Abstract

Zinc is an essential trace element involving in the activity of more than 300 enzymes and proteins of human body. One important role of zinc is to improve metallothionein protein binding heavy metals and functioning as heavy metal detoxification facilities. This research discusses the effect of zinc supplement  on the  improvement of metallothionein protein level. The in vivo test involving 28 rats categorized in 4 groups was performed. The experiments used randomized post test control group design. The 3 groups were daily supplemented by zinc in th concentration of  0.2 mg, 0.4 mg, and 0.8 mg. Whereas, the last group was let without zinc treatment. As an indicator the metallothionein protein level was checked after three weeks. The data was then evaluated by Anova an Bonferroni test in order to know the signicant of protein level difference among the groups. The result showed that the average of metallothionein protein level improved by increasing zinc suppplement with the 0.95 ±0.20; 1.28 ±0.19; 1.39 ±0.09; 1.91 ±0.3 ng/ml metallothionein per 0, 0,2, 0,4, 0,8.mg zinc added. Based on the ANOVA and Bonferroni test,  indicated that the improvement was significant as shown with p value  of 0.00

Abstract

Sepsis remains a health problem because of the high related mortality and morbidity. One of the efforts is the improvement of knowledgeabout manifestation and pathophysiology of sepsis at the level of molecular biology. Nitric oxide (NO) levels increased in sepsis and this isrelated to the occurrence of cell apoptosis. The purpose of this study was to know and to see the clinical picture of sepsis and elevated levels ofNO in mice Balb/C which were injected with lipopolysaccharide (LPS). In this research, the mice were divided into three (3) groups, groupA were injected with saline, group B were injected with LPS 0.1 mg per mouse, while group C was injected with 0.2 mg per mouse. Themice were then measured by clinical parameters from day 0 and day 1, and 3,5 to 7. The levels of NO were assessed by blood sampling asmuch as 1 cc from the peri orbital plexus and examined by Griess method on days 1, 3, 5, 7. The results of the study showed that from theweight parameter there was a decrease in body weight on day 1 in group B and C and the weight continued to decline until day 3. From thetemperature parameter it was shown that the temperature decreased both in group B and C on day 1 but on day 3 and so on varies in bothgroups. From the piloerection parameters from day 1 to day 7 showed that there was piloerection in both groups. Related to the discharge byperi orbital parameters and diarrhoea many variations, occurred on day 1 peri orbital discharge and diarrhoea was found, but not uniformlyin all the mice in each group. On day 5 and 7 the peri orbital discharge and diarrhoea were not visible. NO levels increased mainly on day3 and the percentage was higher in group C and on day 5 and day 7 the levels were negative. In conclusion in this study it was shown, thatthe clinical picture is striking, weight loss and increased levels of NO occurs on day 3 of the observation.

Abstract

Lipopolysaccharide (LPS) is an endotoxin from the outer membrane of Gram-negative bacteria, which has an important role in the occurrence of sepsis. Exposure to LPS will stimulate increase of nitric oxide (NO). Nitric oxide is a bioregulator of apoptosis and has some sepsis prognostic role of apoptosis regulators within the gastrointestinal cells. The objective of the study is to know if endotoxemia induces an increase in NO levels and histopathology scores as well as the existing relationship between them. This study is an observational intervention. The subjects were 48 male mice Balb/C, divided into 2 groups. The samples consisted of 24 tail as control group (group A) and 24 as treatment group (group B). The A group as well as the B group is divided into 4 subgroups according to the time of termination. The levels of NO were examined by Griess method. Histopathology score was examined by HE and read as a score of 0–5. There was a statistically significant difference between the mean NO in the treatment group with the control group at the termination of the group of 12h (p=0.009), 24h (p=0.015), 36h (p=0.014), 48h (p=0.002) and the whole group (p=0.0001), as well as between the mean histopathology score at the termination time of 12 h (p=0.0001), 24h (p=0.0001), 36h (p=0.0001), 48h (p=0465) and the whole group (p=0.0001). Increase in NO and histopathology scores in all groups of mice (r=0.527) showed a statistically significant correlation. NO levels and histopathology scores are increased during endotoxemia and thus have a significant correlation.
