Abstract

Sanjaya, Machmudin D, Kurniawati ND. 2010. Histological study of SlNPV infection on body weight and peritrophicmembrane damage of Spodoptera litura larvae. Nusantara Bioscience 2: 135-140. The effect of SlNPV infection on body weight andperitrophic membrane damage of Spodoptera litura Fab. larvae has been carried out. The method was used Probit analysis, and based onLD 50 the virus was infected to know body weight and post infection damage. The damage of histological structure caused by SlNPV (0,315, 390, 465, 540 dan 615 PIB/mL) was investigated after 0, 12, 24, 72 and 96 hours post infection. The histological material wasprepared by using parafin method after fixation with Bouin Solution, then slice into 7 um and colored with Hematoxilin-Eosin. Theresult showed that the exposure SlNPV cause decreasing food consumption especially on 540 PIB/mL give average rate as amount of0.1675 mg. The descriptive obsevation on structural intact of peritrophic membrane histology caused by SlNPV infection shows atendency to decrease, while in control, there was no damage at all. The longer the exposition of virion in the midgut lumen the moredamage on peritrophic membrane occurred. The severest damage occurred 96 hour after infection. The result prove that haNPV virioncan destroy hystological structure of midgut.Key words: SlNPV, Spodoptera litura, LD50, consumption rate, peritrophic membrane.

Abstract

Fungi has been tested as one of the potential control agents for insect pests, which raises hopes for developing fungi as good biopesticides. The high variation within fungi species made taxonomic identification procedures more complex, thus molecular identification techniques are needed in addition to traditional morphological characteristics currently used as primary methods to classify fungi species. The objective of this research was to identify the species of the most pathogenic fungi to Tetranychus kanzawai Kishida using RAPD-PCR.  The internal transcribed spacer of 5.8s rDNA (ITS-5.8s rDNA) sequence of these fungal isolates were amplified using two sets of universal primers for ITS and then analyzed. Molecular identification showed that these  isolates had a higher  of similarity to Metarhizium anisopliae than Metarhizium flavoviride.

Abstract

The objective of this research was to compare the diversity of insects in chili plant agroecosystem treated with synthetic pesticide versus that treated with biopesticide (venom of spider Nephila sp.) Two plots of chili pepper plants, each sized 600 m2, located at Ciwidey area Bandung –West Java, were taken as the sample plots. The first plot was weekly treated with profenofos insecticide (50% active ingredient, a.i.) while the second plot was treated with  biopesticide (70% a.i. spider venom extracted from Nephila sp.), each at 35 ml/17 L rate of spray solution. Sweep net was used to sample insects along two transects in each plot. Result showed that 14 families of 8 insect orders were found in pesticide-treated plot while 15 families of  9 orders were found in biopesticide-treated plot. The evenness index and diversity index of insects in plot treated with biopesticide were relatively higher than those in pesticide-treated plot. In contrast, dominance index in biopesticide plot was lower than that in synthetic pesticide plot. Thrips sp.(Thysanoptera: Thripidae) was found to be the most dominant species in both plots.

Abstract

Butterflies choose one or several particular plant species which are very closely as a place of food and life, as well asG. evemon. Soursop plant(Annona nuricata) is one of the natural food for the G.evemoncaterpillars. This plant is widely spread both along the highway and in the office complex.Until now there is no research on the life cycle and life table of theG.evemon in the soursopplant. However the need of this information is important to preserve the existence of a butterfly as one of pollinating insects to determine the biotic potential of the G. evemon. The purposes of this study are to determine the life cycle andmorphometric of G.evemon. This study uses rearing method and the subject of the study are 30 eggs G. evemon. The length of time and morphology of each pre-adult stages ofthe G.evemon was recorded. The calculations in the table are based on the life stage of the development and mortality of each stage of the development of G. evemon. The results showed that the average time of the G.evemonlife cycle onsoursop plant was 28 – 50days. The morphology of each stage has a specific characteristic.

Abstract

This study was intended to investigate students’ cognitive and attitude of secondary school in learning global warming using Student Team Achievement Division (STAD) based on gender. This study was conducted at one of bilingual boarding school in Bandung. This study uses the quasi-experimental method with matching pretest-posttest comparison group design while the number of subject in the girl class is 15 (N=15) and in the boy class is 15 (N=15). Data was gained through a cognitive test which measured by Blooms taxonomy cognitive level (C1-C6), a questionnaire which measured by Test of Science Related Attitude (TOSRA). The result shows that student who learns global warming using STAD has an improvement on their cognitive score, especially in the girl class. Meanwhile, girl class has higher students’ attitude result in three scales of TOSRA that are; attitude to scientific inquiry, adoption of a scientific attitude, and enjoyment of science lessons. Along with the students’ cognitive result, girl class also has a better science-related attitude than the boy class. Thus, it can be concluded that there is a significant difference between students’ cognitive and attitude at the girl and boy class in learning global warming using STAD.
