Abstract

Hornstedtia scyphifera var. fusiformis merupakan tumbuhan endemik dari famili Zingiberaceae yang tersebar di Sumatera. Tumbuhan ini  mengandung beberapa metabolit sekunder seperti fenolik, flavonoid, saponin, triterpenoid, steroid dan alkaloid. Pengujian Bioaktifitas meliputi uji antioksidan dan antibakteri. Aktivitas antioksidan ditentukan dengan metoda DPPH. Sedangkan aktivitas antibakteri diuji dengan metode difusi cakram terhadap bakteri Staphylococcus aureus (bakteri gram positif) dan Escherichia coli (bakteri gram negatif). Hasil analisa aktitifitas antioksidan dari ketiga ekstrak, diperoleh bahwa ekstrak metanol menunjukkan aktifitas antioksidan yang paling aktif dengan IC50 = 51,8943 mg/L dibandingkan dua ekstrak lainnya. Ketiga esktrak Hornstedtia scyphifera var. fusiformis memberikan aktivitas antibakteri, selanjutnya esktrak n-heksan dan etil asetat pada konsentrasi 500 ppm memiliki zona hambat 10 mm terhadap bakteri Escherichia coli  dan Staphylococcus aureus memiliki zona hambat 10,3 mm. Untuk mengetahui jumlah senyawa metabolit sekunder  yang terkandung dalam  tumbuhan ini dilakukan analisa dengan HPLC.

Abstract

The anthocyanins in Ficus padana were extracted and identified by using high-performance liquid chromatography/diode array detection and electrospray ionization/mass spectrometry (HPLC-DAD-ESI-MS). The individual anthocyanins were identified by comparing between their mass spectral data and published data. The first compound (peak 1) was identify as a pelargonidin 3-(6”-p-coumarylglucoside)-5-(4”’-Malonylglucoside) and the second compound was identify as a pelargonidin 3-(6’’-Malonylglucoside). The second one is a major compound that taking up to 91% of the total anthocyanin content in F. padana extract. The antioxidant activity was determined with 2,2-diphenyl-1-picrylhydrazyl (DPPH) assays and results showed that extract possessed high antioxidant capacity.

Abstract

Betalains and anthocyanins are classes of natural and water soluble pigments. Now days, these pigments have been developed as a replacement colouring agents. One of betalain resource is dragon fruit and anthocyanin resource is tamarillo. These pigments are relative unstable in some condition, one of them is pH. This research aim is to determine stability of betalain and anthocyanin pigments by pH parameter. These pigments can be isolated by macerated methods, technically processed by chopping the fruit and than soaking in ethanol for ± 24 h. These extracts were condensed by rotary evaporator. These extracts were analysed by ultraviolet-visible spectrophotometer at wavelength (λ) 200-800 nm. The absorption spectrum showed two major absorption peaks at 269 nm and 536 nm for betalain and 531 nm for anthocyanins. The result showed the optimal pH for betalain stability was at pH 5 and pH 3 for anthocyanin

Abstract

Preparative High Performance liquid chromatography (HPLC) today is one of the popular methods to isolate anthocyanin from extract containing anthocyanins. The working principle of preparative-HPLC can be applied to the laboratory standard tupe of HPLC, so it can be used also to isolate the main anthocyanin compounds from anthocyanin mixtures. The function of fraction collector on preparative HPLC equipment were replaced by opening the mobile phase line after the exit of the detector and hold out the solution manually on the standard type of HPLC. Resolution value between the peaks and the computation time when cuts in the chromatogram peak collection mobile phase containing the analyte is of concern in these activities. The appearace of a single peak in the chromatogram of the isolates showed that the technique can be applied for the simple and fast technique of anthocyanins isolation.Keywords : isolation, anthocyanin, Ficus padana Burm.f, Preparative-HPLC, standar type of HPLC.

Abstract

Salix Tetrasperma Roxb. is a plant that found in Indonesia were used as traditional medicine such as diabetes and wound healing. In this study, a flavonoid compound of the ethyl acetate extract of Salix tetrasperma Roxb. leaves was isolated by chromatography technique and the antioxidant activity was determined by DPPH assay. The isolation led to obtain 5,7-dihydroxy-3'-methoxyflavone based on NMR spectra. The ethyl acetate extract exhibited the highest antioxidant activity with the IC50 is 65.89 µg/mL. This study shows that the Salix tetrasperma Roxb. has good potential as source of antioxidant agent.

Abstract

Salix tetrasperma Roxb. (Family Salicacaeae) is a plant that used as traditional medicine for anti-inflammatory, analgesic, reduces fever, and itching medicine. In this study was carried out extraction, isolation, structure elucidation of salicin from Salix tetrasperma Roxb. stem bark and it’s antibacterial activity. The extraction method was used the maceration method by n-hexane, ethyl acetate, and methanol solvents. Isolation of compound from ethyl acetate extract of Salix tetrasperma Roxb. stem bark using chromatography methods and obtained white solid (15 mg). The structure was elucidated using spectroscopic analysis, including Ultraviolet (UV), Infrared (IR), Nuclear Magnetic Resonance (NMR) and comparative literature, identified as salicin compound with molecule formula C13H18O7. Antibacterial activity was evaluated against Escherichia coli and Staphylococcus aureus bacteria using disk diffusion method. This compound has a great an antibacterial activity against Staphylococcus aureus bacteria with clear zone diameter of 10.2 ± 0.3 mm. This shows that the Salix tetrasperma Roxb. stem bark has great potential as a source of antibacterial compound
