PROFIL ALKALOID KUINOLIN PADA KULTUR IN VITRO CINCHONA LEDGERIANA MOENS. (Quinoline Alkaloid Profile in In Vitro Culture Cinchona ledgeriana Moens.)

Abstract

Quinoline alkaloids are alkaloids from Cinchona sp., that harvested from 6-12 years old trees. Plant tissue culture methods can be used as alternative to increase production of quinoline alkaloids. The aims of this reasearch were optimization of the growth of in vitro culture of Cinchona ledgeriana and analysis of its quinoline alkaloids profile. Woody plant medium (WPM) were optimal medium and growth regulator hormons 0.1 mg/LNAA was optimal with added 3 folds of vitamin and myoinositol contains normal. Murashige and Skoog medium (MS) with 0.5 mg/L 2,4D and 0.5 mg/L kinetin as hormones was optimal for the callus induction. Strain ATCC 15834 was the best strain for the hairy root induction with efficiency 67%, compared to strain A4 and 0720001 that only have efficiency 17.39% and 15.5%, respectively. Thin layer chromatography profile showed that stem, root, leaf and callus culture produced alkaloid quinoline i.e.quinine, quinidine, cinchonine and cinchonidine. Quinine was the highest amount alkaloid that produced in stem up to 0.26%, while in root, leaf, and callus were 0.15%, 0.026% and 0.021% respectively and quinidine produces in stem up 0.046% and in root 0.015%.Keywords : in vitro culture, quinoline alkaloid, optimation, Cinchona ledgeriana.AbstrakAlkaloid kuiolin merupakan alkaloid yang dihasilkan tanaman Cinchona sp. yang dipanen pada saat usia tanaman 6-12 tahun. Metode kultur jaringan dapat menjadi alternatif dalam produksi peningkatan kadar alkaloid kuinolin. Penelitian ini dilakukan untuk mengoptimasi pertumbuhan kultur in vitro yang stabil serta menganalisis profil alkaloid kuinolin dari kultur in vitro Cinchona ledgeriana. Optimasi media pertumbuhan kultur kina yang optimal adalah media tanaman berkayu (WPM) serta pada optimasi hormon pengatur tumbuh 1-napthaylactic acid (NAA) 0,1 mg/L optimal dengan penambahan vitamin dan mio-inositol tigakali dari media normal. Induksi kalus paling baik pada media Murashige dan Skoog (MS) yang ditambah hormon pengatur tumbuh 0,5mg/L 2,4-dichlorophenonoxyacetic acid (2,4D) dan 0,5 mg/L kinetin. Pada induksi kultur akar rambut berhasil diinduksi dengan efisiensi 67% menggunakan Agrobacterium rhizogenes galur ATCC-15834, sedangkan galur A4 dan 07200001 masing-masing 17,39% dan 15,5%. Profil alkaloid kuinolin dengan kromatografi lapis tipis (KLT) menunjukan kultur barang, akar, daun dan kalus in vitro mengandung alkaloid kuinolin kinin, kinidin dan sinkonidin dengan kadar kinin tertinggi pada batang yakni 0,26%, sementara pada akar 0,15%, daun 0,026% dan kalus 0,021, sedangkan kinidin pada batang 0,046% dan akar 0,015%.Kata kunci : kultur in vitro, alkaloid kuinolin, optimasi, Cinchona ledgeriana.

Abstract

Research of xantine oxidase enzyme inhibition activity of infusion and ethanol extract of white tea were done. The aim of this research was to evaluate  xantine oxidase inhibitory activity of infusion and ethanol extract of white tea. Phytochemical screening on dry tea, infusion and ethanol extract of white tea showed that all samples contained alkaloid, flavonoid, quinone, tannin and tritherpenoid/ steroide. The IC50 value of samples were 76,64 μg/ml and 31,11μg/ml respectivelly. This study used allopurinol as standard with IC50 was 0,60 μg/ml.

Inhibition activity of α-glucosidase by infusion and ethanol extract of white tea and stevia combination

Abstract

Ethanol extract of white tea and its stevia liquor was observed for α-glukosidase inhibition. The research was divided into some steps;  white tea and stevia combination hedonic sensory test, phytochemical identification, and α-glucosidase inhibition activity. The result of hedonic sensory test incline towards formula 2 which consist 75% of white tea and 25% stevia. Further, this formulation was extracted by ethanol for α-glucosidase inhibition activity test. Formulation 2 liquor tested for enzyme inhibition activity showed IC50 value was 36,96 µg/ml, better than ethanol extract and acarbose. Phytochemical identification presented that both preparations contained alkaloids, flavonoids, saponins, tannins, triterpenes/steroid, and quinones.

Abstract

Green tea has bioactive component, it is polyphenol which has strong antioxidant. As the largest component of polyphenol, flavonoid is very effective as an antioxidant. The purpose of this study was to determine antioxidant activity, phenol total and flavonoid total in green tea from three different estates, they were Cikajang Kab. Garut, Taraju Kab. Tasikmalaya, and Ciwidey Kab. Bandung. The results of this study show that seven samples have good antioxidant activity, but green tea which came from Cikajang (P-IRT No. 810320501698) has the highest IC50 grade it was 21,44 μg/ml. Beside that samples from Cikajang (P-IRT No. 810320501698) has highest total phenol content it was 334,68± SD 0,89/100 g samples and also has the highest total flavonoid it was 0,34 mg Kuersetin/g samples ± SD 0,21. On the contrary, green tea which has the lowest IC50 grade came from Taraju, the grade was 28,03 μg/ml, the samples has the lowest total phenol content it was 208,91± SD 4,47 mgGAE/100 g, and also has the lowest total flavonoid it was 0,207 mg Kuersetin/g samples ± SD 0,00.
