sciences and School of Pharmacy Auburn University, USA. This study conducted in September-December 2011. The materials used are Centella asiatica of Deli Serdang accession, Kabanjahe accession, Berastagi accession and Samosir accession. Equipment needed to support the growth chamber study were digital scales, plastic pots, HPLC system types waters alliance 2695 autosampler, 996 photodiode array detector, coulomb adsorbosphere C18 5μ, size 250 x 4.60 mm, UV absorbance, wavelength of 210 nm, flow rate 1.8 ml / min, injection volume of 20 mL, Empower Pro software, and others. The results that the pattern of centelloside (asiaticoside, madecassoside and asiatic acid), when the content of one of the high content of bioactive, the others bioactive will be lower or biosynthetic pattern toward a compound needed. Centelloside plant age affects the content of Centella asiatica. Centelloside pattern is influenced by the condition of the planting medium, very high levels of phosphorus, biosynthesis of centelloside more toward to asiaticoside.

Abstract

The objectives of this experiment was to observe morphology of leaves and content of asiaticoside in some accessions of Centella asiatica of lowland Sumatera Utara, Medan and Pantai Labu Deli Serdang and the highlands of Sumatera Utara, Berastagi, Kabanjahe and Samosir. The study also examines the link between altitude grow with the content of asiaticoside of Centella asiatica. Domestication research include: exploration and collection. This activity is carried out to collect Centella asiatica from different geographic locations with different altitudes. Later examination asiaticoside levels in the leaves of Centella asiatica by HPLC method and phytochemical test. Analysis of soil at all growing places were also conducted. The highest content of asiaticoside in accessions of Centella asiatica tested is as follows accession Deli Serdang (2.38%), Kabanjahe (1.43%), Medan (1.38%), Berastagi (1.38%), Samosir with shade (0.28%) and accession Samosir (0.24%). Altitude does not affect the content of asiaticoside Centella asiatica because in this study obtained the highest asiaticoside content found on lowland accessions namely (± 4 m asl), namely accession Pantai Labu  Deli Serdang (2.38%).

Abstract

One of the problems of immature rubber plants are non-productive period is too long which is about 5-6 years old. Various efforts have been made to speed up non-productive period, including the application of exogenous growth regulators. This is related to biological natural hormones found in plantduring immature rubber. One of the hormones that affects the auxin hormone . This study aims to analyze content auxin hormone found in some clones and plant age during immature rubber. This research was conducted descriptive / review directly to the field by taking leaf samples at the clones PB 260, PB 340 and RRIM 712 which have been determined. To determine the levels of hormones contained in rubber leaf, leaf samples were analyzed at the Research Center for Biology Indonesian Institute of Sciences in Bogor. Observation auxin hormone levels. The results showed that the type clones and different plant age have different content auxint. The highest content of auxin there at the age of 2 years is the plant on clones PB 260 (116.5 ppm) and RRIM 712 (114.0 ppm).

Abstract

The objective of this research was to know effect of paclobutrazol to the number of chlorophyl,flowering time, and harvesting time of two sorghum varieties. This research was conducted inexperimental field at Jl. Pasar I Tanjung Sari, Medan from October 2013-January 2014,used randomized block design with two factors paclobutrazol concentration (0 g/l, 0,50 g/l,0,75 g/l, and 1 g/l) and varieties (Kawali and Numbu). Parameters measured were the number ofchlorophyl, flowering time, and harvesting time. The result showed that paclobutrazol affectedsignificantly on plant height, stem diameter, and flowering time. Varieties have significant differenton the number of chlorophyl, flowering time, and harvesting time.The interaction of the two factorsnot affected significantly on all parameters. There was no interaction between paclobutrazolconcentration and varieties to all parameters measured.Keywords: paclobutrazol, variety, sorghum

Abstract

The objective of the research was to know effect of treatment low temperature, cryoprotectants,cryopreservation and it’s combination to seed viability of rosele. The research was conducted at theSeed Technology Laboratory, Agriculture Faculty, University of Sumatera Utara, Medan withheight ± 25 meters above sea level, in January to February 2014, using a randomized block designwith 5 level low temperature stratification, cryoprotectants and cryopreservation treatment as wellcombination and control. Parameters observed were moisture content seed (%), germination rate(day), germination normal (%), abnormal sprouts (%), seeds die (%), vigor index (%), wet weight ofsprouts (g), dry weight of sprouts (g). The results showed that rosele seed viability using liquidnitrogen significantly different than without giving liquid nitrogen, provision cryoprotectants andcooling rate -5°C temperature before it immersed in liquid nitrogen not were optimal.Keywords: seed rosele, low temperature stratification, cryoprotectans, cryopreservation

Abstract

The objective of the research was to determine the best medium for micro shoot induction of rubbertree from node explant in WPM medium with combination of Benzylamino purine (BAP) andNaphthalane-acetic acid (NAA). The research was conducted in Microcutting Laboratory,PT. Perkebunan Nusantara III Tebing Tinggi Sumatera Utara Medan from Februari 2014-Juni 2014,used complete random design with sixteen treathments and fifteen replications.The result showedthat combination BAP and NAA in WPM gave significantly different on percent of explant formingshoot. Meanwhile, for parameter life explant combination concentrated BAP and NAA showed nosignificantly different.Keywords: rubber, micro shoot, BAP, NAA, WPM

Abstract

Cryopreservation is a technique for long-term storage nuftah plasma. In this technique cells and meristem or other parts of plants were frozen and stored in liquid nitrogen at a temperature -196°C. The objective of the reseacrh was to know effects of type and duration of soaking cryoprotectant on the viability of roselle seed in cryopreservation. This research was conducted at the Laboratory of Seed Technology Faculty of Agriculture, University of North Sumatra, Medan with a height ± 25 meters above sea level, from September to October 2014, using a randomized block design with 2 factors. The first factor is the type of cryoprotectant with 4 type of PVS1 ; PVS2; PVS3; PVS4 and the second factor of soaking cryoprotectant with 3 levels of 60 minutes; 90 minutes; 120 minutes. Parameter observed were the rate of germination (day), normal seedling (%), abnormal seedling (%), seeds die (%), vigor index, fresh weight of seedling (g), dry weight of seedling (g). The results showed soaking treatment of various types of cryoprotectant significant effect on the observation parameters the rate of germination (day), normal seedling (%), vigor index, fresh weight of seedling (g) and dry weight of seedling (g). Treatment of soaking cryoprotectant significant effect on the observation parameters abnormal seedling (%) and vigor index. Interaction type and duration of soaking cryoprotectant significant effect observation parameters on the rate of germination (day) and vigor index. Abstract

The objective of this research was to get the best progeny based on potential of latex yield characteristic, timber yield characteristic and latex-timber yield resulted from 2008 hand pollination. The research was conducted in ± 54 m above sea level at Balai Penelitian Sungei Putih Pusat Penelitian Karet Galang Deli Serdang Sumatera Utara Medan from February 2014 - July 2014. The research was done by measuring coefisien varians, correlation, regression, path analysis, 10 % and 1 % selection intensity, and two line pattern.The research showed based on selection latex yield are 172/08 (intensity 10% and 1%), based on selection timber yield with intensity 10 % and 1 % are 172/08 and 173/08. Progeny 172/08 potential as latex and timber yielding for 2008 hand pollination.Keywords :Hevea brasiliensis, progeny,path analysis, selection
