Abstract

Abstract. Dengue virus infects 50 to 100 million people every year, however, specific treatment or effective antiviral drugs to treat viral infections has not been found yet. Curcumin known has  perform the inhibition of ubiquitin-proteasome system that causes a decrease of Japanese encephalitis, one kind of flavivirus. Structural modifications was known to increase the biological activity of curcumin. Pentagamavunon-0 (PGV-0) is known have activity similar to or even better than curcumin. This study aims to determine the effect of PGV-0 in the early phase of infection of dengue- virus 2 (one day of infection). This study includes quasi-experimental study. The method used for the detection of Dengue-2 viruswas immunocytochemistry, whichpreviously tested by PGV-0 cytotoxic test against vero cells. Cytotoxic test results indicate safe concentrations (no toxic effects) of PGV-0 against vero cells is 4.44 µM. Calculation of positive rate compared with the positive control(14.55 ± 7.25) showed that the value of positive rate due to one-day Dengue virus-2 infection with PGV-0 treatment was smaller(3.8 ± 3.89). It was concluded that the PGV-0 is able to decrease the positive rate due to Den-2 infection in the initial period of infection. Keywords: dengue, Pentagamavunon-0 (PGV-0),immunocytochemistry, vero cells Abstrak.Virus Dengue menginfeksi 50 sampai 100 juta orangper tahun, namun terapi yang spesifik atau obat antivirus yang efektif belum ditemukan. Kurkumin diketahui mampu melakukan penghambatan system ubiquitin-proteasome yang menyebabkan penurunan produksi salah satu jenis Flavivirus yaitu Japanese encephaitis. Modifikasi struktur kurkumin terbukti meningkatkan aktivitas biologisnya.  Pentagamavunon-0 (PGV-0) diketahui memiliki aktifitas mirip atau bahkan  lebih baik dari kurkumin. Tujuan dari penelitian ini adalah mengetahui pengaruh pemberian PGV-0 pada fase awal infeksi virus Dengue-2 (satu hari infeksi). Penelitian ini termasuk penelitian kuasi-eksperimental. Metode yang digunakan untuk deteksi virus Dengue-2 dalam penelitian ini adalah imunositokimia, setelah sebelumnya dilakukan uji sitotoksik PGV-0 terhadap sel vero. Hasil uji sitotoksik menunjukkan konsentrasi yang aman (tidak memberikan efek toksik) dari PGV-0 terhadap sel Vero adalah 4,44 µM. Hasil uji imunositokimia menunjukkan nilai rata-rata sel positif akibat infeksi Dengue-2 satu hari dengan perlakuan PGV-0 jauh lebih kecil (3.8 ± 3.89) dibandingkan kontrol positifnya (14.55 ± 7.25). Disimpulkan bahwa PGV-0 mampu menurunkan positive rate akibat infeksi Den-2 pada periode awal infeksi. Kata Kunci: dengue, Pentagamavunon-0 (PGV-0), imunositokimia, sel vero

Abstract

The data that have been obtained in Indonesia by using samples of clinical material from the hospital, Staphylocccus aureusis proved to be resistant to penicilin and methicillin, tetracyclin, oxacilin, gentamicin, erytromycin, chloramphenicol, and trimethrophrim-sulfemethoxazole. The high resistant inStaphylocccus aureus bacteria encourage efforts to find drugs that have antibacterial activity. Furfural and its derivates are known to have antibacterial activity. The purpose of this study was to obtain derivate of furfural compound 2,6-bis-(2’-furilidyn)-sikloheksanone, which is expected to have antibacterial activity against drug resistant bacteria Methicillin resistant Stapylococcus aureus (MRSA). 2,6-bis-(2’-furilidyn)-sikloheksanone synthesis was done with starting materials ofcyclohexanone (0,0121 mol)and furfural (0,0121 mol), using KOH 7,5% as catalys in the aquadest solvent. The purity test were carried out with melting distance test, thin layer chromatography, and gas chromatography. Structure elucidation was performed with mass spectrometer analysis, IR, and spectrophotometer, H1-NMR. The syntesis result is then tested for its antibacterial activity againts MRSA bacteria with diffusion method to determine the inhibition diameter. The result of this study showed that 2,6-bis-(2’-furilidyn)-cyclohexanone has been successfully synthesized with an average yield of 73,60% ±0,204. Re-crystallization resulting in pure compound base on its melting range (1,90C), TLC (one spot Rf= 0,33), and GC (100% area). The results of the chemical structure elucidation by mass spectrometry, IR spectrophotometerand H-NMR spectrometer line with the target compound. The synthesis result has antibacterial activity with average of inhibition diameter at a concentration of 3,959mM/ml= 27 mm.   Keywords:synthesis, analog of curcumin, antibacterial

Abstract

Program MKU yang dilaksanakan ini bertujuan memberi pengetahuan keterampilan, dan sikap di bidang jasa~-  vawasan manajemen kewirausahaan di bidang perawatan/pemeliharaan dan perbaikan peralatan pemakai listrik rti     ac,  kulkas,   frezzer, mesin cuci,   trafo,  dan  peralatan rumah tangga  lainnya  bagi  mahasiswa Jurusan-.:_ididikan  Teknik  Elektro  Fakultas  Teknik  Universitas Negeri  Yogyakarta.  Pelaksanaan  kegiatan  meliputi  ::le::daftaran peserta, seleksi peserta, pelaksanaan magang kewirausahaan dan evaluasi program/kegiatan. Kriteria~ ~ dapat mengikuti magang adalah mahasiswa yang minimal duduk di semester VII dan mempunyai motivasi dan-    cat   yang  tinggi,  baik  mahasiswa  yang  belum  atau  sudah  menempuh praktek  industri.  Setelah  mahasiswa-,:. gikuti pembekalan  dengan materi  : menumbuhkan jiwa  wirausaha, upaya  mendapatkan  modal/manajemen=-_angan, liku-liku mendirikan suatu usaha (kewirausahaan), dan manajemen industri, mahasiswa diterjunkan ke~ i  perawatan/pemeliharaan dan  perbaikan {service)    peralatan  pemakai  listrik (alat-alat  rumah  tangga).  Di z+ping   industri sebagai   pembimbing utama, para mahasiswa juga mendapatkan bimbingan secara rutin melalui egraran monitoring dari dosen.Kegiatan mahasiswa di industri mendapat penilaian tentang : kemampuan, keterampilan, disiplin, dan sikapJa   oleh pembimbing  industri. Setelah selesai mengikuti program magang, mahasiswa diwajibkan  menyusun_ cposal  mendirikan bengkel jasa perawatan/pemeliharaan dan perbaikan (service)   peralatan pemakai listrik. Darize      anaan magang, sembilan mahasiswa dinilai sangat baik, dan satu orang mahasiswa  dinilai baik.  Sernua...,r. asiswa peserta magang telah berhasil membuat proposal mendirikan bengkel jasa  perawatan dan perbaikan- _   alan pernakai listrik, dan suatu hal yang lebih menyenangkan dan membanggakan adalah bahwa para peserta:=-    ng secara kelompok sekarang telah membuka usaha jasa perawatan dan perbaikan peralatan pemakai listrik~  rrlah dua buah.-a  kunci   : MKU, Elektro, Servis, Utilindo

Abstract

AbstractMore than 40% of the worlds population who live in tropical and subtropical regions at risk for dengue infection. Specific and effective antiviral therapies to treat dengue infection has not been found yet. Many researches proved that curcumin has preventive activity againts viruses, such as vasicular stomatis (VSV), HSV 1 and 2, parainfluenza-3, reovirus-1, feline corona virus, feline herpes virus. Curcumin also known to perform the inhibition of ubiquitin - proteasome system that decreasethe production of Japanese encephalitis in neuroblastoma cells. Pentagamavunon-0 (PGV-0) is expected to have better activity than curcumin. This study aims to determine the cytotoxic effect andthe potential of curcumin and PGV-0 as antiviral Dengue-2 on vero cells. Including experimental study. Cytotoxic test performed to obtain a safe concentration of curcumin and PGV-0 on vero cells followed by antiviral test using immunocytochemistry SBPC (Streptavidin Biotin Peroxidase Complex). The results showed that the safe concentrations for curcumin is 6.25 ppm and PGV-0 is 1.5625 ppm based on cytotoxic test to vero cell. The positive rate from Immunocytochemistry test showed that no significant difference between curcumin and PGV-0 treatment. However, when compared with the positive control results are significantly different. We concluded both curcumin and PGV-0 can reduce the positive rate caused Dengue-2 infection at one day incubation.Keywords : Dengue-2, curcumin, pentagamavunon-0 (PGV-0), ImmunocytochemistryAbstrakLebih dari 40 % populasi dunia yang tinggal di daerah tropis dan subtropis mempunyai risiko untuk terjangkit infeksi Dengue. Terapi yang spesifik dan efektif untuk mengobati infeksi Dengue belum ditemukan. Kurkumin terbukti memiliki aktivitas preventif terhadap beberapa virus, antara lain:vasicular stomatis (VSV), HSV 1 dan 2, parainfluenza-3, reovirus-1, feline corona virus, feline herpes virus. Kurkumin juga diketahui mampu melakukan penghambatan sistem ubiquitin-proteasome yang menyebabkan penurunan produksi Japanese ensefalitis dari sel neuroblastoma yang sebelumnya terinfeksi. Pentagamavunon-0 (PGV-0) diperkirakan memiliki aktivitas lebih baik daripada kurkumin. Penelitian ini bertujuan untuk mengetahui efek sitotoksik dan mengetahui potensi antiviral Dengue-2 dari kurkumin dan PGV-0 pada sel vero. termasuk penelitian eksperimental. Uji sitotoksik dilakukan untuk mendapatkan konsentrasi yang aman dari kurkumin dan PGV-0 terhadap sel vero dilanjutkan dengan uji antiviral melalui imunositokimia SBPC (Streptavidin Biotin Peroxidase Complex). Hasil uji sitotoksik menunjukkan konsentrasi yang aman terhadap sel vero adalah 6,25 ppm untuk kurkumin dan 1,5625 ppm untuk PGV-0. Perhitungan positive rate dari uji imunositokimia pada sel vero yang diinfeksi Dengue-2 inkubasi 1 hari dan diberi kurkumin dan PGV-0 adalah tidak berbeda nyata, namun jika dibandingkan dengan kontrol positifnya hasilnya berbeda nyata. Disimpulkan baik kurkumin maupun PGV-0 mampu menurunkan positive rate akibat infeksi Dengue-2 inkubasi satu hari.Kata kunci : Dengue-2, kurkumin, pentagamavunon-0 (PGV-0), imunositokimia

Abstract

Curcumin (1,7-bis(3-methoxyphenyl 4hidroksi) -1.6 heptadien, 3,5-dione) is the yellow pigment of Curcuma longa. Curcumin has various biological activities such as antioxidant, antibacterial, antifungal, antiprotozoal and antivirus. Various benefits of curcumin can not be separated from the weakness which is not stable to pH and light. Pentagamavunon-0 (PGV-0) were made by changing the β diketone group on cluster analog of curcumin into monoketon while eliminating active methylene group so it is more stable to pH and light. PGV-0 also has potential as a stronger antioxidant and antiinflammatory agent than other curcumin analogues. The objective of this research was to determine the safe concentrations of curcumin and PGV-0 on vero cells due to the increased use of the two compounds through the cytotoxic test. This research includes experimental research. Cytotoxic test performed with microculture tetrazolium technique (MTT) method. Use of MTT to evaluate the cytotoxic is based on changes of tetrazolium salt into formazan crystals by mitochondrial enzyme succinate dehydrogenase with the help of cellular NADH. The results showed that the safe concentrations of curcumin and PGV-0 on vero cells respectively are 6.25 and 1.5625 ppm. Based on the cytotoxic test the secure concentration of curcumin was higher than PGV-0.

Abstract

Synthesis of Tetrahydrohexagamavunon-5 (THHGV-5) and Tetrahydrohexagamavunone-7 (THHGV-7) were prepared by catalytic hydrogenation reaction on Hexagamvunon-5 (HGV-5) and Hexagamavunone-7 (HGV-7) by using gas H2 as source of hydrogen gas, Pd/C 10 % as metal catalyst and methanol as solvent at room temperature. The products were characterized by IR Spectroscopy, Gas Chromatography-Mass Spectroscopy (GCMS), 1D-NMR (1H-NMR and 13C-NMR) and 2D-NMR (1H-13C HMQC) Spectroscopy to determine the product structure molecules. According to the data of IR, GC-MS, 1H-NMR, 13C-NMR and 1H-13C HMQC spectra, the products are THHGV-5 and THHGV-7 as white crystalline powders. Key words:Tetrahydrohexagamavunon-5, Tetrahydrohexagamavunone-7, Hexagamvunon-5, Hexagamavunone-7

Abstract

Adanya persyaratan minimal atas bahan baku obat tradisional, serta sangat terbatasnya data agronomi atau budidaya dan penanganan pasca panen, merupakan kendala sampai saat ini yang belum dapat diatasi oleh pihak industri dalam memenuhi kebutuhan akan bahan bakunya, karena harus mengusahakan atau membudidayakan sendiri. Kondisi seperti ini telah mendorong pihak industri atau perusahaan obat tradisional untuk melakukan penanganan dan pengendalian dalam pengelolaan dan pengolahan simplisia guna memenuhi kebutuhan bahan bakunya secara kontinyu, dengan cara melakukan a) pengendalian dalam pengadaan bahan baku, b) pengendalian untuk dapat memperoleh keseragaman jenis simplisia yang digunakan, dan c) pengendalian kualitas dengan cara melakukan standardisasi.Makalah ini dibatasi lingkupnya pada pengalaman dalam pengelolaan dan pengolahan simplisia sebagai bahan baku obat tradisional, menuju persyaratan mutu sebagaimana yang tersurat dalam MMI, FI, dan EFI.

Abstract

The synthesis of ciklovalon (HGV-0) analogue was done by substituent modification on aromatic ring. One of the ciklovalon analogues is 2,6-bis(2′-hydroxybenzylidene)cyclohexanone. The objective of this study was obtaining the amount of mol NaOH catalyst in order to find the highest rendement.  The synthesis of 2,6-bis(2′-hydroxybenzylidene)cyclohexanone has been done using aldol condentation reaction from 2′-hydroxybenzaldehyde and cyclohexanone. The variation of mol catalyst NaOH used in this research were 0.2 mole; 0.4 mole and 0.8 mole. The synthesized product was purified by recrystalization with ethanol and cold water. The structure of the synthesized compound was determined by UV-Vis, IR, 1H-NMR, and GC-MS.  The result showed that the average rendement the variation of mol catalyst NaOH 0.2 mole; 0.4 mol and 0.8 mole were (25.12±0.86)%, (11.23±0.40)% and (9.50±0.56)%. The highest rendement was obtained when the 0.2 mole NaOH used. Statistical analysis using tukey test resulted significant differences. The spectroscopic analysis using UV-Vis, IR spectrometry, 1H-NMR spectrometry, and GC-MS showed that synthesized compound was 2,6-bis(2′-hydroxybenzylidene)cyclohexanone.   Keywords: 2,6-bis(2′-hydroxybenzylidene)cyclohexanone, synthesis, catalyst, rendement     ABSTRAK   Sintesis analog siklovalon (HGV-0) dilakukan melalui modifikasi substituen pada cincin aromatiknya. Salah satu analog siklovalon tersebut adalah 2,6-bis(2′-hidroksibenzilidin)sikloheksanon. Tujuan penelitian adalah untuk mengetahui jumlah mol katalis NaOH yang menghasilkan rendemen paling besar.  Sintesis 2,6-bis(2′-hidroksibenzilidin)sikloheksanon dilakukan melalui reaksi kondensasi aldol dari 2-hidroksibenzaldehid dan sikloheksanon dengan menggunakan katalis natrium hidroksida. Variasi jumlah mol katalis NaOH yang digunakan dalam penelitian ini adalah 0,2 mol; 0,4 mol dan 0,8 mol. Pemurnian hasil sintesis dilakukan dengan rekristalisasi menggunakan etanol dan aquades dingin. Struktur senyawa hasil sintesis ditentukan dengan cara spektrofotometri UV-Vis, spektrometri Inframerah, spektrometri Resonansi Magnetik Inti (1H-RMI), dan GC-MS.  Dari hasil penelitian menunjukkan bahwa rerata rendemen dari variasi jumlah mol NaOH 0,2 mol; 0,4 mol dan 0,8 mol berturut-turut adalah (25,12±0,86)%, (11,23±0,40)% dan (9,50±0,56)%. Rendemen paling optimal diperoleh pada jumlah mol katalis NaOH 0,2 mol. Hasil uji Tukey menunjukkan adanya perbedaan yang signifikan pada masing-masing kelompok. Berdasarkan data spektrofotometri UV-Vis, spektrometri Inframerah, spektrometri 1H-RMI, dan GC-MS menunjukkan bahwa senyawa yang dihasilkan adalah 2,6-bis(2′-hidroksibenzilidin) sikloheksanon. Abstract

Kurkumin sejak lama dikenal memiliki aktivitas sebagai antiinflamasi sekaligus antikolesterol. Senyawa-senyawa analog kurkumin seperti Pentagamavunon-0 (PGV-0) dan Gamavuton-0 (GVT-0) yang disintesis oleh Fakultas Farmasi UGM juga dikenal memiliki aktivitas antiinflamasi, bahkan PGV-0 memiliki aktivitas antiinflamasi lebih kuat dibanding kurkumin. Makin meningkatnya penyakit degeneratif khususnya hiperlipidemia mendorong dilakukannya penelitian menggali kemungkinan senyawa analog kurkumin sebagai antikolesterol. PGV-0 dan GVT-0 diberikan kepada tikus jantan yang telah terlebih dahulu diinduksi dengan pakan diet lemak tinggi (DLT) selama 30 hari. PGV-0 (dosis 20 mg/kg bb, GVT-0 (dosis 60 mg/kg bb) serta kontrol positif dengan orlistat (dosis 10,8 mg/kg bb) diberikan kepada tikus pada hari ke-30 hingga 45. Tikus tetap diberi pakan DLT sampai hari ke-45. Pada hari ke-0, 30 dan 45 dilakukan penetapan kadar kolesterol darah tikus serta penetapan aktivitas enzim lipase dari darah tikus. Orlistat, PGV-0 maupun GVT-0 mampu menurunkan kadar kolesterol tikus secara bermakna berkisar 23-33% (P≤0,05) dibandingkan kontrol.

Abstract

Antioxidant is an essential compound to keep man’s health due to its function as radical scavenging. Curcumin analog compounds can function as antioxidant (Sardjiman, 2000). The aim of the experiment was to find out the antioxidant activity of 2,.6-bis-(2’-furilidin)-cyclohexanone, 2,.5-bis-(2’furilidin)-cyclopentanone, and 1,.5-difuril-1.4-pentadien-3-one compounds, and the antioxidant activity of each compound against DPPH  radical with IC50 parameter as well as the correlation of compounds structure’s activities against antioxidant. The antioxidant activity of curcumin analog compounds wereas tested against DPPH free radical. The test was conducted in 5 series of concentrations by adding 4.0 ml test solutions with 1.0 ml DPPH. The antioxidant activity against free radical was measured usingwith spectrophotometer at 517 nm wavelength and determined for the IC50 value. The experiment employed rutin as positive control. The result of the experiment showed that curcumin analog compounds have antioxidant activity with IC50 of rutin, 2,.6-bis-(2’-furilidin)-cyclohexanone, 2,.5-bis-(2’furilidin)-cyclopentanone, and 1,.5-difuril-1.4-pentadien-3-one as follows: 4.93 ppm, 22.73 ppm, 20.67 ppm, and 18.80 ppm  respectively. The highest antioxidant activity belonged to 1,.5-difuril-1.4-pentadien-3-one compound which is 18.80 ppm . Correlation of activity structure of the 3 compounds can be seen from the log p parameter and energy space of HOMO-LUMO.Key words: Antioxidant, DPPH, Analog Curcumin
